Severe obesity and diabetes insipidus in a patient with PCSK1 deficiency.

Non-synonymous mutations affecting both alleles of PCSK1 (proprotein convertase 1/3) are associated with obesity and impaired prohormone processing. We report a proband who was compound heterozygous for a maternally inherited frameshift mutation and a paternally inherited 474kb deletion that encompasses PCSK1, representing a novel genetic mechanism underlying this phenotype. Although pro-vasopressin is not a known physiological substrate of PCSK1, the development of central diabetes insipidus in this proband suggests that PCSK1 deficiency can be associated with impaired osmoregulation.

Long-term stress induced by low bicarbonate alkalinity causes intestinal oxidative stress, gut microbiota imbalance and metabolic disorder in Scylla paramamosain.

To investigate the effects of low bicarbonate alkalinity on intestinal health of Scylla paramamosain, we systematically evaluated the growth survival, intestinal antioxidant enzyme activities, and expression profiles of genes involved in immunity, inflammation, apoptosis and osmoregulation under low alkalinity conditions. Meanwhile, integrated intestinal microbiome and metabolome analyses were performed. The results showed that low bicarbonate alkalinity significantly reduced the survival and molting rates of S. paramamosain. It markedly elevated the activities of intestinal SOD, CAT and GSH-P

Suppression of AVP release by drinking despite hypertonicity during and after gestation.

Water drinking suppresses arginine vasopressin (AVP) release before changes in tonicity or volume. To further characterize this oropharyngeal neuroendocrine reflex, we took advantage of ongoing studies of osmoregulation in pregnancy as follows: five women studied serially in early (8 +/- 1 wk, mean +/- SD) and late (32 +/- 3 wk) pregnancy and 8-10 wk postpartum were slowly infused with hypertonic saline for 150 min while drinking water to satiety (drinking started at 120 min). Plasma osmolality (Posmol) at 120 min was increased (277 +/- 3 to 291 +/- 2, 280 +/- 6 to 295 +/- 5, and 287 +/- 1 to

Effect of cDMEM media containing Ectoine on human periodontal ligament mesenchymal stem cell survival and differentiation.

BACKGROUND/AIM: Ectoine is an amino acid that can preserve osmotic balance and has more preservative activity than other osmoregulators. There are no published reports on the osmoregulators' effects on viability or differentiation of dental stem cells. The aim of this study was to investigate the effect of Ectoine as a storage media on the viability and differentiation potential of human periodontal ligament mesenchymal stem cells (hPDLMSCs). MATERIALS AND METHODS: hPDLMSCs were obtained from impacted third molar teeth. The cells were isolated, submitted to trilineage differentiation, and cha

Renal aquaporins and water balance disorders.

BACKGROUND: Aquaporins (AQPs) are a family of proteins that can act as water channels. Regulation of AQPs is critical to osmoregulation and the maintenance of body water homeostasis. Eight AQPs are expressed in the kidney of which five have been shown to play a role in body water balance; AQP1, AQP2, AQP3, AQP4 and AQP7. AQP2 in particular is regulated by vasopressin. SCOPE OF REVIEW: This review summarizes our current knowledge of the underlying mechanisms of various water balance disorders and their treatment strategies. MAJOR CONCLUSIONS: Dysfunctions of AQPs are involved in disorders ass

Diabetes insipidus.

Water balance is precisely regulated by vasopressin, thirst and the kidneys; plasma osmolality is maintained within a narrow range, despite large variations in normal water intake and loss. Disruption of these finely balanced mechanisms is common, however, and can be precipitated by various disease states. Diabetes insipidus is a disease process with several underlying pathogenic mechanisms--all ultimately resulting in excretion of large volumes of dilute urine. This review will discuss the pathophysiology, diagnosis and treatment of diabetes insipidus, with emphasis on disorders of osmoregula

Patients with disordered water balance. Innovative psychiatric nursing intervention strategies.

1. Patients with disordered water balance (DWB) have difficulty with fluid intake/output and osmoregulation. These difficulties are characterized by polydipsia, polyuria, significant variation in pattern of excretion, abnormal diurnal weight gain, and behavioral changes. 2. Therapeutic milieu-management strategies for patients experiencing acute and chronic phases of DWB are the cornerstone of long-term management of these patients. 3. Implementation of the intervention and milieu-management strategies described in this article resulted in a reduction of patients requiring acute medical treatm

Posterior pituitary function in health and disease.

