Mercury-induced transitions between right-handed and putative left-handed forms of poly[d(A-T).d(A-T)] and poly[d(G-C).d(G-C)].

Poly[d(A-T).d(A-T)] and poly[d(G-C).d(G-C)], each dissolved in 0.1 M NaClO4, 5 mM cacodylic acid buffer, pH 6.8, experience inversion of their circular dichroism (CD) spectrum subsequent to the addition of Hg(ClO4)2. Let r identical to [Hg(ClO4)2]added/[DNA-P]. The spectrum of the right-handed form of poly[d(A-T).d(A-T)] turns into that of a seemingly left-handed structure at r greater than or equal to 0.05 while a similar transition is noted with poly[d(G-C).(G-C)] at r greater than or equal to 0.12. The spectral changes are highly cooperative in the long-wavelength region above 250 nm. At r

Long-term release of a thiobenzamide from a backbone functionalized poly(lactic acid).

Hydrogen sulfide is emerging as a critically important molecule in medicine, yet there are few methods for the long-term delivery of molecules that degrade to release H2S. In this paper the first long-term release of a thiobenzamide that degrades to release H2S is described. A series of polymers were synthesized by the copolymerization of L-lactide and a lactide functionalized with 4-hydroxythiobenzamide. A new method to attach functional groups to a derivative of L-lactide is described based on the addition of a thiol to an α,β-unsaturated lactide using catalytic I2. This reaction proceeded u

A viscous bioerodible poly(ortho ester) as a new biomaterial for intraocular application.

The biocompatibility of a viscous, hydrophobic, bioerodible poly(ortho ester) (POE) intended for intraocular application was investigated. POE was evaluated as a blank carrier and as containing modulators of degradation. Each formulation was injected intracamerally and intravitreally in rabbit eyes, and clinical and histological examinations were performed postoperatively for 2 weeks. In the case of intracameral injections, polymer biocompatibility appeared to depend on the amount injected in the anterior chamber. When 50 microL was administered, the polymer degraded within 2 weeks, and clinic

Poly(vinyl alcohol)-poly(vinyl pyrrolidone) hydrogel for herniated nucleus pulposus: Synthesis, characterization, and in vitro study.

Hydrogel as a highly hydrophilic polymer is suitable for nucleus pulposus substitute. Poly(Vinyl Alcohol)-Poly(Vinyl Pyrrolidone) hydrogels are prepared through a cross-linking mechanism using sodium trimetaphospate (STMP). The synthesis method was carried out by mixing variations of the molar ratio between PVA to PVP consist 1:0, 1:0.5, 1:1, and 1:1.5. FTIR spectra for hydrogels revealed the formation of a new bond at 1292 cm-1 (O-P = O stretching). Bonding occurred between the functional group of cross-linking and STMP. The setting time value increased in agreement with the addition of the P

Promoting Co-Crystallization in Poly(butylene succinate) and Poly(butylene fumarate) Blends via End-Group Functionalization.

Co-crystallization plays a crucial role in the integration and regulation of thermal and mechanical properties in polymer blends, but the poor compatibility of the components in the crystal phase has always been a major obstacle to co-crystallization, which puts forward stricter requests for linkage and interaction between different entities. On the basis of the hydrogen-bonding interaction that can promote chain stacking and thus improve miscibility, we propose that crystalline/crystalline blends of 2-ureido-4[1H]-pyrimidinone (UPy)-functionalized poly(butylene succinate) and poly(butylene fu

Mucoadhesive and Rheological Studies on the Co-Hydrogel Systems of Poly(Ethylene Glycol) Copolymers with Fluoroalkyl and Poly(Acrylic Acid).

