[Mechanism of inhibition by local anesthetics of electron transport at the donor site of the photosystem II].

The mechanism of inhibition by local anaesthetics of the procaine group of electron transport at the donor site of photosystem II (PS II) from pea chloroplasts was investigated. It was found that besides the inactivation of the O2 release system the anaesthetics used at one order of magnitude lesser concentration exert an uncoupling effect. With a rise in pH the inhibiting activity increases; however, this process is not coupled with the protonophore effect but is due to the generation of a neutral form of the amine. The increment of the inhibiting activity of the anaesthetics in the course of

Association of the 17-kDa extrinsic protein with photosystem II in higher plants.

The structural association of the spinach 17-kDa extrinsic protein of photosystem II with other extrinsic and membrane-bound components of the photosystem was investigated by labeling the 17-kDa extrinsic protein with the amino-group-specific reagent N-hydroxysuccinimidobiotin both on intact photosystem II membranes or as a free protein in solution. After isolation of the biotinylated molecules, the modified 17-kDa proteins were allowed to rebind to photosystem II membranes which were depleted of the 17-kDa component. Differential binding of the protein biotinylated in solution compared to unm

Exposure of Synechocystis 6803 cells to series of single turnover flashes increases the psbA transcript level by activating transcription and down-regulating psbA mRNA degradation.

Exposure of Synechocystis sp. PCC 6803 cells to series of single turnover flashes increases specifically the level of psbA and psbD2 messages, encoding the D1 and D2 proteins of photosystem II, as compared to light exposed cells. This increase is due to maintenance the transcription rate as high as in growth light and to the down-regulation of transcript degradation as in darkness. Inhibition of the plastoquinone pool reduction by DCMU or its oxidation by DBMIB does not diminish the transcription of the psbA gene under growth conditions. However, the degradation rate of psbA transcript, as wel

Drought stress-induced changes in PSII functioning in ecologically contrasting plants: chlorophyll fluorescence study of poikilohydric and homoiohydric species.

Dehydration reduces photosynthetic performance in leaves and other photosynthetic organs in response to drought. This decline is linked to changes in chlorophyll fluorescence, which reflects reduced photosystem II activity and increased protective non-photochemical quenching (NPQ). This study examined NPQ dynamics during desiccation across diverse species, including lichens, mosses, conifers, crops, and C3 and C4 plants, using the method of NPQ induction and relaxation curves. Gradual dehydration from a full-hydration state (RWC 100%) to complete dryness (RWC 0%) was applied in order to compar

Tracking photoinhibition and failure dynamics in algae under combined exposure to UVB and water-accommodated oil fractions.

Increasing UV radiation and persistent petroleum-derived contaminants in aquatic environments have raised concerns regarding their combined effects on primary producers. Microalgae, such as Chlorella vulgaris, are particularly vulnerable owing to their surface-dwelling nature and their high sensitivity to impairment of photosystem II (PSII). The hazards associated with photosynthetic failure under ultraviolet (UV) stress in the presence of oil-derived mixtures remain inadequately quantified, particularly under environmentally realistic light spectra. This study addressed this knowledge gap by

Dimethoate delimits both the donor and acceptor side functions of photosystem II of the soil cyanobacterium Anabaena sp. PCC 7119.

The non-target toxicity of organophosphorus (OP) insecticides has affected the distribution and metabolic performances of native microbial communities resulting in loss of soil productivity. Dimethoate (DM) and other OP insecticides are known to decrease physiological performances and change the biochemical composition of cyanobacteria but detailed evaluation of the impact of DM on the photosystem II (PS II), specifically on the major light harvesting and water splitting processes, is lacking. We observed that DM (at 50-200 μM) reduced growth, pigment contents, and photosynthesis, but enhanced

Harmful effects of the semiconductor copper tungstate (CuWO4) on multiple photosynthetic parameters of the green microalga Raphidocelis subcapitata.

The semiconductor copper tungstate (CuWO4) may end up in aquatic ecosystems since it has the potential for water decontamination. The toxic effects of CuWO4 are totally unknown for eukaryotic organisms. In view of this, we aimed to evaluate the toxicity of CuWO4 particles (size of 161.5 nm) on the cosmopolitan green microalga Raphidocelis subcapitata (a standardized test-organism for ecotoxicological assays), analyzing the growth and multiple photosynthetic parameters obtained by pulse amplitude modulated (PAM) fluorometry. At 1.2 mg l-1, the growth was affected, and there was an increase in r

Calcium oxide nanoparticles ameliorate cadmium toxicity in alfalfa seedlings by depriving its bioaccumulation, enhancing photosystem II functionality and antioxidant gene expression.

