Difluoroenol phosphinates as difluoroenolate surrogates: synthesis and applications in defluorination and deoxygenative coupling.

We report defluorinative and deoxygenative functionalization reactions of trifluoromethyl ketones mediated by the phospha-Brook rearrangement, offering a streamlined approach to selectively modifying fluorinated compounds. Trifluoromethyl ketones react with phosphine oxides to undergo a phospha-Brook rearrangement followed by β-fluoride elimination, providing difluoromethyl ketones in good yields. By tuning the reaction conditions, we achieved the selective one-pot synthesis of monofluoromethyl ketones and methyl ketones, demonstrating the method's versatility across a range of fluorine-contai

Electrochemical Halogenation and Etherification of Alcohols Enabled by a Halide-Coupled Phosphine Oxidation.

Phosphine-mediated nucleophilic substitution reactions of alcohol substrates, such as the Appel and Mitsunobu reactions, have found widespread applications in chemical synthesis. However, their reliance on stoichiometric chemical oxidants (e.g., carbon tetrachloride and azodicarboxylate reagents) often results in limited functional group tolerance, environmentally hazardous wastes, and unsatisfactory reaction economy. Herein, we describe a user-friendly electrochemical Appel reaction employing readily available tetrabutylammonium halide salts ( n Bu4N+X-, X = Cl, Br, and I) as the halogen sour

Ruthenium-Catalyzed Enantioselective Propargylic Phosphinylation of Propargylic Alcohols with Phosphine Oxides.

The development of transition metal-catalyzed enantioselective propargylic substitution reactions has gained much progress in recent years, however, no successful example with phosphorus-centered nucleophiles has yet been reported until now. Herein, we report the first successful example of ruthenium-catalyzed enantioselective propargylic substitution reactions of propargylic alcohols with diarylphosphine oxides as phosphorus-centered nucleophiles. This synthetic approach provides a new method to prepare chiral phosphorus-containing organic compounds.

Synthesis and biological evaluation of cyanoaziridine phosphine oxides and phosphonates with antiproliferative activity.

This work reports an efficient diastereoselective synthetic methodology for the preparation of phosphorus substituted cyanoaziridines through the nucleophilic addition of TMSCN, as cyanide source, to the C-N double bond of 2H-azirine derivatives. The aziridine ring, in these novel cyanoaziridines, can be activated by simple N-tosylation or N-acylation. In addition, the cytotoxic effect on cell lines derived from human lung adenocarcinoma (A549) and human embryonic kidney (HEK293) was also screened. N-H and N-Substituted cyanoaziridines showed excellent activity against the A549 cell line in vi

The physiology and toxicology of acute inhalation phosphine poisoning in conscious male rats.

Phosphine (PH3) is a toxidrome-spanning chemical that is widely used as an insecticide and rodenticide. Exposure to PH3 causes a host of target organ and systemic effects, including oxidative stress, cardiopulmonary toxicity, seizure-like activity and overall metabolic disturbance. A custom dynamic inhalation gas exposure system was designed for the whole-body exposure of conscious male Sprague-Dawley rats (250-350 g) to PH3. An integrated plethysmography system was used to collect respiratory parameters in real-time before, during and after PH3 exposure. At several time points post-exposure,

Unusual structural motif in a zwitterionic Fe(II) complex of a tetradentate phosphine.

The reaction of meso-DPPEPM (DPPEPM = bis(diphenylphosphino-ethylphenylphosphino)methane) with one equivalent of FeBr(2) in tetrahydrofuran generates a zwitterionic compound [FeBr(κ(2)-DPPEPM)(κ(3)-DPPEPM-FeBr(3))] (1). Compound 1 exhibits an unusual bonding arrangement with two meso-DPPEPM ligands and one bromide coordinated to a single Fe(II) center. One of the DPPEPM ligands binds to iron in a κ(2) mode via two central phosphorus atoms, leaving the terminal phosphines dangling. The second DPPEPM binds through three phosphines, whereas the fourth one coordinates to the iron center of an exte

Syntheses, and optical, fluorescence, and nonlinear optical characterization of phosphine-substituted terthiophenes.

Earlier studies of phosphine-substituted terthiophenes have demonstrated that some of these materials exhibit nonlinear absorption at 532 nm. However, this wavelength is significantly removed from the linear absorption maxima of the complexes, suggesting that better nonlinear absorption might be observed at wavelengths closer to the linear absorption maxima. To investigate this possibility, a library of compounds has been prepared either by varying the group attached to the nonbonding pair of electrons on the phosphorus atoms of 5,5''-bis(diphenylphosphino)-2,2':5',2''-terthiophene (PT(3)P), o

Dicyclohexyl(2',4',6'-triisopropylbiphenyl-2-yl)phosphine-dichlorophenylborane.

In the crystal structure of the title compound, C(39)H(54)BCl(2)P, the phospho-rus atom is coordinated by a dichloro-phenyl-borane unit. The substituted biphenyl group and the two cyclo-hexyl groups at the phospho-rus atom are arranged in such a way to avoid steric crowding in the mol-ecule as far as possible.

Tertiary arsine-stabilised arsenium salts: syntheses and comparisons with phosphine analogues.

The first tertiary arsine-stabilised arsenium salts, [(L)AsMePh]OTf (L = Ph3As, Me2PhAs, [2-(MeOCH2)C6H4]Ph2As, [2-(MeOCH2)C6H4]Me2As), have been prepared by chloride abstraction from chloromethylphenylarsine with trimethylsilyl triflate in the presence of the arsine. The complexes have been characterised by crystallography and 1H NMR spectroscopy. The chiral cations in the complexes have structures based on the trigonal pyramid in which the arsine is coordinated orthogonally to the prochiral, six-electron MePhAs+ ion that forms the base of the pyramid. The NMR data for the complexes in dichlo

Phosphine-Promoted Ring Opening/Recyclization of Cyclopropyl Ketones to Access Hydrofluorenones.

