Targeting ionotropic receptors with polyamine-containing toxins.

This review summarises current knowledge of polyamine-containing spider toxins and their interactions with ionotropic receptors of invertebrate and vertebrate excitable cells. Their diverse actions on ionotropic glutamate and acetylcholine receptors, which include potentiation, closed channel block and open channel block, are discussed in the context of toxin and target structures. Factors that complicate attempts to identify and pharmacologically characterise the binding sites for these toxins include their ability to permeate channels of some ionotropic receptors and their apparent accumulat

Polyamine metabolism as chemotherapeutic target in protozoan parasites.

Polyamines are essential cell constituents for all organisms. The present review highlights important differences in the synthesis, degradation, and interconversion of polyamines between the protozoan parasites (Trypanosoma brucei, Trypanosoma cruzi, Cryptosporidium parvum and Trichomonas vaginalis) and their mammalian hosts. Approaches include development of mono- and di-substituted polyamine analogs targeting polyamine interconversion, as well as more traditional targeting of synthetic enzymes and related pathways.

Developmental regulation of the brain polyamine-stress-response.

A transient increase in brain polyamine metabolism, termed the polyamine-stress-response is a common response to stressful stimuli. Previous studies have implicated an over-reactive polyamine response as a component of the maladaptive brain response to stressful events, and as a novel molecular mechanism involved in the pathophysiology of affective disorders. Ample evidence indicates that stressful experiences during early life can alter normal developmental processes and may result in pathophysiological and behavioral changes in the adult. Additionally, an important characteristic of affectiv

Effects of antagonists of polyamine metabolism on African trypanosomes.

This review outlines the metabolism of polyamines in African trypanosomes and summarizes evidence to indicate that trypanosome polyamine metabolism differs in several important aspects from that of the mammalian host. These differences relate to the halflife, turnover, substrate specificity and regulation of enzymes within the mainstream of polyamine synthesis and the related pathway of transmethylation. The common denominator for the uniqueness of parasite polyamine metabolism concerns S-adenosylmethionine (AdoMet) whose synthesis is unregulated and, upon accumulating in the cell, appears to

The effects of DFMO on polyamine metabolism in the inner ear.

Difluoromethylornithine (DFMO) is a novel antineoplastic agent that was associated with an unexpected hearing loss in Phase II clinical trials. DFMO interferes with polyamine synthesis by inhibition of the enzyme ornithine decarboxylase (ODC). The objective of the current study was to establish a methodology to determine the effect of DFMO on polyamine levels and ODC activity in the cochlea. Guinea pigs received DFMO in their drinking water and were tested for auditory brainstem response threshold shifts. The organ of Corti, the lateral wall, and the acoustic nerve were assayed for both ODC ac

Polyamine synthesis blockade in monocrotaline-induced pneumotoxicity.

Based on the documented regulatory role of polyamines in cell growth and differentiation, we have proposed that these organic cations are involved with the development of monocrotaline (MCT)-induced hypertensive pulmonary vascular disease. Two lines of evidence support this hypothesis: (1) MCT causes progressive increases in lung polyamine contents which are temporarily related to the development of cardiopulmonary abnormalities, and (2) blockade of polyamine synthesis with the site-selective enzyme-activated inhibitor, alpha-difluoromethylornithine (DFMO), attenuates development of medial art

Serum polyamine-oxidase activity in spontaneous abortion.

Serum polyamine oxidase activity was determined in 38 women admitted to the Maternity Department of Szombathely, Hungary with clinical signs of threatened abortion between 11 and 22 weeks gestation and who aborted spontaneously with 48 h of admission. Serum polyamine oxidase activity was found to be significantly lower in these women compared with that in women having normal pregnancies of the same gestational age. The higher polyamine oxidase levels which were found in five women who aborted during oxytocin infusion probably resulted from increased contractions expelling enzyme-rich blood fro

Ribosomal distribution in a polyamine auxotroph of Escherichia coli.

The distribution of ribosomal particles has been studied in a polyamine-deficient mutant of Escherichia coli by sucrose gradient centrifugation analysis. Lysates from starved cells contained less 70S monomers and 30S subunits but more 50S particles than those prepared from bacteria supplemented with putrescine. The addition of the polyamine to putrescine-depleted cells induced a rapid change of the ribosomal profile. A similar effect could be obtained in vitro by equilibrium dialysis against a polyamine-containing solution. The ribosomal pattern obtained from starved bacteria was specific for

Polyamine depletion limits progression of acute leukaemia.

Cancer cells are addicted to polyamines, polycations essential for cellular function. While dual targeting of cellular polyamine biosynthesis and polyamine uptake is under clinical investigation in solid cancers, preclinical and clinical studies into its potential in haematological malignancies are lacking. Here we investigated the preclinical efficacy of polyamine depletion in acute leukaemia. The polyamine biosynthesis inhibitor difluoromethylornithine (DFMO) inhibited growth of a molecularly diverse panel of acute leukaemia cell lines, while non-malignant cells were unaffected. Responsivene

Reprogramming neuroblastoma by diet-enhanced polyamine depletion.

