Fluorescence Correlation Spectroscopy as a Versatile Method to Define Aptamer-Protein Interactions with Single-Molecule Sensitivity.

Aptamers are folded oligonucleotides that selectively recognize and bind a target and are consequently regarded as an emerging alternative to antibodies for sensing and therapeutic applications. The rational development of functional aptamers is strictly related to the accurate definition of molecular binding properties. Nevertheless, most of the methodologies employed to define binding affinities use bulk measurements. Here, we describe the use of fluorescence correlation spectroscopy (FCS) as a method with single-molecule sensitivity that quantitatively defines aptamer-protein binding. First

Characterisation of a human antibody that potentially links cytomegalovirus infection with systemic lupus erythematosus.

Human cytomegalovirus (HCMV) is a ubiquitous herpesvirus that has been linked with the development of systemic lupus erythematosus (SLE). Thus far, molecular mimicry has been implicated as the principal mechanism that explains this association. In this study, we characterise a potential alternative process whereby HCMV contributes to SLE. In a cohort of SLE patients, we show a significant association between HCMV infection and SLE through a human antibody response that targets UL44. UL44 is an obligate nuclear-resident, non-structural viral protein vital for HCMV DNA replication. The intracell

Aptamers in cancer therapy: Why has clinical translation lagged behind preclinical promise?

Aptamers have attracted substantial interest as cancer therapeutics due to their high target specificity, chemical programmability, and versatility in drug and RNA delivery; however, despite extensive preclinical development, clinical translation in oncology has remained extremely limited, with no aptamer-based therapies approved for cancer indications to date. This gap reflects not a failure of binding affinity, but a fundamental misalignment between aptamer selection and evaluation paradigms and the biological complexity of human tumors. Most aptamers are selected against purified proteins o

Midkine (MDK) as a central regulator of the tumor microenvironment: From developmental cytokine to therapeutic target.

Midkine (MDK) is an oncofetal, heparin-binding cytokine that is re-expressed across diverse cancers and correlates with aggressive disease and treatment resistance. This review synthesizes current evidence on MDK as a coordinator of tumor-intrinsic signaling and microenvironmental remodeling. We summarize MDK structural features, extracellular matrix interactions, and receptor systems that mediate MDK signaling, highlighting LRP1 and PTPRZ1 with context-dependent participation of ALK, nucleolin and integrins. Downstream, MDK engages MAPK, PI3K-AKT, STAT3 and NF-κB pathways to promote tumor cel

APE1/ATP dual-input AND-gated DNA nanoflowers as a colloidal nanoplatform for logic-controlled cancer cell imaging and DNAzyme gene silencing.

Colloidal DNA nanoflowers have emerged as promising biointerface platforms for cancer theranostics owing to their porous architectures, robust nuclease resistance, and favorable cytocompatibility. Integrating molecular logic gates into such bio-colloidal systems enables stringent spatial and temporal control over therapeutic activation, thereby minimizing off-target effects. Herein, we fabricate a APE1/ATP dual-input AND-gated DNA nanoflower (AMDNF) via rolling circle amplification (RCA). The resulting AMDNF features a flower-like organic-inorganic hybrid scaffold co-assembling abundant tandem

Dual Aptamers-Based SETDB1 PROTACs as Effective Anti-Tumor Strategies for Breast Cancer.

PROteolysis TArgeting Chimeras (PROTACs) have emerged as a promising strategy for drug development targeting oncogenic proteins. Here, we report the development and characterization of dual aptamer-based PROTACs targeting SET domain bifurcated histone lysine methyltransferase 1 (SETDB1), a key epigenetic regulator implicated in breast cancer progression, drug resistance, and tumor immune evasion. Using the Systematic Evolution of Ligands by Exponential Enrichment (SELEX) process, we identified a high-affinity single-stranded DNA (ssDNA) aptamer against SETDB1. This aptamer was conjugated to th

Multifunctional DNA nanoplatform based on near-infrared AgInS2 QDs and rolling circle amplification for fluorescence imaging and therapy of breast cancer cells.