This chapter has reviewed briefly the neuroanatomy relevant to the synthesis and release of AVP and OXT. Osmoregulation and baroregulation of AVP secretion has been discussed in detail, emphasizing the importance of osmotic control under normal physiological conditions. The remainder of the text has covered the two major pathophysiological disturbances of AVP secretion. In considering diabetes insipidus a pragmatic approach has been taken in the differentiation of the causes of polyuria and its treatment. A similar approach has been applied to the syndrome of inappropriate antidiuresis. Brief

Annexin in Taenia crassiceps ORF Strain is Localized in the Osmoregulatory System.

PURPOSE: Annexins are proteins with important roles in parasites, some of which are related to excretion-secretion processes, protein traffic, and microvesicle functionality. The participation of annexins in osmoregulation has been reported in tapeworms, including Taenia solium. This study aimed to investigate the localization and expression of annexin in cysticerci of Taenia crassiceps, used as a model of cysticercosis. METHODS: We used an antibody made with a protein, previously employed on Schistosoma bovis, to detect annexin in T. crassiceps proteins extracts used Western blot assay. The

Osmoregulatory thirst in mice lacking the transient receptor potential vanilloid type 1 (TRPV1) and/or type 4 (TRPV4) receptor.

Recent studies suggest the ability of the central nervous system to detect changes in osmolality is mediated by products of the genes encoding the transient receptor potential vanilloid-1 (TRPV1) or vanilloid-4 (TRPV4) channel. The purpose of the present study was to determine whether deletion of TRPV1 and/or TRPV4 channels altered thirst responses to cellular dehydration in mice. Injection of 0.5 or 1.0 M NaCl produced dose-dependent increases in cumulative water intakes of wild-type (WT), TRPV1-/-, TRPV4-/-, and TRPV1-/-V4-/- mice. However, there were no differences in cumulative water intak

Dissolved oxygen thresholds drive adaptive functional trait remodeling in mesophytes via anatomical, physiological, and metabolic synergies.

Climate-exacerbated urban waterlogging imposes severe hypoxic stress on mesophytes. However, whether mesophytes possess dissolved oxygen (DO)-driven mechanisms for adaptive structural and physiological remodeling remains poorly understood. We conducted a hydroponic experiment with four representative mesophyte species (Gardenia jasminoides, Murraya paniculata, Ixora chinensis, and Heptapleurum actinophyllum 'Variegata') across four DO gradients (0-2, 2-4, 4-6, and 6-8 mg/L). Plant growth, physiology, and root anatomy were measured, and structural equation modeling (SEM) was applied to disentan

In vitro effects of 11-deoxycorticosterone on hepatocytes and gill epithelial cells of rainbow trout (Oncorhynchus mykiss).

The 11-deoxycorticosterone (DOC) corticosteroid has been recently described as having a potential role in fish, complementing cortisol action through distinct physiological effects. Although systemic effects of 11-deoxycorticosterone (DOC) have been reported, the specific mechanisms mediated through glucocorticoid (GR) and mineralocorticoid (MR) receptors remain poorly understood. Therefore, we evaluated the DOC effects through both receptors in rainbow trout hepatoma-derived (RTH-149) and gill epithelial (RTgill-W1) cell lines. Cultures were pretreated with GR (Mifepristone) or MR (Eplerenone

Functional and developmental heterogeneity of pituitary lactotropes in medaka.

In fish, prolactin-producing cells (lactotropes) are located in the anterior part of the pituitary and play an essential role in osmoregulation. However, small satellite lactotrope clusters have been described in other parts of the pituitary in several species. The functional and developmental backgrounds of these satellite clusters are not known. We recently discovered two distinct prolactin-expressing cell types in Japanese medaka (Oryzias latipes), a euryhaline species, using single cell transcriptomics. In the present study, we characterize these two transcriptomically distinct lactotrope

[Postoperative polyuria after laparoscopic adrenalectomy for pheochromocytoma].

