Prolonged biocontrol priming sensitizes Aspergillus flavus to cinnamaldehyde via metabolic trade-offs.

The control of Aspergillus flavus and its highly carcinogenic secondary metabolite, aflatoxin B1 (AFB1), remains a major challenge in global agriculture and food safety. In this study, we propose a novel sequential hurdle strategy that combines priming with the biocontrol bacterium Pantoea agglomerans FL1 followed by low-dose cinnamaldehyde (CA) stress, and we further elucidate its underlying mechanisms. Phenotypic analyses revealed that prolonged FL1 priming for 7 days increased the susceptibility of A. flavus upon subsequent CA exposure. Although macroscopic radial growth remained unaffected

Introducing silica-solubilizing and plant growth-promoting bacteria from sugarcane as inducers of drought stress tolerance.

Today’s sugarcane cultivation is faced with abiotic stresses including drought, salinity, heat and water limitation leading to, its reduced growth, yield, and economic productivity. Given the significant role of silica in alleviating these negative effects as well as pathogens, silica solubilizing bacteria opens an eco-friendly and sustainable approach to overcome these challenges. The aim was to find native silicate solubilizing bacteria from sugarcane and assessment their plant growth-promoting potential. Extensive screening was done from five sugarcane fields and symbiont bacterial species

Metabolic syndrome-associated gut microbiota and plasma metabolite profiles in schizophrenia.

Metabolic syndrome (MetS) is a common and clinically important comorbidity in schizophrenia (SCZ), but the biological basis remains unclear. We aimed to characterize the gut microbial and plasma metabolites associated with comorbid MetS in SCZ and to evaluate their potential as integrated biomarkers. A total of 210 participants were enrolled, including 79 patients with SCZ+MetS, 71 patients with SCZ alone, and 60 matched controls. Gut microbiota was analyzed by 16S rRNA gene sequencing, and plasma metabolites were profiled using GC-MS and UPLC-MS. Differential analysis, microbiota-metabolite-c

Effects of feeding a Lactobacillus plantarum JL01 diet on caecal bacteria and metabolites of weaned piglets.

Currently, knowledge is limited concerning the impact of a Lactobacillus plantarum JL01 diet for weaned piglets on caecal bacteria and metabolite profiles. In our experiments, 24 weaned piglets were randomly divided into two groups; each piglet in the treatment groups (Cec-Lac) was fed a basic diet and administered 10 ml of L. plantarum JL01 (1·0 × 109  CFU per ml) every day. The control group (Cec-Con) was fed a basic diet. After feeding for 28 days, we analysed the parameters of the caecal digesta of weaned piglets. We used 16S rDNA gene sequencing and mass spectrometry (MS)-based metabolomi

Improved deferred antagonism technique for detecting antibiosis.

The deferred antagonism technique has been utilized for several decades for detecting antibiosis activity. Most protocols require the elimination of antibiotic-producing cells by exposing them to chloroform vapour, UV radiation or filter sterilizing the filtrate steps that require additional time and expense to complete. We provide a modified approach to current soft agar overlay practices, which involves addition of antibiotics to the soft agar overlay to inhibit growth of the producer but not the indicator strain. This technique can be used to reproducibly and efficiently screen for antibiot

A Thorn in a Haystack: A Rare Case of Septic Arthritis.

Pantoea agglomerans is a facultative anaerobe and environmental bacterium that could be a cause of opportunistic human infections, especially in wound infection with plant material. Arthritis or synovitis secondary to penetrating plant thorn injuries is not frequently reported. We present the case of a 35-year-old otherwise healthy male with a bramble thorn penetrating injury of the left knee. P. agglomerans was isolated from the synovial fluid. The patient was treated with amoxicillin/clavulanate according to sensitivity testing. This case highlights the importance of precise and thorough me

Integrating Host Phylogeny and Bacterial Detection Patterns Reveals Contrasting Associations of Buchnera aphidicola and Other Aphid-Associated Bacteria.

Aphids are important agricultural pests that interact with diverse bacterial taxa living within or associated with their bodies. Some bacteria, such as the obligate symbiont Buchnera aphidicola, provide essential nutritional functions and are predominantly inherited vertically, whereas other aphid-associated bacteria may have more variable distributions and ecological roles. In this study, we investigated patterns of association between aphid evolutionary relationships and selected bacterial taxa. Aphids were identified using morphological characteristics and mitochondrial cytochrome c oxidase

[Farmer's lung antigens in Germany].

Recent studies suggest that besides the long-known farmer's lung antigen sources Saccharopolyspora rectivirgula (Micropolyspora faeni), Thermoactinomyces vulgaris, and Aspergillus fumigatus, additionally the mold Absidia (Lichtheimia) corymbifera as well as the bacteria Erwinia herbicola (Pantoea agglomerans) and Streptomyces albus may cause farmer's lung in Germany. In this study the sera of 64 farmers with a suspicion of farmer's lung were examined for the following further antigens: Wallemia sebi, Cladosporium herbarum, Aspergillus versicolor, and Eurotium amstelodami. Our results indicate

The specificity of bacterial siderophore receptors probed by bioassays.

