Distribution, diversity, and pathogenicity of Oomycete root pathogens affecting Fraser fir (Abies fraseri) Christmas tree production in the Southern Appalachian Mountains.

Fraser fir (Abies fraseri) is the most economically important Christmas tree species in the Southern Appalachian Mountains, especially in North Carolina. However, its production is increasingly threatened by Phytophthora root rot (PRR), caused primarily by Phytophthora cinnamomi and several other Phytophthora species, resulting in significant losses for growers annually. This study surveyed symptomatic Fraser firs across the region and recovered 151 Phytophthora isolates. Six species were identified including: P. cinnamomi (88.7%), P. cryptogea (6%), P. europaea (2%), P. cactorum (1.3%), P. ci

Taxonomy, biogeography and evolution of thirty-four taxa in Phytophthora Clade 7a including eleven new species from natural forests and updated descriptions for P. fragariae, P. rubi and the 'P. ×alni hybrid complex'.

Over the past three decades, surveys in natural and managed ecosystems, driven mainly by forest biosecurity issues, have greatly expanded knowledge of the global diversity and biogeography of the plant pathogen genus Phytophthora. In Phytophthora subclade 7a alone, 16 new species were described raising the number of validly described species in this subclade to 18. Recently, in further surveys conducted in natural forests of Japan, Vietnam, Europe and North America, 11 more new Clade 7a species and one other 7a taxon have now been identified. All are considered to be endemic to the surveyed si

Novel application of ribonucleoprotein-mediated CRISPR-Cas9 gene editing in plant pathogenic oomycete species.

CRISPR-Cas9 gene editing has become an important tool for the study of plant pathogens, allowing researchers to functionally characterize specific genes involved in phytopathogenicity, virulence, and fungicide resistance. Protocols for CRISPR-Cas9 gene editing have already been developed for Phytophthoras, an important group of oomycete plant pathogens; however, these efforts have exclusively focused on agricultural pathosystems, with research lacking for forest pathosystems. We sought to develop CRISPR-Cas9 gene editing in two forest pathogenic Phytophthoras, Phytophthora cactorum and P. ramo

The elicitin β-cryptogein's activity in tomato is mediated by jasmonic acid and ethylene signalling pathways independently of elicitin-sterol interactions.

The level of resistance induced in different tomato genotypes after β-CRY treatment correlated with the upregulation of defence genes, but not sterol binding and involved ethylene and jasmonic acid signalling. Elicitins, a family of small proteins secreted by Phytophthora and Pythium spp., are the most well-known microbe-associated molecular patterns of oomycetes, a lineage of fungus-like organisms that include many economically significant crop pathogens. The responses of tomato plants to elicitin INF1 produced by Phytophthora infestans have been studied extensively. Here, we present studies

Evaluation of methyl bromide alternatives efficacy against soil-borne pathogens, nematodes and soil microbial community.

Methyl bromide (MB) and other alternatives were evaluated for suppression of Fusarium spp., Phytophthora spp., and Meloidogyne spp. and their influence on soil microbial communities. Both Fusarium spp. and Phytophthora spp. were significantly reduced by the MB (30.74 mg kg-1), methyl iodide (MI: 45.58 mg kg-1), metham sodium (MS: 53.92 mg kg-1) treatments. MS exhibited comparable effectiveness to MB in controlling Meloidogyne spp. and total nematodes, followed by MI at the tested rate. By contrast, sulfuryl fluoride (SF: 33.04 mg kg-1) and chloroform (CF: 23.68 mg kg-1) showed low efficacy in

Targeted capture and cloning of Rps6 reveal redundancy in soybean resistance genes.

Effective management of Phytophthora root rot in soybean is compromised by the rapid loss of efficacy of the most widely deployed resistance genes to Phytophthora sojae (Rps). However, some genes such as Rps3a and Rps6 are still offering a strong protection but are nonetheless rare in elite material, probably owing to the fact that they have never been properly characterized. In this study, we have employed RenSeq, and whole genome sequencing to unravel the nature of Rps6 as a complex locus composed of 10 NLR genes. In a cell death assay using soybean protoplasts, we show that one of the candi

Decoupling the "attract-and-kill" strategy: Independent zoospore attraction and ROS-executed killing synergistically drive disease-suppressive intercropping.