The ring-reorganizing transformations of activated cyclopropanes are typically achieved under acidic conditions. This Letter describes the first acid-free and Lewis base-mediated cascade ring opening/recyclization of designed cyclopropyl ketones to access tetrahydrofluorenones. We rationally merged the nucleophilic features of phosphines with the electronically biased cyclopropanes to synthesize several new classes of hydrofluorenones. We have also demonstrated the synthetic utility of the products in accessing highly functionalized molecular scaffolds.

The Biological Impact of Some Phosphonic and Phosphinic Acid Derivatives on Human Osteosarcoma.

Osteosarcoma malignancy currently represents a major health problem; therefore, the need for new therapy approaches is of great interest. In this regard, the current study aims to evaluate the anti-neoplastic potential of a newly developed phosphinic acid derivative (2-carboxyethylphenylphosphinic acid) and, subsequently, to outline its pharmaco-toxicological profile by employing two different in vitro human cell cultures (keratinocytes-HaCaT-and osteosarcoma SAOS-2 cells), employing different techniques (MTT assay, cell morphology assessment, LDH assay, Hoechst staining and RT-PCR). Additiona

Enzymatic Synthesis of Biologically Active H-Phosphinic Analogue of α-Ketoglutarate.

Amino acid analogues with a phosphorus-containing moiety replacing the carboxylic group are promising sources of biologically active compounds. The H-phosphinic group, with hydrogen-phosphorus-carbon (H-P-C) bonds and a flattened tetrahedral configuration, is a bioisostere of the carboxylic group. Consequently, amino-H-phosphinic acids undergo substrate-like enzymatic transformations, leading to new biologically active metabolites. Previous studies employing NMR-based metabolomic and proteomic analyses show that in Escherichia coli, α-KG-γ-PH (the distal H-phosphinic analogue of α-ketoglutarat

Sterically Crowded Tris(2-(trimethylsilyl)phenyl)phosphine - Is it Still a Ligand?

Tris(2-(trimethylsilyl)phenyl)phosphine, P(o-TMSC6 H4 )3 , was synthesised and characterised in solution and in the solid state. The large steric bulk prevents most reactions of the phosphorus donor and makes the compound air stable both in the solid state as well as in solution. This shielded phosphine can still undergo three reactions, namely protonation, oxidation to the phosphine oxide under harsh conditions and complexation to AuI , thus forming a complex with linear coordination. Unexpectedly, complexation was unsuccessful with a range of other metal cations. Neither PdII , PtII , ZnII n

Thermoneutral N-H Bond Activation of Ammonia by a Geometrically Constrained Phosphine.

A geometrically constrained phosphine bearing a tridentate NNS pincer ligand is reported. The effect of the geometric constraint on the electronic structure was probed by theoretical calculations and derivatization reactions. Reactions with N-H bonds result in formation of cooperative addition products. The thermochemistry of these transformations is strongly dependent on the substrate, with ammonia activation being thermoneutral. This represents the first example of a molecular compound that reversibly activates ammonia via N-H bond scission in solution upon mild heating.

Phosphine production in anaerobic wastewater treatment under tetracycline antibiotic pressure.

The influence of tetracycline (TC) antibiotics on phosphine (PH3) production in the anaerobic wastewater treatment was studied. A lab-scale anaerobic baffled reactor with three compartments was employed to simulate this process. The reactor was operated in a TC-absence wastewater and 250μg/L TC-presence wastewater for three months after a start-up period, respectively. The responses of pH, oxidation-reduction potential (ORP), chemical oxygen demand (COD), total phosphorus (TP), enzymes activity (dehydrogenase and acid phosphatase), and microbial community were investigated to reveal the effect

On the catalytic hydrodefluorination of fluoroaromatics using nickel complexes: the true role of the phosphine.

Homogeneous catalytic hydrodefluorination (HDF) of fluoroaromatics under thermal conditions was achieved using nickel(0) compounds of the type [(dippe)Ni(η(2)-C6F6-nHn)] where n = 0-2, as the catalytic precursors. These complexes were prepared in situ by reacting the compound [(dippe)Ni(μ-H)]2 with the respective fluoroaromatic substrate. HDF seems to occur homogeneously, as tested by mercury drop experiments, producing the hydrodefluorinated products. However, despite previous findings by other groups, we found that these HDF reactions were actually the result of direct reaction of the alkylp

(R(P))-Menthyl (1-hydroxy-cyclo-hexyl)phenyl-phosphinate.

The title compound, C(22)H(35)O(3)P, features a tetra-hedral P atom bonded to a phenyl ring, a hydroxy-cyclo-hexyl unit and the O atom of a menthyl group. The axial chirality at phospho-rus is R(P). In the crystal, mol-ecules are connected through O-H⋯O hydrogen bonds involving the hydr-oxy and P=O groups, forming chains along the 2(1) screw axis. The methyl groups of the isopropyl fragment in the menthyl unit are disordered over two sites of equal occupancy.

Gold(I) phosphine mediated selective inhibition of lymphoid tyrosine phosphatase.

Selective protein tyrosine phosphatase (PTP) inhibition is often difficult to achieve owing to the high degree of similarity of the catalytic domains of this family of enzymes. Selective inhibitors of the lymphoid specific tyrosine phosphatase, LYP, are of great interest due to the involvement of LYP in several autoimmune disorders. This manuscript describes a study into the mechanistic details of selective LYP inhibition by a Au(I)-phosphine complex. The complex, [Au((CH(2)CH(2)CN)(2)PPh)Cl], selectively inhibits LYP activity both in vitro and in cells, but does not inhibit other T-cell deriv
