Quantitative method to assess Cryptosporidium oocyst shedding in the chicken model.

An unsophisticated and fairly sensitive quantitative method and its grounds are described, which proved useful to assess Cryptosporidium oocyst shedding in the chicken model. The method is based on the rather slow sedimentation of oocysts. A threshold of sensitivity ranging between 5,000 and 10,000 oocysts/g feces was established for this technique. There was good agreement between triplicate assays over a wide range of oocyst concentrations.

Variations in oocyst output associated with Cryptosporidium baileyi infections in chickens.

Groups of birds, aged 2, 16 and 30 days, were experimentally infected with 400, 20,000 or 1,000,000 oocysts of Cryptosporidium baileyi. The faecal oocyst output for each group was determined by collection of faeces on Days 0, 5, 7 and 13-21 post-infection (p.i.). The duration of patency of infection, as determined by oocyst excretion, declined more rapidly in the older age groups but persisted longer in birds challenged with the lower oocyst numbers.

Experimental Cryptosporidium infections in chickens: oocyst structure and tissue specificity.

Oocysts of an avian isolate of Cryptosporidium were used to inoculate 21 chicks orally and 7 chicks intratracheally to determine the tissue specificity of this organism. Oocysts were passed in the feces 4 to 5 days after inoculation. Oocysts (6.8 by 5.0 microns) were fully sporulated and they were passed for at least 17 days by infected chicks. The mode of inoculation did not influence the distribution of cryptosporidia within the digestive tract. Cryptosporidia were found in the cloaca (100%), bursa of Fabricius (95.7%), terminal portion of the colon (26.1%), and cecum (4.3%) of chicks that w

Inactivation of Cryptosporidium parvum oocysts in fresh apple cider by UV irradiation.

This study evaluated the efficacy of UV irradiation on the inactivation of Cryptosporidium parvum oocysts in fresh apple cider. Cider was inoculated with oocysts and exposed to 14.32 mJ of UV irradiation/cm(2). Oocyst viability was assessed with the gamma interferon gene knockout (GKO) mouse and infant BALB/cByJ mouse models. All GKO mice challenged with UV-treated cider demonstrated no morbidity or mortality, and infant BALB/c mice challenged with treated cider were negative for the presence of C. parvum. In contrast, the GKO mice challenged with non-UV-treated inoculated cider died and the p

Mechanical transport and transmission of Cryptosporidium parvum oocysts by wild filth flies.

Over the course of six months wild filth flies were collected from traps left for 7-10 days in a barn with or without a calf shedding Cryptosporidium parvum Genotype 2 oocysts in diarrheic feces. The oocysts of C. parvum transported on the flies' exoskeletons and eluted from their droplets left on visited surfaces were infectious for mice. The mean number of oocysts carried by a fly varied from 4 to 131, and the total oocyst number per collection varied from 56 to approximately 4.56 x 10(3). Fly abundance and intensity of mechanical transmission of infectious C. parvum oocysts were positively

A most-probable-number assay for enumeration of infectious Cryptosporidium parvum oocysts.

Cryptosporidium is globally established as a contaminant of drinking and recreational waters. A previously described cell culture infectivity assay capable of detecting infectious oocysts was adapted to quantify viable oocysts through sporozoite invasion and clustering of foci. Eight experiments were performed by using oocysts less than 4 months of age to inoculate host HCT-8 cell monolayers. Oocysts were diluted in a standard 5- or 10-fold multiple dilution format, levels of infection and clustering were determined, and the most probable number (MPN) of infectious oocysts in the stock suspens

Improved stool concentration procedure for detection of Cryptosporidium oocysts in fecal specimens.

Epidemiologic and laboratory data suggest that coprodiagnostic methods may fail to detect Cryptosporidium oocysts in stool specimens of infected patients. To improve the efficacy of stool concentration procedures, we modified different steps of the Formalin-ethyl acetate (FEA) stool concentration technique and evaluated these modifications by examining stool samples seeded with known numbers of Cryptosporidium oocysts. Because these modifications failed to improve oocyst detection, we developed a new stool concentration technique that includes FEA sedimentation followed by layering and flotati

Occurrence of Cryptosporidium oocysts in fecal samples submitted for routine microbiological examination.

During a 7-month period, 2,252 fecal samples submitted for routine microbiological examination from 1,621 patients were screened for Cryptosporidium oocysts by the auramine staining method with Kinyoun acid-fast stain as the confirmatory stain. Cryptosporidium oocysts were detected in fecal samples from 19 (1.2%) patients, 18 of whom had gastroenteritis. Diarrheic stools from 14 of these 18 patients were negative for the usual enteropathogens but contained the oocysts in moderate to large numbers. Although Cryptosporidium oocysts were found in patients of all ages, they occurred slightly more

Morphological and autofluorescence assessment of oocysts differentiate live from dead coccidian parasites.

