Changes in parvalbumin immunoreactive retinal ganglion cells and amacrine cells after optic nerve injury.

Parvalbumin (PARV) is a Ca(2+)-binding protein that may offer resistance to cell death as it primarily functions to maintain Ca(2+) homeostasis. The purpose of this study was to investigate whether PARV expressing retinal ganglion cells (RGCs) would be more resistant to cell death than RGCs that do not express PARV. RGCs in Sprague-Dawley rats were retrogradely labeled with Fluorogold (FG). After 2-28 days following an optic nerve crush (ONC) injury immunohistochemistry was performed on the sections using antibodies against PARV and markers of RGCs. The proportion of retinal ganglion cell laye

Cortical parvalbumin and somatostatin GABA neurons express distinct endogenous modulators of nicotinic acetylcholine receptors.

BACKGROUND: Inhibition from GABAergic interneurons in brain circuits is a critical component of cognitive function. This inhibition is regulated through a diverse network of neuromodulation. A number of recent studies suggest that one of the major regulators of interneuron function is nicotinic acetylcholinergic transmission and dysregulation of both systems is common in psychiatric conditions. However, how nicotinic modulation impacts specific subpopulations of diverse GABAergic interneurons remains in question. One potential way of conferring specificity to the convergence of GABAergic and n

PGC-1α provides a transcriptional framework for synchronous neurotransmitter release from parvalbumin-positive interneurons.

Accumulating evidence strongly implicates the transcriptional coactivator peroxisome proliferator-activated receptor γ coactivator 1α (PGC-1α) in the pathophysiology of multiple neurological disorders, but the downstream gene targets of PGC-1α in the brain have remained enigmatic. Previous data demonstrate that PGC-1α is primarily concentrated in inhibitory neurons and that PGC-1α is required for the expression of the interneuron-specific Ca(2+)-binding protein parvalbumin (PV) throughout the cortex. To identify other possible transcriptional targets of PGC-1α in neural tissue, we conducted a

Parvalbumin tunes spike-timing and efferent short-term plasticity in striatal fast spiking interneurons.

Striatal fast spiking interneurons (FSIs) modulate output of the striatum by synchronizing medium-sized spiny neurons (MSNs). Recent studies have broadened our understanding of FSIs, showing that they are implicated in severe motor disorders such as parkinsonism, dystonia and Tourette syndrome. FSIs are the only striatal neurons to express the calcium-binding protein parvalbumin (PV). This selective expression of PV raises questions about the functional role of this Ca(2+) buffer in controlling FSI Ca(2+) dynamics and, consequently, FSI spiking mode and neurotransmission. To study the function

Dissecting local circuits: parvalbumin interneurons underlie broad feedback control of olfactory bulb output.

In the mouse olfactory bulb, information from sensory neurons is extensively processed by local interneurons before being transmitted to the olfactory cortex by mitral and tufted (M/T) cells. The precise function of these local networks remains elusive because of the vast heterogeneity of interneurons, their diverse physiological properties, and their complex synaptic connectivity. Here we identified the parvalbumin interneurons (PVNs) as a prominent component of the M/T presynaptic landscape by using an improved rabies-based transsynaptic tracing method for local circuits. In vivo two-photon-

Hypoglossal motor neurons display a reduced calcium increase after axotomy in mice with upregulated parvalbumin.

Motor neurons that exhibit differences in vulnerability to degeneration have been identified in motor neuron disease and in its animal models. The oculomotor and hypoglossal neurons are regarded as the prototypes of the resistant and susceptible cell types, respectively. Because an increase in the level of intracellular calcium has been proposed as a feature amplifying degenerative processes, we earlier studied the calcium increase in these motor neurons after axotomy in Balb/c mice and demonstrated a correlation between the susceptibility to degeneration and the intracellular calcium increase

Calbindin D-28k, parvalbumin and calcitonin gene-related peptide immunoreactivity in the canine spinal cord.

