The haplo-insufficient p53 knockout (p53+/-) and zetaglobin v-Ha-ras (Tg.AC) transgenic mouse models were compared to the conventional two rodent species carcinogen bioassay by prospectively testing nine chemicals. Seven of the chemicals classified as carcinogens in the conventional bioassay induced tumors in the liver or kidneys of B6C3F1 mice, and one (pentachlorophenol) also induced tumors in other tissues. Only three chemicals, furfuryl alcohol, pyridine, and pentachlorophenol, induced tumors in rats. The tumorigenic effect of pyridine was seen in F344 rats but not in Wistar strain rats. N

The thermal dependence of the fluorescence polarization of 1,6-diphenylhexatriene was recorded upon interaction of 1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane (DDT) and some other pesticides with dipalmitoylphosphatidylcholine liposomes. Female rats were dosed intraperitoneally with 14C-hexaxhlorobenzene. The drug was administered on 2 or 3 occasions. The total doses amounted to 260 and 390 mg/kg 14C-hexachlorobenzene, respectively. We tested the sensitivity of nine submersed macrophyte species to the fungicides chlorothalonil, pentachlorophenol, fluazinam, and carbendazim. Endpoints determined 3 weeks after the start of the treatment were based on shoot and root growth in water. Carbendazim proved not or only moderately toxic to these macrophytes. Hexachlorobenzene alters the hepatic steroid metabolism, and it was suggested that a porphyria was induced by overproduction of 5 beta-H-steroids. Structurally similar chlorinated hydrocarbons (pentachlorobenzene, pentachlorophenol, 2,4,5-trichlorophenol) without porphyrogenic activity did not affect the steroid metabolism in rat liver.

BACKGROUND: The impacts of man-made chemicals, in particular of persistent organic pollutants, are multifactorial as they may affect the integrity of ecosystems, alter biodiversity and have undesirable effects on many organisms. We have previously demonstrated that the belowground mycobiota of forest soils acts as a buffer against the biocide pollutant pentachlorophenol. However, the trade-offs made by mycobiota to mitigate this pollutant remain cryptic.

Male and female F 344 rats were dosed every other day for 103 days with 50 mumole of hexachlorobenzene (HCB)/kg. Females developed a hepatic porphyria, the urine and liver levels of porphyrins being 40- and 310-fold higher respectively than those of males.
