Synthesis of an oleic acid based pH-responsive lipid and its application in nanodelivery of vancomycin.

Stimuli-responsive nano-drug delivery systems can optimize antibiotic delivery to infection sites. Identifying novel lipids for pH responsive delivery to acidic conditions of infection sites will enhance the performance of nano-drug delivery systems. The aim of the present investigation was to synthesize and characterize a biosafe novel pH-responsive lipid for vancomycin delivery to acidic conditions of infection sites. A pH-responsive solid lipid, N-(2-morpholinoethyl) oleamide (NMEO) was synthesized and used to prepare vancomycin (VCM)-loaded solid lipid nanoparticles (VCM_NMEO SLNs). The pa

Effect of the rooster semen enrichment with oleic acid on the quality of semen during chilled storage.

Liquid storage of avian spermatozoa is currently being employed in programs utilizing the artificial insemination to optimize the management of genetically superior males. It is mandatory to use efficient semen storage techniques in order to prevent the reduction of the fertilizing ability of stored semen. The present study was designated to evaluate the effect of oleic acid on rooster semen quality stored at 4°C for 48 h. Semen was collected from 10 roosters twice a week. Good quality ejaculates were pooled and after dilution, the semen was enriched with 0 (control), 0.125 (O 0.125), 0.25 (O

Role of p53 in the cellular response following oleic acid accumulation in Chang liver cells.

Abnormal accumulation of fatty acids triggers the harmful cellular response called lipotoxicity. In this study, we investigated the cellular response following accumulation of oleic acid (OA), a monounsaturated fatty acid, in human Chang liver cells. OA droplets were distributed freely in the cytoplasm and/or degraded within lysosomes. OA exposure increased ATP production and concomitantly dilated mitochondria. At 24h after OA exposure, cell viability decreased slightly and was coupled with a reduction in mitochondrial Ca(2+) concentration, the alteration in cell viability was also associated

SCD1-Mediated Lipid Metabolism Alleviates Neuronal Ferroptosis in Subarachnoid Hemorrhage: Role of Oleic Acid Supplementation.

Subarachnoid hemorrhage (SAH) can cause severe neuronal damage and trigger multiple molecular mechanisms of cell death, among which ferroptosis has garnered significant attention due to its dependence on lipid peroxidation. In this study, ferroptosis-related markers were found to be significantly dysregulated in an in vitro SAH model, confirming the neuronal ferroptosis after SAH. Transcriptomic sequencing results indicated a marked downregulation of stearoyl-CoA desaturase 1 (SCD1), which was further validated by Western blot. Immunofluorescence demonstrated its colocalization with the neuron

Manometric responses of human duodenum during infusion of HCl, hyperosmolar saline, bile and oleic acid.

Duodenal motor activity is incompletely understood. The purpose of this study was to define the contractile patterns of the duodenum that occur in response to rate controlled injection of various solutions. In nine healthy volunteers we placed a six channel perfused catheter, and recorded pressure activity in the antrum, pylorus and duodenum. Volumes of 10 and 20 mL of 0.9% NaCl, 100 mM HCl (pH 1), 5% NaCl (1711 mOsm/kg), human bile and iso-osmolar sodium oleate were randomly injected into the duodenum at 20 ml/min, starting 15 min after phase III migratory motor complex (MMC). Prone position reverses gravitational distribution of perfusion in dog lungs with oleic acid-induced injury.

Although oxygenation improves in patients with the adult respiratory distress syndrome and in animals with oleic acid- (OA) induced acute lung injury when they are turned from the supine to the prone position, the mechanism(s) by which this improvement occurs is not known. Several groups have speculated that this improvement results from preferential edema accumulation in the dorsal lung regions and redistribution of perfusion away from these regions when the patients are turned to the prone position. We used radiolabeled microspheres to measure the regional distribution of perfusion (Qr) to t

Characterization of a novel sn1,3 lipase from Ricinus communis L. suitable for production of oleic acid-palmitic acid-glycerol oleate.

The hydrolysis properties of lipase in castor was evaluated using two different substrate forms (tripalmitic glycerides and trioleic glycerides) to catalyze the reaction under different operational conditions. RcLipase was obtained from castor seeds and results show that RcLipase is a conservative serine lipase with a conserved catalytic center (SDH) and a conserved pentapeptide (GXSXG). This enzyme exhibited the greatest activity and tolerance to chloroform and toluene when it was expressed in Pichia pastoris GS115 at 40 ℃ and pH 8.0. Zn and Cu ions exerted obvious inhibitory effects on the e

Oleic acid and docosahexaenoic acid cause an increase in the paracellular absorption of hydrophilic compounds in an experimental model of human absorptive enterocytes.