This study was conducted as a comparative analysis of the immunohistochemical localization of calbindin D-28k, parvalbumin and the calcitonin gene-related peptide (CGRP) in the cervical through the sacral spinal cord of mongrel dogs, to reveal any distinct patterns of distribution and possible involvement in spinal processing. In laminae I and II of the substantia gelatinosa, both calbindin D-28k and CGRP showed strong immunoreactivity, with calbindin D-28k being positive in both cells and fibres, while CGRP was positive in fibres only. Parvalbumin and CGRP immunoreactive cells were widely dis

Loss of perikaryal parvalbumin immunoreactivity from surviving GABAergic neurons in the CA1 field of epileptic gerbils.

The Mongolian gerbil (Meriones unguiculatus) is known as a genetic model of epilepsy. Seizure behavior ranges from subtle events like arrest of motor activity and facial spasms to grand mal seizures followed by automatisms. Exploratory behavior in a stressful situation represents the most effective environment for provoking seizures in gerbils. Modifications of the inhibitory hippocampal circuits have been suggested as a cause of seizure susceptibility in the gerbil. This study presents a quantitative analysis of the hippocampal parvalbumin (PV)-immunoreactive and gamma-aminobutyric acid (GABA

Parvalbumin-positive small myelinated fiber bundles in anterior marginal areas in the human cervical cord.

During pathological examinations of amyotrophic lateral sclerosis (ALS) patients, we noticed that bundles of dense small myelinated fibers were relatively well preserved in the anterior marginal areas of the cervical cords in a few ALS patients with diffuse myelin pallor. Moreover, these fibers were strongly immunostained by anti-parvalbumin antiserum. These parvalbumin-positive fiber bundles were also seen in other ALS and non-ALS patients, and they were continuously traced up to the spinothalamic tract areas of the medulla oblongata. Therefore, it is suggested that these fiber bundles belong

Transcriptional Profiles of Somatostatin and Parvalbumin Interneuron Subtypes in the Human Dorsolateral Prefrontal Cortex: Implications for Schizophrenia.

OBJECTIVE: Alterations in the somatostatin (SST) and parvalbumin (PVALB) classes of inhibitory GABAergic interneurons in the dorsolateral prefrontal cortex (DLPFC) are thought to contribute to the core cognitive impairments of schizophrenia. Both classes of interneurons are composed of multiple subtypes, but the distinguishing features and relative proportions of these subtypes have not been determined in the DLPFC. The aim of this study is to provide a comprehensive analysis of these subtypes from young and middle-age adults with no known psychiatric disorders. METHODS: The authors used sing

Molecular sensitization profiles and β-parvalbumin sensitization burden associated with clinical fish allergy in children.

BACKGROUND: Fish allergy is a potentially severe condition that is frequently managed with broad elimination diets. Molecular allergology may improve risk stratification by distinguishing clinically relevant fish allergy from asymptomatic sensitization. We aimed to characterize molecular sensitization patterns in fish-sensitized children and identify molecular profiles associated with clinical fish allergy. METHODS: Sixty-one fish-sensitized children (0-18 years) were evaluated using a multiplex molecular allergy assay (ALEX2). Twenty-eight children were classified as fish-allergic based on a

Baicalein alleviates anxiety symptom in Parkinson's disease by targeting Sema3A-mediated parvalbumin interneuron dysfunction.

BACKGROUND: Anxiety is a common non-motor symptom of Parkinson's disease (PD), affecting 20 %-50 % of the patients, with an unmet need for effective therapies. Although perineuronal net (PNN) abnormalities have been implicated in both PD and anxiety, the underlying mechanisms and potential treatments remain poorly characterised. PURPOSE: In this study, we investigated whether baicalein ameliorates PD-associated anxiety via modulation of parvalbumin (PV) interneuron function by targeting semaphorin 3A (Sema3A). Syngap1 regulates the synaptic drive and membrane excitability of Parvalbumin-positive interneurons in mouse auditory cortex.