Cadmium (Cd) is a highly toxic and carcinogenic pollutant that poses significant risks to living organisms and the environment, as it is absorbed by the plant roots and accumulates in different parts of crop during its production. A promising sustainable strategy to counteract these threats to use calcium oxide nanoparticles (CaO-NPs) as soil supplements in fodder crops. This approach has shown notable morpho-physiological and biochemical improvements under metal toxicity conditions. However, the specific mechanisms driving Cd tolerance, particularly at physio-biochemical level and antioxidant

Iron Deficiency Promotes the Lack of Photosynthetic Cytochrome c550 and Affects the Binding of the Luminal Extrinsic Subunits to Photosystem II in the Diatom Phaeodactylum tricornutum.

In the diatom Phaeodactylum tricornutum, iron limitation promotes a decrease in the content of photosystem II, as determined by measurements of oxygen-evolving activity, thermoluminescence, chlorophyll fluorescence analyses and protein quantification methods. Thermoluminescence experiments also indicate that iron limitation induces subtle changes in the energetics of the recombination reaction between reduced QB and the S2/S3 states of the water-splitting machinery. However, electron transfer from QA to QB, involving non-heme iron, seems not to be significantly inhibited. Moreover, iron defici

Effects of sodium hypochlorite treatment on the chlorophyll fluorescence in photosystem II of microalgae.

Chlorophyll fluorescence-based method shows great potentials for on-site assessing the vitality of algae in treated ship's ballast water. However, there is very limited information on the mechanism of chlorophyll fluorescence in photosystem II (PSII) after the NaClO treatment. In this paper, the effects of NaClO treatments with five concentrations (0.01, 0.04, 0.08, 0.12 and 0.15 mg/L) and treating periods (6, 24 and 48 h) on the chlorophyll fluorescence kinetics and spectra of Chlorella vulgaris (C. vulgaris) and Platymonas helgolandica (P. helgolandica) were investigated. Response of gas-exchange characteristics and chlorophyll fluorescence to acute sulfur dioxide exposure in landscape plants.

To explore the toxicity and action mechanism of acute sulfur dioxide (SO2) on urban landscape plants, a simulated SO2 stress environment by using fumigation chamber involving increasing SO2 concentration (0, 25, 50, 100, 200 mg m-3) was carried out among three species. After 72 h of exposure, SO2-induced oxidative damage indicated by electrolyte leakage increased with higher dose of SO2. Meanwhile, SO2 decreased the contents of chlorophyll a, chlorophyll b and carotenoid and increased the contents of sulfur. Net photosynthetic rate (Pn) decreased as a result of stomatal closure when SO2 dose w

Effect of calcium chelators on the formation and oxidation of the slowly relaxing reduced plastoquinone pool in calcium-depleted PSII membranes. Investigation of the F0 yield.

The F(0) fluorescence yield in intact photosystem II (PSII), Ca-depleted PSII (PSII(-Ca/NaCl)), and Mn-depleted PSII membranes was measured before and after dim light treatment (1-2 min), using flash-probe fluorescence and fluorescence induction kinetic measurements. The value of F(0) after the light treatment (F'(0)) was larger than F(0) in dark-adapted PSII membranes and depended on the appearance of the slowly relaxing, reduced plastoquinone pool (t(1/2) = 4 min) formed during preillumination, which was not totally reoxidized before the F'(0) measurement. In PSII(-Ca/NaCl) such a pool also

Analysis of high temperature stress on the dynamics of antenna size and reducing side heterogeneity of Photosystem II in wheat leaves (Triticum aestivum).

This study demonstrates the effect of high temperature stress on the heterogeneous behavior of PSII in Wheat (Triticum aestivum) leaves. Photosystem II in green plant chloroplasts displays heterogeneity both in the composition of its light harvesting antenna i.e. on the basis of antenna size (α, β and γ centers) and in the ability to reduce the plastoquinone pool i.e. the reducing side of the reaction centers (Q(B)-reducing centers and Q(B)-non-reducing centers). Detached wheat leaves were subjected to high temperature stress of 35°C, 40°C and 45°C. The chlorophyll a (Chl a) fluorescence trans

Effect of pH on the semiquinone radical Q(A)- in CN-treated photosystem II: study by hyperfine sublevel correlation spectroscopy.