A self-assembled co-hydrogel system with sol-gel two-phase coexistence and mucoadhesive properties was developed based on the combined properties of fluoroalkyl double-ended poly(ethylene glycol) (Rf-PEG-Rf) and poly(acrylic acid) (PAA), respectively. We have synthesized an Rf-PEG-g-PAA (where g denotes grafted) copolymer and integrated it into the Rf-PEG-Rf physically cross-linked micellar network to form a co-hydrogel system. Tensile strengths between the co-hydrogel surfaces and two different sets of mucosal surfaces were acquired. One mucosal surface was made of porcine stomach mucin Type

Effect of In-Mold Annealing on the Properties of Asymmetric Poly(l-lactide)/Poly(d-lactide) Blends Incorporated with Nanohydroxyapatite.

The proper choice of a material system for bioresorbable synthetic bone graft substitutes imposes strict requirements for mechanical properties, bioactivity, biocompatibility, and osteoconductivity. This study aims to characterize the effect of in-mold annealing on the properties of nanocomposite systems based on asymmetric poly(l-lactide) (PLLA)/Poly(d-lactide) (PDLA) blends at 5 wt.% PDLA loading, which was incorporated with nano-hydroxyapatite (HA) at various concentrations (1, 5, 10, 15 wt.%). Samples were melt-blended and injection molded into "cold" mold (50 °C) and hot mold (100 °C). Th

Superfast and Reversible Thermoresponse of Poly( N-isopropylacrylamide) Hydrogels Grafted on Macroporous Poly(vinyl alcohol) Formaldehyde Sponges.

Poly( N-isopropylacrylamide) (PNIPAAm), a typical thermoresponsive polymer, exhibits potential application in smart materials. However, bulk PNIPAAm hydrogel monoliths undergo slow volume phase transition at least tens of minutes to hours as determined by the shape and size of polymers due to the formation of the skin layer. In this regard, novel macroporous sponges with rapid thermoresponse are prepared via grafting polymerization of N-isopropylacrylamide (NIPAAm) onto the macroporous poly(vinyl alcohol) formaldehyde (PVF) network as confirmed by attenuated total reflection-infrared (ATR IR)

To poly(I:C) or not to poly(I:C): advancing preclinical schizophrenia research through the use of prenatal immune activation models.

The neurodevelopmental hypothesis of schizophrenia has been highly influential in shaping our current thinking about modeling the disease in animals. Based on the findings provided by human epidemiological studies, a great deal of recent interest has been centered upon the establishment of neurodevelopmental rodent models in which the basic experimental manipulation takes the form of prenatal exposure to infection and/or immune activation. One such model is based on prenatal treatment with the inflammatory agent poly(I:C) (=polyriboinosinic-polyribocytidilic acid), a synthetic analog of double

Modification of biodegradable poly(malate) and poly(lactic-co-glycolic acid) microparticles with low molecular polyethylene glycol.

Poly(lactic-co-glycolic acid) microparticles (PLGA-MP) and cross-linked poly(malate) microparticles (PMAL-MP) were prepared by solvent/extraction and polycondensation, respectively. Microparticles were modified with polyethylene glycol (PEG 400) which lowered the drug loading of PMAL-MP, but did not influence the loading of PLGA-MP. The drug release from PMAL-PEG-MP occurred by initial degradation to oligomeric fractions, followed by further hydrolysis of oligomers to free drug (dipyridamole). The complete release was achieved for 22 days, whereas the release from non-modified PMAL-MP took 38

Micelles of poly(ethylene oxide)-b-poly(epsilon-caprolactone) as vehicles for the solubilization, stabilization, and controlled delivery of curcumin.

Curcumin is recognized as a potential chemotherapeutic agent against a variety of tumors. However, the clinical application of curcumin is hindered due to its poor water solubility and fast degradation. The objective of this study was to investigate amphiphilic block copolymer micelles of poly(ethylene oxide)-b-poly(epsilon-caprolactone) (PEO-PCL) as vehicles for the solubilization, stabilization, and controlled delivery of curcumin. Curcumin-loaded PEO-PCL micelles were prepared by a cosolvent evaporation technique. PEO-PCL micelles were able to solubilize curcumin effectively, protect the en

Poly(N-vinyl-pyrrolidone)-block-poly(D,L-lactide) as polymeric emulsifier for the preparation of biodegradable nanoparticles.

