Norway spruce spatiotemporal programs of conifer reproductive development.

Conifers are essential components of forest ecosystems; however, their reproductive development remains largely understudied due to their genomes' complexity. Here, we present a time-resolved spatial transcriptomics (ST) atlas of 88 tissue sections across three time points from developing reproductive and vegetative shoots in wild-type Norway spruce (Picea abies), as well as transition shoots from the acrocona mutant. By studying their different spatiotemporal gene expression dynamics, we identified molecular processes active during the vegetative-to-reproductive shift and their specific spati

Oxytetracycline on the Wetland Macrophyte Bidens laevis L. Seedlings: Uptake, Bioaccumulation, Translocation and Biochemical Stress Responses.

Oxytetracycline (OTC) is a widely used veterinary antibiotic worldwide due to its extensive use in livestock production. However, information regarding its uptake, biochemical and physiological effects in macrophytes remains limited. This study evaluated OTC uptake, bioaccumulation, translocation, and associated biochemical and physiological responses in seedlings of the wetland macrophyte Bidens laevis, exposed for 48 h to environmentally relevant concentrations (0, 0.05, 0.5, 5, 10 and 100 µg L-1). OTC accumulated in the roots and shoots, with bioaccumulation factors exceeding 1 in all treat

A genetic program for rapid initiation of axillary meristems at the onset of flowering.

Shoot branching patterns and their diversity arise from variable activities of axillary meristems (AMs). A common feature of flowering plants is delayed AM initiation and growth during the vegetative phase, followed by a sudden and rapid acceleration of apical AM initiation during the reproductive phase. The mechanisms underlying this pronounced developmental shift are poorly understood. During tomato floral transition, the primary shoot meristem (PSM) terminates in a flower after producing two spatially adjacent AMs: one that will generate a flower and the other a vegetative side shoot. In pl

Knockout of Tobacco Homologs of Arabidopsis Multi-Antibiotic Resistance 1 Gene Confers a Limited Resistance to Aminoglycoside Antibiotics.

To explore a possible recessive selective marker for future DNA-free genome editing by direct delivery of a CRISPR/Cas9-single guide RNA (sgRNA) ribonucleoprotein complex, we knocked out homologs of the Arabidopsis Multi-Antibiotic Resistance 1 (MAR1)/RTS3 gene, mutations of which confer aminoglycoside resistance, in tobacco plants by an efficient Agrobacterium-mediated gene transfer. A Cas9 gene was introduced into Nicotiana tabacum and Nicotiana sylvestris together with an sgRNA gene for one of three different target sequences designed to perfectly match sequences in both S- and T-genome cop

Shoot-derived miR2111 controls legume root and nodule development.

Legumes control their nodule numbers through the autoregulation of nodulation (AON). Rhizobia infection stimulates the production of root-derived CLE peptide hormones that are translocated to the shoot where they regulate a new signal. We used soybean to demonstrate that this shoot-derived signal is miR2111, which is transported via phloem to the root where it targets transcripts of Too Much Love (TML), a negative regulator of nodulation. Shoot perception of rhizobia-induced CLE peptides suppresses miR2111 expression, resulting in TML accumulation in roots and subsequent inhibition of nodule o

Light-dependent activation of HY5 promotes mycorrhizal symbiosis in tomato by systemically regulating strigolactone biosynthesis.

Light quality affects mutualisms between plant roots and arbuscular mycorrhizal fungi (AMFs), which modify nutrient acquisition in plants. However, the mechanisms by which light systemically modulates root colonization by AMFs and phosphate uptake in roots remain unclear. We used a range of approaches, including grafting techniques, protein immunoblot analysis, electrophoretic mobility shift assay, chromatin immunoprecipitation, and dual-luciferase assays, to unveil the molecular basis of light signal transmission from shoot to root that mediates arbuscule development and phosphate uptake in t

ELONGATED HYPOCOTYL5-mediated light signaling promotes shoot regeneration in Arabidopsis thaliana.