To enhance the therapeutic efficacy of breast cancer cells, a multifunctional nanoplatform was developed based on near-infrared AgInS2 quantum dots (AIS QDs) and rolling circle amplification (RCA). The AIS QDs were synthesized via an environmentally benign hydrothermal method using glutathione as a surface ligand, demonstrating favorable optical and photothermal properties. The RCA-generated long DNA strands served as a scaffold to co-assemble AS1411 aptamer-conjugated AIS QDs, gene therapeutic agent (DNAzyme), and doxorubicin (Dox), forming a multimodal nanoplatform (AIS QDs@RD). Multi-Technique Integration Identifies O-GlcNAc Transferase in Fibroblast-Like Synoviocytes as a Therapeutic Target for Rheumatoid Arthritis.

Rheumatoid arthritis (RA) progression is driven by the pathogenic transformation of stromal fibroblast-like synoviocytes (FLSs). Using artificial intelligence-guided analysis of a synovial single-cell transcriptomic dataset, we identify O-GlcNAc transferase (OGT) as a key regulator enriched in RA-FLSs. Gain- and loss-of-function studies demonstrate that OGT is necessary to drive aggressive FLS phenotypes and exacerbate experimental arthritis. Mechanistically, OGT-mediated O-GlcNAcylation stabilizes SAP130, promoting histone deacetylation at the BTG2 promoter. This facilitates the deposition of

CRYβB2 is a biomarker for poor prognosis and response to CDK4/6 inhibitors in breast cancer.

Breast cancer mortality is higher in African American (AA) women than in European American women, but the molecular mechanisms remain poorly understood. Previously, we showed that β-crystallin B2 (CRYβB2) is upregulated in AA breast tumors and promotes oncogenesis via nucleolin (NCL) activation. Presently no biomarkers, other than estrogen receptor (ER)/progesterone receptor (PR) expression, are used to prescribe cyclin-dependent kinases 4/6 inhibitors (CDK4/6i) for women with hormone receptor positive (HR+) metastatic breast cancer. Here, we show that CRYβB2 overexpression drives cell cycle p

Global Profiling of Lysine Lactylation in Prostate Cancer Cells.

BACKGROUND/AIM: Lysine lactylation (Kla) is a recently identified post-translational modification derived from lactate that regulates diverse biological processes. Although Kla has been studied in several cancers, its role in prostate cancer (PC) remains unclear. The objective of this study is to profile Kla in PC in order to explore the mechanisms involved in PC progression. MATERIALS AND METHODS: We performed global Kla profiling in PC-3M prostate cancer cells using affinity enrichment with anti-Kla antibodies, followed by LC-MS/MS. Bioinformatics analyses were conducted to explore the func

RNAi knockdown of Nopp140 induces Minute-like phenotypes in Drosophila.

Nopp140 associates with small nucleolar RNPs to chaperone pre-rRNA processing and ribosome assembly. Alternative splicing yields two isoforms in Drosophila: Nopp140-True is homologous to vertebrate Nopp140 particularly in its carboxy terminus, whereas Nopp140-RGG contains a glycine and arginine-rich (RGG) carboxy terminus typically found in vertebrate nucleolin. Loss of ribosome function or production at critical points in development leads to Minute phenotypes in Drosophila or the Treacher Collins syndrome (TCS) in humans. To ascertain the functional significance of Nopp140 in Drosophila deve

G0S2 modulates homeostatic proliferation of naïve CD8⁺ T cells and inhibits oxidative phosphorylation in mitochondria.

Since its discovery, diverse functions have been attributed to the G0/G1 switch gene 2 (G0S2), from lipid metabolism to control of cell proliferation. Our group showed for the first time that G0S2 promotes quiescence in hematopoietic stem cells by interacting with and retaining nucleolin around the nucleus. Herein, we report the role of G0S2 in the differentiation and function of CD8(+) T cells examined in mice with an embryonic deletion of the G0s2 gene. G0S2 expression in naïve CD8(+) T cells decreased immediately after T-cell receptor activation downstream of the mitogen-activated protein k

Nucleolar localization of RPS19 protein in normal cells and mislocalization due to mutations in the nucleolar localization signals in 2 Diamond-Blackfan anemia patients: potential insights into pathophysiology.

