Isolation of endophytic bacteria from the medicinal, forestal and ornamental tree Handroanthus impetiginosus.

Plant interactions with endophytic bacteria produce mutual benefits and contribute to environmental sustainability. Handroanthus impetiginosus (Mart. ex DC.) Mattos 'pink lapacho' (syn. Tabebuia impetiginosa, Bignoniaceae) is a medicinal, ornamental and forestal native tree from South and Mesoamerica. Plant growth promoting bacteria (PGPB) isolated from pink lapacho are scarcely described. The aim of this work was to isolate and characterize native endophytic bacteria from pink lapacho. Ten bacterial strains were isolated from leaves and six from roots of naturally growing trees in Luján (Cent

The geographic scale of population-level variation in growth and nodulation differs for two species of prairie clover.

PREMISE: Prairies are among the most threatened biomes due to changing patterns of climate and land use, yet information on genetic variation in key species that would inform conservation is often limited. We assessed evidence for the geographic scale of population-level variation in growth of two species of prairie clover and of their symbiotic associations with nitrogen-fixing bacteria. METHODS: Seed representing two species, Dalea candida and D. purpurea, from the same five source populations were planted into an experimental site in Minnesota. We assessed variation within and among source

Arbuscular Mycorrhizal Fungi and Azospirillum brasilense Enhance Growth and Resource Use Efficiency of Tomato Under Resource-Limited Peat-Reduced Cultivation.

The transition towards peat-reduced and resource-efficient horticultural systems presents challenges for maintaining crop productivity because of altered nutrient dynamics and reduced microbial diversity within soilless substrates. Beneficial microbial inoculants may provide a sustainable approach to improve plant performance under reduced-input cultivation; however, their effectiveness in peat-reduced hydroponic environments remains poorly understood. This study evaluated the effects of arbuscular mycorrhizal fungi (AMF; Rhizophagus irregularis) and nitrogen-fixing bacteria (NFB; Azospirillum

Monitoring of antibacterial capabilities of biosynthesized gold nanoparticles facilitated cyanobacterium, Spirulina subsalsa, against MDR pathogenic bacteria.

Gold nanoparticles (AuNPs) were biosynthesized with the non-nitrogen fixing cyanobacterium Spirulina subsalsa (Ss-AuNPs), and their antibacterial properties were monitored. Several analytical techniques, i.e., UV-visible spectroscopy, dynamic light scattering, zeta potential, Fourier transform infrared spectroscopy, and field emission scanning electron microscopy, were employed for characterization. Thereafter, antibacterial efficacies of the biosynthesized AuNPs against multidrug-resistant bacterial strains, Escherichia coli, Acinetobacter baumannii, Staphylococcus aureus, and Streptococcus p

Nitric oxide-forming nitrite reductases in the anaerobic ammonium oxidizer Kuenenia stuttgartiensis.

Anaerobic ammonium-oxidizing (anammox) bacteria contribute to the global nitrogen cycle by removing fixed nitrogen from the environment. They do so via the anaerobic oxidation of ammonium to dinitrogen gas, with nitrite as terminal electron acceptor. The first step in this so-called anammox reaction is the proposed conversion of nitrite to nitric oxide by a nitrite reductase. There is an unusual diversity and redundancy in anammox nitrite reductases and to gain more insight into the puzzling redundancy and diversity, we investigated the putative reductases in the model anammox species 'Candida

Phenotypic and molecular characterisation of efficient nitrogen-fixing Azotobacter strains from rice fields for crop improvement.

Biological nitrogen fixation (BNF) is highly effective in the field and potentially useful to reduce adverse effects chemical fertilisers. Here, Azotobacter species were selected via phenotypic, biochemical and molecular characterisations from different rice fields. Acetylene reduction assay of Azotobacter spp. showed that Azotobacter vinelandii (Az3) fixed higher amount of nitrogen (121.09 nmol C2H4 mg(-1) bacteria h(-1)). Likewise, its plant growth functions, viz. siderophore, hydrogen cyanide, salicylic acid, IAA, GA3, zeatin, NH3, phosphorus solubilisation, ACC deaminase and iron tolerance

The role of proton excreted by Advenella kashmirensis DF12 during ammonium assimilation in phosphate solubilization.