Injured plant somatic tissues regenerate themselves by establishing shoot or root meristems. In Arabidopsis (Arabidopsis thaliana), a two-step culture system ensures regeneration by first promoting the acquisition of pluripotency and subsequently specifying the fate of new meristems. Although previous studies have reported the importance of phytohormones auxin and cytokinin in determining the fate of new meristems, whether and how environmental factors influence this process remains elusive. In this study, we investigated the impact of light signals on shoot regeneration using Arabidopsis hypo

Critical Leaf Magnesium Thresholds and the Impact of Magnesium on Plant Growth and Photo-Oxidative Defense: A Systematic Review and Meta-Analysis From 70 Years of Research.

Magnesium (Mg) deficiency in plants is a widespread problem affecting productivity and quality in agricultural systems and forestry. Although numerous studies addressed the effect of Mg deficiency on biomass and photosynthetic CO2 assimilation, a summary evaluation of the effect of Mg supply on plant growth and photosynthesis is so far missing. We performed a systematic review and meta-analysis to collect and combine all relevant scientifically published data on the relationship between Mg nutrition and parameters that can be related to plant growth such as root and shoot biomass, harvestable

Metabolite Profiles Provide Insights into Underlying Mechanism in Bupleurum (Apiaceae) in Response to Three Levels of Phosphorus Fertilization.

Phosphorus (P) deficiency affects plant yield and quality, yet at the same time, excessive phosphorus application does not necessarily promote the growth of plants. How to maintain a balance between biomass accumulation and phosphorus application is a problem. Therefore, the purpose of this research was to explore the relationship between yield and quality of Bupleurum and phosphorus fertilization, based on three phosphorus fertilization levels (20 kg∙ha-1; 10 kg∙ha-1; and 0 kg∙ha-1). We adopted gas chromatography-mass spectrometry to assess the response of primary metabolites of different pla

Effects of enhancing endogenous and exogenous glutathione in roots on cadmium movement in Arabidopsis thaliana.

Glutathione (GSH) is a thiol-containing compound involved in many aspects of plant metabolism. In the present study, we investigated how enhancing endogenous and exogenous GSH affects cadmium (Cd) movement and distribution in Arabidopsis plants cultured hydroponically. Transgenic Arabidopsis plants with a strong ability to synthesize GSH in roots were generated by transforming the gene encoding the bifunctional γ-glutamylcysteine synthetase-glutathione synthetase enzyme from Streptococcus thermophiles (StGCS-GS). Enhancing endogenous and exogenous GSH decreased the Cd translocation ratio in di

Altered Antioxidant Physiology and Cadmium Partitioning in Arabidopsis F2 Descendants of Chromium-Exposed Ancestors.

Ancestral environmental stress can influence descendant phenotypes, yet the physiological traits underlying such effects remain poorly understood, particularly in response to heterologous stress. We investigated whether chronic chromium (Cr) exposure in the parental generation is associated with persistent physiological differences in Arabidopsis thaliana F2 descendants. Wild-type plants were exposed to 2.5 μM hexavalent chromium [Cr(VI)] supplied as potassium dichromate (K2Cr2O7) from the seedling stage until seed set. The resulting F2 lineage derived from chromium-exposed ancestors (2.5F2) w

The role of nanosilica in antioxidant production, nutrition and photochemical efficiency to mitigate sulfur deficiency in Capsicum annuum L.

Sulfur deficiency limits the development of bell pepper in various cultivation regions worldwide. The use of nanosilica is a promising alternative to mitigate the nutritional impairments caused by stress; however, its efficacy in bell pepper under sulfur-deficiency and its potential benefits for non-stressed plants remain unclear. This study aimed to investigate the impact of sulfur-deficiency on bell pepper and evaluate the effects of nanosilica in mitigating these effects, as well as to explore its benefits under sulfur-sufficient conditions. The experiment was conducted in a greenhouse usin

NH4+-N alleviates iron deficiency in rice seedlings under calcareous conditions.

Drip-irrigated rice (Oryza sativa L.) in calcareous soil exhibits signs of iron (Fe) deficiency. This study aimed to explore whether NH4+ alleviates Fe deficiency in rice seedlings grown under calcareous conditions. Two rice varieties (cv. 'T43' Fe deficiency-tolerant variety and cv. 'T04' Fe deficiency-sensitive variety) were used to carry out two independent experiments with exposure to different nitrogen (N) forms (nitrate (NO3-) or NH4+) under calcareous conditions. In experiment 1, plants were precultured in a nutrient solution with excess Fe (40 µM Fe(II)-EDTA) for 14 d and then supplied

Silicon alleviates Cd stress of wheat seedlings (Triticum turgidum L. cv. Claudio) grown in hydroponics.

