Influence of olfactory innervation on neurogenesis in the developing olfactory bulb of the frog, Xenopus laevis.

Previous research on development of the Xenopus olfactory bulb from our laboratory has shown that mitral cells begin to differentiate after olfactory axons make contact with the olfactory bulb, and the number of olfactory axons is correlated with the number of mitral cells throughout development. In embryos, removal of all afferent innervation before the mitral cells begin to differentiate results in a failure of the bulb to form; removal of half the olfactory axons, results in development of half the normal number of mitral cells. At larval stages, transection of the olfactory nerve results i

Organization of projections from olfactory epithelium to olfactory bulb in the frog, Rana pipiens.

One hypothesis for the coding of olfactory quality is that regions of the olfactory epithelium are differentially sensitive to particular odor qualities and that this regional sensitivity is conveyed to the olfactory bulb in a topographic manner by the olfactory nerve. A corollary to this hypothesis is that there is a sufficiently orderly connection between the epithelium and the olfactory bulb to convey this topographical coding. Thus we examined topography in the projection from epithelium to bulb in the frog, which has been the subject of numerous electrophysiological studies but has not ye

Reinnervation of the olfactory bulb after section of the olfactory nerve in monkey (Saimiri sciureus).

Section of the fila olfactoria in squirrel monkey, a non-human primate, induces rapid degeneration of the sensory axon terminals in the olfactory bulb glomeruli. A population of axons, from newly formed sensory neurons in the olfactory neuroepithelium, regrow, passes the lamina cribrosa and, upon reaching the olfactory bulb, reinnervates the glomeruli. A new set of synaptic contacts is reformed between the sensory terminals and the post-synaptic dendritic processes of the glomeruli. Our observations indicate that this portion of the CNS of a non-human primate can be reinnervated after deaffere

COVID-19-induced anosmia associated with olfactory bulb atrophy.

As the global COVID-19 pandemic evolves, our knowledge of the respiratory and non-respiratory symptoms continues to grow. One such symptom, anosmia, may be a neurologic marker of coronavirus infection and the initial presentation of infected patients. Because this symptom is not routinely investigated by imaging, there is conflicting literature on neuroimaging abnormalities related to COVID-19-related anosmia. We present a novel case of COVID-19 anosmia with definitive olfactory bulb atrophy compared with pre-COVID imaging. The patient had prior MR imaging related to a history of prolactinoma

Decreased olfactory bulb volumes in patients with fibromyalgia syndrome.

Among the other symptoms, impaired olfactory function such as odor identification, threshold, and discrimination have been reported in patients with fibromyalgia syndrome (FMS). To investigate olfactory bulb (OB) volumes in FMS, by using magnetic resonance imaging (MRI), and to make reasonable suggestions are the goals of the present study. The study included 62 individuals as the FMS group (n = 30) and the control group (n = 32). MRI examinations were performed by a 1.5-T scanner and a standard head coil was used for the images. The coronal T2-weighted images were used for to measure OB volum

Rauwolfia vomitoria inhibits olfaction and modifies olfactory bulb cells.

The rising cost of orthodox medication has endeared so many to the use of herbs for the management of neurological conditions. Rauwolfia vomitoria (RV) one of such herbs is a rainforest shrub whose parts are used locally in the management of psychiatry and other medical issues. Its usefulness though not in doubt is wrapped with adverse reports as its active constituents depletes brain monoamine and dopamine stores. This motivated this research on the effects of the root bark extract on olfaction and the olfactory bulb of adult Wistar rats. Eighteen adult Wistar rats (220g average) were divided

Secretagogin-containing neurons in the mouse main olfactory bulb.

Secretagogin (SCGN) is a recently discovered calcium binding protein of the EF hand family. We studied the structural features of SCGN-positive neurons in the mouse main olfactory bulb (MOB). SCGN-positive neurons were localized throughout layers but clustered in the glomerular layer (GL), mitral cell layer (MCL) and granule cell layer (GCL). They were heterogeneous, including numerous juxtaglomerular neurons, granule cells, small to medium-sized neurons in the external plexiform layer (EPL), and a few small cells in the ependymal/subependymal layer. Calretinin and/or tyrosine hydroxylase occa

Synaptogenesis in the mouse olfactory bulb during glomerulus development.

Synaptogenesis is essential for the development of neuronal networks in the brain. In the olfactory bulb (OB) glomeruli, numerous synapses must form between sensory olfactory neurons and the dendrites of mitral/tufted and periglomerular cells. Glomeruli develop from E13 to E16 in the mouse, coincident with an increment of the neuropil in the border between the external plexiform (EPL) and olfactory nerve layers (ONL), coupled to an extensive labelling of phalloidin and GAP-43 from the ONL to EPL. We have tracked synaptogenesis in the OB during this period by electron microscopy (EM) and immuno

Apolipoprotein E immunoreactivity in human and mouse olfactory bulb.

