Time gate, optical layout, and wavelength effects on ballistic imaging.

A method to distinguish a hidden object from a perturbing environment is to use an ultrashort femtosecond pulse of light and a time-resolved detection. To separate ballistic light containing information on a hidden object from multiscattered light coming from the surrounding environment that scrambles the signal, an optical Kerr gate can be used. It consists of a carbon disulfide (CS(2)) cell in which birefringence is optically induced. An imaging beam passes through the studied medium while a pump pulse is used to open the gate. The time-delayed scattered light is excluded from measurements b

Determination of optimal rhodamine fluorophore for in vivo optical imaging.

Optical imaging has the potential to improve the efficacy of surgical and endoscopic approaches to cancer treatment; however, the optimal type of fluorescent probe has not yet been established. It is well-known that rhodamine-core-derived fluorophores offer a combination of desirable properties such as good photostability, high extinction coefficient, and high fluorescence quantum yield. However, despite the ubiquitous use of rhodamine fluorophores for in vivo optical imaging, it remains to be determined if unique chemical properties among individual rhodamine core family members affect fluoro

[Imaging of the anterior eye chamber with optical coherence tomography].

BACKGROUND: Optical coherence tomography (OCT) represents a high-resolution diagnostic method which can be used to precisely image the anterior eye segment. Further developments in scanning technology and data evaluation allowed to visualize the entire anterior eye segment in one image, and the purpose of this study was to clinically evaluate this new system. PATIENTS AND METHODS: The studied OCT system was adapted to a slit-lamp and allowed us to image the anterior segment with an axial resolution of 10 microm at a scanning rate of 200 Hz with a depth of 7 mm and a maximal width of 15 mm. In

Demystifying Experimental Parameters in Optical Photothermal Imaging in Aqueous Media.

Optical photothermal infrared (O-PTIR) spectroscopy is an indirect mid-infrared spectral imaging technique that enables label-free chemical analysis with submicrometer resolution. The technique is often used for analyzing cells and tissues and is discussed as capable of measuring in aqueous media, yet a quantitative understanding of its signal formation in liquids remains incomplete. Here, we present a systematic experimental study of transmission-mode O-PTIR supported by detailed simulations on the underlying signal generating process, focusing on O-PTIR applied to a water filled cuvette comp

Optical Coherence Tomography for Quantitative Point-of-Care Ear Imaging.

OBJECTIVE: To determine whether an optical coherence tomography (OCT) otoscope can provide quantitative morphological measurements of the tympanic membrane (TM) that distinguish abnormal ears from normal controls. METHODS: Patients with ear disease and normal volunteers were imaged using an OCT otoscope. Integrated video otoscopy with simultaneous OCT imaging guided probe positioning. OCT volumes (< 5 s each) were acquired and fused to generate wide-field volumetric images of the TM and middle ear (ME). Datasets were analyzed using semi-automated segmentation to quantify TM thickness and surf

Cardiac Microanatomy Imaging Using Forward-Viewing Optical Coherence Tomography Endoscope.

OBJECTIVE: Due to limitations in current imaging technologies detecting subtle cardiac microstructural changes that can lead to sudden cardiac death is a significant clinical challenge. To address this problem, we developed a forward-viewing optical coherence tomography (OCT) endoscope for the detection of relevant cardiac microstructures in the subendocardium, including Purkinje fibers, scar tissue, surviving myocytes, and adipose tissue. METHODS: An endoscopic probe based on the scanning fiber principle was developed for OCT measurements in contact. The probe was evaluated in freshly excise

Automated dual-camera DMD system for stereo optical sectioning imaging.

We present a compact, automated, stereo-optical-sectioning imaging system for clinical assessment of the anterior segment, using a digital micromirror device (DMD) as a spatial light modulator. The platform synchronizes two CMOS cameras with the DMD, which projects programmable slit beams to produce depth-resolved cross-sectional views of the anterior eye. By adjusting the micromirror activation time, the system can also extend the dynamic range of the acquired images, accommodating both dim (e.g., the cornea) and highly reflective regions (i.e., the iris). In a pilot study, the prototype inst

Optical Redox Imaging Predicts Post-Loading Cartilage Mitochondrial Membrane Potential.

PURPOSE: Disrupted cellular redox balance is associated with various diseases, including osteoarthritis. Although mitochondrial (MT) membrane potential is a proxy for redox balance, the translational potential of this method is limited by exogenous dye. Therefore, the objective of this study was to predict changes in MT membrane potential in response to mechanical loading using dye-free optical redox imaging (ORI). A secondary objective was to determine the effect of loading on ORI metrics. METHODS: Full-thickness porcine cartilage strips were subjected to tensile loading at one of two strain

Optical imaging of the small intestine immune compartment across scales.

