Effects of chelidonic acid, a secondary plant metabolite, on mast cell degranulation and adaptive immunity in rats.

The present study evaluated the immunomodulatory effects of chelidonic acid, a secondary plant metabolite, with therapeutic potential in allergic disorders, in experimental animals. In mast cell degranulation studies, ovalbumin immunized and challenged rats, chelidonic acid (1, 3 and 10mg/kg, i.p.) dose relatedly prevented ovalbumin challenge induced mast cell degranulation by differing degrees when compared with vehicle treated group, and these effects were comparable with prednisolone (10mg/kg). A reduction in post-challenge mortality was also observed in all treated groups. Further, there w

Novel plant virus-based vaccine induces protective cytotoxic T-lymphocyte-mediated antiviral immunity through dendritic cell maturation.

Currently used vaccines protect mainly through the production of neutralizing antibodies. However, antibodies confer little or no protection for a majority of chronic viral infections that require active involvement of cytotoxic T lymphocytes (CTLs). Virus-like particles (VLPs) have been shown to be efficient inducers of cell-mediated immune responses, but administration of an adjuvant is generally required. We recently reported the generation of a novel VLP system exploiting the self-assembly property of the papaya mosaic virus (PapMV) coat protein. We show here that uptake of PapMV-like part

Generation of protective immune response against anthrax by oral immunization with protective antigen plant-based vaccine.

In concern with frequent recurrence of anthrax in endemic areas and inadvertent use of its spores as biological weapon, the development of an effective anthrax vaccine suitable for both human and veterinary needs is highly desirable. A simple oral delivery through expression in plant system could offer promising alternative to the current methods that rely on injectable vaccines extracted from bacterial sources. In the present study, we have expressed protective antigen (PA) gene in Indian mustard by Agrobacterium-mediated transformation and in tobacco by plastid transformation. Putative trans

Emerging Roles of Receptor-Like Proteins in Plant Immunity: Crosstalk, Signalling Networks and Prospects for Disease Resistance Breeding.

Receptor-like proteins (RLPs) are cell-surface receptors that recognise pathogen- or damage-associated molecular patterns to activate immune responses, and some also participate in plant development and modulation of defence signalling. Recent advances in bioinformatics and high-throughput technologies have begun to shed light on the functional roles of RLPs, their interactions with other immune receptors, and their involvement in plant defence mechanisms. This review summarises the emerging roles of RLPs in plant immunity, with a particular focus on their dynamic interplay with receptor-like

Azospirillum spp. as bio-innovative defenders: Metabolomic signatures and molecular defense reprogramming in plant immunity against fungal phytopathogens.

Fungal diseases accounts for nearly 30-40% of global crop yield losses, underscoring the need for sustainable alternatives to chemical fungicides. Azospirillum spp., traditionally recognized for plant growth promotion, have emerged as potent biocontrol agent with multifaceted defense functions. Beyond niche competition and siderophore production, Azospirillum primes host immunity through reprogramming primary and secondary metabolism and activating systemic defense pathways such as salicylic acid (SA), jasmonic acid (JA), and induced systemic resistance (ISR). Metabolomic analyses reveal its i

Genome-wide Characterization of Small Secreted Peptides in Nicotiana tabacum and Functional Assessment of NtLTP25 in Plant Immunity.

Small secreted peptides (SSPs), serving as signaling molecules for intercellular communication, play significant regulatory roles in plant growth, development, pathogen immunity, and responses to abiotic stress. Despite several SSPs, such as PIP, PSK, and PSY having been identified to participate in plant immunity, the majority of SSPs remain understudied, necessitating the exploration and identification of SSPs regulating plant immunity from vast genomic resources. Here we systematically characterized 756 putative SSPs across the genome of Nicotiana tabacum. 173 SSPs were further annotated as

Seeing is believing: Understanding functions of NPR1 and its paralogs in plant immunity through cellular and structural analyses.

