Molecular Characterization of Lineage IV Peste des Petits Ruminants Viruses in Sudan Isolated from Outbreaks Between 2015 and 2018 Suggests the Existence of the North-East Africa Episystem.

Multiple outbreaks with high mortality rates and a pattern of disease occurrence similar to peste des petits ruminants (PPR) were observed in sheep and goats across many states of Sudan between 2015 and 2018. Therefore, this study aimed to investigate and identify the cause of these disease outbreaks. A total of 276 blood and lung tissue samples were collected from infected sheep (n = 223) and goats (n = 53). Sample analysis in an IC-ELISA revealed the presence of the PPRV antigen in 56.9% of sheep and 62.3% of goat samples. Twenty-seven [sheep (18) and goats (9)] PPRV-positive samples from IC

Establishment of a blocking enzyme-linked immunosorbent assay based on tandem expression of dominant antigenic epitopes of the nucleocapsid protein from attenuated and virulent peste des petits ruminants virus.

Peste des petits ruminants (PPR) is an acute, contagious disease caused by the Peste des Petits Ruminants virus (PPRV), mainly affecting small ruminants such as goats and sheep. PPR poses a serious threat to the healthy development of the livestock industry and wildlife safety. To develop a rapid method for detecting antibodies against the PPRV N protein, we analyzed the dominant antigenic epitopes of the N protein of virulent and attenuated PPRV strains using bioinformatics. The dominant antigenic epitopes of the N gene were linked in tandem via overlapping PCR and cloned into a prokaryotic e

Simultaneous detection of Rift Valley Fever, bluetongue, rinderpest, and Peste des petits ruminants viruses by a single-tube multiplex reverse transcriptase-PCR assay using a dual-priming oligonucleotide system.

The aim of this study was to develop a highly sensitive and specific one-step multiplex reverse transcriptase PCR assay for the simultaneous and differential detection of Rift Valley Fever virus (RVFV), bluetongue virus (BTV), rinderpest virus (RPV), and Peste des petits ruminants virus (PPRV). These viruses cause mucosal lesions in cattle, sheep, and goats, and they are difficult to differentiate from one another based solely on their clinical presentation in suspected disease cases. In this study, we developed a multiplex reverse transcriptase PCR to detect these viruses using a novel dual-p

Evaluation of the Safety and Immunogenicity of a Peste Des Petits Ruminants (PPR), Sheep and Goat Pox (SGP) and Contagious Caprine Pleuropneumonia (CCPP) Multivalent Vaccine (JOVAC-Combo®) Under Field Conditions in Somalia.

Peste des petits ruminants (PPR), sheep and goat pox (SGP), and contagious caprine pleuropneumonia (CCPP) are diseases of small ruminants and are the major constraints to livestock trading in Africa, the Middle East, and Asia. Vaccination against these diseases individually is challenging to veterinary authorities due to high cost and logistics. In this study, we evaluate the safety and immunogenicity of a multivalent vaccine (JOVAC-Combo®) in goats in Somalia that induced seroconversion against all three pathogens, PPR, SGP, and CCPP. A total of 120 apparently healthy, seronegative young goat

[Serological survey of small ruminants in 4 districts of Niger].

In 1990, a serosurvey on 1,474 small ruminants was conducted in four districts of Niger (Maradi, Zinder, Diffa and Dosso). The epidemiology and seroprevalence of eight diseases were studied: Brucella melitensis and B. ovis brucellosis, chlamydiosis, coxiellosis, contagious caprine pleuropneumonia, type A pasteurellosis, peste des petits ruminants, Rift Valley fever, Crimée-Congo haemorragic fever. The main health problem for the development of small ruminants farming, seems to be both pasteurellosis and peste des petits ruminants.

Genetic fusion of peste des petits ruminants virus haemagglutinin and fusion protein domains to the amino terminal subunit of glycoprotein B of bovine herpesvirus 1 interferes with transport and function of gB for BHV-1 infectious replication.

