Influence of xylem fluid chemistry on planktonic growth, biofilm formation and aggregation of Xylella fastidiosa.

Xylella fastidiosa is the causal agent of Pierce's disease in grapevines. The mechanisms of pathogenicity are largely due to occlusion of xylem vessels by aggregation of X. fastidiosa and biofilm formation. Xylella fastidiosa was subjected to xylem fluids with varying chemistries to examine the effects of nutritional components on bacterial growth in vitro. The exposure of X. fastidiosa to xylem fluids collected from different Vitis genotypes resulted in highly significant differences in both planktonic growth and biofilm formation. Planktonic growth of X. fastidiosa in Vitis xylem fluid was c

In vitro effects of antimicrobial agents on planktonic and biofilm forms of Staphylococcus lugdunensis clinical isolates.

Staphylococcus lugdunensis is an atypically virulent coagulase-negative staphylococcal species associated with acute and destructive infections that often resemble Staphylococcus aureus infections. Several types of infection caused by S. lugdunensis (e.g., native valve endocarditis, prosthetic joint infection, and intravascular catheter infection) are associated with biofilm formation, which may lead to an inability to eradicate the infection due to the intrinsic nature of biofilms to resist high levels of antibiotics. In this study, planktonic MICs and MBCs and biofilm bactericidal concentrat

Use and detection of a vitamin B1 degradation product yields new views of the marine B1 cycle and plankton metabolite exchange.

Vitamin B1 (thiamin) is a vital nutrient for most cells in nature, including marine plankton. Early and recent experiments show that B1 degradation products instead of B1 can support the growth of marine bacterioplankton and phytoplankton. However, the use and occurrence of some degradation products remains uninvestigated, namely N-formyl-4-amino-5-aminomethyl-2-methylpyrimidine (FAMP), which has been a focus of plant oxidative stress research. We investigated the relevance of FAMP in the ocean. Experiments and global ocean meta-omic data indicate that eukaryotic phytoplankton, including picoe

Spatial and temporal variations of total and methylmercury concentrations in plankton from a mercury-contaminated and eutrophic reservoir in Guizhou Province, China.

Total mercury (THg) and methylmercury (MeHg) concentrations in four size fractions of plankton from three sampling stations in the Hg-contaminated and eutrophic Baihua Reservoir, Guizhou, China, were investigated for biomagnification and trophic transfer of Hg at different sites with various proximity to the major point sources of nutrients and metals. Total Hg concentrations in plankton of the various size fractions varied from 49 to 5,504 ng g(-1) and MeHg concentrations ranged from 3 to 101 ng g(-1). The percentage of Hg as MeHg varied from 0.16 to 70%. Total Hg and MeHg concentrations in p

Multiple regression analysis to assess the role of plankton on the distribution and speciation of mercury in water of a contaminated lagoon.

Spatial and seasonal variation of mercury species aqueous concentrations and distributions was carried out during six sampling campaigns at four locations within Laranjo Bay, the most mercury-contaminated area of the Aveiro Lagoon (Portugal). Inorganic mercury (IHg(II)) and methylmercury (MeHg) were determined in filter-retained (IHgPART, MeHgPART) and filtered (<0.45μm) fractions (IHg(II)DISS, MeHgDISS). The concentrations of IHgPART depended on site and on dilution with downstream particles. Similar processes were evidenced for MeHgPART, however, its concentrations increased for particles ri

Seasonal and evolutionary patterns of planktonic Hikarchaeia reveal ecological niches of marine rare biosphere constituents.

Members of Marine Group IV, now termed Hikarchaeia, hold an important transitional evolutionary position among Archaea but remain uncultivated, and their ecology remains uncharacterized. Over a 5-year period, we performed monthly profile sampling in the North Atlantic Subtropical Gyre and sequenced V4 16S rRNA gene amplicons, while also collecting additional size-fractionated and metagenomic samples. Hikarchaeal relative abundance annually peaked at ~1% of prokaryotic amplicons. Their relative abundances were significantly higher after deep winter mixing when thermal stratification isolated ex

Species-Specific Susceptibility of Planktonic and Biofilm Forming Candida Strains to Cyclodextrin-Encapsulated Essential Oils.

