Candida tropicalis and Penicillium marneffei mixed fungaemia in a patient with Waldenström's macroglobulinaemia.

A patient with Waldenstrom's macroglobulinaemia who had previously been treated with cladribine presented with septic arthritis caused by Escherichia coli. The patient's condition subsequently deteriorated, and he succumbed to pneumonia and mixed fungaemia due to Candida tropicalis and Penicillium marneffei. Profound lymphopenia coexisted with fungaemia. This is the first reported case of mixed Penicillium marneffei and Candida fungaemia and penicilliosis marneffei in a patient with Waldenstrom's macroglobulinaemia. Penicilliosis marneffei should be considered as a potential complication in pa

[Extrinsic allergic alveolitis caused by Penicillium frequentans. Review and presentation of a case].

We present a case of extrinsic allergic alveolitis due to Penicillium frequentans exposure (suberosis) at a cork factory. Diagnosis was based on clinical history, a restrictive spirometric pattern, evidence of reticular-nodular lung infiltrates on the X-ray, intradermal skin tests with dual reaction and precipitins-positive serum samples in the presence of the P. frequentans fungus. Although the patient refused to submit to a specific bronchial challenge, two natural challenges occurred when he once again happened to handle wet cork after leaving his previous job. The literature referring to t

Nephrotoxicity of Penicillium aurantiogriseum, a possible factor in the aetiology of Balkan endemic nephropathy.

Water-soluble components of a nephrotoxic isolate of Penicillium aurantiogriseum have been fractionated by sequential ion-exchange, size-exclusion gel filtration, reverse-phase silica chromatography and HPLC. Nephrotoxicity in the rat was confined to a size-exclusion fraction approximating to 1,500 daltons, which also inhibited DNA synthesis in cultured kidney cells. The more sensitive in vitro assay allowed toxicity to be followed to a sub-fraction from gradient-elution HPLC which in further HPLC resolved into a small group of glycopeptides. Recent Yugoslavian P. aurantiogriseum isolates, fro

Discovery of previously undescribed specialized metabolites from the endophytic fungus Penicillium griseofulvum.

Eight previously undescribed specialized metabolites were obtained and characterised from Penicillium griseofulvum, a root endophyte from Delphinium grandiflorum, namely three aryl ether derivatives (1-3), three 2,5-DKP derivatives (7-9), two aryl esters (10-11), along with 22 known compounds. Their structures were fully characterised by comprehensive spectroscopic analyses. Notably, compounds 25, 28, and 30 exhibited moderate cytotoxic activities against triple-negative breast cancer (TNBC) cells (MDA-MB-231). Compound 29 demonstrated potent broad-spectrum antibacterial activity against clini

Disseminated Penicillium labradoris (synonym: P. labradorum) in three dogs from Western Australia.

INTRODUCTION: This case series presents three new cases of disseminated Penicillium labradoris (syn. P. labradorum) infection in unrelated dogs from Western Australia, illustrating varied clinical presentations and outcomes. P. labradoris infections are rare in dogs, with only three prior cases reported worldwide. Typically considered low-virulence mycotic pathogens, Penicillium species can occasionally cause severe disease in dogs likely affected by some genetically programmed immune defect. CASE HISTORIES: Case one was a three-year-old Staffordshire bull terrier crossbreed who presented for

Origins, Structures, and Bioactivities of Secondary Metabolites from Marine-derived Penicillium Fungi.

In recent years, marine-derived Penicillium fungi have received remarkable interest as a valuable source of novel natural products encompassing diverse chemical structures and bioactive properties. Mangroves, sediments, algae, and sponges are the four main sources of marine-derived Penicillium fungi. As of 2014, more than 390 novel natural products have been isolated from the marine- derived Penicillium fungi, mainly including polyketides, alkaloids, terpenoids, and macrolides. Biological investigations have shown that these compounds possess antimicrobial, anti-inflammatory, cytotoxic, and ot

Identification and functional characterization of Penicillium marneffei major facilitator superfamily (MFS) transporters.

is a thermally dimorphic fungus that causes penicilliosis, and become the third-most-common opportunistic fungal infection in immunocompromised patients in Southeast Asia. Azoles and amphotericin B have been introduced for the treatment, however, it is important to investigate possible mechanisms of azole resistance for future treatment failure. We identified 177 putative MFS transporters and classified into 17 subfamilies. Among those, members of the Drug:H+ antiporter 1 subfamily are known to confer resistance to antifungals. Out of 39 paralogs, three (encoded by PmMDR1, PmMDR2, and PmMDR3)

Auranthine, a Benzodiazepinone from Penicillium aurantiogriseum: Refined Structure, Absolute Configuration, and Cytotoxicity.

The structure of the known Penicillium aurantiogriseum-derived secondary metabolite auranthine was refined using a combination of synthetic, spectroscopic, and X-ray diffractometric approaches. Thus, auranthine was shown to be a fused quinazolino benzodiazepinedione (2) bearing an acyclic aliphatic nitrile moiety, thereby significantly differing from the originally proposed structure 1 published in 1986. Its absolute configuration was confirmed by CD spectroscopy and DFT calculations. The cultivation of P. aurantiogriseum was optimized, allowing high production of auranthine. The cytotoxicity

Comparative metabolomic study of Penicillium chrysogenum during pilot and industrial penicillin fermentations.