To assess and mitigate foodborne risk, regulatory agencies and produce growers require the means not merely to detect but moreover determine the viability of foodborne eukaryotic pathogens such as Cyclospora cayetanensis. Viability assessment would also aid those employing live attenuated vaccines against coccidiosis, a major problem in poultry production. Therefore, we sought to identify morphological changes differentiating viable from non-viable coccidian oocysts, employing Eimeria acervulina as a tractable model, enabling empirical validation by means of in vivo challenge infections in the

Anticoccidial activities of Piper betle L essential oil on Eimeria tenella oocysts.

Coccidiosis poses a significant threat to the poultry industry, with synthetic antibiotics and disinfectants being the primary tools for control. This study investigated the potential of Piper betle L essential oil (PBEO) as a natural alternative against Eimeria tenella, one of the most pathogenic Eimeria species affecting poultry. Our findings revealed that PBEO exhibits significant anticoccidial effects through two primary mechanisms: (i) oocysticidal activity by disintegrating oocyst walls and (ii) inhibition of the sporulation process. PBEO demonstrated oocysticidal activities ranging from

Detection of Infectious Cryptosporidium parvum Oocysts from Lamb's Lettuce: CC-qPCR's Intake.

Cryptosporidium spp. is responsible for several food and waterborne disease outbreaks worldwide. Healthier lifestyles attract consumers to eat, notably, fresh food like fruits and vegetables. The consumption of raw or under-cooked food increases the risk of foodborne transmission of Cryptosporidiosis. The assessment of the consumer's exposure to Cryptosporidium danger is crucial for public health. Still, the standardized method to detect this parasite in fresh leafy greens and berry fruits has only been available since 2016 and suffers from weaknesses. Consequently, in this study, we propose a

A protocol to count Cryptosporidium oocysts by flow cytometry without antibody staining.

Cryptosporidiosis caused by the protozoan parasites Cryptosporidium hominis and C. parvum, threatens the lives of young children in developing countries. In veterinary medicine, C. parvum causes life-threatening diarrhea and dehydration in newborn dairy calves. Protocols to detect Cryptosporidium spp. oocysts using flow cytometry have been reported; however, these protocols use antibodies against the parasite and typically focus on detection of oocysts, not quantification. These techniques are not well-suited for studies that generate large variations in oocyst burdens because the amount of an

Surface binding properties of aged and fresh (recently excreted) Toxoplasma gondii oocysts.

The surfaces of aged (10 years) and fresh (recently excreted) oocysts of Toxoplasma gondii were investigated using monoclonal antibody (mAb) and lectin-binding assays. Fresh oocysts bound a wall-specific mAb labelled with fluorescein isothiocyanate while aged oocysts did not. In contrast, the walls of aged oocysts bound a lectin (wheat germ agglutinin, WGA), but not the walls of fresh oocysts. Exposure of oocysts to detergent solutions or trypsin did not affect the binding properties of the walls of the oocysts. However, exposure of fresh oocysts to acidified pepsin enabled labelling of the wa

Land use and soil contamination with Toxoplasma gondii oocysts in urban areas.

Because soil contaminated with Toxoplasma gondii oocysts is increasingly recognized as a major source of infection for humans, in this study, we investigated the spatial pattern of soil contamination with T. gondii oocysts in urban area of northeastern Mainland China. From April 2014 to May 2015, more than 9000 soil samples were collected. Detection of T. gondii oocysts was performed applying real-time quantitative PCR. Sensitivity was improved by analyzing four replicates for each sampling point. T. gondii was detected in 30.3% of all samples. Subsequently, a maximum entropy model was used to

Experimental infection of dogs (Canis familiaris) with sporulated oocysts of Neospora caninum.

Neospora caninum is widely distributed in the world and this parasite is one of the major causes of abortion in cattle. Dogs and coyotes are definitive hosts of N. caninum and several species of domestic and wild animals are intermediate hosts. Dogs can become infected by the ingestion of tissues containing cysts and then excrete oocysts. It is not yet known whether sporulated oocysts are able to induce a patent infection in dogs, i.e. a shedding of N. caninum oocysts in feces. The objective of this study was to experimentally examine the infection of dogs by sporulated oocysts. The oocysts us

Quantitative comparison of different purification and detection methods for Cryptosporidium parvum oocysts.

UNLABELLED: Cryptosporidium parvum (C. parvum) is the causal agent of cryptosporidiosis in many animals, mainly cattle, and possesses a high zoonotic potential. It occurs worldwide and ubiquitously. Detection of C. parvum is mainly performed directly but purification of the oocysts is useful to increase sensitivity and to obtain oocyst material for further use. The study was designed to compare (a) three different direct diagnostic methods, namely modified Ziehl-Neelsen staining, carbol fuchsin staining and conventional PCR, and (b) three routine oocyst purification methods, in particular flot

Lack of detectable shedding of Cryptosporidium parvum oocysts by periparturient dairy cattle.

We examined whether periparturient dairy cattle shed Cryptosporidium parvum oocysts within 12 hr of calving on 3 commercial dairy farms endemic for calfhood cryptosporidiosis. Using a diagnostic method that can detect as few as 1 oocyst per gram of feces, we found no evidence of C. parvum oocysts in 86 fecal samples collected within 12 hr of calving from 43 dairy cows.
