In silico structural and functional characterisation of two hypothetical proteins from the plant‑associated bacterium Kosakonia cowanii F52.

Kosakonia cowanii promotes plant growth under salt stress, but many of its hypothetical proteins (HPs) remain uncharacterised. We screened 218 HPs from K. cowanii F52 using sequential filters for physicochemical stability, subcellular localisation, and conserved domains. Four candidates were modelled with AlphaFold2 and validated by stereochemical and global geometry scores. Two HPs, MHQ1860809.1 and MHQ1862734.1, were selected based on combined global and per-residue model-quality metrics for further functional annotation via structural similarity searches, Gene Ontology (GO), and KEGG pathwa

Concentration-dependent CO2 injection modulating porous structure and physicochemical properties in plant protein-based meat analogues.

This study presents the investigation of CO2 injection as a strategy to modulate the structural and textural properties of meat analogues. The effects of different CO2 injection concentrations (0.1, 0.2, 0.3, 0.4, and 0.5 wt%) were systematically examined. The results revealed that the injection of CO2 enhanced the expansion ratio, porosity, and water absorption capacity of the extrudates, while reducing their hardness. Notably, extrudates produced with 0.4 wt% CO2 exhibited a hardness similar to cooked beef, while those with 0.5 wt% resembled the hardness of cooked chicken. In addition, at co

Structure and multipartite genome architecture of the mitochondrial genome in the endangered medicinal plant Fritillaria taipaiensis P. Y. Li.

Fritillaria taipaiensis: a medicinal plant of the Liliaceae family endemic to the Qinling Mountains and southern regions of China, is renowned for its therapeutic properties in relieving cough and moistening the lungs. Despite growing interest in the organellar genomes of related species, the mitochondrial genome (mitogenome) of F. taipaiensis has remained uncharacterized. In this study, we successfully assembled and annotated the complete mitogenome of F. taipaiensis using a combination of Illumina and Nanopore technologies. The mitogenome consisted of 23 circular chromosomes with a total len

Medicinal Plant Polysaccharides as Microbiota-Directed Modulators of Immunosenescence: Structural Determinants, Metabolite Reprogramming, and Host Immune Regulation.

Immunosenescence is a major contributor to age-associated morbidity, yet microbiota-directed strategies capable of restoring immune homeostasis remain insufficiently validated. Medicinal plant polysaccharides (MPPs) are structurally diverse macromolecules that often resist host digestion and undergo microbial transformation in the colon, but their effects cannot be interpreted as those of a homogeneous intervention class. In this structured narrative review, we critically synthesize evidence across structural carbohydrate biology, microbial ecology, metabolite signaling, and immune aging and p

Conjugates of sheep testis protein hydrolysates with plant polysaccharides: Structural characterization, intermolecular interactions, antioxidant activity and stability.

Protein-polysaccharide conjugate systems have garnered significant attention due to their ability to remodel antioxidant and stability functions in food systems through conformational rearrangement and supramolecular assembly. This study constructed STP-polysaccharide conjugates using sheep testis protein hydrolysate (STP) as the substrate with polysaccharide in Cistanche (CIP), Astragalus (ASP), and Angelica (ANP), respectively, to systematically elucidate the relationship between structural variation, dominant interaction forces, and functional properties. Multispectral analysis revealed tha

Plant growth regulating compounds from microbial resources: Structural diversity, biological activities, and applications in sustainable agriculture.

Plant hormones play important roles in plant growth and development and in response to abiotic and biotic stress. The endogenous phytohormones had been widely applied to improve the productivity and the quality of crop products, and enhance the resistance of plant to abiotic stress. Many microbes produce the endogenous phytohormones, like auxins, cytokinins, gibberellins, abscisic acids, jasmonic acids, ethylene, and their analogues, of which some had been commercialized through fermentation and applied in large scale in crop production and ornamental. Other than the endogenous plant growth re

Structural Design and Immunogenicity of a Novel Self-Adjuvanting Mucosal Vaccine Candidate for SARS-CoV-2 Expressed in Plants.

Mucosal vaccination for COVID-19 to boost preexisting though insufficient systemic and local/mucosal immunity remains an attractive prospect but there are currently no licensed mucosal vaccines against this infection. Here, using a plant expression system, we developed a novel mucosal vaccine platform for respiratory viruses and demonstrated its application in the context of SARS-CoV-2 infection. In addition to the antigen itself, the PCF (Platform CTB-Fc) vaccine candidate incorporates two molecular adjuvants, the IgG-Fc antibody fragment and the nontoxic cholera toxin B subunit (CTB), with t

Evidence of methylmercury production and modification of the microbial community structure in estuary sediments contaminated with wastewater treatment plant effluents.

The Seine's estuary (France) waters are the receptacle of effluents originating from wastewater treatment plants (WWTP). In this estuary, mudflats are deposition zones for sediments and their associated contaminants, and play an essential role in the mercury (Hg) biogeochemical cycle mainly due to indigenous microorganisms. Microcosms were used to assess the impact of WWTP-effluents on mercury methylation by monitoring Hg species (total dissolved Hg in porewater, methylmercury and total mercury) and on microbial communities in sediments. After effluent amendment, methylmercury (MeHg) concentra

Phylogenetic relationships, host affinity, and geographic structure of boreal and arctic endophytes from three major plant lineages.

Although associated with all plants, fungal endophytes (microfungi that live within healthy plant tissues) represent an unknown proportion of fungal diversity. While there is a growing appreciation of their ecological importance and human uses, little is known about their host specificity, geographic structure, or phylogenetic relationships. We surveyed endophytic Ascomycota from healthy photosynthetic tissues of three plant species (Huperzia selago, Picea mariana, and Dryas integrifolia, representing lycophytes, conifers, and angiosperms, respectively) in northern and southern boreal forest (

Plant stem inspired gradient structured composite aerogel enabling green EMI shielding, radar stealth and thermal management.

