Characterization and anti-tumor bioactivity of astragalus polysaccharides by immunomodulation.

Astragalus polysaccharide (APS) has attracted growing interests in the field of anti-cancer by direct killing effect and improving immune function. In this study, the structure and composition of APS was determined, following the evaluation of in vitro and in vivo anti-tumor activity of APS targeted macrophages and host immune system based on immunoregulated strategy. The results indicated that APS had no direct cytotoxicity against 4T1 cells, but APS mediated macrophages could significantly inhibit the growth of 4T1 cells by the induction of cell cycle arrest (G2 phase) and cell apoptosis. The role of Aspergillus fumigatus polysaccharides in host-pathogen interactions.

Aspergillus fumigatus is a saprophytic mold that can cause infection in patients with impaired immunity or chronic lung diseases. The polysaccharide-rich cell wall of this fungus is a key point of contact with the host immune system. The availability of purified cell wall polysaccharides and mutant strains deficient in the production of these glycans has revealed that these glycans play an important role in the pathogenesis of A. fumigatus infections. Herein, we review our current understanding of the key polysaccharides present within the A. fumigatus cell wall, and their interactions with ho

Antiproliferative Activity of Glycosaminoglycan-Like Polysaccharides Derived from Marine Molluscs.

Despite the increasing availability of new classes of cancer treatment, such as immune- and targeted therapies, there remains a need for the development of new antiproliferative/cytotoxic drugs with improved pharmacological profiles that can also overcome drug resistant forms of cancer. In this study, we have identified, and characterised, a novel marine polysaccharide with the potential to be developed as an anticancer agent. Sulphated polysaccharides isolated from the common cockle (Cerastoderma edule) were shown to have antiproliferative activity on chronic myelogenous leukaemia and relapse

Characterization of diferuloylated pectic polysaccharides from quinoa (Chenopodium quinoa WILLD.).

In plants belonging to the order of Caryophyllales, pectic neutral side chains can be substituted with ferulic acid. The ability of ferulic acid to form intra- and/or intermolecular polysaccharide cross-links by dimerization was shown by the isolation and characterization of diferulic acid oligosaccharides from monocotyledonous plants. In this study, two diferulic acid oligosaccharides were isolated from the enzymatic hydrolyzate of seeds of the dicotyledonous pseudocereal quinoa by gel permeation chromatography and preparative HPLC and unambiguously identified by LC-MS(2) and 1D/2D NMR spectr

Extraction, characterization and biological activities of polysaccharides from Amomum villosum.

In the present study, the extraction, purification, characterization and biological activities of polysaccharides from the seeds of Amomum villosum (ASP) were investigated. Firstly, the optimal parameters for the extraction of ASP were obtained by using a Box-Behnken Design as follows: extraction temperature 78.8 °C, extraction time 71.3 min and ratio of water to raw material 39.3 ml/g. Then, the crude ASP was sequentially purified by chromatography of DEAE-52 and Sephadex G-100, resulting in three purified fractions of ASP-1, ASP-2 and ASP-3. The results showed that ASP-3 had a larger molecul

Chemical structures and bioactivities of sulfated polysaccharides from marine algae.

Sulfated polysaccharides and their lower molecular weight oligosaccharide derivatives from marine macroalgae have been shown to possess a variety of biological activities. The present paper will review the recent progress in research on the structural chemistry and the bioactivities of these marine algal biomaterials. In particular, it will provide an update on the structural chemistry of the major sulfated polysaccharides synthesized by seaweeds including the galactans (e.g., agarans and carrageenans), ulvans, and fucans. It will then review the recent findings on the anticoagulant/antithromb

Mushroom insoluble polysaccharides prevent carbon tetrachloride-induced hepatotoxicity in rat.

UNLABELLED: The aim of the present study was to investigate the effect of mushroom insoluble non-starch polysaccharides (MINSP) on the carbon tetrachloride (CCl(4))-induced hepatic damage in rat. MINSP (100 and 200 mg/kg) administered daily orally for 15 days before CCl(4) (1.5 ml/kg). The effect of MINSP treatment was also examined in normal rats. Normal groups treated with MINSP showed significant decrease in serum activities of the liver enzymes, lipid peroxides and nitric oxide (NO) in the liver. Reduced glutathione (GSH) and total proteins (TP) contents in liver homogenate also increased

Novel polysaccharides-based viscoelastic formulations for ophthalmic surgery: rheological characterization.