In the past 30 years, our knowledge of how nonexpressor of pathogenesis-related genes 1 (NPR1) serves as a master regulator of salicylic acid (SA)-mediated immune responses in plants has been informed largely by molecular genetic studies. Despite extensive efforts, the biochemical functions of this protein in promoting plant survival against a wide range of pathogens and abiotic stresses are not completely understood. Recent breakthroughs in cellular and structural analyses of NPR1 and its paralogs have provided a molecular framework for reinterpreting decades of genetic observations and have

A Bacterial Type III Effector Targets the Master Regulator of Salicylic Acid Signaling, NPR1, to Subvert Plant Immunity.

Most plant bacterial pathogens rely on type III effectors to cause diseases. Although it is well known that the plant hormone salicylic acid (SA) plays an essential role in defense, whether the master regulator of SA signaling, NPR1, is targeted by any plant pathogen effectors is unknown. SA facilitates the reduction of cytosolic NPR1 oligomers into monomers, which enter the nucleus and function as transcriptional coactivators of plant defense genes. We show that SA promotes the interaction between the Pseudomonas syringae type III effector AvrPtoB and NPR1. In the presence of SA, AvrPtoB medi

Grapevine rust fungus Neophysopella tropicalis deploys protein effectors with plant immunity-suppressing activity during sequential stages of pathogenesis.

Germination of urediniospores marks the onset of infection by Neophysopella tropicalis, the causal agent of Asian grapevine leaf rust, yet the molecular mechanisms enabling this obligate biotrophic fungus to establish infection remain largely unknown. We combined transcriptomics, histological analyses, and functional assays to identify and characterize candidate effector proteins deployed during the earliest stages of pathogenesis. RNA sequencing of in vitro-germinated urediniospores identified 303 genes encoding predicted secreted proteins bearing hallmarks of fungal effectors, grouped into 1

High Light Intensity Potentiates Bacillus amyloliquefaciens PMB05-Associated Plant Immunity and Bacterial Soft Rot Control in Arabidopsis thaliana.

Bacterial soft rot, caused by Pectobacterium carotovorum subsp. carotovorum, affects numerous vegetable crops. Beneficial microbes like Bacillus amyloliquefaciens PMB05 offer a sustainable alternative to traditional copper- or antibiotic-based controls, which risks the development of pathogen resistance. Bacillus amyloliquefaciens PMB05 enhances disease resistance to soft rot by intensifying PAMP-triggered immunity (PTI). Although our previous research showed that PMB05-intensified PTI correlates with a reduced photosynthetic electron transport rate under high light conditions, it remains uncl

The Arabidopsis tex1-4 mutant exhibits altered mRNA export and hormonal crosstalk in plant immunity against Pseudomonas syringae.

TEX1 contributes to mRNA export and SA-mediated defense while modulating ABA-driven susceptibility, linking RNA processing to immune regulation in Arabidopsis TEX1, a subunit of the multi-protein THO/TREX complex conserved in eukaryotes, facilitates the synthesis, splicing, and nuclear-cytoplasmic export of mRNAs. However, its mechanism in plants remains to be thoroughly established, particularly its contribution to plant immunity and the mRNA export mechanism in response to pathogen challenges. Here, we showed that TEX1 contributes to mRNA export and hormonal signaling during pathogen infecti

The ABI4-SEN1-TRXm1 module integrates ABA and H2S signaling to coordinate redox homeostasis, senescence, and plant immunity.

Plants undergo metabolic reprogramming, including senescence, in response to pathogen attack. Although senescence-associated genes are critical for this process, the genes that integrate plant defense and senescence processes remain largely unknown. Here, we identified and characterized SENESCENCE ASSOCIATED GENE 1 (SEN1) as a central regulator that coordinates these processes in Arabidopsis. SEN1 is transcriptionally activated by abscisic acid (ABA) through the ABA signaling component ABA INSENSITIVE 4 (ABI4). SEN1 interacts with TRXm1, and this interaction modulates the sulfotransferase acti

Flagellin-derived peptide flg22 from non-pathogenic Pseudomonas fragi Sneb1990 triggers plant immunity and reduces Meloidogyne incognita infestation.