Ribosomal protein S19 (RPS19) is frequently mutated in Diamond-Blackfan anemia (DBA), a rare congenital hypoplastic anemia. Recent studies have shown that RPS19 expression decreases during terminal erythroid differentiation. Currently no information is available on the subcellular localization of normal RPS19 and the potential effects of various RPS19 mutations on cellular localization. In the present study, using wild-type and mutant RPS19 cDNA, we explored the subcellular distribution of normal and mutant proteins in a fibroblast cell line (Cos-7 cells). RPS19 was detected primarily in the n

Etomidate vs Propofol in Older Surgical Patients.

PURPOSE: There are currently no means available for the efficient delivery of recombinant proteins into retinal cells in vivo. Although cell-penetrating peptides have been somewhat effective in protein delivery to the retina, they generally require conjugation chemistry with the payload, negatively impacting function of the therapeutic protein. In this study, we developed a novel peptide (Nuc1) that acts as a chaperone for delivery of small and large molecules, including steroids, peptides, antibodies, recombinant proteins, and viruses (adeno-associated viruses [AAVs]) across biological membra

Properties and Potential Antiproliferative Activity of Thrombin-Binding Aptamer (TBA) Derivatives with One or Two Additional G-Tetrads.

In this paper, we study the biological properties of two TBA analogs containing one and two extra G-tetrads, namely TBAG3 and TBAG4, respectively, and two further derivatives in which one of the small loops at the bottom (TBAG41S) or the large loop at the top (TBAG4GS) of the TBAG4 structure has been completely modified by replacing all loop residues with abasic site mimics. The therapeutical development of the TBA was hindered by its low thermodynamic and nuclease stability, while its potential as an anticancer/antiproliferative molecule is also affected by the anticoagulant activity, being a

Characterization of putative Japanese encephalitis virus receptor molecules on microglial cells.

Japanese encephalitis virus (JEV) a mosquito-borne flavivirus is a major cause of viral encephalitis in Asia. While the principle target cells for JEV in the central nervous system are believed to be neurons, microglia are activated in response to JEV and have been proposed to act as a long lasting virus reservoir. Viral attachment to a host cell is the first step of the viral entry process and is a critical mediator of tissue tropism. This study sought to identify molecules associated with JEV entry to microglial cells. Virus overlay protein-binding assay (VOPBA) and liquid chromatography-mas

P2P-R protein localizes to the nucleolus of interphase cells and the periphery of chromosomes in mitotic cells which show maximum P2P-R immunoreactivity.

P2P-R is a nuclear protein that can bind both p53 and Rb1. Its functions include roles in the control of RNA metabolism, apoptosis, and p53-dependent transcription. The expression of P2P-R also is repressed in G1 arrested terminally differentiated cells. The current studies therefore evaluated if P2P-R undergoes cell cycle-associated changes in its abundance and/or localization. Western blots show that relative to G0 quiescent cells, P2P-R protein levels are higher in populations of G2/M cells prepared by the physiological parasynchronization technique of serum deprivation followed by serum st

Next-Generation Liposomal and Nanoparticle-Based Systems for Glioblastoma: Molecular Targeting Strategies to Overcome the Blood-Brain Barrier.

Glioblastoma (GBM) remains one of the most lethal and intractable central nervous system malignancies, characterized by aggressive cellular proliferation, profound hypoxia, and an immunosuppressive microenvironment. The primary pharmacokinetic obstacle for systemic interventions is the blood-brain barrier (BBB), which restricts the infiltration of free therapeutic agents. To overcome this limitation, translational nanomedicine has driven the design of next-generation delivery systems, including liposomal vectors and synthetic organic, inorganic, and hybrid nanoparticles. This study establishes

Aptamer-Functionalized Liposomes for Targeted Delivery of Anticancer Drugs in Lung Cancer.

Aptamer-functionalized liposomes are a promising strategy to improve the selectivity of anticancer therapies. AT11-L2 is a G-quadruplex (G4)-forming aptamer with high affinity for nucleolin (NCL), which is overexpressed at the surface of non-small cell lung cancer (NSCLC) cells. Here, we developed AT11-L2-functionalized liposomes loaded with doxorubicin (DOX) or BRACO-19 for targeted delivery to NSCLC cells. The effects of both compounds on AT11-L2 stability and the ability of the aptamer to retain G4 folding after liposome conjugation were evaluated. Liposomes were characterized for size, pol
