Spleen-tumor dual-targeting vaccines reshape anticancer landscape by activating both innate and adaptive immunity.

Owing to the reticular structure and dense lymphocyte concentration within the spleen, spleen-targeted vaccines can overcome the challenges faced by conventional vaccines, such as inefficient antigen delivery and delayed immune activation. However, the existing spleen-targeted vaccines are ineffective in preventing and treating immunologically cold tumors. Herein, we develop a spleen/tumor dual-target hybrid vaccine that combines red blood cell membrane-derived vesicles (RBCVs) and bacterial outer membrane vesicles (OMVs) to deliver tumor antigens. This hybrid vaccine utilizes RBCVs to enhance

Heterologous Prime-Boost Vaccination with GRA35-Encoding DNA and mRNA Vaccines Enhances Protective Immunity Against Toxoplasma gondii Infection in Mouse Models.

Toxoplasma gondii is an obligate intracellular protozoan parasite that causes toxoplasmosis, posing a significant threat to human health and livestock production worldwide. Although monovalent DNA or mRNA vaccines often confer only partial protection, whether these platforms can be effectively integrated into a heterologous prime-boost regimen against T. gondii remains to be fully elucidated. Here, we constructed GRA35-encoding DNA and mRNA vaccines and evaluated their immunogenicity and protective efficacy, administered either alone or in heterologous prime-boost combinations, in C57BL/6 and

Advances and Challenges in Nanodelivery Systems for RNA Vaccines.

RNA vaccines have become a transformative technology in infectious disease prevention and cancer treatment, thanks to their rapid development, high immunogenicity, and scalability in production. However, the clinical application of RNA vaccines faces significant challenges, including the inherent instability and susceptibility to degradation of naked RNA, its poor permeability across cell membranes, and the potential for nonspecific immune responses following systemic administration. Nanodelivery systems are key to addressing these challenges; by providing physical protection, enabling targete

Sequence Analysis of Egyptian Foot-and-Mouth Disease Virus Field and Vaccine Strains: Intertypic Recombination and Evidence for Accidental Release of Virulent Virus.

In spite of annual mass vaccination programs with polyvalent inactivated vaccines, the incidence and economic impact of foot-and-mouth disease (FMD) in Egypt is high. Viruses of the A, O and SAT 2 serotypes are endemic and repeated incursions of new lineages from other countries lead to an unstable situation that makes the selection of appropriate vaccine antigens very difficult. In this study, whole genome sequencing of a 2016 serotype A isolate from Egypt revealed a recombination event with an African serotype O virus. Based on available vaccine matching data, none of the vaccines currently

Efficacy of an optimal vaccination strategy for H120 and NNA vaccines against the novel HX strain of the IBV GVI-1 genotype.

The infectious bronchitis virus (IBV) causes significant economic losses to the global poultry industry. Recently, there has been a rapid spread of the GVI-1 lineage of IBV in Asia, particularly in China. However, to date there have been few studies that have assessed the immune protection efficacy of commonly used IB vaccines against the GVI-1 lineage strains. In this study, we evaluated the protective efficacy of two commonly used vaccines, H120 and NNA, against the GVI-1 lineage HX strain based on serological neutralization tests and animal challenge protection experiments. The protective e

A Recombinant Subunit Based Zika Virus Vaccine Is Efficacious in Non-human Primates.

Zika Virus (ZIKV), a virus with no severe clinical symptoms or sequelae previously associated with human infection, became a public health threat following an epidemic in French Polynesia 2013-2014 that resulted in neurological complications associated with infection. Although no treatment currently exists, several vaccines using different platforms are in clinical development. These include nucleic acid vaccines based on the prM-E protein from the virus and purified formalin-inactivated ZIKV vaccines (ZPIV) which are in Phase 1/2 clinical trials. Using a recombinant subunit platform consistin

Characterizing influenza A virus lineages and clinically relevant mutations through high-coverage wastewater sequencing.

Influenza A virus poses significant public health challenges, causing seasonal outbreaks and pandemics. Its rapid evolution motivates continuous monitoring of circulating influenza genomes to inform vaccine and antiviral development. Wastewater-based surveillance offers an unbiased, cost-effective approach for genomic surveillance. We developed a novel tiling amplicon primer panel that covers diversity of influenza A virus, targeting segments of the surface proteins HA, NA, and M of subtypes H1N1 and H3N2. Using this panel, we sequenced nucleic acid extracts from 59 Swiss wastewater samples co

Quantification of two variant strains contained in freeze-dried Japanese BCG vaccine preparation by real-time PCR.

Japanese bacillus Calmette-Guerin (BCG) vaccine preparation contains two types of variant strains, Type I, which has a 22-base-pair deletion in the RD16 region, and Type II, which has an identical sequence to those of other BCG strains. In this study, we established a method to quantify the percentage of variant strain Type II contained in freeze-dried BCG product with real-time PCR. With this method we examined the master seed lot Tokyo 172, two secondary seed lots, Tokyo 172-1 and Tokyo 172-2, which were produced from Tokyo 172, and four commercial lots produced form Tokyo 172-1. Messenger RNA-based vaccines: progress, challenges, applications.