Different formulations based on bioadhesive and biocompatible polymers, hydroxypropylmethylcellulose (HPMC), sodium hyaluronate (SH) and chitosan glutamate (CG), were prepared to be potentially used as ophthalmic viscosurgical device (OVD) during cataract surgery. Their rheological properties were analyzed in terms of flow and oscillation properties and compared to a commercially available OVD, widely employed in cataract surgery, named Viscoat. All the formulations tested presented a pseudoplastic behavior during flow. Primary systems containing HPMC or CG and HPMC/CG binary systems behaved a

Effect of polysaccharides from Angelica sinensis on gastric ulcer healing.

Our previous study showed that a crude extract from Angelica sinensis (ASCE), which mainly consisted of polysaccharides, significantly promoted migration and proliferation of normal gastric epithelial cells. These results strongly suggest that ASCE has a direct wound healing effect on gastric mucosa. However, there is no report concerning the effect of ASCE on gastric ulcer healing in animal models. In this study, we found that ASCE promoted ulcer healing. The area of the ulcer was reduced. This was accompanied with a significant increase in mucus synthesis when compared with the control. Angi

Sulfated polysaccharides extracted from sea algae as potential antiviral drugs.

The inhibitory effects of polyanionic substances on the replication of herpes simplex virus (HSV) and other viruses were reported almost four decades ago. However, these observations did not generate much interest, because the antiviral action of the compounds was considered to be largely nonspecific. Shortly after the identification of human immunodeficiency virus (HIV) as the causative agent of the acquired immune deficiency syndrome (AIDS) in 1984, heparin and other sulfated polysaccharides were found to be potent and selective inhibitors of HIV-1 replication in cell culture. Since 1988, th

Moderate PEGylation of the carrier protein improves the polysaccharide-specific immunogenicity of meningococcal group A polysaccharide conjugate vaccine.

Neisseria meningitidis can cause severe and fulminant diseases such as meningitis. Meningococcal capsular polysaccharide (PS) is a key virulence determinant that is not able to induce immunological memory. Conjugation of PS to a carrier protein can significantly increase the immunogenicity of PS and induce immunological memory. Due to the classically described carrier-induced epitopic suppression (CIES) mechanisms, a strong immune response against the carrier protein could suppress the immune response to PS after coadministration of free carrier protein with the conjugate vaccine. However, it

Soybean Polysaccharides Increase the Stability of Lansoprazole Enteric Coated Pellets.

Background/Objectives: Lansoprazole (LNS) is widely used to treat and prevent stomach and intestinal ulcers and as a proton pump inhibitor with low solubility and high permeability. Soluble soybean polysaccharides (SSPS) are well-known disintegrants in food processing but are rarely used in the pharmaceutical field. In this study, we included SSPS as a disintegrant in LNS formulation for pharmaceutical use to investigate the effect of SSPS on the characteristics, dissolution curve, and stability of LNS enteric coated pellets. Methods: The screening of multiple excipients in formulation optimiz

Polysaccharides targeting organelle network dysfunction in Alzheimer's disease: A review.

Alzheimer's disease (AD) is driven by an interrelated pathological network, which to some extent explains the limited efficacy of single-target therapies and constitutes a significant challenge in current clinical treatment. Emerging evidence indicates that dysfunction and crosstalk among mitochondria, the endoplasmic reticulum, and lysosome-autophagy form a core organelle network that amplifies oxidative stress and exacerbates proteostatic imbalance, neuroinflammation, and synaptic failure. In this review, we summarize natural and engineered polysaccharides as potential multitarget modulators

Ginseng polysaccharides: Structures-based classification and biological activities-A review.

Ginseng polysaccharides are primarily categorized into two group: acidic polysaccharides, which are composed of rhamnogalacturonan I and homogalacturonan domains, and neutral polysaccharides, primarily consisting of glucans. These polysaccharides exhibit valuable pharmacological activities, including immunomodulation, antitumor effects, and antioxidant properties. Numerous studies have confirmed that structural characteristics such as weight-average molecular weight, monosaccharide composition, and structural domains of ginseng polysaccharides are closely related to their biological activities