The limitations of 2D microscopy constrain our ability to observe and understand tissue-wide networks that are, by nature, 3-dimensional. Optical projection tomography (OPT) enables the acquisition of large volumes (ranging from micrometres to centimetres) in various tissues. We present a multi-modal workflow for the characterization of both structural and quantitative parameters of the mouse small intestine. As proof of principle, we evidence its applicability for imaging the mouse intestinal immune compartment and surrounding mucosal structures. We quantify the volumetric size and spatial di

All-optical ultrasound catheter for rapid B-mode oesophageal imaging.

All-optical ultrasound (OpUS) is an imaging paradigm that uses light to both generate and receive ultrasound, and has progressed from benchtop to in vivo studies in recent years, demonstrating promise for minimally invasive surgical applications. In this work, we present a rapid pullback imaging catheter for side-viewing B-mode ultrasound imaging within the upper gastrointestinal tract. The device comprised an ultrasound transmitter configured to generate ultrasound laterally from the catheter and a plano-concave microresonator for ultrasound reception. This imaging probe was capable of genera

Optical recognition of constructs using hyperspectral imaging and detection (ORCHID).

Challenges to deep sample imaging have necessitated the development of special techniques such as spatially offset optical spectroscopy to collect signals that have travelled through several layers of tissue. However, these techniques provide only spectral information in one dimension (i.e., depth). Here, we describe a general and practical method, referred to as Optical Recognition of Constructs Using Hyperspectral Imaging and Detection (ORCHID). The sensing strategy integrates (1) the spatial offset detection concept by computationally binning 2D optical data associated with digital offsets

Guidelines on Optical Coherence Tomography Angiography Imaging: 2020 Focused Update.

Optical coherence tomography angiography (OCTA) has significantly expanded our knowledge of the ocular vasculature. In this review, we provide a discussion of the fundamental principles of OCTA and the application of this imaging modality to study the retinal and choroidal vessels. These guidelines are focused on 2020, and include updates since the 2019 publication. Importantly, we will comment on recent findings on OCTA technology with a special focus on the three-dimensional (3D) OCTA visualization.

Plasmonic Optical Imaging of Gold Nanorods Localization in Small Animals.

Gold nanoparticles (GNP) have been intensively investigated for applications in cancer imaging and therapy. Most imaging studies focused on microscopic imaging. Their potential as optical imaging probes for whole body small animal imaging has rarely been explored. Taking advantage of their surface plasmon resonance (SPR) properties, we aim to develop a noninvasive diffuse optical imaging method to map the distribution of a special type of GNP, gold nanorods (GNR), in small animals. We developed an integrated dual-modality imaging system capable of both x-ray computed tomography (XCT) and diffu

Enhanced Depth Imaging Optical Coherence Tomography of Posterior Globe Rupture.

Occult globe rupture is a rare but potentially devastating result of eye trauma. The authors present a case of a woman who sustained blunt trauma to the right eye resulting in a peripapillary circumferential posterior globe rupture. Clinically, the eye maintained good vision and normal intraocular pressure with a clear media, though subretinal fluid and retinal hemorrhage were present overlying the scleral defect. Enhanced depth imaging spectral-domain optical coherence tomography (EDI SD-OCT) revealed a sharp step-off of the sclera, choroid, and Bruch's membrane/retinal pigment epithelium, wi

Optical and nuclear imaging of glioblastoma with phosphatidylserine-targeted nanovesicles.

Multimodal tumor imaging with targeted nanoparticles potentially offers both enhanced specificity and sensitivity, leading to more precise cancer diagnosis and monitoring. We describe the synthesis and characterization of phenol-substituted, lipophilic orange and far-red fluorescent dyes and a simple radioiodination procedure to generate a dual (optical and nuclear) imaging probe. MALDI-ToF analyses revealed high iodination efficiency of the lipophilic reporters, achieved by electrophilic aromatic substitution using the chloramide 1,3,4,6-tetrachloro-3α,6α-diphenyl glycoluril (Iodogen) as the

Retinal Imaging of Infants on Spectral Domain Optical Coherence Tomography.

Spectral domain coherence tomography (SD OCT) has become an important tool in the management of pediatric retinal diseases. It is a noncontact imaging device that provides detailed assessment of the microanatomy and pathology of the infant retina with a short acquisition time allowing office examination without the requirement of anesthesia. Our understanding of the development and maturation of the infant fovea has been enhanced by SD OCT allowing an in vivo assessment that correlates with histopathology. This has helped us understand the critical correlation of foveal development with visual

In vivo imaging of mouse cochlea by optical coherence tomography.

HYPOTHESIS: Cochlear pathology can be evaluated in living animals using optical coherence tomography (OCT). BACKGROUND: The current imaging methods available for the detailed analysis of cochlear pathology in a clinical setting provide only limited information. Thus, a cochlear imaging modality with high definition is needed for improving the diagnosis of cochlear pathology. OCT has been used in other fields for obtaining high-resolution subsurface images, and its use could potentially be extended to the analysis of cochlear pathogenesis. METHODS: Slc26a4(-/-) mice, which generate endolympha
