Fos expression in gonadotropin-releasing hormone neurons by naloxone or bicuculline in intact male rats.

Double immunocytochemistry for Fos and GnRH was performed after the intravenous infusion of naloxone or bicuculline. Either naloxone or bicuculline significantly increased serum LH concentrations. After the naloxone infusion, some Fos-positive GnRH neurons were found from the preoptic area to the basal hypothalamic area, whereas those were found from the diagonal band of Broca to the preoptic area after the bicuculline infusion. These results suggest the distribution of GnRH neurons activated by naloxone and bicuculline in intact male rats.

Cytoplasmic progestin-receptors in guinea pig brain: characteristics and relationship to the induction of sexual behavior.

The synthetic progestin, R 5020, was used to measure cytoplasmic progestin receptors in the brain and pituitary gland of ovariectomized guinea pigs. Progestin receptors with a dissociation constant of 0.1--0.3 nM were measured by gel filtration in all brain regions studied, pituitary gland and the uterus. The receptor is progestin-specific; biologically potent progestins compete well against [3H]R 5020 for binding, but androgens, glucocorticoids and estrogens do not. The concentration of the cytoplasmic progestin receptor in hypothalamus-preoptic area-septum and midbrain is decreased in vivo b

Regulation of stress-induced sleep fragmentation by preoptic glutamatergic neurons.

Sleep disturbances are detrimental to our behavioral and emotional well-being. Stressful events disrupt sleep, in particular by inducing brief awakenings (microarousals, MAs), resulting in sleep fragmentation. The preoptic area of the hypothalamus (POA) is crucial for sleep control. However, how POA neurons contribute to the regulation of MAs and thereby impact sleep quality is unknown. Using fiber photometry in mice, we examine the activity of genetically defined POA subpopulations during sleep. We find that POA glutamatergic neurons are rhythmically activated in synchrony with an infraslow r

Possible role of cyclooxygenase-2 in the brain vasculature in febrile response.

These results support the hypothesis that the brain vasculature is the site of PGE2 production responsible for LPS-induced fever. LPS seems to increase the PGE2 level in the entire brain via the induction of COX-2. Fever may be mediated by PGE2 which is produced in the blood vessels in the preoptic area or which is produced in other parts of the brain and transported to the preoptic area through the ventricular system.

The luteinizing hormone-releasing hormone (LH-RH) neuronal networks of the guinea pig brain. I. Intra- and extra-hypothalamic projections.

In the guinea pig brain, LH-RH-containing cell bodies are located not only within the classical hypophysiotrophic area but also in the medial preoptic area, septum and olfactory tubercle. LH-RH fiber tracts project not only to the primary portal plexus in the median eminence but also throughout the limbic forebrain and limbic midbrain regions. Using radiofrequency lesions in different brain regions, the projections of LH-RH cell bodies were determined. Cells in the medial preoptic area project ot the organum vasculosum of the lamina terminalis (OVLT), the suprachiasmatic nucleus, the mammillar

Relative enlargement of the medial preoptic nucleus in the Etruscan shrew, the smallest torpid mammal.

Endothermy is a key feature of mammalian biology and enables mammals to maintain stable body temperature and homeostatic functions in the face of a rapidly changing environment. However, when faced with harsh environmental conditions, certain mammalian species enter torpor, a state characterized by reduced metabolism, body temperature, and activity, to minimize energy loss. Etruscan shrews are the smallest mammals, with a surface-to-volume ratio that is very unfavorable for endothermic animals. As a result, Etruscan shrews have an extremely high metabolic rate and are known to enter torpor fre

Arginine vasotocin neuronal phenotypes and their relationship to aggressive behavior in the territorial monogamous multiband butterflyfish, Chaetodon multicinctus.