Comparative metabolomics was carried out to investigate the metabolic differences of Penicillium chrysogenum in the pilot and industrial fermentations that resulted from the scale-up. By principal component analysis, the early stages of two fermentation processes were clearly distinguished, whereas the middle and final stages were clustered together. It indicated that the different metabolisms of cells in the pilot and industrial fermentations mainly existed during the early stage. Furthermore, the levels of polyamines, polyols, glycolysis, and tricarboxylic acid cycle intermediates, which cha

Nonlinear biosynthetic gene cluster dose effect on penicillin production by Penicillium chrysogenum.

Industrial penicillin production levels by the filamentous fungus Penicillium chrysogenum increased dramatically by classical strain improvement. High-yielding strains contain multiple copies of the penicillin biosynthetic gene cluster that encodes three key enzymes of the β-lactam biosynthetic pathway. We have analyzed the gene cluster dose effect on penicillin production using the high-yielding P. chrysogenum strain DS17690 that was cured from its native clusters. The amount of penicillin V produced increased with the penicillin biosynthetic gene cluster number but was saturated at high copy

Cutaneous manifestations of disseminated Penicillium marneffei mycosis in five HIV-infected patients.

Five cases of disseminated Penicillium marneffei mycosis of Thai HIV patients, presented between November 1990 and April 1991, have been investigated. All patients were male and had bone marrow involvement. They showed mucocutaneous lesions with papules predominantly on the faces and upper extremities (5 cases), palatal papules (3 cases), and chronic genital ulcer (2 cases). P. marneffei was cultured, and the specific tissue form of the fungus was demonstrated histopathologically from skin and bone marrow specimens. In future, with increasing awareness of mucocutaneous lesions, disseminated pe

Some properties of acid protease from the thermophilic fungus, Penicillium duponti K1014.

A purified acid protease from a true thermophilic fungus, Penicillium duponti K1014, was most active at pH 2.5 for milk casein and at pH 3.0 for hemoglobin. The enzyme was stable at a pH range of 2.5 to 6.0 at 30 C for 20 h. The acid protease retained full activity after 1 h at 60 C at a pH range between 3.5 and 5.5. At the most stable pH of 4.5, more than 65% of its activity remained after heat treatment for 1 h at 70 C. These thermal properties show the enzyme as a thermophilic protein. The enzyme activity was strongly inhibited by sodium lauryl sulfate and oxidizing reagents such as potassi

Rhizosphere Penicillium and phosphorus availability drive bioactive compound accumulation in Amomum villosum.

INTRODUCTION: Amomum villosum (A. villosum) , a medicinal herb rich in bioactive metabolites (borneol, bornyl acetate), exhibits superior quality in its core production region Yangchun (YC). However, the key habitat factors influencing the quality of A. villosum, as well as the underlying mechanisms, remain unclear. METHODS: This study employed an integrated multi-omics approach, incorporating metabolomics, transcriptomics, soil physicochemical property analysis, and metagenomics, to systematically compare the core production region with non-core regions. By identifying potential factors infl

Antiosteoclastogenic Indole Alkaloids from the Mangrove Endophytic Fungus Penicillium brefeldianum GXIMD 02511.

Eight new prenylated indole alkaloids, brefeldindoles A-F (1-6), G (8), and H (11), together with 17 known analogues, were obtained from the Beibu Gulf mangrove-derived fungal strain Penicillium brefeldianum GXIMD 02511. Their structures, including absolute configurations, were determined by analysis of spectroscopic data and electronic circular dichroism (ECD) calculations. Brefeldindoles A-F (1-6) are characterized as a rare class of indole-diterpenoid derivatives with uncommon 3-methyl-3-hydroxybutyl substituents in the benzene ring. Brefeldindole H (11) is the first example of an unprecede

Optimized Protocol for RNA Isolation from Penicillium spp. and Aspergillus fumigatus Strains.

Efficient RNA isolation from filamentous fungi is crucial for gene expression studies, but it poses significant technical challenges due to the robust cell walls and susceptibility of RNA to degradation by ribonucleases. This study presents the effectiveness of two RNA isolation protocols for four species of filamentous fungi: Penicillium crustosum, Penicillium rubens, Penicillium griseofulvum, and Aspergillus fumigatus. Both protocols utilized Fenzol Plus for cell lysis but varied in the mechanical disruption methods: bead-beating versus manual vortexing. The results show that the bead-beater

Cellulose amendment promotes P solubilization by Penicillium aculeatum in non-sterilized soil.

Successful application of microbial biofertilizers, such as phosphorus (P) solubilizing fungi to agroecosystems, is constrained from the lack of knowledge about their ecology; for example in terms of how they respond to an external input of carbon (C) to get established in the soil. In two soil incubation experiments we examined the performance of the P solubilizing fungus Penicillium aculeatum in non-sterile and semi-sterile (γ-irradiated) soil with different C and P sources. Results from the first experiment with C sources showed that starch and cellulose generally improved P solubilization
