Nocturnal light exposure impairs affective responses in a wavelength-dependent manner.

Life on earth is entrained to a 24 h solar cycle that synchronizes circadian rhythms in physiology and behavior; light is the most potent entraining cue. In mammals, light is detected by (1) rods and cones, which mediate visual function, and (2) intrinsically photosensitive retinal ganglion cells (ipRGCs), which primarily project to the suprachiasmatic nucleus (SCN) in the hypothalamus to regulate circadian rhythms. Recent evidence, however, demonstrates that ipRGCs also project to limbic brain regions, suggesting that, through this pathway, light may have a role in cognition and mood. Therefo

The dietary flavonoids naringenin and quercetin acutely impair glucose metabolism in rodents possibly via inhibition of hypothalamic insulin signalling.

Secondary metabolites of herbs and spices are widely used as an alternative strategy in the therapy of various diseases. The polyphenols naringenin, quercetin and curcumin have been characterised as anti-diabetic agents. Conversely, in vitro, naringenin and quercetin are described to inhibit phosphoinositide-3-kinase (PI3K), an enzyme that is essential for the neuronal control of whole body glucose homoeostasis. Using both in vitro and in vivo experiments, we tested whether the inhibitory effect on PI3K occurs in neurons and if it might affect whole body glucose homoeostasis. Quercetin was fou

Appetite regulation and seasonality: implications for obesity.

High circulating concentrations of leptin in obesity are associated with an apparent loss of its characteristic anorexic action within the hypothalamic region of the brain. Central insensitivity to leptin may therefore contribute to the aetiology of this disease, and an increased understanding of the underlying mechanisms will identify potential means of prevention and/or therapeutic targets. Seasonal animals such as sheep and Siberian hamsters (Phodopus sungorus) exhibit annual photoperiod-driven cycles of appetite and body weight. Increased food intake and weight gain in long days (summer) a

The effects of dietary cholesterol on metabolism and daily torpor patterns in Siberian hamsters.

The concentrations of fatty acids in the diet influence torpor in numerous species of mammals. Much less is known, however, about the potential effects of other types of dietary lipids. One study demonstrated that increasing dietary cholesterol levels during fall feeding increased torpor bout length and also decreased minimum body temperatures during hibernation by chipmunks. Another hibernation study with ground squirrels revealed that the cholesterol contents of both the cerebral cortex and the microsomes were significantly greater during arousal episodes than during torpor bouts, suggesting

Co-localization of serine/threonine kinase 33 (Stk33) and vimentin in the hypothalamus.

We investigate the immunoreactivity of serine/threonine kinase 33 (Stk33) and of vimentin in the brain of mouse, rat and hamster. Using a Stk33-specific polyclonal antibody, we show by immunofluorescence staining that Stk33 is present in a variety of brain regions. We found a strong staining in the ependymal lining of all cerebral ventricles and the central canal of the spinal cord as well as in hypothalamic tanycytes. Stk33 immunoreactivity was also found in circumventricular organs such as the area postrema, subfornical organ and pituitary and pineal glands. Double-immunostaining experiments

Exogenous T₃ elicits long day-like alterations in testis size and the RFamides Kisspeptin and gonadotropin-inhibitory hormone in short-day Siberian hamsters.

Siberian hamsters (Phodopus sungorus) exhibit robust seasonal rhythms of reproduction driven by changes in day length. Day length is encoded endogenously by the duration of nocturnal melatonin (Mel) secretion from the pineal gland. Short duration Mel signals stimulate whereas long duration Mel signals inhibit reproduction. The mechanism by which Mel regulates the reproductive axis has not been fully characterized. In Siberian hamsters, the thyroid hormone triiodothyronine (T₃) is thought to be part of the photoperiodic mechanism. The availability of T₃ is decreased in hamsters housed in short

Melatonin mediates seasonal transitions in aggressive behavior and circulating androgen profiles in male Siberian hamsters.