Surface active compounds present in food possibly have the ability to enhance the absorption of water soluble toxic agents. Therefore, we investigated whether fatty acids such as oleic acid and docosahexaenoic acid (DHA), both commonly present in food, negatively affect the integrity of tight junctions (TJ) in the intestinal epithelium and thereby increase the absorption of poorly absorbed hydrophilic substances. Caco-2 cells, which are derived from human absorptive enterocytes, were grown on permeable filters for 20-25 days. Differentiated cell monolayers were apically exposed for 90min to ma

Oleic acid promotes lung injury in hypertriglyceridaemia-associated acute pancreatitis via the PIEZO1/NR4A1/CPT1A axis impairing endothelial fatty acid oxidation.

BACKGROUND: Hypertriglyceridaemia-induced acute pancreatitis (HTG-AP) is frequently complicated by acute lung injury (ALI), which worsens prognosis. Oleic acid (OA), a major circulating free fatty acid, may play a key role, but the underlying mechanism remains unclear. OBJECTIVE: To investigate the relationship between plasma OA and HTG-AP-associated ALI and to explore the mechanism by which OA disrupts endothelial barrier through PIEZO1-mediated impairment of fatty acid oxidation (FAO). DESIGN: This study used clinical sample analysis, an HTG-AP mouse model and OA-stimulated human umbilical

Role of fatty acid transport protein 4 in oleic acid-induced glucagon-like peptide-1 secretion from murine intestinal L cells.

The antidiabetic intestinal L cell hormone glucagon-like peptide-1 (GLP-1) enhances glucose-dependent insulin secretion and inhibits gastric emptying. GLP-1 secretion is stimulated by luminal oleic acid (OA), which crosses the cell membrane by an unknown mechanism. We hypothesized that L cell fatty acid transport proteins (FATPs) are essential for OA-induced GLP-1 release. Therefore, the murine GLUTag L cell model was used for immunoblotting, [(3)H]OA uptake assay, and GLP-1 secretion assay as determined by radioimmunoassay following treatment with OA ± phloretin, sulfo-N-succinimidyl oleate,

Pomegranate peel-derived Cu/hydrochar nanocomposites: a sustainable catalyst for biodiesel production via oleic acid esterification.

Transforming pomegranate peel waste into a biodiesel catalyst advances waste-to-resource circularity. A facile hydrothermal carbothermal reduction method was used to synthesize copper-decorated hydrochar nanocomposites derived from peel bio-waste and CuSO4, with Cu(II) concentrations of 10, 25, and 50 mmol/L (denoted as Cu10/HC, Cu25/HC, and Cu50/HC, respectively). The physicochemical properties of the nanocomposites were characterized by FTIR, XRD, BET, XPS, SEM-EDX, and NH₃-TPD, and their catalytic performance was evaluated for biodiesel production via the esterification of oleic acid (OA) w

Gestational diabetes severity stratification during pregnancy: role of plasma oleic acid as a possible early marker.

Normal pregnancy is characterized by changes in lipid metabolism with significant implications for the health of both mother and offspring. When these changes develop into maternal dyslipidemia, a significant association with adverse pregnancy outcomes has been observed, including the development of gestational diabetes (GD), modulation of the inflammatory response, and excessive fetal growth. In the present study, we performed a lipidomic assessment of patients at GD diagnosis (24-28 weeks of gestation) and 12 weeks after diagnosis. We found higher levels of esterified oleic acid in plasma at

Oleic Acid Prevents Isoprenaline-Induced Cardiac Injury: Effects on Cellular Oxidative Stress, Inflammation and Histopathological Alterations.

The present study was designed to assess the cardio-protective role of oleic acid in myocardial injury (MI) induced by intra-peritoneal injection of isoprenaline (ISO) in rats for 2 consecutive days. Oleic acid (OA) was administered orally (@ 5 mg/kg b.wt and 10 mg/kg b.wt) for 21 days before inducing MI. Pre-exposure to OA at higher dose significantly improved the HW/BW ratio, myocardial infarct size, lipid profiles (total cholesterol, HDL-C) and cardiac injury biomarkers (LDH, CK-MB, cardiac troponin-I, MMP-9), thus suggesting its cardio-protective role. The ameliorative potential of the hig

Oleic Acid Protects against Hepatic Ischemia and Reperfusion Injury in Mice by Inhibiting AKT/mTOR Pathways.

Hepatic ischemia-reperfusion (I/R) injury is a serious complication in patients who have undergone hepatic surgery such as orthotopic liver transplantation and partial hepatectomy. Recently, a new cytoprotective agent, ursodeoxycholyl lysophosphatidylethanolamide (UDCA-LPE), was reported to protect against hepatic I/R injury. However, the protective mechanism of UDCA-LPE is not fully understood. Therefore, we conducted this study to explore its underlying mechanism. We used liquid chromatography-tandem mass spectrometry (LC-MS/MS) to analyze the liver lipid metabolism changes in mice during I/
