The effective treatment of bacterial infections requires rapid and selective enrichment of antibacterial agents on bacterial surfaces within infectious microenvironments. Transition metal ion-based antibacterial agents show broad-spectrum bactericidal activity, but they usually rely on passive diffusion and stochastic bacterial contact, limiting their ability to establish sufficient local doses within short treatment windows. Herein, an M13 phage-mediated targeting strategy was developed for the rapid enrichment and delivery of copper ion-protocatechuic acid nanoparticles (CP NPs). Through pha

Background: Bacteriophages (phages) are viruses that infect and kill bacteria. The antimicrobial resistance crisis has driven renewed interest in phage therapy, including the use of phages to treat chronic orthopaedic infections. Methods: Here, we present the results of the first three orthopaedic patients treated with phage therapy in the United Kingdom. Results: The first patient was treated in May 2023 and received phages active against Staphylococcus aureus. At nine months follow-up, the patient's wound remained healed, the C-reactive protein normal and the patient was walking independentl

Phage therapy has emerged as a promising alternative to conventional antibiotics for combating intestinal bacterial infections, especially in the era of rising antimicrobial resistance. Despite its therapeutic potential, the clinical translation of phage therapy remains hindered by limited large-scale trial data and incomplete mechanistic understanding. This review systematically evaluates the efficacy of phage therapy in animal models of intestinal diseases, encompassing bacterial infection-induced diarrhea (e.g., cholera, typhoid fever), bacterial enteritis, and sepsis. By synthesizing evide

BACKGROUND: Pseudomonas aeruginosa has the ability to exhibit resistance to a broad range of antibiotics, highlighting the importance of identifying alternative or adjunctive treatment options, such as phages.
PATIENTS AND METHODS: We report the case of a 25-year-old male who experienced an accidental electrocution resulting in exposed calvarium in the left parieto-temporal region, complicated by a difficult-to-treat P. aeruginosa (DTR-P. aeruginosa) infection. Cefiderocol was the sole antibiotic with consistent activity against six bacterial isolates obtained from the infected region over a

Ability of a cocktail of five lytic vibriophages to combatting Vibrio cholerae O1 infection in rabbit model was examined. In one group, rabbits were administered 1 × 10(8) plaque forming unit of phage cocktail 6 and 12 h prior to the administration of V. cholerae O1, while in the other group, same procedure was applied 6 and 12 h post infection. It was observed that oral administration of the phage cocktail after oral bacterial challenge lowered the shedding of bacteria significantly (p < 0.01). In contrast phage treatment prior to bacterial challenge had no such effect (p > 0.05). Results sug

The treatment of three patients suffering from chronic bacterial prostatitis who were qualified for an experimental phage therapy protocol managed at the Phage Therapy Unit in Wrocław is described. They had previously been treated unsuccessfully with long-term targeted antibiotics, autovaccines, and laser biostimulation. Rectal application of phage lysates targeted against Enterococcus faecalis cultured from the prostatic fluid gave encouraging results regarding bacterial eradication, abatement of clinical symptoms of prostatitis, and lack of early disease recurrence.

Klebsiella pneumoniae (K. pneumoniae) is a critical ESKAPE pathogen with rapidly escalating resistance to last-line antibiotics, for which phages and their derived endolysins have emerged as promising alternatives. Phages specifically recognize bacterial surface structures, including capsular polysaccharide (CPS) and lipopolysaccharide (LPS), with their depolymerases degrading these barriers and disrupting biofilms to attenuate virulence. In parallel, endolysins directly cleave the peptidoglycan layer to induce rapid bacterial lysis. Animal model studies confirm that both phages and endolysins

Biofilms, a major cause of chronic bacterial infections, present significant treatment challenges due to their protective extracellular matrix that shields bacteria from both antibiotics and host immune defenses. To treat biofilms more effectively, here we discovered a biofilm-binding peptide from a phage library and verified that it selectively bound the polysaccharides on the biofilm. We then engineered M13 phage into trifunctional nanofibers displaying the biofilm-binding peptide at the tip and carrying gold nanoparticles (AuNPs, as photothermal agents) and tetrakis(4-carboxyphenyl) porphyr

AIM: Methicillin-resistant Staphylococcus aureus (MRSA) is a global public health concern owing to its resistance to conventional antibiotics. To overcome this challenge, innovative strategies such as nanotechnology and phage therapy have emerged as promising alternatives to conventional antimicrobial treatments.
METHODS: This review explores the dual approach of using nanoparticles (NPs) and bacteriophages to treat MRSA. NPs, such as silver, gold, and zinc oxide, exhibit antimicrobial effects through mechanisms including membrane disruption, the generation of reactive oxygen species (ROS), a

