Biochemical indicators of hepatotoxic effects of pesticides.

Pesticides can cause damage to man and beneficial organisms. Some sub-lethal effects of pesticides were studied in birds with a view to identifying characteristic biochemical responses that may be useful for the monitoring of exposure to sub-lethal levels in the field. Pesticides were used: demeton-S-methyl, (DSM), chlorpyriphos, chlorfenviphos, triazophos, pirimicarb, methiocarb and permethrin. Blood was collected before dosing, and 2, 6, 24, 48 and 72 hours after the treatment from the brachial vein of birds. Enzyme activities were assayed in the plasma or serum samples obtained. The assays

Occupational exposure to pesticides and pancreatic cancer.

BACKGROUND: An increased risk of exposure to pesticides for pancreatic cancer has been suggested in a number of epidemiologic studies. METHODS: Cases (N = 484), aged 30-79 years, were diagnosed in 1986-1989. Controls (N = 2,095) were a random sample of the general population. Information on usual occupation and potential confounding factors was obtained. A job-exposure matrix (JEM) approach was used to estimate the level of occupational exposure to pesticides. RESULTS: A significant trend in risk with increasing exposure level of pesticides was observed, with ORs of 1.3 and 1.4 for low and m

Genotoxicity from domestic use of organophosphate pesticides.

This report describes the types of chromosome damage in peripheral blood found in patients exposed to domestic application of organophosphate pesticides. These changes serve as a biomarker of cumulative toxic exposure. Susceptible individuals show DNA damage as chromosome alterations. The importance of these findings is that the apparent genotoxic changes occurred from domestic application of two of the commonly used organophosphate pesticides in America and adds one more adverse potential effect from these hazardous chemicals. Peripheral blood from eight patients exposed to the domestic spray

Dietary exposures to selected metals and pesticides.

Average daily dietary exposures to 11 contaminants were estimated for approximately 120,000 U.S. adults by combining data on annual diet, as measured by a food frequency questionnaire, with contaminant residue data for table-ready foods that were collected as part of the annual U.S. Food and Drug Administration Total Diet Study. The contaminants included in the analysis were four heavy metals (arsenic, cadmium, lead, mercury), three organophosphate pesticides (chlorpyrifos, diazinon, malathion), and four organochlorine pesticides (dieldrin, p,p'-DDE, lindane, heptachlor epoxide). Dietary expos

Effects of pesticides on human leukocyte functions.

Aroclor 1254 and twelve pesticides of different chemical classes were tested for their in vitro effects on lymphocyte mitogenic responses to PHA, rosette formation with sheep red blood cells (E-rosette) and neutrophil chemotaxis. By using a diluted whole blood culture, butoxide at 10 micrometers inhibit lymphocyte mitogenic response to PHA by 26%. Inhibition by other pesticides were less than 20% (5--20(). Higher degrees of inhibition were observed if the lymphocytes were pretreated with pesticides in the absence of serum: pp-DDT inhibited by 73% trithion by 50% and benzylthiocyanate by 30%. E

Mortality in 1969 from pesticides, including aerosols.

Among the 87 deaths in the United States in 1969 for which a causal relationship to pesticides could not be excluded, there were 62 valid accidents, 20 intentionally inflicted deaths, and five deaths incorrectly attributed to pesticides. The reduction in the number of such deaths compared to the decade and a half prior to 1962 probably involves a trend. As in the past, a disproportionate fraction of the victims were children, boys and men, and nonwhites. Findings in four cases (three in 1969) were consistent with the conclusion that intensive inhalation of an aerosol can sensitize the heart to

Broad-scale pesticide screening finds anticoagulant rodenticide and legacy pesticides in Australian frogs.

Pesticide contamination poses a significant threat to non-target wildlife, including amphibians, many of which are already highly threatened. This study assessed the extent of pesticide exposure in dead frogs collected during a mass mortality event across eastern New South Wales, Australia between July 2021 and March 2022. Liver tissue from 77 individual frogs of six species were analysed for >600 legacy and contemporary pesticides, including rodenticides. More than a third (36 %) of the liver samples contained at least one of the following pesticides: brodifacoum, dieldrin, DDE, heptachlor/he

Increasing pesticide-resistant ectoparasitic infections may increase pesticide poisoning risks in children.

Head louse and scabies mite infestations are common among pre-school and school-age children, and topical pesticides are frequently prescribed to treat such conditions. Ectoparasite resistance to the safest and most commonly prescribed pyrethrin/pyrethroid pesticides for ectoparasitic infections has, however, been increasing since the 1980s. The increasing resistance of these arthropods to the safest pesticides may lead to greater use of more toxic, alternative pesticides to control infestations and to prevent institutional outbreaks. MEDLINE and Cochrane searches, 1966-2008, were conducted to

Public attitude toward pesticides. A random survey of pesticide use in Allegheny County, Pa.