Peste des petits ruminants is an emerging, often fatal viral disease of domestic and wild small ruminants caused by peste des petits ruminants virus. The haemagglutinin and the fusion protein are viral envelope glycoproteins and essential for the infection process and both induce a protective immune response in infected or vaccinated animals. Attempts to generate pseudotyped bovine herpesvirus 1 recombinants firstly by integration of expression cassettes for PPRV-H and PPRV-F into the herpesviral genome or secondly to generate pseudotyped BHV-1 by genetically fusing relevant parts of both PPRV

Determining the decreasing trend of maternal immunity against small ruminant morbillivirus in goat kids.

BACKGROUND: Small ruminant morbillivirus (SRMV) is the etiological agent of Peste des petits ruminants (PPR) disease. PPR is one of the most important viral diseases of small ruminant husbandry. In the endemic countries, vaccination is the main way to control this disease. Administering the first PPR vaccine in goat kids requires decreased maternal immunity. OBJECTIVE: The aim of this study was to determine the decreasing trend of maternal immunity against SRMV in goat kids born from vaccinated goats. METHODS: Twenty Saanen goat kids were studied in two groups including control (n = 5, recei

Exchange of C-Terminal Variable Sequences within Morbillivirus Nucleocapsid Protein Are Tolerated: Development and Evaluation of Two Marker (DIVA) Vaccines (Sungri/96 DIVA, Nigeria/75/1 DIVA) against PPR.

Across Africa, the Middle East, and Asia, peste des petits ruminants virus (PPRV) places a huge disease burden on agriculture, affecting, in particular, small ruminant production. The recent PPR outbreaks in Northern Africa, the European part of Turkey, and Bulgaria represent a significant threat to mainland Europe, as a source of disease. Although two safe and efficacious live attenuated vaccines (Sungri/96 and Nigeria/75/1) are available for the control of PPR, current serological tests do not enable the differentiation between naturally infected and vaccinated animals (DIVA). The vaccinated

Early transcriptome profile of goat peripheral blood mononuclear cells (PBMCs) infected with peste des petits ruminant's vaccine virus (Sungri/96) revealed induction of antiviral response in an interferon independent manner.

Sungri/96 vaccine strain is considered the most potent vaccine providing long-term immunity against peste des petits ruminants (PPR) in India. Previous studies in our laboratory highlighted induction of robust antiviral response in an interferon independent manner at 48 h and 120 h post infection (p.i.). However, immune response at the earliest time point 6 h p.i. (time taken to complete one PPRV life cycle), in PBMCs infected with Sungri/96 vaccine virus has not been investigated. This study was taken up to understand the global gene expression profiling of goat PBMCs after Sungri/96 PPRV vac

Development of reverse genetics system for small ruminant morbillivirus: Rescuing recombinant virus to express Echinococcus granulosus EG95 antigen.

Peste des petits ruminants and cystic hydatidosis may be simultaneously endemic in a given area. Their pathogens are small ruminant morbillivirus (SRMV) and Echinococcus granulosus (E. granulosus), respectively. The SRMV, formerly called peste-des-petits-ruminants virus (PPRV), is classified into the genus Morbillivirus in the family Paramyxoviridae. This virus is an ideal vaccine vector to deliver immunogenic proteins. In this study, a reverse genetics system was developed to rescue a recombinant SRMV (Nigeria 75/1 strain) expressing E. granulosus EG95 antigen in vitro. The recombinant SRMV,

How fast can PPRV spread? Analyzing the results of an experimental PPRV infection of goats equipped with Ultra-Wideband sensors.

Peste des Petits Ruminants (PPR) is a highly contagious disease affecting goats and sheep. The speed and extent of its spread depend on contact patterns and the virus's intrinsic characteristics. To estimate its propagation speed and the role of inter-individual contacts, we conducted PPR virus transmission experiments in goats that were equipped with Ultra-Wideband (UWB) sensors to continuously track inter-individual distances alongside clinical and virological monitoring. Using Bayesian modeling, we integrated contact data and health status to estimate the basic reproduction number (R0), the
