Further investigation of the alpha-adrenoceptor-mediated actions of chloroethylclonidine in rat aorta.

We have investigated the interaction between chloroethylclonidine and alpha-adrenoceptors in rat aorta. Chloroethylclonidine has two actions on rat aorta: reduction of the contraction to low concentrations of noradrenaline by alpha1-adrenoceptor antagonism and irreversible partial agonism in combination with high concentrations of noradrenaline. The former antagonist action was found to be more marked in vessels from immature rats (1 month). We have examined further the latter agonist actions in adult rats (3 month). In the absence of chloroethylclonidine, exposure to phenoxybenzamine (10 micr

Changes in calcitonin release during sympathetic nerve degeneration after superior cervical ganglionectomy of rats.

To assess the role of peripheral sympathetic nerves in the regulation of calcitonin release, rats subjected to superior cervical ganglionectomy (SCGx) 16-28 h earlier were used. The time periods selected allowed us to examine C cell response during the supraliminal release of sympathetic transmitter that accompanies anterograde degeneration of nerve varicosities as well as during the neural paralysis that ensues thereafter. At the time intervals examined, SCGx did not result in significant changes of basal serum calcitonin or Ca levels. The intraperitoneal administration of CaCl2 brought about

Opioidergic, dopaminergic and adrenergic regulation of LH secretion in prepubertal heifers.

Studies have shown inhibitory effects of endogenous opioids on LH secretion in early post-natal heifers. However, it is not clear whether these effects change during the rest of the prepubertal period or whether the inhibitory influences on the GnRH neurones are direct or by way of other neuronal systems. Two experiments were performed in heifer calves to study the developmental patterns of opioidergic, dopaminergic and adrenergic regulation of LH and the possible interactions between opioids and dopaminergic and adrenergic neuronal systems, in the regulation of LH secretion. In Expt 1 four gr

Sulfhydryl reagents have different effects on substance P and neurokinin B binding sites on cortical synaptosomes in the rat.

The effects of several protein modifying reagents, including phenoxybenzamine, N-ethylmaleimide (NEM), 5,5'-dithiobis (2-nitro)--benzoic acid (DTNB), p-chloromercuryphenyl sulfonic acid (PCMP) and p-bromophenacylbromide (PBPB), on the binding of 125I-Bolton Hunter substance P (125I-BHSP), 125I-Bolton Hunter eledoisin (125I-BHELE) and 3H-neurokinin B (3H-NKB)4 to cortical synaptosomes were examined. PCMP (10(-4) M), DTNB (10(-4) M) and NEM (10(-3) M) were without effect on the 125I-BHSP binding but reduced markedly the specific binding of 125I-BHELE or 3H-NKB. Although phenoxybenzamine and PBPB

The role of adrenergic receptors in the regulation of gastric motility in the rat.

Gastric motility was investigated under the effect of adrenergic receptor agonist and antagonist substances in albino rats of both sexes and weighing 150 to 180 g. Gastric evacuation was studied, after introducing 2 ml/100 g of Novobarium suspension in the stomach, by radioscopy and radiography. Epinephrine (Tonogen) was found to inhibit gastric motility, this inhibition being effective even after pylorotomy. The dominantly alpha adrenergic agonist phenylephrine (Sympathomim) had not any effect; the alpha blocker phenoxybenzamine (Dibenzyline) slightly depressed gastric evaluation. A marked in

Methyldopa produces central inhibition of parasympathetic activity in the cat.

alpha-Methyldopa (10-100 mg/kg i.v.) produced a dose-dependent pupillary dilation in anaesthetized cats which was antagonized by subsequent administration of yohimbine hydrochloride (0.5 mg/kg i.v.). The peak effects were observed approximately 2-3 h after injection. This alpha-methyldopa-induced mydriasis was present only when the parasympathetic innervation to the iris was intact. Prior treatment with yohimbine (0.5 mg/kg i.v.) 30 min before alpha-methyldopa also antagonized the mydriatic effect, whereas pretreatment with phenoxybenzamine (2.5 mg/kg i.v.) did not. In contrast, phenoxybenzami

Contractor effect of barbiturates on smooth muscle.

