Chiral Artificial Antigen-Presenting Cells Enhancing Tumor Immunotherapy: Evasion of Macrophage Uptake through Inhibition of Serum Protein Corona Formation.

Biomimetic artificial antigen-presenting cells (aAPCs) have emerged as a promising platform for the immune system activation and modulation. However, abundant serum proteins in the living body prefer to adsorb on the aAPC surface to form a protein corona, leading to rapid clearance of aAPCs by mononuclear phagocytes, causing inefficient tumor immunotherapy by aAPCs. To achieve long-lasting antitumor immunity, chiral aAPCs with left-handed (aAPCs-L) or right-handed (aAPCs-D) helices are constructed via coassembly, chiral induction, chiral memory, and a click reaction. aAPCs-L can efficiently pr

Bovine serum albumin-gold nanoclusters protein corona stabilized polystyrene nanoparticles as dual-color fluorescent nanoprobes for breast cancer detection.

Breast cancer is the most prevalent malignancy and the first leading cause of cancer-related mortality among the female population worldwide. Approaches for precise and reliable detection of breast cancer cells, particularly in the nascent state, are desperately needed for elevating the survival rate of patients bearing the breast tumor. In this work, we successfully performed the sensitive, precise, and reliable breast cancer cell detection using facilely fabricated bovine serum albumin-gold nanocluster (BSA-AuNCs) protein corona stabilized, epithelial cell adhesion molecule (EpCAM) aptamer l

Differential Roles of Plasma Protein Corona on Immune Cell Association and Cytokine Secretion of Oligomeric and Fibrillar Beta-Amyloid.

Alzheimer's disease (AD) is a primary neurological disease with no effective cure. A hallmark of AD is the presence of intracellular tangles and extracellular plaques derived from the aberrant aggregation of tau- and beta-amyloid (Aβ). Aβ presents in the brain as well as in cerebrospinal fluid and the circulation, and Aβ toxicity has been attributed to amyloidosis and inflammation, among other causes. In this study, the effects of the plasma protein corona have been investigated with regard to the blood cell association and cytokine secretion of oligomeric (Aβo) and fibrillar Aβ1-42(Aβf), two

Synthetic virus-like particles prepared via protein corona formation enable effective vaccination in an avian model of coronavirus infection.

The ongoing battle against current and rising viral infectious threats has prompted increasing effort in the development of vaccine technology. A major thrust in vaccine research focuses on developing formulations with virus-like features towards enhancing antigen presentation and immune processing. Herein, a facile approach to formulate synthetic virus-like particles (sVLPs) is demonstrated by exploiting the phenomenon of protein corona formation induced by the high-energy surfaces of synthetic nanoparticles. Using an avian coronavirus spike protein as a model antigen, sVLPs were prepared by

The importance of selecting a proper biological milieu for protein corona analysis in vitro: Human plasma versus human serum.

Nanoparticle (NP) exposure to biological fluids in the body results in protein binding to the NP surface, which forms a protein coating that is called the "protein corona". To simplify studies of protein-NP interactions and protein corona formation, NPs are incubated with biological solutions, such as human serum or human plasma, and the effects of this exposure are characterized in vitro. Yet, how NP exposure to these two different biological milieus affects protein corona composition and cell response has not been investigated. Here, we explore the differences between the protein coronas tha

The Microenvironment in an Experimental Model of Acute Pancreatitis Can Modify the Formation of the Protein Corona of sEVs, with Implications on Their Biological Function.

A considerable number of the physiological functions of extracellular vesicles are conditioned by the protein corona attached to their surface. The composition of this corona is initially defined during their intracellular synthesis, but it can be subsequently modified by interactions with the microenvironment. Here, we evaluated how the corona of small extracellular vesicles exposed to the inflammatory environment generated in acute pancreatitis is modified and what functional changes occur as a result of these modifications. Small extracellular vesicles obtained from a pancreatic cell line w

The impact of Zn doping on CdTe quantum dots-protein corona formation and the subsequent toxicity at the molecular and cellular level.

Understanding the formation of protein corona (PC) is of vital importance for exploring the toxicity of nanoparticles and promoting their safe applications. In this study, CdTe QDs doping with 0, 1%, 5% and 10% Zn were synthesized using one-pot hydrothermal methods. Afterwards, this study explored and compared the formation of pure and Zn doped-QDs PC as well as the subsequent molecular and cellular toxicity. Result found that Zn doping regulated the toxicity of Cd-QDs by controlling their ability to adsorb serum proteins. The adsorption to Cd-QDs induced the dispersion, unfolding, secondary s

Serum Protein Corona Abolishes Changes in the Expression of Proinflammatory Genes Induced by Quantum Dots in Human Blood Mononuclear Cell.

