Optogenetic restoration of retinal ganglion cell activity in the living primate.

Optogenetic therapies for vision restoration aim to confer intrinsic light sensitivity to retinal ganglion cells when photoreceptors have degenerated and light sensitivity has been irreversibly lost. We combine adaptive optics ophthalmoscopy with calcium imaging to optically record optogenetically restored retinal ganglion cell activity in the fovea of the living primate. Recording from the intact eye of a living animal, we compare the patterns of activity evoked by the optogenetic actuator ChrimsonR with natural photoreceptor mediated stimulation in the same retinal ganglion cells. Optogeneti

Towards the optical cochlear implant: optogenetic approaches for hearing restoration.

Cochlear implants (CIs) are considered the most successful neuroprosthesis as they enable speech comprehension in the majority of half a million CI users suffering from sensorineural hearing loss. By electrically stimulating the auditory nerve, CIs constitute an interface re-connecting the brain and the auditory scene, providing the patient with information regarding the latter. However, since electric current is hard to focus in conductive environments such as the cochlea, the precision of electrical sound encoding-and thus quality of artificial hearing-is limited. Recently, optogenetic stimu

Optogenetic-induced sympathetic neuromodulation of brown adipose tissue thermogenesis.

The brown adipose tissue (BAT) is a thermogenic organ that plays a major role in energy balance, obesity, and diabetes due to the potent glucose and lipid clearance that fuels its thermogenesis, which is largely mediated via sympathetic nervous system activation. However, thus far there has been little experimental validation of the hypothesis that selective neuromodulation of the sympathetic nerves innervating the BAT is sufficient to elicit thermogenesis in mice. We generated mice expressing blue light-activated channelrhodopsin-2 (ChR2) in the sympathetic nerves innervating the BAT using tw

Optogenetic Termination of Cardiac Arrhythmia: Mechanistic Enlightenment and Therapeutic Application?

Optogenetic methods enable selective de- and hyperpolarization of cardiomyocytes expressing light-sensitive proteins within the myocardium. By using light, this technology provides very high spatial and temporal precision, which is in clear contrast to electrical stimulation. In addition, cardiomyocyte-specific expression would allow pain-free stimulation. In light of these intrinsic technical advantages, optogenetic methods provide an intriguing opportunity to understand and improve current strategies to terminate cardiac arrhythmia as well as for possible pain-free arrhythmia termination in

Optogenetic Analysis of Depolarization-Dependent Glucagonlike Peptide-1 Release.

Incretin hormones play an important role in the regulation of food intake and glucose homeostasis. Glucagonlike peptide-1 (GLP-1)-secreting cells have been demonstrated to be electrically excitable and to fire action potentials (APs) with increased frequency in response to nutrient exposure. However, nutrients can also be metabolized or activate G-protein-coupled receptors, thus potentially stimulating GLP-1 secretion independent of their effects on the plasma membrane potential. Here we used channelrhodopsins to manipulate the membrane potential of GLUTag cells, a well-established model of GL

Visceral hypersensitivity induced by optogenetic activation of the amygdala in conscious rats.

In vivo optogenetics identifies brain circuits controlling behaviors in conscious animals by using light to alter neuronal function and offers a novel tool to study the brain-gut axis. Using adenoviral-mediated expression, we aimed to investigate whether photoactivation with channelrhodopsin (ChR2) or photoinhibition with halorhodopsin (HR3.0) of fibers originating from the central nucleus of the amygdala (CeA) at the bed nucleus of the stria terminalis (BNST) had any effect on colonic sensitivity. We also investigated whether there was any deleterious effect of the adenovirus on the neuronal

Optogenetic control of thalamus as a tool for interrupting penicillin induced seizures.

Penicillin epilepsy model, whose discharge resembles that of human absence epilepsy, is one of the most useful acute experimental epilepsy models. Though closed-loop optogenetic strategy of interrupting seizures was proved sufficient to switch off epilepsy by controlling thalamus in the post-lesion partial chronic epilepsy model, doubts still exist in absence epilepsy attenuation through silencing thalamus. Here we directly arrested the thalamus to modulate penicillin-induced absence seizures through pseudorandom responsive stimulation on eNpHR-transfected rats. Our data suggested that the dur

Optogenetic control of targeted peripheral axons in freely moving animals.

