Pasteurella multocida: from zoonosis to cellular microbiology.

In a world where most emerging and reemerging infectious diseases are zoonotic in nature and our contacts with both domestic and wild animals abound, there is growing awareness of the potential for human acquisition of animal diseases. Like other Pasteurellaceae, Pasteurella species are highly prevalent among animal populations, where they are often found as part of the normal microbiota of the oral, nasopharyngeal, and upper respiratory tracts. Many Pasteurella species are opportunistic pathogens that can cause endemic disease and are associated increasingly with epizootic outbreaks. Zoonotic

Molecular pathogenicity of the oral opportunistic pathogen Actinobacillus actinomycetemcomitans.

Periodontitis is mankind's most common chronic inflammatory disease. One severe form of periodontitis is localized aggressive periodontitis (LAP), a condition to which individuals of African origin demonstrate an increased susceptibility. The main causative organism of this disease is Actinobacillus actinomycetemcomitans. A member of the Pasteurellaceae, A. actinomycetemcomitans produces a number of interesting putative virulence factors including (a) an RTX leukotoxin that targets only neutrophils and monocytes and whose action is influenced by a novel type IV secretion system involved in bac

Atypical actinobacillosis affecting hind limbs and lungs in a single beef cattle herd.

Actinobacillosis usually is a sporadic infection that affects the tongue in cattle ("wooden tongue") with possible spread to the digestive tract. Two 4-year-old Rouge-des-Prés cows from a single French beef herd were referred for chronic (2-6 months) swelling and cutaneous nodules in the distal hind limbs. In addition to cutaneous signs, physical examination disclosed cachexia, lameness, lymphadenitis of the hind limbs, and pneumonia in both cows. Cytologic examination of direct skin smears was inconclusive, and no parasites were observed in examination of multiple skin scrapings. Histopatholo

Development of a 23S rRNA-based PCR assay for the identification of Pasteurella multocida.

AIMS: The aim of this work was to develop a rapid diagnostic test for Pasteurella multocida. METHODS AND RESULTS: A polymerase chain reaction (PCR) assay using primers derived from the 23S rRNA gene sequence of Past. multocida was developed. The PCR assay correctly identified all 144 isolates of Past. multocida tested, including type strains of the three subspecies as well as the reference strains for the Heddleston and Carter typing schemes. Of 20 closely related bacteria from the family Pasteurellaceae tested, only the type strains of Past. canis biovar 2 and Past. avium biovar 2 were posit

Biology of Haemophilus ducreyi.

The etiological agent of the sexually transmitted genital ulcer disease chancroid was first described in 1889 by Auguste Ducrey following repeated autoinoculation of purulent ulcer material from a series of patients. The organism was isolated on artificial media a decade later but has remained difficult to isolate consistently, resulting in controversy over its characteristics and role as the causative agent of chancroid. Because of its fastidious growth requirements, including unknown components in blood, the organism was included in the original description of the genus Haemophilus. Requirem

A recombinant subunit vaccine for bovine RSV and Histophilus somni protects calves against dual pathogen challenge.

Bovine respiratory syncytial virus (BRSV) and Histophilus somni synergize to cause respiratory disease in cattle. These pathogens cause enhanced disease during dual-infection and an IgE response to antigens of H. somni in dual-infected but not singly infected calves. Vaccines containing whole inactivated BRSV or H. somni have been associated with IgE responses A vaccine strategy that avoids stimulation of IgE antibodies would provide superior protection from dual infection. We hypothesized that a subunit vaccine consisting of the nucleoprotein (NP) from BRSV and the recombinant antigen IbpA DR

Hepatic Abscess Caused by Aggregatibacter kilianii in an Immunocompetent Active Duty Male, Case Report.

Bacterial translocation from normal oral flora, such as Aggregatibacter spp., can rarely lead to endocarditis, bloodstream infections, and abscesses. We present a case of an immunocompetent active duty service member with a hepatic abscess from a presumed routine dental hygiene cleaning with Aggregatibacter kilianii bacteremia. Aggregatibacter kilianii is a relatively novel species, closely related to the well-reported Aggregatibacter aphrophilus but genetically distinct. This appears to be one of the first described cases of a hepatic abscess caused by this species.

Bovine peritonitis associated with Mannheimia haemolytica serotype 2 in a three-day-old Japanese Black calf.

A Japanese Black calf became dehydrated on the first day of life and died on the third day. Gross examination revealed a large amount of yellowish-brown serous fluid in the abdominal cavity and whitish-yellow fibrin in the serosa of the abdominal organs. Patchy red spots were observed throughout the peritoneum, and the outer membrane of the umbilical arteries was dark red. Bacteriologically, Mannheimia haemolytica serotype 2 was isolated from the umbilical arteries and vein, liver, and kidney. Histopathology revealed inflammation with M. haemolytica serotype 2 in the outer membrane of the umbi

Vaccination schedules in small ruminant farms.

Development and implementation of health management plans is the cornerstone of profitable farms; prevention of microbial diseases by means of vaccination is an integral part of such a plan. In every production type and management system in small ruminants, microbial diseases have a major significance, hence their proper control must be based in good health management practices, including use of effective and safe vaccines. Development of various types of vaccines is evolving very quickly in recent years and the improvement of new type of vaccines offers prospects. The article reviews and disc

Genetic diversity and tetracycline resistance genes of Histophilus somni.