Phosphate-solubilizing bacteria (PSB) can solubilize soil fixed phosphorus (P) to plant available forms. In previous studies, the mechanisms of inorganic phosphate solubilization by PSB mostly focused on the acidolysis of organic acids. Here we screened a highly efficient PSB, Advenella kashmirensis DF12, with the maximum P solubilization of 590 mg L- 1 at 6 days. In addition to its P solubilizing ability, DF12 also showed a tolerance to pH from 5 to 10 and a nitrogen fixation potential. The multiple functions of DF12 and its wide adaptability to various environmental conditions make it a prom

The phosphate-solubilising fungi in sustainable agriculture: unleashing the potential of fungal biofertilisers for plant growth.

Phosphate-solubilising fungi (PSF) are beneficial microorganisms that play a pivotal role in plant growth by increasing the availability of phosphorus (P) in soil. Although phosphorus is an essential nutrient for plants, it often becomes inaccessible as it binds into insoluble forms. PSF effectively facilitate the release of this bound phosphorus through diverse mechanisms. Numerous fungal species demonstrate the ability to solubilise various types of phosphate compounds. Among the commonly researched PSF are Penicillium, Aspergillus, Rhizopus, Fusarium, Trichoderma, and Sclerotium. Moreover,

Rapid cultivation and enrichment of anammox bacteria solely using traditional activated sludge as inoculum and biocarrier in low-strength real sewage treatment.

In low-ammonia sewage anammox process, cultivation and enrichment of anammox bacteria (AnAOB) is a challenge especially from traditional activated sludge. To this end, a novel strategy solely using activated sludge as inoculum and biocarrier in a dynamic fixed-bed reactor was proposed in this study. During 115-day operation, excellent performance was achieved with influent total inorganic nitrogen (TIN) and effluent TIN of 55.3 mg·L-1 and 4.1 mg·L-1, respectively. Rapid enrichment of AnAOB (doubling time: 8.5 days) was demonstrated by augmented specific anammox activity (trace value to 1.85 mg

Managing Friends and Foes: Sanctioning Mutualists in Mixed-Infection Nodules Trades off With Defense Against Antagonists.

Successful plant growth requires plants to minimize harm from antagonists and maximize benefit from mutualists. However, these outcomes may be difficult to achieve simultaneously, since plant defenses activated in response to antagonists can compromise mutualism function, and plant resources allocated to defense may trade off with resources allocated to managing mutualists. Here, we investigate how antagonist attack affects plant ability to manage mutualists with sanctions, in which a plant rewards cooperative mutualists and/or punishes uncooperative mutualists. We studied interactions among w

Harnessing chickpea bacterial endophytes for improved plant health and fitness.

Endophytic bacteria live asymptomatically inside the tissues of host plants without inflicting any damage. Endophytes can confer several beneficial traits to plants, which can contribute to their growth, development, and overall health. They have been found to stimulate plant growth by enhancing nutrient uptake and availability. They can produce plant growth-promoting substances such as auxins, cytokinins, and gibberellins, which regulate various aspects of plant growth and development. Endophytes can also improve root system architecture, leading to increased nutrient and water absorption. So

Cobalt: An Essential Micronutrient for Plant Growth?

Cobalt is a transition metal located in the fourth row of the periodic table and is a neighbor of iron and nickel. It has been considered an essential element for prokaryotes, human beings, and other mammals, but its essentiality for plants remains obscure. In this article, we proposed that cobalt (Co) is a potentially essential micronutrient of plants. Co is essential for the growth of many lower plants, such as marine algal species including diatoms, chrysophytes, and dinoflagellates, as well as for higher plants in the family Fabaceae or Leguminosae. The essentiality to leguminous plants is

Endophytic microbes: biodiversity, plant growth-promoting mechanisms and potential applications for agricultural sustainability.