SYNGAP1 haploinsufficiency-related intellectual disability (SYNGAP1-ID) is characterized by moderate to severe ID, generalized epilepsy, autism spectrum disorder, sensory processing dysfunction, and other behavioral abnormalities. While numerous studies have highlighted a role of Syngap1 in cortical excitatory neurons development, recent studies suggest that Syngap1 plays a role in GABAergic inhibitory neuron development as well. However, the molecular pathways by which Syngap1 acts on GABAergic neurons, and whether they are similar or different from the mechanisms underlying its effects in ex

Parvalbumin interneurons regulate rehabilitation-induced functional recovery after stroke and identify a rehabilitation drug.

Motor disability is a critical impairment in stroke patients. Rehabilitation has a limited effect on recovery; but there is no medical therapy for post-stroke recovery. The biological mechanisms of rehabilitation in the brain remain unknown. Here, using a photothrombotic stroke model in male mice, we demonstrate that rehabilitation after stroke selectively enhances synapse formation in presynaptic parvalbumin interneurons and postsynaptic neurons in the rostral forelimb motor area with axonal projections to the caudal forelimb motor area where stroke was induced (stroke-projecting neuron). Functional Alterations in Parvalbumin-Positive Interneurons after Traumatic Brain Injury in Somatosensory Cortex of Mice.

Traumatic brain injury (TBI) can lead to long-lasting cognitive deficits in the human brain, with a considerable contribution of secondary morphological and functional sequela in cortical regions distant to the lesion site. In order to uncover the role of early functional alterations in parvalbumin-positive basket cells (PV-BCs), an interneuron population required to maintain inhibition of neocortical circuits, to this dysfunctional plasticity, we investigated anatomical and electrophysiological properties of PV-BCs in PV-IRES-Cre-tdTomato mice of both sexes 24 h after a cortical impact. Hyperactivity of subicular parvalbumin interneurons drives early amyloid pathology and cognitive deficits in Alzheimer's disease.

Atrophy of the subiculum is the earliest hippocampal anatomical marker of Alzheimer's disease (AD) and is closely associated with early cognitive decline. However, the underlying mechanisms driving this vulnerability remain unclear. In this study, using the 5×FAD mouse model, we identified significant amyloid-beta (Aβ) accumulation in the subiculum during the early stages of AD. Through a combination of laser microdissection and proteomic analysis, we uncovered early dysregulation of GABAergic neurons in the subiculum. Further investigation revealed that parvalbumin (PV) interneurons (PV-INs)

Prefrontal parvalbumin neurons as a target for enhancing cognition in non-pathological and 22q11.2 microdeletion syndrome mice.

To achieve goals in a dynamic environment, the prefrontal cortex (PFC) facilitates various cognitive functions that allow an organism to detect and discriminate information, identify changes in context, and apply this information to representations of known rules and internal states. A failure of organized communication in the PFC is thought to contribute to the emergence of cognitive impairments in psychiatric diseases, with attentional deficits occurring as a fundamental symptom across various conditions. The 22q11.2 microdeletion syndrome is a rare genetic condition that confers a high risk

Localization of Calretinin, Parvalbumin, and S100 Protein in Nothobranchius guentheri Retina: A Suitable Model for the Retina Aging.

Calcium-binding proteins (CaBPs) are members of a heterogeneous family of proteins able to buffer intracellular Ca2+ ion concentration. CaBPs are expressed in the central and peripheral nervous system, including a subpopulation of retinal neurons. Since neurons expressing different CaBPs show different susceptibility to degeneration, it could be hypothesized that they are not just markers of different neuronal subpopulations, but that they might be crucial in survival. CaBPs' ability to buffer Ca2+ cytoplasmatic concentration makes them able to defend against a toxic increase in intracellular

Activation of TrkB in Parvalbumin interneurons is required for the promotion of reversal learning in spatial and fear memory by antidepressants.

Critical period-like plasticity (iPlasticity) can be reinstated in the adult brain by several interventions, including drugs and optogenetic modifications. We have demonstrated that a combination of iPlasticity with optimal training improves behaviors related to neuropsychiatric disorders. In this context, the activation of TrkB, a receptor for BDNF, in Parvalbumin-positive (PV+) interneurons has a pivotal role in cortical network changes. However, it is unknown if the activation of TrkB in PV+ interneurons is important for other plasticity-related behaviors, especially for learning and memory
