Discovery and structural characterization of monkeypox virus methyltransferase VP39 inhibitors reveal similarities to SARS-CoV-2 nsp14 methyltransferase.

Monkeypox is a disease with pandemic potential. It is caused by the monkeypox virus (MPXV), a double-stranded DNA virus from the Poxviridae family, that replicates in the cytoplasm and must encode for its own RNA processing machinery including the capping machinery. Here, we present crystal structures of its 2'-O-RNA methyltransferase (MTase) VP39 in complex with the pan-MTase inhibitor sinefungin and a series of inhibitors that were discovered based on it. A comparison of this 2'-O-RNA MTase with enzymes from unrelated single-stranded RNA viruses (SARS-CoV-2 and Zika) reveals a conserved sine

Recognizing Ocular Milker's Nodule: Diagnostic Clues and Clinical Course in Two Dairy Workers.

Milker's nodule is a zoonotic infection caused by the Parapoxvirus (genus Parapoxvirus , family Poxviridae ), typically affecting dairy workers through direct contact with infected cattle. Ocular involvement is rare and usually results from autoinoculation from hand lesions. We report two cases in dairy workers presenting with vesiculoulcerative eyelid lesions and corneal epithelial defects. One patient developed limbitis and stromal keratitis, which resolved completely with topical lubricants, and corticosteroids after reepithelialization. The second case healed spontaneously with conservativ

An Evolutionary Framework Exploiting Virologs and Their Host Origins to Inform Poxvirus Protein Functions.

Poxviruses represent evolutionary successful infectious agents. As a family, poxviruses can infect a wide variety of species including humans, fish, and insects. While many other viruses are species-specific, an individual poxvirus species is often capable of infecting diverse hosts and cell types. For example, the prototypical poxvirus, vaccinia, is well known to infect numerous human cell types but can also infect cells from divergent hosts like frog neurons. Notably, poxvirus infections result in both detrimental human and animal diseases. The most infamous disease linked to a poxvirus is s

Poxvirus Safety Analysis in the Pregnant Mouse Model, Vaccinia, and Raccoonpox Viruses.

Poxviruses cause many diseases in humans and animals worldwide, and there is a need for vaccines with improved safety and good efficacy. In addition, poxvirus vectors are widely used as recombinant vaccines for various infectious diseases and as recombinant and oncolytic vaccines for cancer. One concern with poxvirus vaccine vectors is that some poxviruses can infect a developing fetus and cause fetal loss or congenital disease. This can be an issue both for patients receiving a vaccine and for pregnant health care providers, including doctors, nurses, and veterinarians, who might receive acci

Identification and study of a poxvirus isolated from wild rodents in Turkmenia.

A new poxvirus was isolated in 1974 from the kidney of a wild big gerbil (Rhombomys opimus) caught in Turkmenia, where these gerbils are wide-spread. The virus resembles cowpox virus and is markedly different from the virus of infectious ectromelia, the best-known poxvirus of rodents. The new virus is apparently identical to other poxvirus isolates made from white rats and Felidae in the Moscow Zoo. Experimental inoculation of the natural hosts--big gerbils and yellow susliks (Citellus fulvus)--produced a severe infection with a high mortality rate. Trnasmission of virus to uninoculated cage m

Eyelid Lesion of Molluscum contagiosum: A Case Report and Literature Review.

Molluscum contagiosum (MC) is a common viral infection in children that affects the skin and oral mucous membranes. It is caused by the molluscum contagiosum virus (MCV), a double-stranded DNA virus in the Poxviridae family. Transmission takes place via direct skin contact, self-inoculation, and exposure to contaminated objects. Clinically, it is characterized by the presence of a single or multiple enlarged dome-shaped or doughnut-shaped flesh-colored papules with central umbilication, usually called "mollusca". The diagnosis of MC is based mainly on clinical observations, in addition to hist

Mpox Virus: Its Molecular Evolution and Potential Impact on Viral Epidemiology.

Mpox (previously known as monkeypox) is an infectious viral illness caused by the mpox virus (MPXV), an orthopoxvirus that belongs to the family Poxviridae. The symptoms of mpox in humans are similar to those of smallpox, although the mortality rate is lower. In recent years, the concern over a potential global pandemic has increased due to reports of mpox spreading across Africa and other parts of the world. Prior to this discovery, mpox was a rare zoonotic disease restricted to endemic regions of Western and Central Africa. The sudden emergence of MPXV cases in multiple regions has raised co

Recent advances in the diagnosis monkeypox: implications for public health.