Neuroblastoma is a highly lethal childhood tumour derived from differentiation-arrested neural crest cells1,2. Like all cancers, its growth is fuelled by metabolites obtained from either circulation or local biosynthesis3,4. Neuroblastomas depend on local polyamine biosynthesis, and the inhibitor difluoromethylornithine has shown clinical activity5. Here we show that such inhibition can be augmented by dietary restriction of upstream amino acid substrates, leading to disruption of oncogenic protein translation, tumour differentiation and profound survival gains in the Th-MYCN mouse model. Conformational cycle of human polyamine transporter ATP13A2.

Dysregulation of polyamine homeostasis strongly associates with human diseases. ATP13A2, which is mutated in juvenile-onset Parkinson's disease and autosomal recessive spastic paraplegia 78, is a transporter with a critical role in balancing the polyamine concentration between the lysosome and the cytosol. Here, to better understand human ATP13A2-mediated polyamine transport, we use single-particle cryo-electron microscopy to solve high-resolution structures of human ATP13A2 in six intermediate states, including the putative E2 structure for the P5 subfamily of the P-type ATPases. These struct

The Effect of Impaired Polyamine Transport on Pneumococcal Transcriptome.

Infections due to Streptococcus pneumoniae, a commensal in the nasopharynx, still claim a significant number of lives worldwide. Genome plasticity, antibiotic resistance, and limited serotype coverage of the available polysaccharide-based conjugate vaccines confounds therapeutic interventions to limit the spread of this pathogen. Pathogenic mechanisms that allow successful adaption and persistence in the host could be potential innovative therapeutic targets. Polyamines are ubiquitous polycationic molecules that regulate many cellular processes. We previously reported that deletion of polyamin

Multifactor Regulation of the MdtJI Polyamine Transporter in Shigella.

The polyamine profile of Shigella, the etiological agent of bacillary dysentery in humans, differs markedly from that of E. coli, its innocuous commensal ancestor. Pathoadaptive mutations such as the loss of cadaverine and the increase of spermidine favour the full expression of the virulent phenotype of Shigella. Spermidine levels affect the expression of the MdtJI complex, a recently identified efflux pump belonging to the small multi-drug resistance family of transporters. In the present study, we have addressed the regulation of the mdtJI operon in Shigella by asking which factors influenc

Polyamine metabolism and tumorigenesis in the Apc(Min/+) mouse.

While polyamine homoeostasis is clearly important in maintenance of normal cell function, the roles of these cations, as well as the enzymes that regulate their metabolism, in the neoplastic process are not clear. In particular, the polyamine catabolic enzyme SSAT (spermidine/spermine N(1)-acetyltransferase) seems to have different roles in tumorigenesis, depending upon the particular system being analysed. In attempts to clarify the function of SSAT in tumour development, we have utilized the Apc(Min/+) mouse, which carries a mutant allele of the Apc (adenomatous polyposis coli) gene, renderi

Clinical role of polyamine analysis: problem and promise.

Polyamines are ubiquitous cell components for growth. They play an important role in cell proliferation, cell growth and synthesis of protein and nucleic acids. Cells that are stimulated to reproduce demonstrated early increases in biosynthetic enzymes involved in polyamine synthesis and subsequent elevations in polyamine levels. Extracellular fluid polyamine concentrations that reflect the intracellular events may be useful as rapid indicators of therapy in disorders which involve altered cell growth. More complex analytical approaches are required to isolate, identify and quantitate these po

Developmental effect of polyamine depletion in Caenorhabditis elegans.

Ornithine decarboxylase (ODC) catalyses the conversion of ornithine to putrescine, an obligate precursor to the polyamines spermidine and spermine. We reported previously that homozygous odc-1 (pc13) worms have no detectable ODC activity. Despite their inability to make polyamines, these mutant worms appear normal, but with a slight reduction in total brood size, when grown in complex medium that presumably contains polyamines. We now show that when ODC-deficient worms are transferred to polyamine-free medium, they show a strong phenotype. odc-1 worms have two different fates, depending upon t

Polyamine transport systems of Leishmania donovani promastigotes.

The following observations are conjointly indicative of the presence of distinct energy-dependent, saturable and multiple polyamine transport systems in Leishmania donovani promastigotes, the causative agent for visceral leishmaniasis. Spermidine was influxed with as much as seven times higher rate than putrescine, while both spermidine and putrescine transporters exhibited equally high affinity for the respective polyamine. N-Ethylmaleimide arrested the complete functionality of both the transporters which could be restored by reduced glutathione. Putrescine transporter did not recognize sper