Optogenetic control of the peripheral nervous system (PNS) would enable novel studies of motor control, somatosensory transduction, and pain processing. Such control requires the development of methods to deliver opsins and light to targeted sub-populations of neurons within peripheral nerves. We report here methods to deliver opsins and light to targeted peripheral neurons and robust optogenetic modulation of motor neuron activity in freely moving, non-transgenic mammals. We show that intramuscular injection of adeno-associated virus serotype 6 enables expression of channelrhodopsin (ChR2) in

Multi-Frequency Interpolation X-talk Removal Algorithm: Enabling Combinations of Concurrent Optogenetics and Lock-in Amplification Fiber Photometry via Removal of Optogenetic Stimulation Crosstalk.

Simultaneous fiber photometry and optogenetics is a powerful emerging technique for precisely studying the interactions of neuronal brain networks. However, spectral overlap between photometry and optogenetic components has severely limited the application of an all-optical approach. Due to spectral overlap, light from optogenetic stimulation saturates the photosensor and occludes photometry fluorescence, which is especially problematic in physically smaller model organism brains like mice. Here, we demonstrate the multi-frequency interpolation X-talk removal algorithm (MuFIX or μFIX) for reco

Strength of activation and temporal dynamics of bioluminescent-optogenetics in response to systemic injections of the luciferin.

BioLuminescent OptoGenetics ("BL-OG") is a chemogenetic method that can evoke optogenetic reactions in the brain non-invasively. In BL-OG, an enzyme that catalyzes a light producing reaction (i.e., a luciferase) is tethered to an optogenetic element that is activated in response to bioluminescent light. Bioluminescence is generated by injecting a chemical substrate (luciferin, e.g., h-Coelenterazine; h-CTZ) that is catalyzed by the luciferase. By directly injecting the luciferin into the brain, we show that bioluminescent light is proportional to spiking activity, and this relationship scales

Non-invasive phenotyping and drug testing in single cardiomyocytes or beta-cells by calcium imaging and optogenetics.

Identification of drug induced electrical instability of the heart curtails development, and introduction, of potentially proarrhythmic drugs. This problem usually requires complimentary contact based approaches such as patch-clamp electrophysiology combined with field stimulation electrodes to observe and control the cell. This produces data with high signal to noise but requires direct physical contact generally preventing high-throughput, or prolonged, phenotyping of single cells or tissues. Combining genetically encoded optogenetic control and spectrally compatible calcium indicator tools

Opto- μECoG array: a hybrid neural interface with transparent μECoG electrode array and integrated LEDs for optogenetics.

Electrocorticogram (ECoG) recordings, taken from electrodes placed on the surface of the cortex, have been successfully implemented for control of brain machine interfaces (BMIs). Optogenetics, direct optical stimulation of neurons in brain tissue genetically modified to express channelrhodopsin-2 (ChR2), enables targeting of specific types of neurons with sub-millisecond temporal precision. In this work, we developed a BMI device, called an Opto- μECoG array, which combines ECoG recording and optogenetics-based stimulation to enable multichannel, bi-directional interactions with neurons. Temporal and spatial control of phosphatidylinositides using optogenetics ameliorates behavioral deficits in an Alzheimer's disease mouse model.

Deficits in levels of phosphatidylinositol-4,5-bisphosphate [PI(4,5)P2] mediate Alzheimer's disease (AD) pathogenesis and etiology. The depletion of PI(4,5)P2 has been reported in AD in both human brain and animal models. Repletion of the synaptic pool of PI(4,5)P2, through haploinsufficiency of the degrading enzyme, Synaptojanin 1, ameliorated behavioral deficits in a mouse model of AD, in spite of accumulating amyloid. In order to refine the contribution of PI(4,5)P2 to AD, we used optogenetic translocation of the PI(4,5)P2-synthesizing enzyme, phosphoinositide phosphate 4 kinase2A (PIP4K2A)

From optical control to translational readiness: an evidence-tiered framework for optogenetics in neuromuscular and neurological disorders.

