Liquid-liquid phase separation (LLPS) is a fundamental mechanism that drives biomolecular assembly. The concentration of biomolecules within phase-separated condensates is a critical parameter that reflects their physical state, modulates their biological functions, and provides insights into their formation and aging. However, current analytical methods, such as spectroscopy, fluorescence imaging, quantitative phase imaging, and bulk measurements, have limitations in quantifying biomolecule concentrations, involving measurement inaccuracies, sample perturbation, or requirements for large samp

Mutations in the ATRX genes cause alpha-thalassemia X-linked intellectual disability (ATR-X) syndrome. Here, we show that ATRX influences the fate of human neural progenitor cells (hNPCs) by forming condensates through liquid-liquid phase separation (LLPS). The intrinsically disordered region (IDR) of ATRX is essential for LLPS and enables ATRX to form dynamic condensates that recruit co-activators. These condensates are necessary for ATRX localization at super-enhancers (SEs) in hNPCs, linking its compartmentalization to transcriptional regulation. Disruption of ATRX condensates alters gene e

Although it is now recognized that specific RNAs and protein families are critical for the biogenesis of ribonucleoprotein (RNP) condensates, how these molecular constituents determine condensate size and morphology is unknown. To circumvent the biochemical complexity of endogenous RNP condensates, the use of programmable tools to reconstitute condensate formation with minimal constituents can be instrumental. Here we report a methodology to form RNA-containing condensates in living cells programmed to specifically recruit a single RNA species. Our bioengineered condensates are made of ArtiGra

Sulfolane is explored as a novel stationary phase for use in analytical separations by capillary column gas chromatography with flame ionization detection (GC-FID). Stainless steel capillaries were found to provide a good substrate for coating and retaining a sulfolane phase, whereas fused silica tubing did not perform well for this. In general, the phase was found to be stable for several hours of use when using elevated carrier gas pressures (90 psi) and a small restriction (25 μm I.D. tubing) at the outlet. This normally provided good performance at temperatures up to about 200 °C with very

The wings of the black butterfly, Pachliopta aristolochiae, are covered by micro- and nanostructured scales that harvest sunlight over a wide spectral and angular range. Considering that these properties are particularly attractive for photovoltaic applications, we analyze the contribution of these micro- and nanostructures, focusing on the structural disorder observed in the wing scales. In addition to microspectroscopy experiments, we conduct three-dimensional optical simulations of the exact scale structure. On the basis of these results, we design nanostructured thin photovoltaic absorbers

The phase separation behavior of bisphenol-A-polycarbonate (PC), dissolved in N-methyl-2-pyrrolidone and dichloromethane solvents in coagulant water, was studied by the cloud point method. The respective cloud point data were determined by titration against water at room temperature and the characteristic binodal curves for the ternary systems were plotted. Further, the physical properties such as viscosity, refractive index, and density of the solution were measured. The critical polymer concentrations were determined from the viscosity measurements. PC/NMP and PC/DCM membranes were fabricate

Immobilized polysaccharide chiral stationary phases (CSPs) are a new development in chiral chromatography with increasing importance for the resolution of racemic compounds. These CSPs can be used with solvents traditionally applied for the classical coated polysaccharide CSPs as well as solvents that were "forbidden" for the coated phases such as tetrahydrofuran, chloroform, dichloromethane, acetone, 1,4-dioxane, ethylacetate, and certain ethers. However, these forbidden solvents may be required for the determination of the chiral recognition mechanism. Therefore, the immobilized CSPs are eff

Liquid-liquid phase separation (LLPS) is an emerging biophysical principle that governs subcellular organization through the formation of dynamic, membraneless biomolecular condensates. This review aims to elucidate the multifaceted mechanisms by which dysregulated LLPS drives cancer drug resistance and to explore therapeutic strategies targeting oncogenic biomolecular condensates for improved anticancer outcomes. We synthesize evidence demonstrating that dysregulated LLPS drives cancer drug resistance through diverse mechanisms, including sustaining oncogenic transcription despite targeted th

The transcriptional regulation of cell fate plays a central role in eukaryotic cell differentiation. In the human fungal pathogen Candida albicans, the white-to-opaque cell fate switch is controlled by an interconnected network of eight transcription factors (TFs). These include Ssn6, a negative regulator of the opaque state that can function with its co-repressor Tup1; together, they form a global repressor complex that is conserved from yeast to mammals. Here, we evaluated the roles of four Ssn6 domains (N, TPR, M, and CTD) in white-opaque switching. Loss of the prion-like N or M domains had

IFI16 is an interferon-inducible protein that senses viral DNA and can also restrict RNA virus replication. Here, using IFI16 knockout cellular models, we identify IFI16 as a host restriction factor that limits SARS-CoV-2 replication. Upon SARS-CoV-2 infection, IFI16 relocalizes from the nucleus to the cytoplasm, where it binds both the nucleocapsid protein and the viral genome. This impairs SARS-CoV-2 replication by inhibiting viral RNA-induced condensate formation of the nucleocapsid protein. Finally, we extend our analysis to other human coronaviruses and observe that IFI16 depletion also e