In the drug-resistance era, phage therapy has received considerable attention from worldwide researchers. Phage therapy has been given much attention in public health but is rarely applied to control plant diseases. Herein, we discuss phage therapy as a biocontrol approach against several plant diseases. The emergence of antibiotic resistance in agriculturally important pathogenic bacteria and the toxic nature of different synthetic compounds used to control microbes has driven researchers to rethink the century-old strategy of phage therapy''. Compared to other treatment strategies, phage the

Bacteriophage therapy is considered one of the most promising tools to control zoonotic bacteria, such as Salmonella, in broiler production. Phages exhibit high specificity for their targeted bacterial hosts, causing minimal disruption to the niche microbiota. However, data on the gut environment's response to phage therapy in poultry are limited. This study investigated the influence of Salmonella phage on host physiology through caecal microbiota and metabolome modulation using high-throughput 16S rRNA gene sequencing and an untargeted metabolomics approach. We employed 24 caecum content sam

A phage PEf771 that specifically infects and lyses pathogenic Enterococcus faecalis YN771 in patients with refractory periapical periodontitis was used to investigate resistance against E. faecalis infection in vitro and in vivo. PEf771 completely lysed YN771 within 3 h, with a multiplicity of infection of 1. Compared with ten routinely used clinical antibiotics, PEf771 demonstrated the highest bacteriostatic effect within 72 h. The antibacterial effect of PEf771 on extracted teeth within 72 h was better than that of conventional root canal disinfectants such as camphorated phenol, formaldehyd

INTRODUCTION: Salmonella enterica serotype Enteritidis (S. Enteritidis) is a cause of food-borne human illness. Given the prevalence of antibiotic resistance of Salmonella Enteritidis and the lack of antibiotic efficacy in future years, its replacement with other agents is necessary. One of the most useful agents is bacteriophages.
METHODS: S. Enteritidis was identified using a multiplex polymerase chain reaction assay. The effective bacteriophages were isolated from hospital wastewater samples. The effects of the bacteriophages were evaluated both in vitro and in vivo.
RESULTS: The phage SE

Phage therapy is a promising antibacterial approach but it has limited efficacy against enteric bacterial infections, such as Salmonella enterica Typhimurium (STm). Here we hypothesized that establishing high concentrations of a pathogen-targeting phage in the gut before infection would prophylactically protect against future infection. To this end, we engineered non-pathogenic Escherichia coli to encode a prophage that produces obligately lytic phage progeny targeting STm while overcoming its host defence systems, including restriction-modification systems. We show that prophylactic gut colon

The increase in multidrug-resistant (MDR) Enterobacter cloacae complex (ECC) infections, particularly those resistant to carbapenems, underscores the urgent need for alternative therapies. Phage therapy, with its specific bactericidal action, offers a promising solution. However, there remains a shortage of well-characterized ECC-targeting phages, and dosing and timing optimization for ECC-specific phage cocktails is largely unexplored. In this study, we isolated and characterized three novel lytic phages with diverse genome sizes and host ranges. Notably, ФEBU8 demonstrated broad-spectrum act

Pulmonary infections of patients with cystic fibrosis (CF) or in intensive care units are frequently caused by the Gram-negative opportunistic pathogen Pseudomonas aeruginosa. Since these bacteria are commonly inherently multidrug-resistant (MDR) and hence, antibiotic treatment options are limited, bacteriophages may provide alternative therapeutic and prophylactic measures in the combat of pneumonia caused by P. aeruginosa. This prompted us to perform a comprehensive literature survey of current knowledge regarding effects of phages applied against pulmonary P. aeruginosa infections. The incl

Patients with cystic fibrosis (CF) are prone to developing life-threatening lung infections with a variety of pathogens that are difficult to eradicate, such as Burkholderia cepacia complex (Bcc), Hemophilus influenzae, Mycobacterium abscessus (Mab), Pseudomonas aeruginosa, and Staphylococcus aureus. These infections still remain an important issue, despite the therapy for CF having considerably improved in recent years. Moreover, prolonged exposure to antibiotics in combination favors the development and spread of multi-resistant bacteria; thus, the development of alternative strategies is cr

Bordetella bronchiseptica is a respiratory animal pathogen that shows growing resistance to commonly used antibiotics, which has necessitated the examination of new antimicrobials, including bacteriophages. In this study, we examined the previously isolated and partially characterized B. bronchiseptica siphoviruses of the genus Vojvodinavirus (LK3, CN1, CN2, FP1 and MW2) for their ability to inhibit bacterial growth and biofilm, and we examined other therapeutically important properties through genomic analysis and lysogeny experiments. The phages inhibited bacterial growth at a low multiplici