The ability to utilize siderophores of bacterial and fungal origin has been studied in wild-type and mutant strains of the enterobacterial genera Salmonella, Escherichia, Shigella, Moellerella, Klebsiella, Enterobacter, Hafnia, Pantoea, Ewingella, Tatumella, Yersinia, and in the non-enterics Aeromonas, Pseudomonas and Aureobacterium. Although only a few representative strains were tested, the results show characteristic genus-specific differences in the utilization of hydroxamate and catecholate siderophores. Moreover, the different response to structural alterations of certain siderophore cla

Gammaproteobacteria as essential primary symbionts in the striped shield bug, Graphosoma Lineatum (Hemiptera: Pentatomidae).

Many members of suborder Heteroptra harbor heritable symbiotic bacteria. Here we characterize the gut symbiotic bacterium in Graphosoma lineatum (Hemiptera: Pentatomidae) by using molecular phylogeny, real-time PCR analysis as well as light and electron microscopy observations. The microscopy observations revealed the presence of a large number of rod-shaped bacterial cells in the crypts. A very high prevalence (98 to 100%) of the symbiont infection was found in the insect populations that strongly supports an intimate association between these two organisms. Real-time PCR analysis also showed

ESBL-producing Gram-negative organisms in the healthcare environment as a source of genetic material for resistance in human infections.

BACKGROUND: The increasing prevalence of extended-spectrum β-lactamase (ESBL)-producing Enterobacteriaceae in the healthcare setting and in the community despite established infection control guidelines indicates that these microorganisms may possess survival strategies that allow them to persist in the environment. AIMS: To determine the extent and variation in endemic ESBL-carrying species in different ward environments, and to investigate the potential for cephalosporin resistance to be transferred from environmental isolates to human pathogens. METHODS: Conventional microbiological metho

Potential for Nezara viridula (Hemiptera: Pentatomidae) to transmit bacterial and fungal pathogens into cotton bolls.

Recently, we showed that the southern green stink bug (SGSB), Nezara viridula (L.), can transmit Pantoea agglomerans (Ewing and Fife), an opportunistic bacterium, into green cotton bolls resulting in plant disease. Here, we hypothesized that our established model could be used to determine if the SGSB was a general, non-discriminate vector by using two other opportunistic bacterial pathogens of bolls (Pantoea ananatis [Serano] and Klebsiella pneumoniae [Schroeter]) and the known fungal pathogen Nematospora coryli (Peglion). Variants of P. ananatis (strain Pa-1R) and K. pneumoniae (strain Kp 5-

Post-flexible Bronchoscopy Escherichia coli Empyema: A Rare Complication or an Inflammatory Bacterial Translocation?

Empyema is an unusual complication of diagnostic bronchoscopy. We report a case of a male patient in his late 70s with bronchiectasis who developed a right lower lobe empyema 10 days after undergoing flexible bronchoscopy for incidentally detected lung nodules. Bronchial washings grew Staphylococcus aureus (S. aureus) and the atypical Gram-negative bacillus Pantoea septica (P. septica), while subsequent pleural fluid culture yielded Escherichia coli (E. coli). This case highlights the risk of post-bronchoscopy infectious complications in patients with structural lung disease and illustrates ho

The immune factors involved in the rapid clearance of bacteria from the midgut of the tick Ixodes ricinus.

Ticks are obligate hematophagous arthropods that transmit a wide range of pathogens to humans as well as wild and domestic animals. They also harbor a non-pathogenic microbiota, although our previous study has shown that the diverse bacterial microbiome in the midgut of Ixodes ricinus is quantitatively poor and lacks a core. In artificial infections by capillary feeding of ticks with two model bacteria (Gram-positive Micrococcus luteus and Gram-negative Pantoea sp.), rapid clearance of these microbes from the midgut was observed, indicating the presence of active immune mechanisms in this orga

The Therapeutic Efficacy of Inhaled Vitamin D3 Metabolites Initiated During the Acute Phase of Hypersensitivity Pneumonitis: An In Vivo Study.

Our earlier studies have revealed the beneficial impact of vitamin D3 (VD3) metabolites on the development of pulmonary fibrosis in a murine model of hypersensitivity pneumonitis (HP). Despite the discovery of the great antifibrotic potential of both the bioactive form of VD3, 1,25(OH)2-VD3 and its direct precursor, 25(OH)-VD3, study designs based on the co-administration of VD3-metabolites and fibrosis-inducers do not reflect the clinical situation. Thus, the presented study aims to examine the effectiveness of therapeutic interventions based on the mentioned inhalations, initiated during the

Comprehensive genomic insights into Mixta calida isolated from the faecal sample of a tuberculosis patient in India.

Mixta calida, formerly known as Pantoea calida, is a motile Gram-negative, facultatively anaerobic bacterium with coccoid rod morphology. Although previously considered non-pathogenic, emerging case studies indicate its potential role in causing serious infections, including bacteraemia, meningitis, sepsis, and implant-associated infections. This study presents the first whole-genome sequence of M. calida of Indian origin, isolated from the stool sample of a tuberculosis patient undergoing treatment. Sequencing was performed using the Illumina NextSeq 2000 and Oxford Nanopore PromethION platfo

Successful treatment of infertility in a female Sumatran orangutan Pongo abelii.

In 2011, a female Sumatran orangutan housed at Durrell Wildlife Conservation Trust became infertile following a massive antepartum hemorrhage in labor and the delivery of a stillborn infant. The placenta was infected with Pantoea sp. Hysterosalpingography (HSG) revealed blocked fallopian tubes, and pressurized fallopian tube perfusion was used to reverse the tubal occlusion. She subsequently conceived and following an intensive training program, we were able to measure umbilical artery waveform analysis for fetal well-being and placental localization to exclude placenta previa, which could com