Endophytic microbes are known to live asymptomatically inside their host throughout different stages of their life cycle and play crucial roles in the growth, development, fitness, and diversification of plants. The plant-endophyte association ranges from mutualism to pathogenicity. These microbes help the host to combat a diverse array of biotic and abiotic stressful conditions. Endophytic microbes play a major role in the growth promotion of their host by solubilizing of macronutrients such as phosphorous, potassium, and zinc; fixing of atmospheric nitrogen, synthesizing of phytohormones, si

Expression cloning and biochemical characterization of a Rhizobium leguminosarum lipid A 1-phosphatase.

Lipid A of Rhizobium leguminosarum, a nitrogen-fixing plant endosymbiont, displays several significant structural differences when compared with Escherichia coli. An especially striking feature of R. leguminosarum lipid A is that it lacks both the 1- and 4'-phosphate groups. Distinct lipid A phosphatases that attack either the 1 or the 4' positions have previously been identified in extracts of R. leguminosarum and Rhizobium etli but not Sinorhizobium meliloti or E. coli. Here we describe the identification of a hybrid cosmid (pMJK-1) containing a 25-kb R. leguminosarum 3841 DNA insert that di

Plant growth promotion mechanisms of bacteria isolated from a long-term reclaimed smelter waste deposit.

The use of beneficial bacterial strains is proposed as a nature based practice to support sustainable crop production. Strains exposed to extreme environmental stress may have developed robust stress resistance and the capacity to enhance plant growth under unfavorable conditions. Our study provides the new aspect in characterising bacteria from polluted soil. The novelty of our study was isolation of bacteria from a long-term contaminated site and their testing for plant growth promoting mechanisms. The aim of this research was to characterize bacterial strains, collected from the root zone o

Exploring the diversity and evolutionary strategies of prophages in Hyphomicrobiales, comparing animal-associated with non-animal-associated bacteria.

The Hyphomicrobiales bacterial order (previously Rhizobiales) exhibits a wide range of lifestyle characteristics, including free-living, plant-association, nitrogen-fixing, and association with animals (Bartonella and Brucella). This study explores the diversity and evolutionary strategies of bacteriophages within the Hyphomicrobiales order, comparing animal-associated (AAB) with non-animal-associated bacteria (NAAB). We curated 560 high-quality complete genomes of 58 genera from this order and used the PHASTER server for prophage annotation and classification. For 19 genera with representativ

Pontiella agarivorans sp. nov., a novel marine anaerobic bacterium capable of degrading macroalgal polysaccharides and fixing nitrogen.

Marine macroalgae produce abundant and diverse polysaccharides, which contribute substantially to the organic matter exported to the deep ocean. Microbial degradation of these polysaccharides plays an important role in the turnover of macroalgal biomass. Various members of the Planctomycetes-Verrucomicrobia-Chlamydia (PVC) superphylum are degraders of polysaccharides in widespread anoxic environments. In this study, we isolated a novel anaerobic bacterial strain NLcol2T from microbial mats on the surface of marine sediments offshore Santa Barbara, CA, USA. Based on 16S ribosomal RNA (rRNA) gen

Genetic Diversity and Symbiotic Efficiency of Indigenous Common Bean Rhizobia in Croatia.

Nodule bacteria (rhizobia) in symbiotic associations with legumes enable considerable entries of biologically fixed nitrogen into soil. Efforts are therefore made to intensify the natural process of symbiotic nitrogen fixation by legume inoculation. Studies of field populations of rhizobia open up the possibility to preserve and probably exploit some indigenous strains with hidden symbiotic or ecological potentials. The main aim of the present study is to determine genetic diversity of common bean rhizobia isolated from different field sites in central Croatia and to evaluate their symbiotic e

YOLO-based high-throughput phenotyping pipeline for soybean nodulation traits in genomic research.

The symbiotic interaction between soybean (Glycine max) and Bradyrhizobium japonicum results in the formation of root nodules, specialized organs that house nitrogen-fixing bacteria converting atmospheric N2 into plant-accessible ammonia (NH3). Accurate quantification of nodule traits is essential for understanding host-microbe interactions and genetic determinants of nodulation. However, traditional manual or semi-quantitative approaches are labor-intensive, subjective, and unsuitable for large-scale studies. Here, we present a high-throughput phenotyping pipeline based on the YOLO deep learn