Some seasonally-breeding animals are more aggressive during the short, "winter-like" days (SD) of the non-breeding season, despite gonadal regression and reduced circulating androgen levels. While the mechanisms underlying SD increases in aggression are not well understood, previous work from our lab suggests that pineal melatonin (MEL) and the adrenal androgen dehydroepiandrosterone (DHEA) are important in facilitating non-breeding aggression in Siberian hamsters (Phodopus sungorus). To characterize the role of MEL in modulating seasonal transitions in aggressive behavior, we housed male hams

Chronic citalopram treatment ameliorates depressive behavior associated with light at night.

Chronic exposure to light at night (LAN) is a circadian disruptor and may be linked to various health risks, including mood disorders. We recently demonstrated that chronic exposure to dim (5 lux) LAN provokes depressive-like behaviors and reduced hippocampal CA1 dendritic spine density in female hamsters. Whether this model is responsive to selective serotonin reuptake inhibitors remains unspecified. In this study, we exposed hamsters to 5 lux LAN and treated with citalopram to determine effects on depressive-like behavior and CA1 dendritic spine density. Female hamsters were ovariectomized a

HIOMT drives the photoperiodic changes in the amplitude of the melatonin peak of the Siberian hamster.

In the pineal, melatonin (Mel) is synthesized from serotonin by arylalkylamine-N-acetyltransferase (AA-NAT) and hydroxyindole-O-methyltransferase (HIOMT). Although it is clear that AA-NAT drives the daily rhythm in Mel synthesis, the mechanisms involved in the photoperiodic changes of the amplitude of the Mel peak, as observed in the Siberian hamster, remain to be determined. We investigated the characteristics of AA-NAT and HIOMT in Siberian hamsters kept either under a short (SP) or a long photoperiod (LP). The amplitude of the nocturnal peak of Mel was about two times higher under SP than u

Melatonin synthesis in the retina and pineal gland of Djungarian hamsters at different times of the year.

Daily profiles of melatonin content in the retina of Djungarian hamsters were measured throughout the course of a year. Peak levels of retinal melatonin were found at various times of the day at different seasons. In a subsequent study we determined concurrently daily profiles of nine indole metabolites (tryptophan, 5-hydroxytryptophan, 5-hydroxytryptamin, 5-hydroxyindoleacetic acid, 5-hydroxytryptophol, N-acetylserotonin, melatonin, 5-methoxyindoleacetic acid and 5-methoxytryptophol) in samples of hamster retina and pineal gland at the four different seasons. Perinatal influences of melatonin on testicular development and photoperiodic memory in Siberian hamsters.

We assessed the influence of perinatal melatonin on reproductive development and adult responsiveness to melatonin. Testicular growth in an intermediate day length (14 : 10 h light/dark cycle) was substantially reduced in Siberian hamsters gestated by pinealectomised compared to pineal-intact females; gonadal development was normalised in offspring of pinealectomised dams that were pinealectomised at 3-4 days of age. Hamsters deprived of melatonin only during gestation, or both pre- and postnatally, underwent testicular involution during treatment with melatonin in adulthood. Photoperiodic his

Independence of circadian entrainment state and responses to melatonin in male Siberian hamsters.

BACKGROUND: Seasonal fluctuations in physiology and behavior depend on the duration of nocturnal melatonin secretion programmed by the circadian system. A melatonin signal of a given duration, however, can elicit different responses depending on whether an animal was previously exposed to longer or shorter photoperiod signals (i.e., its photoperiodic history). This report examined in male Siberian hamsters which of two aspects of photoperiod history--prior melatonin exposure or entrainment state of the circadian system - is critical for generating contingent responses to a common photoperiodic

Monosodium glutamate-induced arcuate nucleus damage affects both natural torpor and 2DG-induced torpor-like hypothermia in Siberian hamsters.