Four different convulsant barbiturates and also thiopental, methohexital and pentobarbital were tested for in vitro contractor effects on rabbit jejunal segments, aortic strips and tracheal chains. The two convulsants that elicited lethal, tonic seizures in vivo contracted all three types of isolated preparation; however, two other convulsants that elicited intense but nonlethal, clonic seizures lacked contractor effect. Methohexital and pentobarbital also lacked contractor effect. In aortic strips, contractions induced by the two convulsants were not impaired by concentrations of phenoxybenza

Persistence of sympathetic-mediated forearm vasoconstriction after alpha-blockade in hypertensive patients.

Sympathetic vasoconstriction not mediated by alpha-adrenoceptors has been identified in vitro and in animals but not in humans. We evaluated the effect of alpha-adrenoceptor blockade on either endogenous vascular sympathetic activation (obtained through the application of a nonhypotensive lower-body negative pressure, -10 mm Hg for 5 minutes) or selective postsynaptic alpha-adrenoceptor stimulation by exogenous norepinephrine (0.005 micrograms/100 ml forearm tissue/min for 3 minutes) in the presence of beta-blockade by propranolol (10 micrograms/100 ml forearm tissue/min for 15 minutes). Immune cell-specific genetic architecture of Alzheimer's disease revealed by multi-omics analysis for therapeutic target discovery and prioritization.

Alzheimer's disease (AD) is a multifactorial neurodegenerative condition in which accumulating genetic and molecular evidence implicates dysregulation of peripheral immune processes in disease pathogenesis. Nevertheless, the contribution of distinct peripheral immune cell subsets and associated gene regulatory landscapes to AD risk remains incompletely defined. To address this gap, we integrated single-cell expression quantitative trait loci (sc‑eQTL) data from the OneK1K cohort with AD GWAS summary statistics. We systematically interrogated immune cell-specific genes for their contributions t

A comparative study of postsynaptic alpha2-adrenoceptors of the dog mesenteric and rat femoral veins.

The aim of the present study was to compare the subtype of postjunctional alpha2-adrenoceptors of canine mesenteric and rat femoral veins. To check whether the presence of alpha1-adrenoceptors in both tissues might interfere with alpha2-adrenoceptor-mediated effects, the experiments were carried out under two experimental conditions: without and with alpha1-adrenoceptor blockade. The selective and irreversible alpha1-adrenoceptor antagonist phenoxybenzamine (30 nM) was used to eliminate alpha1-adrenoceptors. The pA2 values for the antagonism exerted by eight alpha-adrenoceptor antagonists (rau

Sexual differences in hypothalamic adenosine 3',5'-cyclic monophosphate content.

Hypothalamic adenosine 3',5'-cyclic monophosphate (cAMP) concentration was measured in the morning (10:00) and afternoon (16:00) in castrated and intact rats of both sexes to evaluate a possible sexual difference. Castration produced different effects on the hypothalamic cAMP concentration. In females, ovariectomy significantly reduced total hypothalamic cAMP concentration, whereas in males, orchidectomy elevated hypothalamic cAMP content. This observation was independent of the time of day at which the animals were killed. On the other hand, administration of the alpha-adrenoreceptor blocking

Non-adrenergic sympathetic vasoconstriction in the eye and some other facial tissues in the rabbit.

The effects of unilateral sympathetic nerve stimulation (SNS) on regional blood flow in the rabbit were studied with radioactive microspheres. SNS at 10 or 4 Hz caused an approximately 60% reduction in choroidal blood flow, which was partly resistant to alpha-adrenoceptor blockade with phenoxybenzamine. The vasoconstriction evoked by SNS at 2 Hz was completely abolished by alpha-adrenoceptor blockade. A similar response was seen in the iris, ciliary body, masseter muscle and lacrimal gland. In the harderian gland, however, SNS (2 Hz) after alpha-adrenoceptor blockade caused a significant reduc

Renin release by pentobarbital anesthesia in the rat: a role for vascular mechanisms.