Pesticide use was examined by means of a random survey in Allegheny County, Pa., in October and November 1973. The objectives included gaining insight into the need for a new community pesticide program and estimating its public acceptance. The 110 survey sites were grouped as single-family dwellings, commercial and recreational lawns, institutions, farms, rights-of-way, and wasteland. In the single-family dwellings, most householders (85 percent) used a pesticide in the previous 12 months, usually an aerosol insecticide (76 percent) or herbicide (55 percent). Their pesticide selections were m

Pesticide-Induced Inflammation at a Glance.

The increasing number of studies reporting the risks of the exposure to pesticides aligned with the intensified use of such hazardous chemicals has emerged as a pressing contemporary issue, notably due to the potential effects to both the environment and human health. Pesticides, while broadly applied in modern agriculture for pest control and crop protection, have raised concerns due to their unintended effects on non-target organisms. The immune system exerts a key role in the protection against the exposome, which could result in cellular imbalances and tissue damage through the inflammator

Carbofuran pesticide toxicity to the eye.

Pesticide exposure to eyes is a major source of ocular morbidities in adults and children all over the world. Carbofuran (CF), N-methyl carbamate, pesticide is most widely used as an insecticide, nematicide, and acaricide in agriculture, forestry, and gardening. Contact or ingestion of carbofuran causes high morbidity and mortality in humans and pets. Pesticides are absorbed in the eye faster than other organs of the body and damage ocular tissues very quickly. Carbofuran exposure to eye causes blurred vision, pain, loss of coordination, anti-cholinesterase activities, weakness, sweating, naus

Pesticide Toxicity Associated with Infertility.

Pesticides have benefited mankind in many ways like agriculture, industrial and health sectors. On the other hand, conversely their deleterious effects in both, humans and animals are also alarming. Pesticides including organophosphates, organochlorines, carbamates, pyrethrins and pyrethroids are found sufficiently in the environment resulting in everyday human exposure. This is of a huge concern because most of the pesticides are known to target all the physiological functions of both humans and animals. Indeed, reproduction, being one of the most important physiological processes, that is af

Fungicides: An Overlooked Pesticide Class?

Fungicides are indispensable to global food security and their use is forecasted to intensify. Fungicides can reach aquatic ecosystems and occur in surface water bodies in agricultural catchments throughout the entire growing season due to their frequent, prophylactic application. However, in comparison to herbicides and insecticides, the exposure to and effects of fungicides have received less attention. We provide an overview of the risk of fungicides to aquatic ecosystems covering fungicide exposure (i.e., environmental fate, exposure modeling, and mitigation measures) as well as direct and

The evolutionary origins of pesticide resistance.

Durable crop protection is an essential component of current and future food security. However, the effectiveness of pesticides is threatened by the evolution of resistant pathogens, weeds and insect pests. Pesticides are mostly novel synthetic compounds, and yet target species are often able to evolve resistance soon after a new compound is introduced. Therefore, pesticide resistance provides an interesting case of rapid evolution under strong selective pressures, which can be used to address fundamental questions concerning the evolutionary origins of adaptations to novel conditions. We ask:

Pesticide toxicity: a mechanistic approach.

Pesticides are known for their high persistence and pervasiveness in the environment, and along with products of their biotransformation, they may remain in and interact with the environment and living organisms in multiple ways, according to their nature and chemical structure, dose and targets. In this review, the classifications of pesticides based on their nature, use, physical state, pathophysiological effects, and sources are discussed. The effects of these xenobiotics on the environment, their biotransformation in terms of bioaccumulation are highlighted with special focus on the molecu

Applications of Metabonomics in Pesticide Toxicology.

Metabonomic studies quantitatively measure the small molecule metabolites and their intermediates in the biological samples (serum, urine or tissue extracts) and have gained wide applications in many fields, especially in toxicology. Pesticides are extensively used around the world and pesticide toxicity has become a serious threat to human health. Metabonomic approach has been applied in many aspects of pesticide toxicology research such as eco-environmental toxicity studies, biomarker identification, and mechanism of toxicity studies. Both whole organism animal models and cell culture models

Curious about pesticide action.

The safe and effective use of pesticides requires knowledge of their mode of action in pests and adverse effects in nontarget organisms coupled with an understanding of their metabolic activation and detoxification. The author and his laboratory colleagues were privileged to observe, participate in, and sometimes influence these developments for the past six decades. This review considers contributions of the Berkeley and Madison laboratories to understanding insecticides acting at voltage-gated sodium and GABA-gated chloride channels and the nicotinic receptor and at serine hydrolases and oth

Pesticides and Parkinson's disease--is there a link?

Parkinson's disease (PD) is an idiopathic disease of the nervous system characterized by progressive tremor, bradykinesia, rigidity, and postural instability. It has been postulated that exogenous toxicants, including pesticides, might be involved in the etiology of PD. In this article we present a comprehensive review of the published epidemiologic and toxicologic literature and critically evaluate whether a relationship exists between pesticide exposure and PD. From the epidemiologic literature, there does appear to be a relatively consistent relationship between pesticide exposure and PD. T