We studied changes in the transcription of genes encoding cytokines (TNF, IL-6, IL-10, and IL-32), cell activation markers (ICAM1, CD38, Fas, and FCGRIII), ROS production catalyst (NOX2), autophagy (Beclin 1, LC3B, and p62) and apoptosis (BAX, BCL2) regulators in peripheral blood mononuclear cells upon contact with quantum dots CdSe/ZnS-MPA and CdSe/CdSeZnS/ZnS-PTVP. Up-regulation of TNF, ICAM1, Fas, p62 mRNA and down-regulation of the FCGRIII and NOX2 mRNA in response to the presence of quantum dots were revealed. The formation of serum protein corona on the surface of quantum dots abolished

A fluorogenic peptide containing the processing site of human SARS corona virus S-protein: kinetic evaluation and NMR structure elucidation.

Human severe acute respiratory syndrome coronavirus (hSARS-CoV) is the causative agent for SARS infection. Its surface glycoprotein (spike protein) is considered to be one of the prime targets for SARS therapeutics and intervention because its proteolytic maturation by a host protease is crucial for host-virus fusion. Using intramolecularly quenched fluorogenic (IQF) peptides based on hSARS-CoV spike protein (Abz-(755)Glu-Gln-Asp-Arg-Asn-Thr-Arg-Glu-Val-Phe-Ala-Gln(766)-Tyx-NH(2)) and in vitro studies, we show that besides furin, other PCs, like PC5 and PC7, might also be involved in this clea

Protein Corona Formation on Colloidal Polymeric Nanoparticles and Polymeric Nanogels: Impact on Cellular Uptake, Toxicity, Immunogenicity, and Drug Release Properties.

The adsorption of biomolecules to the surface of nanoparticles (NPs) following administration into biological environments is widely recognized. In particular, the "protein corona" is well understood in terms of formation kinetics and impact upon the biological interactions of NPs. Its presence is an essential consideration in the design of therapeutic NPs. In the present study, the protein coronas of six polymeric nanoparticles of prospective therapeutic use were investigated. These included three colloidal NPs-soft core-multishell (CMS) NPs, plus solid cationic Eudragit RS (EGRS), and anioni

Plasma Gelation as an Overlooked Determinant of Transfection Efficiency in Protein Corona-Coated Lipid Nanoparticles: Implications for In Vitro-In Vivo Translation.

A common observation in drug delivery studies is that preincubation of nanoparticles (NPs) with high concentrations of human plasma (HP) markedly reduces in vitro transfection efficiency (TE). This effect is commonly attributed to the formation of a protein corona (PC), which is assumed to impair cellular uptake or intracellular trafficking of gene delivery systems such as lipid nanoparticles (LNPs). However, the evidence presented here suggests an alternative mechanism underlying this phenomenon. To reproduce conditions in which LNPs encounter an excess of circulating proteins, NPs were incub

Fate of polystyrene micro- and nanoplastics in zebrafish liver cells: Influence of protein corona on transport, oxidative stress, and glycolipid metabolism.

Micro- and nanoplastics (MNPs) form protein corona (PC) upon contact with biological fluids, but their impact on the intracellular transport, distribution, and toxicity of MNPs remains unclear. Fetal bovine serum (FBS) and bovine serum albumin (BSA) were used to simulate in vivo environment, this study explored their influence on the transport and toxicity of polystyrene (PS) MNPs in zebrafish liver (ZFL) cells. Results showed PS MNPs were wrapped by proteins into stable complexes. Nanoparticles (NP, 50 nm) and their protein complexes (NP@PC) were internalized by cells within 6 h, with PC form

CdxZn1-xS nanoparticles-protein corona: Kinetics and temperature dependent interaction along with detection of the denaturation and unfolding of serum albumin.

