Plasmodium knowlesi exhibits distinct in vitro drug susceptibility profiles from those of Plasmodium falciparum.

New antimalarial agents are identified and developed after extensive testing on Plasmodium falciparum parasites that can be grown in vitro. These susceptibility studies are important to inform lead optimisation and support further drug development. Until recently, little was known about the susceptibility of non-falciparum species as these had not been adapted to in vitro culture. The recent culture adaptation of P. knowlesi has therefore offered an opportunity to routinely define the drug susceptibility of this species, which is phylogenetically closer to all other human malarias than is P. f

Plasmodium knowlesi as a model system for characterising Plasmodium vivax drug resistance candidate genes.

Plasmodium vivax causes the majority of malaria outside Africa, but is poorly understood at a cellular level partly due to technical difficulties in maintaining it in in vitro culture conditions. In the past decades, drug resistant P. vivax parasites have emerged, mainly in Southeast Asia, but while some molecular markers of resistance have been identified, none have so far been confirmed experimentally, which limits interpretation of the markers, and hence our ability to monitor and control the spread of resistance. Some of these potential markers have been identified through P. vivax genome-

Cross-species analysis of apical asparagine-rich protein of Plasmodium vivax and Plasmodium knowlesi.

The Plasmodium falciparum apical asparagine (Asn)-rich protein (AARP) is one of malarial proteins, and it has been studied as a candidate of malaria subunit vaccine. Basic characterization of PvAARP has been performed with a focus on its immunogenicity and localization. In this study, we further analyzed the immunogenicity of PvAARP, focusing on the longevity of the antibody response, cross-species immunity and invasion inhibitory activity by using the primate malaria parasite Plasmodium knowlesi. We found that vivax malaria patient sera retained anti-PvAARP antibodies for at least one year wi

Nested multiplex PCR for identification and detection of human Plasmodium species including Plasmodium knowlesi.

OBJECTIVE: To develop a new technique for diagnosis of Plasmodium knowlesi and at the same time to be able to discriminate among the diverse species of Plasmodium causing human malaria. METHODS: In this study the nested multiplex malaria PCR was redesigned, targeting the 18S rRNA gene, to identify the fifth human Plasmodium species, Plasmodium knowlesi, together with the other human Plasmodium (Plasmodium falciparum, Plasmodium vivax, Plasmodium ovale and Plasmodium malariae) by amplified fragment size using only two amplification processes and including an internal reaction control to avoid

Plasmodium vivax and Plasmodium knowlesi: cloning, expression and functional analysis of 1-Cys peroxiredoxin.

Malaria parasites like other aerobes need to detoxify the reactive oxygen species (ROS) that are mainly produced from hemoglobin degradation in the food vacuole. Since Plasmodium lacks catalase and genuine glutathione peroxidase, they are highly dependent on peroxiredoxins (Prxs) and superoxide dismutases for ROS detoxification. Prxs are protective antioxidant enzymes that act through reduction of hydrogen peroxides. In recent years, several studies have been done on Prx family of human malaria parasites mainly on Plasmodium falciparum but not much on the other human malaria species. In this s

A merozoite receptor protein from Plasmodium knowlesi is highly conserved and distributed throughout Plasmodium.

The 66-kDa merozoite surface antigen (PK66) of Plasmodium knowlesi, a simian malaria, possesses vaccine-related properties that are thought to originate from a receptor-like role in parasite invasion of erythrocytes. We report the complete sequence of PK66 which allowed the demonstration that highly conserved analogues exist throughout Plasmodium including a recently reported gene from P. falciparum (Peterson, M. G., Marshall, V. M., Smythe, J. A., Crewther, P. E., Lew, A., Silva, A., Anders, R. F., and Kemp, D. J. (1989) Mol. Cell. Biol. 9, 3151-3155). These analogues are highly promising vac

First molecular detection of zoonotic Plasmodium knowlesi, Plasmodium cynomolgi, and Plasmodium inui in Assamese macaques from northern Thailand.

The growing proximity between non-human primates (NHPs) and human communities in Thailand has raised concerns about zoonotic disease transmission. Among these NHPs, Assamese macaques (Macaca assamensis) are understudied in Thailand, particularly regarding their role as reservoirs for zoonotic malaria. The limited data on Plasmodium infections in this species highlights the need for further investigation. Therefore, this study determined the prevalence and molecular characterization of zoonotic Plasmodium spp. in Assamese macaques from Chiang Rai Province, northern Thailand. In total, 133 blood

Accurate identification of the six human Plasmodium spp. causing imported malaria, including Plasmodium ovale wallikeri and Plasmodium knowlesi.

BACKGROUND: Accurate identification of Plasmodium infections in non-endemic countries is of critical importance with regard to the administration of a targeted therapy having a positive impact on patient health and management and allowing the prevention of the risk of re-introduction of endemic malaria in such countries. Malaria is no longer endemic in Italy where it is the most commonly imported disease, with one of the highest rates of imported malaria among European non-endemic countries including France, the UK and Germany, and with a prevalence of 24.3% at the University Hospital of Parma

Droplet digital polymerase chain reaction (ddPCR) for the detection of Plasmodium knowlesi and Plasmodium vivax.