Disturbances of osmoregulation leading to polyuria are well known complications after operations in the sella region. In contrast, increased urinary output following adrenalectomy is rare. We report a case of postoperative polyuria after laparoscopic adrenalectomy for pheochromocytoma with urine output up to 15 l per day. Treatment included careful monitoring of the patient and intravenous and oral replacement of fluids and electrolytes. In our case a normalisation of the urine output was observed after few days. A further treatment with vasopressin was not undertaken as the osmolality at all

Amino acids of Diclidophora merlangi (Monogenea).

The level of free amino acids in Diclidophora merlangi is high, comprising over 500 mu moles/g ethanol extracted dry weight. A single amino acid, proline, constitutes some 70% of the total pool. Analysis of parasite protein and host blood and mucus revealed low proline levels, suggesting that the high free pool content was not related to a requirement for protein systhesis or to its abundance in the diet of the worm. Experiments revealed that proline was not involved specifically in osmoregulation, and the reasons for the large amounts present in Diclidophora remain unknown.

Engineering citrus for psyllid control: virus-induced silencing of osmoregulatory genes reduces survival and development of Diaphorina citri.

BACKGROUND: The Asian citrus psyllid (Diaphorina citri) is the insect vector of 'Candidatus Liberibacter asiaticus', the causal agent of huanglongbing (HLB), the most destructive citrus disease worldwide. Current management depends largely on repeated insecticide applications, underscoring the need for sustainable alternatives. Osmoregulation is indispensable for D. citri because the insect feeds exclusively on sucrose-rich phloem sap and must continuously regulate carbohydrate metabolism and water balance to maintain physiological homeostasis. RNA interference (RNAi)-mediated silencing of suc

Cyprinid herpesvirus-3 (CyHV-3) disturbs osmotic balance in carp (Cyprinus carpio L.)--A potential cause of mortality.

Cyprinid herpesvirus-3 (CyHV-3) causes a fatal disease in carp (Cyprinus carpio) and its ornamental koi varieties which seriously affects production and trade of this fish species globally. Up to now, the pathophysiology of this disease remains unclear. Affected individuals develop most prominent lesions in gills, skin and kidney, in tissues which are involved in the osmotic regulation of freshwater teleosts. Therefore, here serum and urine electrolyte levels were examined during the course of an experimental infection of carp with CyHV-3. In infected carp an interstitial nephritis with a prog

Osmotic gradients and transretinal water flow-a quantitative elemental microanalytical study of frozen hydrated chick eyes.

Optical clarity and efficient phototransduction are necessary for optimal vision, however, how the associated processes of osmoregulation and continuous fluid drainage across the whole eye are achieved remains relatively unexplored. Hence, we have employed elemental microanalysis of planed surfaces of light-adapted bulk frozen-hydrated chick eyes to determine the unique intracellular elemental localization, compositions, and hydration states that contribute to maintaining osmotic gradients and water flow from the vitreous, across the retina, retinal pigment epithelium (RPE), to choroid and scl

Hepatocyte Nuclear Factor-1β Regulates Urinary Concentration and Response to Hypertonicity.

The transcription factor hepatocyte nuclear factor-1β (HNF-1β) is essential for normal kidney development and function. Inactivation of HNF-1β in mouse kidney tubules leads to early-onset cyst formation and postnatal lethality. Here, we used Pkhd1/Cre mice to delete HNF-1β specifically in renal collecting ducts (CDs). CD-specific HNF-1β mutant mice survived long term and developed slowly progressive cystic kidney disease, renal fibrosis, and hydronephrosis. Compared with wild-type littermates, HNF-1β mutant mice exhibited polyuria and polydipsia. Before the development of significant renal str

CaCP15 Gene Negatively Regulates Salt and Osmotic Stress Responses in Capsicum annuum L.

Salt and osmotic stress seriously restrict the growth, development, and productivity of horticultural crops in the greenhouse. The papain-like cysteine proteases (PLCPs) participate in multi-stress responses in plants. We previously demonstrated that salt and osmotic stress affect cysteine protease 15 of pepper (Capsicum annuum L.) (CaCP15); however, the role of CaCP15 in salt and osmotic stress responses is unknown. Here, the function of CaCP15 in regulating pepper salt and osmotic stress resistance was explored. Pepper plants were subjected to abiotic (sodium chloride, mannitol, salicylic ac