The semiquinone radical Q(A)- has been studied by electron spin echo envelope modulation (ESEEM) spectroscopy in Photosystem II membranes treated with CN- at various pH values. Two protein 14N nuclei (N(I) and N(II)) were found to be magnetically coupled with the Q(A)- spin. N(I) is assigned to an amide nitrogen from the protein backbone while N(II) is assigned to the amino nitrogen, N(epsilon), of an imidazole. Above pH 8.5 only the N(I) coupling is present while both N(I) and N(II) couplings are present at lower pH values. These results are interpreted in terms of a model based on the struct

Transcriptome-based investigation of the response and repair mechanisms in the photosynthetic system of Cycas panzhihuaensis under dual high-temperature and drought stress.

BACKGROUND: Cycas panzhihuaensis, as a national first-class protected plant, has long been threatened by dual high-temperature and drought stress. Photosynthesis is a key target of dual high-temperature and drought stress. However, the specific processes underlying post-injury recovery remain poorly characterized. Utilizing chlorophyll fluorescence parameters, chloroplast ultrastructure and transcriptome sequencing techniques, this investigation analyzes the photosynthetic damage of C. panzhihuaensis under dual high-temperature and drought stress and the repair mechanism of photosynthetic syst

An additional water is introduced into the manganese cluster during the formation of the S3 state of photosystem II.

During photosynthetic water oxidation, the Mn4Ca cluster in Photosystem II progresses through five intermediate Si (i = 0-4) states. X-ray crystallography studies have reported the insertion of one new O ligand during the formation of the S3 state, but recent studies question the presence of this additional ligand based on cryo-EM and earlier room-temperature crystallography data. There is also controversy about whether the O-O bond interaction already occurs in the S3 state or in the subsequent S3 to S0 transition. Here we report conventional high-resolution data for the S1, S2, and S3 states

Using multiple endpoints to assess the toxicity of cadmium and cobalt for chlorophycean Raphidocelis subcapitata.

Metals may cause damage to the biota of contaminated environments. Moreover, using multiple endpoints in ecotoxicological studies is useful to better elucidate the mechanisms of toxicity of these compounds. Therefore, this study aimed to evaluate the effects of cadmium (Cd) and cobalt (Co) on growth, biochemical and photosynthetic parameters of the microalgae Raphidocelis subcapitata, through quantification of lipid classes composition, chlorophyll a (Chl a) content, maximum (ΦM) and effective (Φ'M) quantum yields and efficiency of the oxygen-evolving complex (OEC). Structure-Activity Relationships of Hierarchical Three-Dimensional Electrodes with Photosystem II for Semiartificial Photosynthesis.

Semiartificial photosynthesis integrates photosynthetic enzymes with artificial electronics, which is an emerging approach to reroute the natural photoelectrogenetic pathways for sustainable fuel and chemical synthesis. However, the reduced catalytic activity of enzymes in bioelectrodes limits the overall performance and further applications in fuel production. Here, we show new insights into factors that affect the photoelectrogenesis in a model system consisting of photosystem II and three-dimensional indium tin oxide and graphene electrodes. Comparative analyses of H2 photoproduction in magnesium- and sulfur-starved Chlamydomonas reinhardtii cultures.

Magnesium (Mg)-deprived Chlamydomonas reinhardtii cells are capable to sustain hydrogen (H2 ) photoproduction at relatively high photosystem II (PSII) activity levels for an extended time period as compared with sulfur (S)-deprived cells. Herein, we present a comparative study of H2 photoproduction induced by Mg and S shortage to unravel the specific rearrangements of the photosynthetic machinery and cell metabolism occurring under the two deprivation protocols. The exhaustive analysis of photosynthetic activity and regulatory pathways, respiration and starch metabolism revealed the specific r

Suppression of both ELIP1 and ELIP2 in Arabidopsis does not affect tolerance to photoinhibition and photooxidative stress.

ELIPs (early light-induced proteins) are thylakoid proteins transiently induced during greening of etiolated seedlings and during exposure to high light stress conditions. This expression pattern suggests that these proteins may be involved in the protection of the photosynthetic apparatus against photooxidative damage. To test this hypothesis, we have generated Arabidopsis (Arabidopsis thaliana) mutant plants null for both elip genes (Elip1 and Elip2) and have analyzed their sensitivity to light during greening of seedlings and to high light and cold in mature plants. In particular, we have e