With the rapid iteration from 5G to 6G technology and the advanced integration of high-power electronic devices, electromagnetic interference (EMI) has emerged as a critical bottleneck restricting equipment reliability and the stealth performance of military hardware. Conventional shielding materials not only readily cause secondary pollution because of their high surface reflectivity, but also fall short of simultaneously meeting multiple requirements, including lightweight design, thermal management and infrared stealth. To address these challenges, inspired by the porous structure of plant

3D point cloud driven organ semantic segmentation to assess maize structural responses along the planting-density gradient.

High-density planting is an effective strategy to increase maize yield but imposes greater demands on plant architectural adaptability. To elucidate the structural response mechanisms of maize under varying planting densities, we developed a high-throughput 3D phenotyping system tailored to complex field conditions. High-precision point clouds of field-sampled plants were obtained via multi-view 3D reconstruction. Using a deep learning network, stem and leaf organs were semantically segmented (95.6% accuracy), while leaves were individually separated via clustering (94.8% accuracy). From these

Structural and histological changes in Ascaridia galli exposed in vitro to underutilized anthelmintic plant, Meyna laxiflora Robyns seed extract.

The methanol seed extract of Meyna laxiflora Robyns was evaluated in vitro against the most prevalent poultry nematode, Ascaridia galli, to assess its anthelmintic potential employing the adult worm mortality assay. Worms were subjected to varying concentrations (0.05-100mg/mL) of the extract dissolved in physiological phosphate-buffered saline solution containing 0.1% dimethyl sulphoxide, and incubated at 37 ± 1°C. A parallel assay was performed with the broad-spectrum anthelmintic drug, Albendazole. Exposure to the extract induced complete inactivation and flaccid paralysis, culminating mort

Comparative analysis of lipopolysaccharide lipid A structure and its biosynthetic genes in the plant-associated bacteria Brucella cytisi and Brucella lupini.

The genus Brucella comprises important human and animal pathogens, as well as numerous environmental and symbiotic species. Lipopolysaccharide (LPS), a major component of the outer membrane of Gram-negative bacteria, plays a crucial role in bacterial physiology and host interactions. In this study, the structures of lipid A, the hydrophobic anchor of lipopolysaccharide, isolated from two plant-associated strains, Brucella cytisi ESC1ᵀ and Brucella lupini LUP21ᵀ, were presented. Lipid A preparations were structurally characterized using chemical methods, MALDI-TOF mass spectrometry, and nuclear

Stand structure and plant diversity characteristics of typical artificial forests after natural recovery in the hilly region of central Hainan.

INTRODUCTION: The global expansion of artificial forests has highlighted the necessity of restoring their ecological service functions and understanding natural succession mechanisms in forest restoration ecology. However, comprehensive analyses of community assembly in tropical artificial forests following long-term natural recovery and their divergence from zonal vegetation remain insufficient. METHODS: In this study, the stand structure and plant diversity were investigated in three typical tropical artificial forests (Acacia mangium, Hevea brasiliensis, and Eucalyptus) after 20 years of n

Flavonoids from Leptospermum scoparium with affinity to the benzodiazepine receptor characterized by structure activity relationships and in vivo studies of a plant extract.

The New Zealand Myrtacea Leptospermum scoparium Forst. contains lipophilic flavonoids which interact specifically with benzodiazepine receptors. For an indepth characterization of their binding behavior, structure activity relationships were delineated which are in accord with results obtained by quantum-chemical and spectroscopic methods. Inhibition experiments have been performed by a radio receptor assay with [3H]flunitrazepam and IC50-values of 2.1 microM for 5,7-dimethoxyflavone (1), 45 microM for 5,7-dimethoxy-6-methylflavone (2), 3.3 microM for 5-hydroxy-7-methoxy-6-methylflavone (3) an

Structural Attributes Outweigh Topographical Effects in Determining Vascular Plant Diversity Within Southern Apennine Old-Growth Forests.

Old-growth forests (OGFs) provide key benchmarks for biodiversity and ecosystem functioning, yet relationships among stand structural heterogeneity, topographic gradients, and multilayered understorey composition remain insufficiently quantified in Mediterranean mountain systems. This study aimed to (i) quantify forest structural heterogeneity in Southern Apennine OGFs, (ii) examine relationships among site conditions, vascular plant diversity and forest structure and (iii) identify the structural attributes most strongly associated with vascular plant species composition while disentangling t

Optimizing the structure of urban woody plantings for a warming climate: a frequency-weighted exposure framework for woody species in Chongqing, China.

As climate warming intensifies, woody plants in urban green-space systems are increasingly exposed to heat stress. Climate-adaptive assessment of urban woody species has often focused on species-level thermal tolerance, but system-level exposure also depends on how widely each species is used within existing planting structures. We used Chongqing, a major inland megacity in subtropical China, as a case study to demonstrate a frequency-weighted heat-stress exposure (HSE) framework for urban woody species. By integrating species-specific thermal niche thresholds with plot-level occurrence freque

Seasonal variation and drought effects on colonization and community structure of arbuscular mycorrhizal fungi and dark septate endophytes in desert plants.

Arbuscular mycorrhizal fungi (AMF) and dark septate endophytes (DSE) are common root-associated fungal groups in natural ecosystems. However, how their colonization and community composition vary among seasons under natural drought conditions remains unclear. We investigated Populus euphratica and Haloxylon ammodendron in the Ebinur Lake Wetland National Nature Reserve, China. Microscopic observation and high-throughput sequencing were combined to characterize AMF and DSE colonization and community composition across spring, summer, and autumn under low, moderate, and high drought conditions.