Stress granules (SGs) are biomolecular condensates that form transiently in the cytosol of mammalian cells in response to stress. Dysregulation of their assembly or disassembly is implicated in human age-related diseases. While phase separation is the key process underlying SG assembly, understanding of their function, composition and regulation in response to physiological stimuli is limited. This knowledge gap reflects the challenge of gaining comprehensive and quantitative insights into the dynamic regulation of the complex composition of SGs at the single-cell level. Here we present an emu

Food-derived bioactive peptides (FDBPs), released from dietary proteins during processing, fermentation, or digestion, exhibit diverse physiological activities that are not fully explained by classical receptor- or enzyme-centered models. Depending on their amino acid composition, physicochemical properties, and assembly state, some FDBPs may interact directly with cellular membranes and modulate lipid organization. Membrane phase separation provides an emerging biophysical framework for interpreting the actions of FDBPs. This review presents a prospective analysis of FDBP-membrane phase inter

Nuclear enriched abundant transcript 1 (NEAT1) has two isoforms that serve as core structural components of paraspeckles. Although the regulatory functions of NEAT1 as a noncoding RNA are well established, it has remained unclear whether the short polyadenylated isoform, NEAT1_1, encodes a functional microprotein. Herein, we identify NEAT1_1 encodes a microprotein, which we name PSNEAT1 (phase-separating NEAT1 microprotein). PSNEAT1 localizes to the fibrillar center of nucleolus, where it enhances ribosomal DNA (rDNA) transcription, ribosome biogenesis, and tumor growth. Notably, depletion of

This study demonstrates the sustainable conversion of agricultural waste into high-performance fluorescent epoxy resins through direct blending of coumaric acid epoxy (CE) and ferulic acid epoxy (FE) derived from corn stover. These bio-based resins exhibit tunable photoluminescence, with emission maxima shifting from 299 nm to 841 nm as FE content increases from 0 to 33 wt%. In contrast, blends based on commercial bisphenol A diglycidyl ether (BADGE) reach emission saturation around 550 nm and exhibit phase separation at higher FE concentrations, as confirmed by light microscopy. The synthesiz

The impact of missense genetic variations on protein function is often enigmatic, especially for mutations that map to intrinsically disordered regions (IDRs). Given the functional importance of phase separation of IDRs, it has been proposed that mutations that modulate phase separation might preferentially lead to disease. To examine this idea, we used the robust predictability of phase-separating (PS) IDRs and annotation of disease-associated proteins and mutations to map the correlation between disease and phase separation. Consistent with previous work linking phase separation to cancer an

Cell fate decisions in eukaryotes are regulated by interconnected networks of transcription factors (TFs) that drive heritable changes in identity. However, much is unknown about how TFs act together to control cell fate, despite links to cellular dysfunction and disease when TF function is aberrant. Here, we addressed the interplay between TFs that control heritable switching in the diploid fungal pathogen Candida albicans. This species can propagate in two distinct cell states, white and opaque, with epigenetic transitions between states regulated by a core network of eight TFs plus >100 aux

Enriching trace biomarkers (e.g., proteins, nucleic acids) is critical for biomedical applications; yet conventional methods often lack versatility, limiting their effectiveness to specific biomarker types. To address this, the phase separation-assisted pre-enrichment (PSAP) technology is presented that exploits differential polymer-polymer partitioning to achieve 47-fold antigen and 44-fold RNA enrichment simultaneously. Through systematic optimization of interfacial chemistry, including pH modulation, polymer hydrophilicity, mass fraction, and molecular weights, the protocol is refined to en

The rapid advancement of synthetic biology has enabled the construction of artificial cells that closely mimic the morphology and functionality of their natural counterparts. However, significant limitations remain in engineering artificial cells capable of regulated protein expression. Here, we demonstrate that engineered polymers containing multivalent association motifs can reversibly regulate translational activity through liquid-liquid phase separation (LLPS)-induced protein aggregation, enabling precise temporal control of cell-free protein synthesis (CFPS) activity. This aggregation mec

Understanding the impact of liquid-liquid phase separation (LLPS) on drug and biological molecules is key to advancing drug delivery and biomolecular engineering. This study investigates how polyethylene glycol (PEG)-induced LLPS modulates the photophysical behavior of topotecan (TPT) and the structural organization of calf thymus DNA (ctDNA) in binary (PEG:TPT and PEG:ctDNA) and ternary (PEG:TPT:ctDNA) systems. In the PEG:TPT system, LLPS induced a shift from the emissive Z-form to the more stable C-TPT form, with enhanced fluorescence and a longer ∼4.0 ns lifetime, suggesting reduced water a

The methyltransferase complex (MTC) deposits N6-adenosine (m6A) onto RNA, whereas the microprocessor produces microRNA. Whether and how these two distinct complexes cross-regulate each other has been poorly studied. Here we report that the MTC subunit B tends to form insoluble condensates with poor activity, with its level monitored by the 20S proteasome. Conversely, the microprocessor component SERRATE (SE) forms liquid-like condensates, which in turn promote the solubility and stability of the MTC subunit B, leading to increased MTC activity. Consistently, the hypomorphic lines expressing SE