Twenty years after the demonstration that messenger RNA (mRNA) was expressed and immunogenic upon direct injection in mice, the first successful proof-of-concept of specific protection against viral infection in small and large animals was reported. These data indicate wider applicability to infectious disease and should encourage continued translation of mRNA-based prophylactic vaccines into human clinical trials. At the conceptual level, mRNA-based vaccines-more than other genetic vectors-combine the simplicity, safety, and focused immunogenicity of subunit vaccines with favorable immunologi

Zika Virus Vaccine Development.

The emergence of Zika virus in Brazil and its association with microcephaly and Guillain-Barré syndrome led to accelerated vaccine development efforts. Based on prior flavivirus vaccine development programs, knowledge of flavivirus particle structure, definition of E dimers as the key antigenic target, and deep understanding of neutralizing mechanisms, multiple vaccine strategies have advanced to the stage of clinical evaluation with unprecedented speed. These include nucleic acid (DNA and messenger RNA), whole-inactivated virus, live-attenuated or chimeric virus, and protein or viruslike part

Recent Advances in Cancer Vaccines: Challenges, Achievements, and Futuristic Prospects.

Cancer is a chronic disease, and it can be lethal due to limited therapeutic options. The conventional treatment options for cancer have numerous challenges, such as a low blood circulation time as well as poor solubility of anticancer drugs. Therapeutic cancer vaccines emerged to try to improve anticancer drugs' efficiency and to deliver them to the target site. Cancer vaccines are considered a viable therapeutic technique for most solid tumors. Vaccines boost antitumor immunity by delivering tumor antigens, nucleic acids, entire cells, and peptides. Cancer vaccines are designed to induce lon

Bidirectional Interplay Between Tumor Vaccines and the Tumor Microenvironment: Mechanisms, Cold-to-Hot Conversion, and Combination Strategies.

Therapeutic cancer vaccines are designed to initiate tumor-specific immunity, yet their clinical success depends not only on antigen selection but also on the capacity to overcome the profoundly suppressive tumor microenvironment. Within tumors, abnormal vasculature, hypoxia, nutrient competition, acidic pH, and suppressive myeloid and stromal cells collectively constrain antigen presentation, T-cell priming, trafficking, and effector function, often converting otherwise immunogenic vaccination into an ineffective immune stimulus. Recent advances in neoantigen discovery, dendritic cell enginee

Preventative and therapeutic vaccines for cervical cancer.

"High-risk" genotypes of the human papillomavirus (HPV), most commonly HPV genotype 16, are the primary etiologic agents of cervical cancer. Indeed HPV DNA is detected in 99% of cervical carcinomas. Thus, cervical cancer and other HPV-associated malignancies might be prevented or treated by the induction of the appropriate viral-antigen-specific immune responses. Transmission of papillomavirus may be prevented by the generation of antibodies to capsid proteins L1 and L2 that neutralize viral infection. HPV L1 virus-like particles (VLPs) show great promise as prophylactic HPV vaccines in ongoin

A point-of-care testing platform for pathogen nucleic acid analysis utilizing paper-based extraction and portable qPCR.

Rapid and reliable diagnostic methods are crucial for controlling animal disease outbreaks and reducing mortality, especially when effective drugs or vaccines are unavailable. While real-time quantitative polymerase chain reaction (qPCR) remains the gold standard for nucleic acid detection due to its superior sensitivity and specificity, conventional workflows are inherently cumbersome. Traditional extraction and amplification processes typically require 1.5 to 3 h, with further delays incurred by transporting samples to centralized laboratories. To overcome these limitations, this study intro

Advances in lipids design for LNP-mediated DNA and RNA delivery.

Gene therapy represents a promising strategy for treating a range of diseases. Non-viral gene delivery systems, including lipid nanoparticles (LNPs), polymer micelles, and liposomes, enable the tissue-specific delivery of genetic sequences and promote the expression of functional proteins in target cells. Compared with traditional carriers, LNPs possess distinct, mechanistically supported advantages: high biosafety with less than 5% cytotoxicity in most primary cells and minimal systemic inflammation in preclinical models, good reproducibility with a coefficient of variation less than 10% for

Between defence and delivery: the DNA sensing response to gene electrotransfer.

BACKGROUND: Gene therapy has emerged as a transformative biomedical approach, offering new therapeutic possibilities from many so far uncurable diseases through the introduction of recombinant nucleic acids into target cells. Among non-viral delivery techniques, gene electrotransfer (GET) has become one of the frequently applied methods in clinical trials. It is based on the application of short, high-intensity electric pulses that transiently permeabilize cell membranes and enable the efficient transfer of plasmid DNA or other types of recombinant nucleic acids into various cell types. Beyond