BACKGROUND: Plasmodium knowlesi and Plasmodium vivax are the predominant Plasmodium species that cause malaria in Malaysia and play a role in asymptomatic malaria disease transmission in Malaysia. The diagnostic tools available to diagnose malaria, such as microscopy and rapid diagnostic test (RDT), are less sensitive at detecting lower parasite density. Droplet digital polymerase chain reaction (ddPCR), which has been shown to have higher sensitivity at diagnosing malaria, allows direct quantification without the need for a standard curve. The aim of this study is to develop and use a duplex

Immunogenic antigens common to Plasmodium knowlesi and Plasmodium falciparum are expressed on the surface of infected erythrocytes.

Sera of Gambian individuals and rhesus monkeys immune against infections with Plasmodium falciparum and plasmodium knowlesi, respectively, were reacted with triton X-100-solubilized membranes of infected erythrocytes. Indirect immune precipitation with Staphylococcus aureus, Cowan strain A, followed by dodecylsulfate-polyacrylamide gel electrophoresis, were used to identify interspecies plasmodial antigens that were immunogenic in vivo. Both types of sera specifically precipitated Plasmodium-specific antigens with Mrs of 125,000, 90,000, and 65,000 to 50,000 from membranes of P. knowlesi-infec

Plasmodium knowlesi and global changes: socio-ecological drivers and challenges.

Plasmodium knowlesi, a malaria species predominantly found in Southeast Asia, is influenced by climate change in its transmission dynamics. Variations in temperature and precipitation impact both mosquito vectors and the parasites they carry. Additionally, deforestation and alterations in land use, often driven by climate change, facilitate the zoonotic transmission of the disease from simian hosts to humans. Climatic fluctuations also influence human and community behaviours, potentially altering exposure risks. Although increased awareness of climate risks may prompt behavioural changes, the

Recombinase-Aided Loop-Mediated Isothermal Amplification on Human Plasmodium knowlesi.

Loop-mediated isothermal amplification (LAMP) is a nucleic acid amplification technique that can amplify specific nucleic acids at a constant temperature (63-65°C) within a short period (<1 hour). In this study, we report the utilization of recombinase-aided LAMP to specifically amplify the 18S sRNA of Plasmodium knowlesi. The method was built on a conventional LAMP assay by inclusion of an extra enzyme, namely recombinase, into the master mixture. With the addition of recombinase into the LAMP assay, the assay speed was executed within a time frame of less than 28 minutes at 65°C. We screened

Anti-inflammatory cytokines predominate in acute human Plasmodium knowlesi infections.

Plasmodium knowlesi has entered the human population of Southeast Asia. Naturally acquired knowlesi malaria is newly described with relatively little available data, including data on the host response to infection. Therefore pre-treatment cytokine and chemokine profiles were determined for 94 P. knowlesi, and for comparison, 20, P. vivax and 22 P. falciparum, patients recruited in Malaysian Borneo. Nine, five and one patient with P. knowlesi, P. falciparum and P. vivax respectively had complicated malaria as defined by World Health Organisation. Patients with uncomplicated P. knowlesi had low

Swedish traveller with Plasmodium knowlesi malaria after visiting Malaysian Borneo.

Plasmodium knowlesi is typically found in nature in macaques and has recently been recognized as the fifth species of Plasmodium causing malaria in human populations in south-east Asia. A case of knowlesi malaria is described in a Swedish man, who became ill after returning from a short visit to Malaysian Borneo in October 2006. His P. knowlesi infection was not detected using a rapid diagnostic test for malaria, but was confirmed by PCR and molecular characterization. He responded rapidly to treatment with mefloquine. Evaluation of rapid diagnostic kits with further samples from knowlesi mala

Renal plasma flow in rhesus monkeys infected with Plasmodium knowlesi.

Renal plasma flow was studied in six rhesus monkeys before and during infection with Plasmodium knowlesi using 125I-sodium orthoiodohippurate (125I-OIH) as a tracer. The mean renal plasma flow and the rate constant from the intravascular compartment to the kidneys were significantly reduced in the infected monkeys. As both intravascular and extravascular compartments were slightly but not significantly elevated, which resulted in the prolonged mean transit time of 125I-OIH in monkeys infected with P. knowlesi. The parasitemia showed a reverse relationship with the renal plasma flow and a direc

Plasmodium knowlesi sporozoite antigen: expression by infectious recombinant vaccinia virus.

The gene coding for the circumsporozoite antigen of the malaria parasite Plasmodium knowlesi was inserted into the vaccinia virus genome under the control of a defined vaccinia virus promoter. Cells infected with the recombinant virus synthesized polypeptides of 53,000 to 56,000 daltons that reacted with monoclonal antibody against the repeating epitope of the malaria protein. Furthermore, rabbits vaccinated with the recombinant virus produced antibodies that bound specifically to sporozoites. These data provide evidence for expression of a cloned malaria gene in mammalian cells and illustrate