We investigated the potential role of silicon in improving tolerance and decreasing cadmium (Cd) toxicity in durum wheat (Triticum turgidum L. durum) either through a reduced Cd uptake or exclusion/sequestration in non-metabolic tissues. For this, plants were grown in hydroponic conditions for 10 days either in presence or absence of 1 mM Si and for 11 additional days in various Cd concentrations (0, 0.5, 5.0 and 50 μM). After harvesting, morphological and physiological parameters as well as elemental concentrations were recorded. Cadmium caused reduction in growth parameters, photosynthetic p

In Vitro Propagation of Eleutherine palmifolia (L.) Merr.: Optimization of Surface Sterilization and Effects of BA and NAA on Shoot and Root Induction.

Eleutherine palmifolia (L.) Merr., or Wan Hom Daeng, is a medicinal plant valued for its antioxidant, anti-inflammatory, and antibacterial properties. Its bulbs contain flavonoids, phenolics, and naphthoquinones, but slow natural propagation and low seed viability hinder large-scale cultivation and conservation. This study evaluated the optimal Clorox concentration and exposure time for sterilizing E. palmifolia bulb explants and examined the effects of BA and NAA on in vitro shoot and root development. The results revealed that surface sterilization with 20% Clorox for 20 min yielded the high

Calcium affects uranium responses in Arabidopsis thaliana: From distribution to toxicity.

Uranium, a heavy metal and primordial radionuclide, is present in surface waters and soils both naturally and due to industrial activities. Uranium is known to be toxic to plants and its uptake and toxicity can be influenced by multiple factors such as pH and the presence of different ions. However, the precise role of the different ions in uranium uptake is not yet known. Here we investigated whether calcium influences uranium uptake and toxicity in the terrestrial plant Arabidopsis thaliana. To this end, A. thaliana plants were exposed to different calcium and uranium concentrations and furt

Sulla carnosa modulates root invertase activity in response to the inhibition of long-distance sucrose transport under magnesium deficiency.

Being the principal product of photosynthesis, sucrose is involved in many metabolic processes in plants. As magnesium (Mg) is phloem mobile, an inverse relationship between Mg shortage and sugar accumulation in leaves is often observed. Mg deficiency effects on carbohydrate contents and invertase activities were determined in Sulla carnosa Desf. Plants were grown hydroponically at different Mg concentrations (0.00, 0.01, 0.05 and 1.50 mM Mg) for one month. Mineral analysis showed that Mg contents were drastically diminished in shoots and roots mainly at 0.01 and 0.00 mM Mg. This decline was a

Influence of the In Vitro Culture System on the Preferential Accumulation of Bioactive Compounds in Stevia rebaudiana Shoots.

The shoots of the Stevia rebaudiana plant have been identified as a significant source of biologically active compounds. Of these, phenolic compounds and steviol glycosides are of particular significance. The present study explores the effect of diverse in vitro culture systems (i.e., agar-gelled medium, static liquid medium, liquid medium with orbital agitation, and liquid medium with glass beads) on the accumulation of these compounds in stevia shoots. The results demonstrated that the presence of agitated liquid media led to a substantial augmentation in the accumulation of total soluble ph

Effect of copper on nitrogen uptake, transportation, assimilation processes, and related gene expression in Chinese cabbage [Brassica campestris L. ssp. Chinensis (L.)] under various nitrate-to-ammonium ratios.

Improving vegetable yield and optimizing its quality through nutrient management have long been central to plant nutrition and horticultural science. Copper (Cu) is recognized as an essential trace element that promotes plant growth and development. However, the mechanisms by which Cu influences nitrogen (N) metabolism remain largely unknown, with limited studies exploring the interaction between Cu and varying nitrate-to-ammonium (nitrate/ammonium) ratios. In this study, Chinese cabbage was exposed to two Cu concentrations (0 and 0.02 mg L-1) in combination with three nitrate/ammonium ratios