Nasal swabs were collected at three time points from 2378 calves in four feedlots and cultured for Histophilus somni to assess genetic relatedness and tetracycline resistance. The proportions of animals carrying tetracycline resistant isolates were 0.32% at arrival, 14.82% at interim, and 0.80% at exit. The 606 H. somni isolates recovered were compared by pulsed-field gel electrophoresis (PFGE), screened for the presence of plasmids, and assessed for the tetracycline resistance genes tet(A), tet(B), tet(C), tet(E), tet(G), tet(H), tet(K), tet(L), tet(M) and tet(O) using multiplex polymerase ch

NTDB: a curated database of natural transformation machinery and regulatory genes in prokaryotes.

Natural transformation, genetically controlled by specialized machinery and regulatory genes, drives evolution and adaptation in prokaryotes. This process facilitates the spread of clinical traits such as antibiotic resistance. The DNA uptake specificity is mediated by conserved motifs found in certain naturally transformable species. Despite its evolutionary and clinical significance, dedicated resources for natural transformation have been lacking. Here, we present the Natural Transformation DataBase (NTDB) (https://bioinfo-mml.sjtu.edu.cn/NTDB/), which compiles information on machinery and

Pulmonary microbial profiles in slaughtered Duolang sheep from Kashgar stratified by gross lung lesions: implications for respiratory disease surveillance.

Ovine pneumonia is a multifactorial respiratory disease in which complex microbial communities and antimicrobial resistance complicate disease control. This study characterized pulmonary microbial profiles in 49 slaughtered Duolang sheep from Kashgar stratified by gross lung lesions (apparently healthy, n = 10; pneumonic, n = 39) using pathology, bacterial isolation, Mycoplasma ovipneumoniae PCR, antimicrobial susceptibility testing, and 16S rRNA gene sequencing. Suppurative pneumonia predominated in pneumonic lungs (31/39, 79.5%), and M. ovipneumoniae positivity was higher than in apparently

Rodentibacter abscessus sp. nov. isolated from pet hamster subcutaneous abscess.

Five strains related to the genus Rodentibacter were isolated from five pet hamsters kept in Japan. Isolates were genotypically and phenotypically distinct from previously described species within the genus Rodentibacter (family Pasteurellaceae). The five strains were the most genetically similar to Rodentibacter haemolyticus 1625-19T, exhibiting a 16S rRNA gene sequence similarity of 96.5%, an average nucleotide identity value of 90.8% and a digital DNA-DNA hybridization value of 42.7%. Phenotypic characteristics further distinguish the isolated strains from other Rodentibacter species. Altho

Development and comparison of challenge models for three serovars of Avibacterium paragallinarum.

Infectious coryza (IC), caused by Avibacterium paragallinarum (Av. paragallinarum), imposes a substantial economic burden on global poultry production. This study established and standardized comparative challenge models employing infraorbital sinus injection and intranasal inoculation to evaluate the pathogenicity of Av. paragallinarum serovars A, B, and C in SPF chickens. Infraorbital sinus injection leads to rapid onset, stable incidence, and more severe disease progression. The clinical symptom scores at 1 days post-infection (dpi) were higher in the infraorbital sinus injection group comp

Genomic and pathogenic characterization of RTX toxin producing Rodentibacter sp. that is closely related to Rodentibacter haemolyticus.

Rodentibacter spp. are opportunistic pathogens that are often isolated from the upper respiratory tracts of laboratory rodents. In particular, R. pneumotropicus and R. heylii require considerable caution in rodent colonies, as they cause lethal pneumonia in rodents. A new species, R. haemolyticus, has recently been classified in the genus, and a very closely related strain, Rodentibacter sp. strain JRC, has been isolated in Japan. This study focused on strain JRC by performing genomic and pathogenic analyses. Draft genome sequencing of strain JRC identified several genes coding for putative vi

Host Immune Response to Histophilus somni.

Histophilus somni is known to cause several overlapping syndromes or to be found in genital or upper respiratory carrier states in ruminants. Vaccines have been used for decades, yet efficacy is controversial and mechanisms of protective immunity are not well understood. Since H. somni survives phagocytosis, it has sometimes been considered to be a facultative intercellular parasite, implying that cell-mediated immunity would be critical in protection. However, H. somni not only inhibits phagocyte function, but also is cytotoxic for macrophages. Therefore, it does not live for long periods in

A tissue cage model in calves for studies on pharmacokinetic/pharmacodynamic interactions of antimicrobials.

An in vivo model for studies of pharmacokinetic/pharmacodynamic (PK/PD) interactions of antimicrobials was developed. Tissue cages with a constant surface area but with different volumes were implanted in calves and infected with Mannheimia haemolytica. Penicillin was injected directly into the cages. With this procedure, different concentration-time profiles could be simulated so that the effect of a range of PK/PD indices on the infection could be monitored. The area under the curve to minimum inhibitory concentration (MIC) and time above MIC were equally predictive for effect, but Cmax to M

[Brain abscess due to Aggregatibacter aphrophilus].

Brain abscess is a focal suppurative process produced in most cases by bacterial agents. Aggregatibacter aphrophilus is a gram-negative bacteria belonging to the HACEK group, which causes infective endocarditis, liver abscesses, among others. Brain abscesses secondary to this germ are rare and, in most cases, it is associated with contact with pets, poor dental hygiene or dental procedures. Treatment consists of drainage of the abscess (greater than 2.5 cm) combined with antibiotic therapy, ideally beta-lactams. The case of a 64-year-old male patient with no relevant history is here presented.