Siberian hamsters (Phodopus sungorus) have the ability to express daily torpor and decrease their body temperature to approximately 15 degrees C, providing a significant savings in energy expenditure. Daily torpor in hamsters is cued by winterlike photoperiods and occurs coincident with the annual nadirs in body fat reserves and chronic leptin concentrations. To better understand the neural mechanisms underlying torpor, Siberian hamster pups were postnatally treated with saline or MSG to ablate arcuate nucleus neurons that likely possess leptin receptors. Body temperature was studied telemetri

Sleep regulation in the Djungarian hamster: comparison of the dynamics leading to the slow-wave activity increase after sleep deprivation and daily torpor.

STUDY OBJECTIVES: Emerging from daily torpor, Djungarian hamsters (Phodopus sungorus) show an initial increase in electroencephalographic slow-wave activity (power density between 0.75 and 4.0 Hz) during sleep that gradually declines. This feature is typical for sleep following prolonged waking and supports the hypothesis that sleep pressure increases during daily torpor. After hamsters were subjected to sleep deprivation or partial non-rapid eye movement sleep deprivation immediately following torpor, slow-wave activity remained high and decreased only when sleep was allowed. An analysis of t

Siberian hamster: a new indoor source of allergic sensitization and respiratory disease.

BACKGROUND: The identification of 'Нет данных' allergic sensitizations may determine the prognosis and treatment of patients with respiratory airway disease. Currently, the presence in homes of 'exotic' animals as pets is increasing. In this article the Siberian hamster or dwarf hamster (Phodopus sungorus) was identified as a new indoor source of aeroallergens and respiratory disease. METHODS: The subjects were six outpatients who were treated for asthma and rhinitis. Siberian hamster hair extract was prepared with a standard wt/vol method, and patients were skin-prick tested with the extract. S

Neural network-assisted humanisation of COVID-19 hamster transcriptomic data reveals matching severity states in human disease.

BACKGROUND: Translating findings from animal models to human disease is essential for dissecting disease mechanisms, developing and testing precise therapeutic strategies. The coronavirus disease 2019 (COVID-19) pandemic has highlighted this need, particularly for models showing disease severity-dependent immune responses. METHODS: Single-cell transcriptomics (scRNAseq) is well poised to reveal similarities and differences between species at the molecular and cellular level with unprecedented resolution. However, computational methods enabling detailed matching are still scarce. Here, we prov

Autologous fat transplants influence compensatory white adipose tissue mass increases after lipectomy.

Direct tests of the hypothesized total body fat regulatory system have been accomplished by partial surgical lipectomy. This usually results in the restoration of the lipid deficit through compensatory increases in nonexcised white adipose tissue (WAT) masses of ground squirrels, laboratory rats, and mice, as well as Siberian and Syrian hamsters. We challenged this hypothesized total body fat regulatory system by testing the response of Siberian hamsters to 1) lipid deficits [lipectomy; primarily bilateral epididymal WAT (EWAT) removal], 2) lipid surfeits (addition of donor EWAT with no lipect

Prolonged immune activation in post-acute sequelae of SARS-CoV-2: neutrophil dynamics and therapeutic insights.

Post-acute sequelae of SARS-CoV-2 (PASC) is characterized by persistent symptoms such as fatigue, respiratory complications and cognitive dysfunction, affecting approximately 13.5% of SARS-CoV-2-infected individuals. Despite its clinical significance, the mechanisms driving PASC remain poorly understood. Here, to address this, we utilized a Phodopus roborovskii hamster model to investigate the long-term effects of SARS-CoV-2 infection compared with influenza A virus. While 46.25-47.50% of hamsters survived SARS-CoV-2 or influenza A virus H1N1 infection, 13.75% of SARS-CoV-2 survivors exhibited

Time course and role of the pineal gland in photoperiod control of innate immune cell functions in male Siberian hamsters.

The time course of select phagocyte and natural killer activities to short days was determined. In advance of testes regression, circulating granulocyte and monocyte cell numbers in hamsters decreased while lymphocyte numbers increased; phagocytosis and oxidative burst activity also decreased. To determine whether the pineal gland influences these innate immune cell functions, hamsters were exposed to constant light. Photoperiod control of testes weight and basal oxidative burst activity was abolished by treatment with constant light; other phagocyte activities and leukocyte proportions in cir