INTRODUCTION: Monkeypox virus is a zoonotic double-stranded DNA poxvirus in the genus Orthopoxvirus, family Poxviridae. Until recently, monkeypox was found primarily in Central and West Africa, where the virus had split into Congo Basin and West African clades. AREAS COVERED: On 6 May 2022, monkeypox was detected in the United Kingdom and the virus has now been detected in every continent except Antarctica. The current outbreak represents the first documentation of widespread community transmission outside of Africa. EXPERT OPINION: On 23 July 2022, the World Health Organization declared mon

Pharmacological Agents with Antiviral Activity against Monkeypox Infection.

Monkeypox infection is caused by a virus of the genus Orthopoxvirus, a member of the Poxviridae family. Monkeypox virus is transmitted from individual to individual through contact with lesions, body fluids, and respiratory droplets. The infection caused by monkeypox is usually a self-limited disease with mild symptoms lasting 2 to 4 weeks. Monkeypox typically presents with fever, rash, and enlarged lymph nodes. New vaccines have recently been authorized for the prevention of monkeypox infection, whereas there are no specific pharmacological antiviral treatments for monkeypox infection. Howeve

Human monkeypox coinfection with syphilis in an immunocompromised patient.

Monkeypox is a viral zoonosis from the Poxviridae family that spreads at an unprecedented rate. It is transmitted through contact with skin lesions, respiratory droplets, body fluids, and sexual contact. The diverse presentation of the disease leads to misdiagnosis. Thus, clinicians should have a high index of suspicion, mainly with diseases with skin lesions. The most vulnerable group to developing this disease are individuals with risky sexual relationships, sexually transmitted infections, or human immunodeficiency virus/acquired immunodeficiency syndrome (HIV/AIDS). To date, only one case

First report of a chemokine from camelids: Dromedary CXCL8 is induced by poxvirus and heavy metal toxicity.

Low molecular weight proteins, known as chemokines, facilitate the migration and localization of immune cells to the site of infection and injury. One of the first chemokines identified, CXCL8 functions as a key neutrophil activator, recruiting neutrophils to sites of inflammation. Several viral infections, including zoonotic coronaviruses and poxviruses, have been reported to induce the expression of CXCL8. Dromedary camels are known to harbor several potentially zoonotic pathogens, but critical immune molecules such as chemokines remain unidentified. We report here the identification of CXCL

An unusual poxvirus from Nigeria.

A poxvirus of the variola-vaccinia subgroup was isolated from the lesions of a female African refugee suffering from a smallpox-like illness. The virus is interesting because it is neither variola nor vaccinia but combines some properties of each. These properties are described and the possible origin of the virus is discussed.

Human monkeypox.

Between October 1970 and May 1971, six cases of human infection with monkeypox virus were identified in Liberia, Nigeria, and Sierra Leone. Four of the cases were confirmed by virus isolation and two were diagnosed on the basis of epidemiological and serological investigations. All the cases occurred in unvaccinated individuals.Post-infection serological studies showed high haemagglutination-inhibition and neutralizing titres to pox group virus in four of the cases. Repeated challenge vaccination of all cases with potent smallpox vaccine resulted in equivocal reactions. In all, 24 susceptible h

Poxviruses isolated from clinically ill and asymptomatically infected monkeys and a chimpanzee.

Poxviruses were isolated from the kidneys of an outwardly healthy chimpanzee trapped in an area of the Democratic Republic of the Congo where a case of monkeypox had recently occurred in man, from the kidneys of clinically healthy cynomolgus monkeys in a colony in the Netherlands, and from monkeys suffering from monkeypox during outbreaks in colonies in the USA. It was established that two of the three viruses isolated from animals asymptomatically infected-namely, strain Chimp-9 from the chimpanzee and strain 64-7255 from the cynomolgus monkeys-although similar to one another differed markedl

Working Safely with Vaccinia Virus: Laboratory Technique and Review of Published Cases of Accidental Laboratory Infections with Poxviruses.

Vaccinia virus, the prototype Orthopoxvirus, is widely used in the laboratory as a model system to study various aspects of viral biology and virus-host interactions, as a protein expression system, as a vaccine vector, and as an oncolytic agent. The ubiquitous use of vaccinia viruses in laboratories around the world raises certain safety concerns because the virus can be a pathogen in individuals with immunological and dermatological abnormalities, and on occasion can cause serious problems in normal hosts. This chapter reviews standard operating procedures when working with vaccinia virus an

Research on substances with activity against orthopoxviruses.

Although smallpox was eradicated over 30 years ago, the disease remains a major threat. High mortality, high infectivity and low resistance of the contemporary population make the smallpox virus very attractive to terrorists. The possible presence of illegal stocks of the virus or risk of deliberate genetic modifications cause serious concerns among experts. Hence, it is reasonable to seek effective drugs that could be used in case of smallpox outbreak. This paper reviews studies on compounds with proven in vitro or in vivo antipoxviruses potential, which show various mechanisms of action. Nuc