Background/Objectives: Essential oils (EOs) have multi-target antifungal activity, but their translation is limited by volatility and poor aqueous dispersibility. Randomly methylated β-cyclodextrin (RAMEB) inclusion may enhance effective exposure and thereby alter susceptibility, stress responses, and biofilm outcomes in a species-dependent manner. This study quantified species-specific planktonic and biofilm susceptibility to four EOs and their RAMEB complexes across clinically relevant Candida species. Methods: Lavender (L), lemon balm (B), peppermint (P), and thyme (T) oils and their RAMEB

Comparative fermentation performance of biofilm and planktonic Saccharomyces cerevisiae under standard and high-carbon media.

Biofilms are structured microbial communities with wide-ranging industrial applications. In Saccharomyces cerevisiae, biofilm formation can influence fermentation, improve microbial stability, and enhance production efficiency. This study evaluates the fermentation performance of S. cerevisiae in both planktonic and biofilm states under standard and high-carbon media. During a 96-hour fermentation, planktonic cultures consumed glucose within 24 h, whereas biofilm-associated cells required 48 h due to diffusion constraints within the extracellular polymeric substance (EPS) matrix. Under standar

Ecological risks of olivine with different particle sizes and dosages on planktonic ecosystems during ocean alkalinization.

With industrialization, global climate change has intensified. Ocean alkalinization via olivine addition enhances seawater alkalinity and carbon sequestration but its ecological risks remain poorly understood. In this study, Chlorella vulgaris and Brachionus plicatilis were used as model organisms. Olivine of different particle sizes (≤38 μm, 50-74 μm, ≥154 μm) and dosages (1‰, 2‰) was applied in laboratory experiments with molecular analyses and model evaluation to assess effects on planktonic ecosystems and carbon sequestration. Results indicate strong particle-size dependence, with finer pa

The Effect of Medium-Chain Triglycerides Oil on Candida albicans and Streptococcus mutans in Planktonic and Mucosal Models.

Despite MCT oil's potential antimicrobial benefits for gastrointestinal health, its effects on disrupting cariogenic pathogens on oral mucosal surfaces remain underexplored. This study evaluated the impact of MCT oil on the adhesion and invasion of Candida albicans and Streptoccocus mutans using planktonic and mucosal models. First, a planktonic model was used to assess the impact of various concentrations of MCT on the growth of S. mutans and C. albicans. Subsequently, a mucosal model was established by seeding TR-146 human buccal mucosal epithelial cells on a 3 µm porous transwell membrane,

Reduced Pseudomonas aeruginosa Cell Size Observed on Planktonic Cultures Grown in the International Space Station.

Bacterial growth and behavior have been studied in microgravity in the past, but little focus has been directed to cell size despite its impact on a myriad of processes, including biofilm formation, which is impactful regarding crew health. To interrogate this characteristic, supernatant aliquots of P. aeruginosa cultured on different materials and media on board the International Space Station (ISS) as part of the Space Biofilms Project were analyzed. For that experiment, P. aeruginosa was grown in microgravity-with matching Earth controls-in modified artificial urine medium (mAUMg-high Pi) o

A new genus and species of oceanic planktonic Tisbidae (Crustacea, Copepoda, Harpacticoida) with enlarged modified eyes.

Both sexes of a new monotypic genus of Tisbidae (Copepoda, Harpacticoida) are described from the epi- or mesopelagic zone in the Kuroshio region, Japan. Gyoromeguttatumgen. et sp. nov. belongs to a monophyletic lineage of deepwater holoplanktonic genera defined by a suite of characters. Within this clade, Gyoromegen. nov. appears most closely related to Neotisbella Boxshall, 1979. The most distinguishable feature of G.guttatumgen. et sp. nov. is the presence of large, paired, frontal modified eyes, each consisting of a baculiform ocellus, a globular (Gicklhorn's?) organ, and a semi-parabolic p

Concentration of mercury and other metals in an Arctic planktonic food web under a climate warming scenario.