We evaluated the distribution of apolipoprotein E (apoE) immunoreactivity in mouse and human olfactory bulbs. ApoE immunoreactivity was present in the olfactory nerve and around the glomeruli. Immunoreactivity was seen in somata that appeared to be glial. No neuronal staining was seen. The apoE immunoreactivity was also present in the mouse olfactory bulb subependymal layer. The specificity of immunoreactivity was confirmed with apoE-deficient mice (apoE gene knock-out mice, apoE KO) which did not display any immunoreactivity. The presence of apoE around glomeruli and nerve suggest that apoE i

Glomeruli of the Mouse Olfactory Bulb: Numbers, Sizes, Shapes.

The discovery of olfactory bulb glomeruli was reported by the neuroanatomist Camillo Golgi in 1875, but the number of glomeruli in a mammalian olfactory bulb remains unknown. The 1141 intact odorant receptor genes in the mouse genome are key determinants of the coalescence of axons of olfactory sensory neurons into glomeruli at recognizable positions in the olfactory bulb. Here, we devised a methodology for quantifying the numbers, sizes, and shapes of the glomeruli of mouse olfactory bulbs. Using serial two-photon tomography, we generated complete image stacks of 12 mouse olfactory bulbs labe

Ca2+-permeable AMPA receptors in mouse olfactory bulb astrocytes.

Ca2+ signaling in astrocytes is considered to be mainly mediated by metabotropic receptors linked to intracellular Ca2+ release. However, recent studies demonstrate a significant contribution of Ca2+ influx to spontaneous and evoked Ca2+ signaling in astrocytes, suggesting that Ca2+ influx might account for astrocytic Ca2+ signaling to a greater extent than previously thought. Here, we investigated AMPA-evoked Ca2+ influx into olfactory bulb astrocytes in mouse brain slices using Fluo-4 and GCaMP6s, respectively. Bath application of AMPA evoked Ca2+ transients in periglomerular astrocytes that

Olfactory bulb volume in Taiwanese patients with posttraumatic anosmia.

BACKGROUND: Olfactory bulb (OB) volume has been shown to be an indicator of olfactory function. However, few studies have been done in Asia to investigate the influence of different disorders on OB volume. METHODS: Data from patients with posttraumatic anosmia were collected in our department. Their olfactory thresholds were assessed by the phenyl ethyl alcohol threshold test. They were treated with a course of high-dose steroid, and followed up for at least 3 months without any olfactory improvement. Magnetic resonance imaging was subsequently used to measure patients' OB volumes. Subjects w

Detecting activity in olfactory bulb glomeruli with astrocyte recording.

In the olfactory bulb, axons of olfactory sensory neurons (OSNs) expressing the same olfactory receptor converge on specific glomeruli. These afferents form axodendritic synapses with mitral/tufted and periglomerular cell dendrites, whereas the dendrites of mitral/tufted cells and periglomerular interneurons form dendrodendritic synapses. The two types of intraglomerular synapses appear to be spatially isolated in subcompartments delineated by astrocyte processes. Because each astrocyte sends processes into a single glomerulus, we used astrocyte recording as an intraglomerular detector of neur

Sublaminar organization of the mouse olfactory bulb nerve layer.

Olfactory sensory neuron (OSN) axons coalesce to form the olfactory nerve (ON) and then grow from the olfactory epithelium to the olfactory bulb (OB), enter the olfactory nerve layer (ONL), reorganize extensively, and innervate specific glomeruli. Within the ON and ONL a population of glial cells, the olfactory ensheathing cells (OECs), surround OSN axon fascicles. To better understand the relationship between OECs and axon fascicles in the ONL of the adult mouse, we used confocal microscopy and antibodies to the low affinity nerve growth factor receptor p75 (p75), glial fibrillary acidic prot

Mapping at glomerular resolution: fMRI of rat olfactory bulb.

The rat olfactory bulb contains approximately 2000 functional units called glomeruli which are used to recognize specific characteristics of odorants. Activity localization of individual glomerulae ( approximately 0.002 microL) has important consequences for understanding mechanisms in olfactory information encoding. High-resolution functional MRI (fMRI) data from the rat olfactory bulb are presented using the blood oxygenation level dependent (BOLD) method at 7 T. Either individual or clusters of fMRI voxels suggestive of activity in the olfactory nerve and glomerular layers were reproducibly

Septo-optic dysplasia with olfactory tract and bulb hypoplasia.

Optic nerve hypoplasia can be associated with other central nervous system malformations. When associated with absence of the septum pellucidum or thinning of the corpus callosum, it is termed septo-optic dysplasia. Considered to be a first trimester event, the embryologic timing and etiology of this syndrome still is not clearly defined. This report represents the first documented case of septo-optic dysplasia with associated hypoplasia of the olfactory tract. Optic nerve hypoplasia is attributed to an abnormal developmental process during gestation. Generally accepted to be due to a first tr