Intra and interspecific comparisons of arginine vasotocin (AVT) and its mammalian homolog arginine vasopressin (AVP) demonstrate several relationships between these neuropeptides and aggression/dominance behaviors. Prior studies in coral reef butterflyfishes and other fishes indicate that features of AVT neurons in the gigantocellular preoptic area (gPOA) and axon varicosities within the ventral nucleus of the ventral telencephalon should have a positive relationship with aggressive behavior, whereas AVT-ir neuronal features in the parvocellular preoptic area (pPOA) should have a negative rela

Modifications in gonadotropin control and reproductive behavior in the female rat by hypothalamic and preoptic lesions.

Effects of small hypothalamic and preoptic area lesions on vaginal cyclicity, ovulation and ovarian weights of female rats were examined. Animals were then gonadectomized and tested for lordosis behavior following injections of estrogen alone and estrogen plus progesterone. Male sex behavior was also measured during daily treatment with testosterone. Relative to sham operated rats, lesions in the dorsal preoptic area produced a significant increase in lordosis behavior, virtual elimination of male sex behavior, and only marginal effects on ovarian function. Animals with lesions in the ventral

Effect of intrahypothalamic injection of neostigmine on the secretion of epinephrine and norepinephrine and on plasma glucose level.

We previously reported that the injection of neostigmine, an inhibitor of acetylcholinesterase, into the third cerebral ventricle of fasted rats produced hyperglycemia associated with the secretion of epinephrine and norepinephrine. However, the central nervous system site of action of neostigmine by which the plasma catecholamine and glucose concentrations were increased is not known. In this study we injected neostigmine into the ventromedial hypothalamus, lateral hypothalamus, paraventricular hypothalamus, median site of the lateral-preoptic area, lateral site of the lateral-preoptic area,

Interleukin-1 beta acts at hypothalamic sites to inhibit gastric acid secretion in rats.

It has been established that interleukin-1 beta (IL-1 beta) injected into the cerebrospinal fluid inhibits gastric acid secretion in rats. Brain sites of action of IL-1 beta were investigated in conscious rats implanted unilaterally with chronic hypothalamic cannula. Gastric acid secretion was monitored 2 h after pylorus ligation. Human recombinant IL-1 beta (10 ng) microinjected into the medial preoptic area, anterior hypothalamus, and paraventricular nucleus inhibited gastric acid secretion by 76-83%. IL-1 beta microinjected into the ventromedial hypothalamus and other hypothalamic sites out

Atrial natriuretic factor in central nervous system regulatory mechanisms: effect of experimental alterations in water and salt homeostasis and blood pressure.

Atrial natriuretic factor (ANF) is widely distributed in the preoptic area and the hypothalamus, it is present there both in cell bodies and nerve terminals. Effect of experimental alterations in the salt and water balance was examined on preoptic-hypothalamic ANF levels measured in ten microdissected nuclei. Immunohistochemical analysis was also performed to confirm radioimmunological results. Following interventions were performed in adult male rats: adrenalectomy (5 days), daily 0.9% NaCl, aldosterone (5 micrograms/100 g) and dexamethasone (2 micrograms/ml drinking water) treatments in both

Hormonal regulation of androgen, estrogen receptor-α, and progesterone receptor expression in the adult male rabbit brain.

Sex hormones regulate a broad range of brain functions beyond reproduction via specific receptors, whose expression can be modulated by hormones. However, the regulation of sex hormone receptors across different brain regions in male rabbits remains unknown. We investigated the effects of two hormone treatments in castrated male rabbits: testosterone propionate (TP) and dihydrotestosterone plus estradiol benzoate (DHT + EB), on androgen receptor (AR), estrogen receptor alpha (ERα), and progesterone receptor (PR) expression in the brain. Adult male New Zealand White rabbits were bilaterally cas

Expression, distribution, and function of sex hormone receptors in the rabbit brain.

Sex hormones such as estradiol, progesterone, and testosterone are crucial for vertebrate brain functions by interacting with their specific receptors. In rabbits, these hormones regulate sexual behavior, scent marking, nest building, and nipple searching across different brain regions. However, information on sex hormone receptors in the rabbit brain is limited. We examined intracellular progesterone (PR), estrogen (ER), and androgen receptors (AR) in the rabbit brain. PR activation by progesterone is associated with pregnancy and maternal behaviors like nest building and nipple searching. PR

Role of progesterone receptors during postpartum estrus in rats.