Arctic marine ecosystems act as a global sink of mercury (Hg) and other metals, and high concentrations of these have been measured in higher trophic-level organisms. Nevertheless, the concentrations of metals at the basis of the marine food web in the Arctic is less known despite the likelihood of biomagnification from dietary sources. We investigated the concentrations of mercury (Hg) and other metals in different size fractions of plankton in West Greenland. All size fractions contained detectable levels of Hg (ranging from 4.8 to 241.3 ng g dw-1) at all stations, although with high geograp

Antifungal Activity of Chitosan against Histoplasma capsulatum in Planktonic and Biofilm Forms: A Therapeutic Strategy in the Future?

Histoplasmosis is a respiratory disease caused by Histoplasma capsulatum, a dimorphic fungus, with high mortality and morbidity rates, especially in immunocompromised patients. Considering the small existing therapeutic arsenal, new treatment approaches are still required. Chitosan, a linear polysaccharide obtained from partial chitin deacetylation, has anti-inflammatory, antimicrobial, biocompatibility, biodegradability, and non-toxicity properties. Chitosan with different deacetylation degrees and molecular weights has been explored as a potential agent against fungal pathogens. In this stud

Pomegranate sarcotesta lectin (PgTeL) inhibits planktonic growth and disrupts biofilm formed by Cryptococcus neoformans.

AIMS: We investigated the putative fungistatic and fungicidal activities of pomegranate sarcotesta lectin (PgTeL) against Cryptococcus neoformans B3501 (serotype D), specifically the ability of PgTeL to inhibit yeast capsule and biofilm formation in this strain. METHODS AND RESULTS: PgTeL showed a minimum inhibitory concentration of 172.0 μg ml-1, at which it did not exhibit a fungicidal effect. PgTeL concentrations of 4.0-256.0 μg ml-1 reduced biofilm biomass by 31.0%-64.0%. Furthermore, 32.0-256.0 μg ml-1 PgTeL decreased the metabolic activity of the biofilm by 32.0%-93.0%. Multi-Strain and -Species Investigation of Volatile Metabolites Emitted from Planktonic and Biofilm Candida Cultures.

Candida parapsiliosis is a prevalent neonatal pathogen that attains its virulence through its strain-specific ability to form biofilms. The use of volatilomics, the profiling of volatile metabolites from microbes is a non-invasive, simple way to identify and classify microbes; it has shown great potential for pathogen identification. Although C. parapsiliosis is one of the most common clinical fungal pathogens, its volatilome has never been characterised. In this study, planktonic volatilomes of ten clinical strains of C. parapsilosis were analysed, along with a single strain of Candida albica

Hydroxybiphenylamide GroEL/ES Inhibitors Are Potent Antibacterials against Planktonic and Biofilm Forms of Staphylococcus aureus.

We recently reported the identification of a GroEL/ES inhibitor (1, N-(4-(benzo[ d]thiazol-2-ylthio)-3-chlorophenyl)-3,5-dibromo-2-hydroxybenzamide) that exhibited in vitro antibacterial effects against Staphylococcus aureus comparable to vancomycin, an antibiotic of last resort. To follow up, we have synthesized 43 compound 1 analogs to determine the most effective functional groups of the scaffold for inhibiting GroEL/ES and killing bacteria. Our results identified that the benzothiazole and hydroxyl groups are important for inhibiting GroEL/ES-mediated folding functions, with the hydroxyl e

Evolution of Antibiotic Resistance in Biofilm and Planktonic Pseudomonas aeruginosa Populations Exposed to Subinhibitory Levels of Ciprofloxacin.

The opportunistic Gram-negative pathogen Pseudomonas aeruginosa, known for its intrinsic and acquired antibiotic resistance, has a notorious ability to form biofilms, which often facilitate chronic infections. The evolutionary paths to antibiotic resistance have mainly been investigated in planktonic cultures and are less studied in biofilms. We experimentally evolved P. aeruginosa PAO1 colony biofilms and stationary-phase planktonic cultures for seven passages in the presence of subinhibitory levels (0.1 mg/liter) of ciprofloxacin (CIP) and performed a genotypic (whole-bacterial population se
