Optical characterization of a laser speckle contrast imaging system for intraoperative blood flow monitoring.

We present the experimental characterization of a laser speckle contrast imaging system for blood flow monitoring under variable imaging conditions. A tissue phantom setup was used to evaluate system performance across multiple parameters, including laser power, exposure time, vessel depth, and flow velocity. Additionally, two optical configurations were tested to assess their robustness to the variation of these parameters: the conventional arrangement, where the camera is positioned perpendicular to the sample, and an alternative geometry, where the laser is perpendicular to the sample and t

An AR 3D Tracking and Registration Method That Integrates Optical Flow Tracking and Mean Shift.

Augmented reality (AR) enhances the real world scene by overlaying virtual information onto it. Vision-based 3D tracking and registration is the key technology for ensuring the fusion of virtual and real content in monocular AR systems. Existing mainstream visual tracking and registration methods are susceptible to illumination variations, motion blur, target occlusion, and dynamic background interference in complex scenarios. They also suffer from low computational efficiency, cumulative pose errors, and insufficient stability, making them difficult to deploy on low power edge devices such as

The effect of optical flow perturbations on walking foot placement control in people with multiple sclerosis.

People with multiple sclerosis (PwMS) experience elevated fall risk, yet traditional gait measures such as step variability provide limited insight into underlying control mechanisms. We examined step-to-step regulation of foot placement in PwMS versus age-matched controls during treadmill walking with and without anteroposterior (AP) and mediolateral (ML) optical flow perturbations. Fifteen PwMS and fifteen matched controls completed three-minute walking trials while kinematics were recorded. Detrended fluctuation analysis (DFA) assessed the temporal organization of step width (SW) and step l

Just Go with the Optical Flow: Enhanced Axon Tracing by Modeling Fiber Connections as Motion.

In this work, we explore the extension of motionbased techniques from computer vision into the realm of biological structure detection. Specifically, we propose using optical flow as a feature for guiding dense axon centerline detection and tracing. Optical flow is a technique that describes the apparent movement of objects over time, as in a video. Our experiments show that interpreting 3D volumes as videos, by interchanging the time axis of optical flow with the x, y, and z axes of the volumes, extracts useful spatial relationships in the context of axon centerline detection. This approach o

Combined Dielectric-Optical Characterization of Single Cells Using Dielectrophoresis-Imaging Flow Cytometry.

In this paper, we present a microfluidic flow cytometer for simultaneous imaging and dielectric characterization of individual biological cells within a flow. Utilizing a combination of dielectrophoresis (DEP) and high-speed imaging, this system offers a dual-modality approach to analyze both cell morphology and dielectric properties, enhancing the ability to analyze, characterize, and discriminate cells in a heterogeneous population. A high-speed camera is used to capture images of and track multiple cells in real-time as they flow through a microfluidic channel. A wide channel is used, enabl

Measurement of retinal blood flow precision in the human eye with multimodal adaptive optics imaging.

Impaired retinal blood flow (RBF) autoregulation plays a key role in the development and progression of several ocular diseases, including glaucoma and diabetic retinopathy. Clinically, reproducible RBF quantitation could significantly improve early diagnosis and disease management. Several non-invasive techniques have been developed but are limited for retinal microvasculature flow measurements due to their low signal-to-noise ratio and poor lateral resolution. In this study, we demonstrate reproducible vessel caliber and retinal blood flow velocity measurements in healthy human volunteers us

Overcoming fixation and permeabilization challenges in flow cytometry by optical barcoding and multi-pass acquisition.

The fixation and permeabilization of cells are essential for labeling intracellular biomarkers in flow cytometry. However, these chemical treatments often alter fragile targets, such as cell surface and fluorescent proteins, and can destroy chemically-sensitive fluorescent labels. This reduces measurement accuracy and introduces compromises into sample workflows, leading to losses in data quality. Here, we demonstrate a novel multi-pass flow cytometry approach to address this long-standing problem. Our technique utilizes individual cell barcoding with laser particles, enabling sequential analy

Blood flow rate estimation in optic disc capillaries and vessels using Doppler optical coherence tomography with 3D fast phase unwrapping.