Studies were undertaken in intact rats to characterize the renin response to pentobarbital anesthesia and the mechanisms involved in this response. Aortic and peritoneal cavity cannulas were previously implanted to allow drug infusion, blood sampling and anesthesia to be performed without stress. A sustained 2-3-fold increase in plasma renin concentration (PRC) and a 10-15 mm Hg depression of mean arterial pressure were found in pentobarbital anesthesia. Circulating levels of epinephrine and norepinephrine were unchanged. Sympathetic stimulation by tyramine did not decrease and chronic renal d

alpha-Adrenoceptor blocking properties of a new antihypertensive agent, 2-[4-(n-butyryl)-homopiperazine-1-yl]-4-amino-6,7-dimethoxyquinazoline (E-643).

Postsynaptic alpha-receptor blocking properties of E-643 were studied in vivo and in vitro and compared with these same properties of phentolamine and phenoxybenzamine. In anesthetized rats, E-643 (i.v.) attenuated pressor response to adrenaline dose-dependently and an adrenaline-reversal was seen with large doses. The in vivo alpha-adrenoreceptor blocking effect of E-643 was 3.4 times more potent than that of phentolamine. On the other hand, hypotensive action of E-643 was 9.4 times more potent than that of phentolamine. In the isolated rabbit aorta, E-643 blocked noradrenaline-induced contra

Adenosine causes dilatation and constriction of hypothalamic blood vessels.

The effect of adenosine on intraparenchymal cerebral blood flow was examined in conscious rabbits with the 133Xe clearance technique. Perivascular application of 10(-3) and 10(-4) M adenosine to hypothalamic blood vessels increased hypothalamic blood flow by approximately 50% (p less than 0.005). This vasodilatation was attenuated by the intrahypothalamic injection of the beta-adrenergic receptor antagonist propranolol, but was unaffected by alpha-adrenoreceptor blockade with phenoxybenzamine, or depression of neuronal activity with barbiturate. 2-Chloroadenosine, a stable analogue of adenosin

Hypertension in Pheochromocytoma and Paraganglioma: Evaluation and Management in Pediatric Patients.

PURPOSE OF REVIEW: The rare catecholamine-secreting tumors, pheochromocytomas and paragangliomas (PPGL), account for a minority of cases of secondary hypertension in pediatrics. As such, perioperative blood pressure (BP) management in pediatric patients presents a distinct challenge. This review will expand the practitioner's knowledge of antihypertensive treatment options for the pediatric patient with PPGL with a focus on literature in the past several years. RECENT FINDINGS: There continue to be only small case series and single-center experiences to provide guidelines regarding BP managem

[Pheochromocytoma: blood volume and hemodynamics].

In 12 patients with phaeochromocytoma total blood volume (TBV) prior to and after treatment with phenoxybenzamine and the haemodynamic changes during surgery were determined. Our results were analysed with regard to the standard of our anaesthesiological management. The mean TBV was reduced in 6 of 12 patients. Great individual variations were found after treatment with phenoxybenzamine. In 6 patients alpha-blockade induced an increase of TBV (+ 10%), 2 patients showed no changes and 4 a decrease of TBV (-18%). Haemodynamic measurements prior to induction of anaesthesia revealed a reversal of

Inhibition of post-ganglionic motor transmission in vas deferens by indirectly acting sympathomimetic drugs.

1. Using field stimulation with short trains of pulses (< 10 per train), the post-ganglionic motor transmission in the mammalian vas deferens has been further analysed pharmacologically.2. In preparations taken from guinea-pigs, rats and rabbits the effects of the indirectly sympathomimetic drugs, tyramine and cocaine, could be explained entirely on the basis of the actions of released, endogenous noradrenaline.3. Tyramine produced a contraction in vasa taken from normal rats but not from normal guinea-pigs. The tyramine contraction was due to release of endogenous noradrenaline because it was

Multiple paragangliomas in a pregnant patient with a succinate dehydrogenase B mutation.

In this case report, we document the clinical course, laboratory findings, and imaging findings of a 21-year-old pregnant woman with multiple paragangliomas due to a succinate dehydrogenase B (SDHB) mutation. We also review the literature on previously reported cases. The patient presented with nausea, palpitations, angina, and amenorrhea. Her blood pressure was 170/100 mm Hg and her beta-human chorionic gonadotropin was positive. Her blood pressure remained high despite phenoxybenzamine titration. A 24-hour urine analysis revealed elevated plasma metanephrines. Imaging was initially deferred