In this study, the interaction and corona formation of HSA with nanostructured CdxZn1-xS have been analysed through microscopic, spectroscopic and structural investigations. Binding kinetics showed corona formation time for CdS NRs, Cd0.5Zn0.5S NPs and ZnS NPs are 60,45,70 min, respectively. The observed time for surface reorganization (85,130,110 min) was greater than corona formation time (60, 45, 70 min, respectively). The HRTEM showed that during corona formation of HSA, CdS NRs are attached each other and Cd0.5Zn0.5S NPs leads aggregation of HSA through positive cooperative reaction at ro

Proteomic Atlas of Notochordal Cell-Derived Extracellular Vesicles Highlights EV-Specific NF-κB Modulation and Functional Implications of the Protein Corona.

The notochord is a central signalling hub during embryonic development. After regression of the notochord, notochordal cells (NCs) reside within the developing intervertebral disc. The NC-secretome, including matrix, proteins and extracellular vesicles (NC-EVs), promotes regeneration of degenerate intervertebral discs and cartilage. This study aims to unravel the NC-EV-associated proteome and modulatory role in regenerative processes. NC-EVs were isolated from conditioned medium from porcine NC-rich tissue using differential centrifugation, size exclusion chromatography and density gradient ce

Liquid chromatography-based metallomics and transmission electron microscopy reveal gold nanoparticle surface treatment with vicinal dithiols to abolish protein corona formation.

AIMS: While gold nanoparticles (AuNPs) should allow the delivery of surface immobilized drugs to intended target tissues via the bloodstream, their interactions with plasma proteins may induce their aggregation and thus impede an effective delivery of chemotherapeutic agents to target tissues. The deliberate surface treatment of AuNPs has the potential to overcome this inherent limitation. METHODS: To probe interactions between surface treated AuNPs in blood plasma, we employed a size-exclusion chromatography (SEC)-based metallomics tool together with transmission electron microscopy (TEM).

Protein Coronas Derived from Mucus Act as Both Spear and Shield to Regulate Transferrin Functionalized Nanoparticle Transcellular Transport in Enterocytes.

The epithelial mucosa is a key biological barrier faced by gastrointestinal, intraoral, intranasal, ocular, and vaginal drug delivery. Ligand-modified nanoparticles demonstrate excellent ability on this process, but their efficacy is diminished by the formation of protein coronas (PCs) when they interact with biological matrices. PCs are broadly implicated in affecting the fate of NPs in vivo and in vitro, yet few studies have investigated PCs formed during interactions of NPs with the epithelial mucosa, especially mucus. In this study, we constructed transferrin modified NPs (Tf-NPs) as a mod

House Dust Mite Proteins Adsorb on Multiwalled Carbon Nanotubes Forming an Allergen Corona That Intensifies Allergic Lung Disease in Mice.

The increasing use of multiwalled carbon nanotubes (MWCNTs) could increase the risk of allergic lung disease in occupational or consumer settings. We previously reported that MWCNTs exacerbated allergic lung disease in mice induced by extract from house dust mites (HDM), a common cause of asthma in humans. Because MWCNTs avidly bind biomolecules to form protein coronas that can modify immunotoxicity, we hypothesized that exacerbation of allergic lung disease in mice caused by coexposure to MWCNTs and HDM extract was due to the formation of an allergen corona. In a first set of experiments, mal

Impact of Surface Chemistry of Ultrasmall Superparamagnetic Iron Oxide Nanoparticles on Protein Corona Formation and Endothelial Cell Uptake, Toxicity, and Barrier Function.

Ultrasmall superparamagnetic iron oxide nanoparticles (USPIONs) have been investigated for biomedical applications, including novel contrast agents, magnetic tracers for tumor imaging, targeted drug delivery vehicles, and magneto-mechanical actuators for hyperthermia and thrombolysis. Despite significant progress, recent clinical reports have raised concerns regarding USPION safety related to endothelial cell dysfunction; however, there is limited information on factors contributing to these clinical responses. The influence of USPION surface chemistry on nanoparticle interactions with protein

Glycopolymeric Nanoparticles Enrich Less Immunogenic Protein Coronas, Reduce Mononuclear Phagocyte Clearance, and Improve Tumor Delivery Compared to PEGylated Nanoparticles.

Nanoparticles (NPs) offer significant promise as drug delivery vehicles; however, their in vivo efficacy is often hindered by the formation of a protein corona (PC), which influences key physiological responses such as blood circulation time, biodistribution, cellular uptake, and intracellular localization. Understanding NP-PC interactions is crucial for optimizing NP design for biomedical applications. Traditional approaches have utilized hydrophilic polymer coatings like polyethylene glycol (PEG) to resist protein adsorption, but glycopolymer-coated nanoparticles have emerged as potential al