The retinal volumetric flow rate contains useful information not only for ophthalmology but also for the diagnosis of common civilization diseases such as diabetes, Alzheimer's disease, or cerebrovascular diseases. Non-invasive optical methods for quantitative flow assessment, such as Doppler optical coherence tomography (OCT), have certain limitations. One is the phase wrapping that makes simultaneous calculations of the flow in all human retinal vessels impossible due to a very large span of flow velocities. We demonstrate that three-dimensional Doppler OCT combined with three-dimensional fo

Color Doppler imaging evaluation of blood flow parameters in ophthalmic and posterior ciliary arteries and optic nerve diameter in chronic optic neuritis in multiple sclerosis patients.

BACKGROUND AND PURPOSE: Optic neuritis (ON) is a common clinical manifestation of Multiple Sclerosis (MS), which is associated with reduced visual acuity, blurred vision, blindness and retro-bulbar pain. In several studies, specific relations between optic nerve diameter and hemodynamics of the eye bulb arteries with ON have been reported. However, no consensus has been reached in this regard. We aim at determining the alterations in optic nerve diameter and eye bulb arteries hemodynamics in ON in MS patients. METHODS: This case-control study includes 40 patients, who at least had experienced

Effects of visual cues on scene-relative object motion judgments and concurrent heading estimation from optic flow.

During self-movement, the visual system uses optic flow to identify scene-relative object motion and estimate the direction of self-movement (heading). Although both processes rely on optic flow, their relationship and the conditions under which independent object motion biases heading estimation remain unclear. The causal inference model predicts that misjudging object motion leads to its integration into heading estimation, causing errors in heading estimation, whereas correct judgments reduce these errors. However, most studies have examined these processes independently. Here we used a dua

Area PEc Neurons Use a Multiphasic Pattern of Activity to Signal the Spatial Properties of Optic Flow.

The cortical representation of visual perception requires the integration of several-signal processing distributed across many cortical areas, but the neural substrates of such perception are largely unknown. The type of firing pattern exhibited by single neurons is an important indicator of dynamic circuitry within or across cortical areas. Neurons in area PEc are involved in the spatial mapping of the visual field; thus, we sought to analyze the firing pattern of activity of PEc optic flow neurons to shed some light on the cortical processing of visual signals. We quantified the firing activ

Multichannel Integrated Fiber-Optic End-Face Sensor for Deuterium Detection from Hydrogen Via Flow-Temperature Synergistic Regulation.

Inspired by the selective adsorption and separation technology of isotope gases using porous materials, this work reports the first successful realization of optical fiber-based deuterium (D2) sensing in the presence of hydrogen (H2). Initially, we propose a novel three-core fiber (TCF) end-face sensor integrating two microtips and a plasmonic metasurface on its end-face, enabling simultaneous sensing of three parameters: temperature, flow velocity, and gas concentration. Specifically, the two microprobes form two Fabry-Pérot (FP) cavities; the glass microprobe is used for flow velocity sensin

Evaluation of blood flow in arteritic anterior ischemic optic neuropathy using laser speckle flowgraphy: A case series.

BACKGROUND: Arteritic anterior ischemic optic neuropathy (AAION), primarily caused by giant cell arteritis, is a leading cause of blindness. This disease results in significant ocular blood flow (BF) impairment, though data on ocular hemodynamics are limited. METHODS: This observational case series enrolled four patients treated for AAION (age: 81.8 ± 7.8 years; male to female ratio: 3:1) who underwent laser speckle flowgraphy (LSFG) scanning at the initial visit and after steroid treatment in both eyes. Mean blur rate (MBR), an LSFG parameter that represents BF velocity, was obtained in the

Visual pursuit behavior in mice maintains the pursued prey on the retinal region with least optic flow.

Mice have a large visual field that is constantly stabilized by vestibular ocular reflex (VOR) driven eye rotations that counter head-rotations. While maintaining their extensive visual coverage is advantageous for predator detection, mice also track and capture prey using vision. However, in the freely moving animal quantifying object location in the field of view is challenging. Here, we developed a method to digitally reconstruct and quantify the visual scene of freely moving mice performing a visually based prey capture task. By isolating the visual sense and combining a mouse eye optic mo