We studied the role of progesterone receptor (PR) in the display of female sexual behavior during postpartum estrus in rats. Adult female rats were treated with the PR antagonist, RU486 (1.25 and 5 mg), 3 h after parturition and sexual behavior was evaluated throughout the first postpartum day. Estradiol and progesterone serum levels changed during the first 24 h postpartum. The highest estradiol and progesterone levels were found at 9 and 12 h postpartum, respectively. The predominant PR isoform in the hypothalamus and the preoptic area was PR-A during postpartum day. The content of PR-A incr

Subpopulations of migrating neurons express different levels of LHRH in quail and chick embryos.

LHRH neurons of the septal-preoptic area originate in the olfactory placode and migrate in the olfactory nerve into the brain during embryonic development. In adult birds, LHRH neurons have been found in the septal-preoptic area, mesencephalon and more recently in the lateral anterior nucleus of the thalamus (LA). LHRH neurons of the LA do not originate in the olfactory placode. Using immunocytochemistry, we examined the distribution of LHRH neurons in the embryonic and adult quail nervous system. The pattern of LHRH immunostaining in quail embryos was similar to that seen in chick embryos. Ho

Prostaglandin E receptor EP3 subtype is involved in thermal hyperalgesia through its actions in the preoptic hypothalamus and the diagonal band of Broca in rats.

The effects of microinjection of prostaglandin E2 (PGE2) (0.5 fg-500 pg/0.2 microl) into the medial part of the preoptic area (MPO) on nociception were studied using a hot-plate test in rats. The intraMPO microinjection of PGE2 only at non-pyrogenic doses (5-50 fg) reduced the paw-withdrawal latency, suggesting hyperalgesia. Maximal reduction was obtained 15 min after the injection of PGE2 at 50 fg. Subsequently, to determine which types of prostanoid receptors are involved in the hyperalgesic effect of PGE2 in the MPO, we administered PGE2 receptor agonists, i.e., 17-phenyl-omega-trinor PGE2

Hypothermia evoked by stimulation of medial preoptic nucleus protects the brain in a mouse model of ischaemia.

Therapeutic hypothermia at 32-34 °C during or after cerebral ischaemia is neuroprotective. However, peripheral cold sensor-triggered hypothermia is ineffective and evokes vigorous counteractive shivering thermogenesis and complications that are difficult to tolerate in awake patients. Here, we show in mice that deep brain stimulation (DBS) of warm-sensitive neurones (WSNs) in the medial preoptic nucleus (MPN) produces tolerable hypothermia. In contrast to surface cooling-evoked hypothermia, DBS mice exhibit a torpor-like state without counteractive shivering. Like hypothermia evoked by chemoge

The hypothalamic neurosecretory system of the marine teleost fish, Mugil auratus risso.

The hypothalamic neurosecretory system of the marinefish, Mugil auratus, consists of two nuclei, viz., the nucleus praeopticus and the nucleus laterlis tuberis. Both are paired, and while those of the nucleus praeopticus are vertically arranged as L-shaped bodies, their strand-like counterparts in the nucleus lateralis tuberis extend in an antero-posterior direction. The two constituent bodies of the nucleus praeopticus lie on both sides of the third cerebral ventricle. Each is differentiated into a dorsal pars magnocellularis and a ventral pars parvocellularis. Sex differences in the distribution of estrogen receptors in the septal area and hypothalamus of the domestic pig (Sus scrofa).

Recently in the pig hypothalamus a vasopressin- and oxytocin-containing nucleus was identified which, like the supraoptic nucleus, becomes sexually dimorphic after puberty. Following the increase in circulating steroids at puberty, the vasopressin- and oxytocin-containing nucleus becomes twice as large in both males and females. In adulthood, the vasopressin- and oxytocin-containing nucleus of females is approximately twice as large as that in males. Because these alterations are possibly due to an influence of gonadal steroids, i.e. estrogens, the vasopressin- and oxytocin-containing nucleus