BACKGROUND: Biological control is widely recognized for its environmental benefits and has gained increasing attention. The peptide flg22 derived from biocontrol bacteria Pseudomonas fragi Sneb1990 (flg22Pf) exhibits significant efficacy against Meloidogyne incognita, but its mechanism is still unknown. RESULTS: In this study, we cloned the full-length flagellin gene from P. fragi Sneb1990 and found that it shares 32.02% sequence identity with the flagellin from non-pathogenic Pseudomonas syringae pv. tomato DC3000 (Pst DC300). Compared to flg22 derived from Pst DC3000 (flg22Ps), flg22Pf conta

Activation of Plant Innate Immunity by Extracellular High Mobility Group Box 3 and Its Inhibition by Salicylic Acid.

Damage-associated molecular pattern molecules (DAMPs) signal the presence of tissue damage to induce immune responses in plants and animals. Here, we report that High Mobility Group Box 3 (HMGB3) is a novel plant DAMP. Extracellular HMGB3, through receptor-like kinases BAK1 and BKK1, induced hallmark innate immune responses, including i) MAPK activation, ii) defense-related gene expression, iii) callose deposition, and iv) enhanced resistance to Botrytis cinerea. Infection by necrotrophic B. cinerea released HMGB3 into the extracellular space (apoplast). Silencing HMGBs enhanced susceptibility

Salicyloyl-aspartate synthesized by the acetyl-amido synthetase GH3.5 is a potential activator of plant immunity in Arabidopsis.

Salicylic acid (SA) plays a critical role in plant immunity responses against pathogen infection, especially in the establishment of systemic acquired resistance. Whether other forms of salicylates also function in plant immunity has not been explored. Our previous study has revealed that salicyloyl-aspartate (SA-Asp), the only reported endogenous SA-amino acid conjugate in plants, was highly accumulated in the Arabidopsis activation-tagged mutant gh3.5-1D after pathogen infection. In this study, we dissected SA-Asp production in Arabidopsis. In vitro biochemical experiments showed that the GH

A plant based protective antigen [PA(dIV)] vaccine expressed in chloroplasts demonstrates protective immunity in mice against anthrax.

The currently available anthrax vaccines are limited by being incompletely characterized, potentially reactogenic and have an expanded dosage schedule. Plant based vaccines offer safe alternative for vaccine production. In the present study, we expressed domain IV of Bacillus anthracis protective antigen gene [PA(dIV)] in planta (by nuclear agrobacterium and chloroplast transformation) and E. coli [rPA(dIV)]. The presence of transgene and the expression of PA(dIV) in planta was confirmed by molecular analysis. Expression levels up to 5.3% of total soluble protein (TSP) were obtained with AT ri

Iron homeostasis and plant immune responses: Recent insights and translational implications.

Iron metabolism and the plant immune system are both critical for plant vigor in natural ecosystems and for reliable agricultural productivity. Mechanistic studies of plant iron home-ostasis and plant immunity have traditionally been carried out in isolation from each other; however, our growing understanding of both processes has uncovered significant connections. For example, iron plays a critical role in the generation of reactive oxygen intermediates during immunity and has been recently implicated as a critical factor for immune-initiated cell death via ferroptosis. Moreover, plant iron s

Neutralizing immune responses induced by oligomeric H5N1-hemagglutinins from plants.

Plant-based transient expression is an alternative platform to produce hemagglutinin-based subunit vaccines. This production system provides not only fast and effective response in the context of a pandemic but also enables the supply of big volume vaccines at low cost. Crude plant extracts containing influenza hemagglutinin are considered to use as vaccine sources because of avoidance of related purification steps resulting in low cost production allowing veterinary applications. Highly immunogenic influenza hemagglutinins are urgently required to meet these pre-conditions. Here, we present a

The insect effector CSPs activate an atypical plant TNL immune receptor.

CSP-mediated activation of RCSP involves noncanonical structural and catalytic mechanisms-including BB-loop deletion, α5α6-loop insertion, and atypical catalytic residues within the RCSP-TIR domain. Chemosensory proteins (CSPs), a conserved effector family in phloem-feeding insects such as aphids and the brown planthopper (Nilaparvata lugens), induce dwarfism in Nicotiana benthamiana through recognition by the Toll-interleukin-1 receptor (TIR) domain-containing nucleotide-binding leucine-rich repeat receptor (TNL) RCSP. However, CSP-mediated RCSP activation and enzymatic mechanism remain unkno