Soilborne Phytophthora diseases pose a major threat to agricultural sustainability. However, how nonhost roots disrupt the transmission of soilborne Phytophthora pathogens without relying solely on classical antimicrobial exudates remains poorly understood. Through a decade-long field study, we demonstrate that strip intercropping can sustainably suppress disease incidence by up to 46.85% by leveraging nonhost roots as ecological barriers that intercept zoospore transmission. Moving beyond the conventional focus on antimicrobial exudates, we resolve the "attract-and-kill" strategy into two dis

Exploring the Biocontrol Potential of Pochonia chlamydosporia: Antifungal and Nematicidal Activities of Culture Filtrates and Secondary Metabolites.

This study investigated the biocontrol potential of culture filtrates and secondary metabolites (SMs) from the nematophagous fungus Pochonia chlamydosporia. The fungus exhibited antifungal activity against pathogens such as Pythium myriotylum, Macrophomina sp., Colletotrichum sp., Phytophthora sp., Fusarium sp., and Exerohilum rostratum. The highest percentage inhibition of radial growth values was observed with P. myriotylum (81.42%) and Phytophthora sp. (82.71%), while Macrophomina sp. showed the lowest inhibition (40%). The study also aimed to identify the SMs responsible for the biocontrol

Smartphone-integrated RPA-CRISPR/Cas12a detection system with microneedle sampling for early point-of-care diagnosis of potato late blight.

Potato late blight, caused by the oomycete pathogen Phytophthora infestans (P. infestans), is one of the most devastating diseases threatening global potato production. Conventional plant disease detection methods rely on a labor-intensive and time-consuming workflow and require bulky and expensive benchtop equipment, limiting their in-field applications. Here, we report a portable RPA-CRISPR/Cas12a-based diagnostic platform integrated with a polyvinyl alcohol (PVA) microneedle (MN) patch, which allows rapid in-field sampling, and smartphone-based fluorescence acquisition and analysis to detec

Trichoderma-Based Biopreparation with Prebiotics Supplementation for the Naturalization of Raspberry Plant Rhizosphere.

The number of raspberry plants dying from a sudden outbreak of gray mold, verticillium wilt, anthracnosis, and phytophthora infection has increased in recent times, leading to crop failure. The plants suffer tissue collapse and black roots, symptoms similar to a Botrytis-Verticillium-Colletotrichum-Phytophthora disease complex. A sizeable number of fungal isolates were acquired from the root and rhizosphere samples of wild raspberries from different locations. Subsequent in vitro tests revealed that a core consortium of 11 isolates of selected Trichoderma spp. was the most essential element fo

Centuries of Potato Late Blight: Tracking Global Epidemics and Managing Future Outbreaks.

Phytophthora infestans killed the potato crop in Ireland in 1845, leading to widespread famine and the death of more than one million people. Historic herbarium specimens from the famine era were used to understand the pathogen's biology and track its global spread, providing a valuable resource for research. Historic outbreaks in the United States and Europe were caused by the FAM-1 lineage, whereas the US-1 lineage spread later. The famine lineage was basal in the phylogeny and ancestral to modern US-1, Mexican, and globally aggressive lineages. An admixture between the famine lineage and th

Inhibition of Oomycetes by the Mixture of Maleic Acid and Copper Sulfate.

Since the protective activity of the Bordeaux mixture against plant disease caused by oomycetes was discovered, copper compounds have been used for more than a century as an effective plant protection strategy. However, the application of excessive copper can cause adverse effects through long-term heavy metal accumulation in soils. Therefore, it is necessary to develop new strategies to reduce or replace copper in pesticides based on organic and low-input farming systems. Organic acids are eco-friendly. In this study, we tested the antifungal and anti-oomycete activity of maleic acid (MA) and

Differential roles of melatonin in plant-host resistance and pathogen suppression in cucurbits.

Since the 1950s, research on the animal neurohormone, melatonin, has focused on its multiregulatory effect on patients suffering from insomnia, cancer, and Alzheimer's disease. In plants, melatonin plays major role in plant growth and development, and is inducible in response to diverse biotic and abiotic stresses. However, studies on the direct role of melatonin in disease suppression and as a signaling molecule in host-pathogen defense mechanism are lacking. This study provides insight on the predicted biosynthetic pathway of melatonin in watermelon (Citrullus lanatus), and how application o

Teratogenic potential of blighted potato concentrate in rabbits, hamsters, and miniature swine.

Freeze-dried preparations of potatoes infected with phytophthora infestans or Alternaria solani were fed to pregnant hamsters, rabbits, and miniature swine. No evidence was observed that these diets or the extracts of respective blights were capable of producing teratologic effects in hamsters. Lesions of neurulation and anencephaly were noticed in rabbits and swine, respectively. The need for further pharmacological and toxicological evaluation in sensitive species is emphasized.