Optogenetics provides exceptional spatial, temporal, and cell-type specificity for manipulating biological function, but the ability to produce a light-evoked response does not by itself establish therapeutic relevance. Disease-oriented studies now span molecular assembly, neuronal excitability, neuromuscular transmission, circuit modulation, engineered human tissues, closed-loop control, and early human intervention, yet these outcomes support fundamentally different levels of inference. Here, we critically examine optogenetic applications across neuromuscular and neurological disorders and p

Precise rewiring of corticospinal axons and spinal interneurons via near-infrared optogenetics for spinal cord injury treatment.

To date, precise restoration of proper connections between posttrauma axons and neurons following spinal cord injury (SCI) remains a substantial challenge. Here, we developed glutamate-linked upconversion nanoparticles (Glu-UCNP) to facilitate optogenetic control of axonal sprouting in SCI mice. After being specifically uptaken by the postsynaptic interneurons innervated by corticospinal tract (CST) axons, Glu-UCNP not only serves as internal light transducers that convert near-infrared light to visible light but also acts as nanobeacons that guide axonal sprouting toward postsynaptic neurons

Fabrication and utility of a transparent graphene neural electrode array for electrophysiology, in vivo imaging, and optogenetics.

Transparent graphene-based neural electrode arrays provide unique opportunities for simultaneous investigation of electrophysiology, various neural imaging modalities, and optogenetics. Graphene electrodes have previously demonstrated greater broad-wavelength transmittance (∼90%) than other transparent materials such as indium tin oxide (∼80%) and ultrathin metals (∼60%). This protocol describes how to fabricate and implant a graphene-based microelectrocorticography (μECoG) electrode array and subsequently use this alongside electrophysiology, fluorescence microscopy, optical coherence tomogra

A Monolithically Integrated Multimodal Low-Artifact Neural Probe for Optogenetic Applications.

In optogenetic neural probes, stimulus light intensity monitoring, thermal safety assessment, and stimulation artifact suppression are of critical importance for achieving cell-type-specific optical modulation and high-fidelity neural signal recording. Here, micro light-emitting diode (µLED) stimulation sources and photodetectors (PDs) were fabricated on a GaN-based blue LED epitaxial structure on a sapphire substrate, and resistance temperature detectors (RTDs) and metal recording electrodes were monolithically integrated to form a multimodal neural probe. Optogenetic insights into short- and long-duration random noise electrical stimulation.

Previous research by Paulus's group in humans showed that 10 min of transcranial random noise stimulation (tRNS) enhances motor-evoked potentials for over 1 h. In contrast, 4 min of tRNS attenuates task-evoked blood oxygen level-dependent (BOLD) responses, suggesting that shorter durations reduce task-related cortical activity. Importantly, this effect occurs during task execution and reflects attenuation of evoked activity rather than direct suppression of baseline cortical excitability. In this context, prior human neuroimaging findings highlight a conceptual bridge between facilitatory and

Unrestrained and hedonically motivated mouse psychophysics compatible with wireless optogenetic modulation.

BACKGROUND: Visual neuroscience research has adopted the mouse as one of its primary animal models. Collection of perceptually driven behavior in these mice often involves head fixation to present the visual stimulus under controlled conditions and water restriction to motivate responses. However, both head fixation and water restriction can induce physiological stress. NEW METHOD: Here, we adapted a popular open-source two-alternative-forced-choice (2AFC) visual psychophysics apparatus to allow unrestrained mice to maintain consistent stimulus viewing position while performing trials for hed

Photonic biosensing and optogenetic technologies: Emerging therapeutic strategies in neurodegenerative diseases.

The rising burden of neurodegenerative diseases has exposed critical gaps in the development of diagnostic and therapeutic tools, particularly regarding their limited sensitivity, poor spatiotemporal resolution, and lack of treatment specificity. These shortcomings highlight the need for technologies that enable early disease detection, real-time monitoring of regenerative processes, and precise therapeutic interventions. This review focuses on recent advances in photonic biosensing and optogenetics, examining how these technologies are reshaping neural regeneration research and assessing thei