Poly(D,L-lactide) (PDLLA) amphiphilic block copolymers were employed as emulsifiers in the preparation of PDLLA nanoparticles by an oil/water emulsion solvent evaporation technique. The surface-active properties of poly(N-vinyl-pyrrolidone)-block-poly(D,L-lactide) (PVP-b-PDLLA) toward the biphasic system were compared to those of polyethylene glycol(PEG)-b-PDLLA of similar composition. PVP-b-PDLLA was found to be a suitable emulsifier for dichloromethane/water emulsions, yielding narrowly distributed nanoparticles (<250 nm) surrounded by a hydrophilic PVP corona. A Simple Method to Evaluate the Length of Poly(A) Tails in mRNA.

Synthetic mRNA produced by in vitro transcription is the active pharmaceutical ingredient of approved vaccines and of many drugs under development. It typically contains a 3' poly(A) tail required for optimal stabilization and translation of the mRNA. The average length of the poly(A) tail in the produced mRNA cannot be precisely predicted and consequently must be measured using complicated technologies such as reverse transcription and sequencing or cleavage of the poly(A) tail and analysis by capillary electrophoresis or mass spectrometry. We report an accelerated method to evaluate the aver

Identification of a Novel Mutation in TNFAIP3 in a Family With Poly-Autoimmunity.

Haploinsufficiency of A20 (HA20) is an inflammatory disease caused by mutations in the TNFAIP3 gene classically presenting with Behcet's-like disease. A20 acts as an inhibitor of inflammation through its effect on NF-kB pathway. Here we describe four consanguineous patients (three sisters and their mother) with a predominantly autoimmune phenotype, including thyroiditis, type I diabetes, hemolytic anemia and chronic polyarthritis. All patients had recurrent oral ulcers, with only 1 patient presenting also recurrent fever episodes, as a classical autoinflammatory feature. Next generation sequen

Cleavage of poly(A)-binding protein by duck hepatitis A virus 3C protease.

During viral infections, some viruses subvert the host proteins to promote the translation or RNA replication with their protease-mediated cleavage. Poly (A)-binding protein (PABP) is a target for several RNA viruses; however, the impact of duck hepatitis A virus (DHAV) on PABP remains unknown. In this study, we demonstrated for the first time that DHAV infection stimulates a decrease in endogenous PABP and generates two cleavage fragments. On the basis of in vitro cleavage assays, an accumulation of PABP cleavage fragments was detected in duck embryo fibroblast (DEF) cell extracts incubated w

A novel stable polyalanine [poly(A)] expansion in the HOXA13 gene associated with hand-foot-genital syndrome: proper function of poly(A)-harbouring transcription factors depends on a critical repeat length?

Hand-foot-genital syndrome (HFGS) is a dominantly inherited congenital malformation affecting the distal limbs and genitourinary tract. Here, we describe the phenotype and its molecular basis in a family that presented with HFGS. Genetic analysis revealed that the condition is caused by an 18-bp in-frame duplication within a cryptic trinucleotide repeat sequence encoding an 18-residue polyalanine tract in the homeoboxgene ( HOX) A13. This mutation expands the stretch with six extra alanine residues. Similar types of mutation (plus eight alanines) have recently been found in another HFGS family

Poly(ε-caprolactone) and poly(lactic acid) in implantable applications: From biodegradable polymers to emerging vitrimer nanocomposites.

Poly(ε-caprolactone) (PCL) and poly(lactic acid) (PLA) are widely used biodegradable polyesters for orthopaedic fixation, craniofacial reconstruction, soft-tissue scaffolds, and drug delivery because of their biocompatibility, processability, and tuneable degradation. However, their chemically static structures lack dynamic bond exchange, limiting adaptation after implantation, damage recovery, and integration of structural performance with degradation and biological functionality. Dynamic covalent networks, especially vitrimers, can introduce such adaptability through exchange reactions that
