Effect of static magnetic field (200 mT) on biofilm formation in Pseudomonas aeruginosa.

Several studies have investigated the effects of ionizing and non-ionizing radiations on microorganisms. However, the interaction between the magnetic field radiations and bacteria is less studied. The aim of our study was to study the effect of static magnetic field on the biofilm formation in Pseudomonas aeruginosa and its isogenic sod mutants. Our results revealed that the exposure to the static magnetic field (200 mT) increases significantly the swarming in the wild strain. The fliC gene expression did not show significant difference after 6 h exposure of the wild-type strain. The release

Thermostable xylanase inhibits and disassembles Pseudomonas aeruginosa biofilms.

Pseudomonas aeruginosa biofilms are problematic and play a critical role in the persistence of chronic infections because of their ability to tolerate antimicrobial agents. In this study, various cell-wall degrading enzymes were investigated for their ability to inhibit biofilm formation of two P. aeruginosa strains, PAO1 and PA14. Xylanase markedly inhibited and detached P. aeruginosa biofilms without affecting planktonic growth. Xylanase treatment broke down extracellular polymeric substances and decreased the viscosity of P. aeruginosa strains. However, xylanase treatment did not change the

The Fem cell-surface signaling system is regulated by ExsA in Pseudomonas aeruginosa and affects pathogenicity.

Bacterial interspecies interactions shape microbial communities and influence the progression of polymicrobial infections. FemI-FemR-FemA, a cell-surface signaling system, in Pseudomonas aeruginosa, is involved in the uptake of iron-chelating mycobactin produced by Mycobacterium spp. In this report, we present the data that indicates the femA-PA1909 operon is positively regulated by ExsA, a master regulator for the type three secretion system (T3SS), connecting the Fem system with T3SS. Intriguingly, the Fem system also influenced virulence factors in P. aeruginosa, including the quorum sensin

Synthesis and Biological Evaluation of Cajaninstilbene Acid and Amorfrutins A and B as Inhibitors of the Pseudomonas aeruginosa Quorum Sensing System.

The quorum sensing (QS) system inhibitors of Pseudomonas aeruginosa are thought to attenuate bacterial pathogenicity and drug resistance by inhibiting biofilm formation and the production of virulence factors. In this study, a synthetic approach to the natural products cajaninstilbene acid (1) and amorfrutins A (2) and B (3) has been developed and was characterized by the Heck reaction, which was used to obtain the stilbene core and a Pinick oxidation to give the O-hydroxybenzoic acid. The biological activities of these compounds against the P. aeruginosa quorum sensing systems were evaluated.

CRISPR-driven enhanced hydrocarbon emulsification in an environmental Pseudomonas aeruginosa strain.

BACKGROUND: Oil spills are a major concern due to the economic impact and severe effects on the ecosystem. To mitigate oil spills, hydrocarbon dispersion through emulsification is a promising approach, as it makes oil more susceptible to degradation by microorganisms. Environmental strains of Pseudomonas aeruginosa have demonstrated significant potential for producing rhamnolipids (RMLs) and pyocyanin (PYO), secondary metabolites associated to hydrocarbon emulsification. In this study, we isolated and characterized an environmental strain from an oil-contaminated site in the Gulf of Mexico. Up

Lysine polyphosphate modifications contribute to virulence factors in Pseudomonas aeruginosa.

Inorganic polyphosphate (polyP) is a universally conserved polymer involved in various biological processes, but its role as a direct protein regulator remains largely unexplored. Lysine polyphosphate modification (KPM), a strong but non-covalent interaction between polyP and lysine-rich protein sequences, has not been functionally characterized until now. In this study, we present the first investigation into KPM's biological significance using Pseudomonas aeruginosa, a critical priority pathogen known for its antibiotic resistance and virulence. We identified two essential bacterial proteins

Exploration of anti-quorum sensing properties of eicosyl heptafluorobutyrate against a clinical strain of Pseudomonas aeruginosa.

Pseudomonas aeruginosa is a multidrug-resistant (MDR) pathogen. It causes life-threatening diseases through quorum sensing (QS)-associated virulence. Thus, the attenuation of QS may be an effective strategy to combat the pathogen. In this study, Shewanella indica SU1 isolated from the marine environment showed QS inhibitory activity against biosensor strain Chromobacterium violaceum MTCC 2656. The lead compound was purified and tentatively identified as eicosyl heptafluorobutyrate (EPB). P. aeruginosa MCC 3457 was treated with EPB, and results revealed a reduced production of pyocyanin (79%),

Macrolides impact the growth ability of clinical Pseudomonas aeruginosa through quorum-sensing systems.

The aim of the present study was to examine the impact of macrolides on the expression of virulence factors and QS-associated genes in clinical P. aeruginosa isolates. Among 60 clinical P. aeruginosa, pyocyanin production was detected in 27 (45%) isolates, which belonged to various STs. Erythromycin inhibited the production of pigments in 12 out of 27 isolates. Other antibiotic categories didn't have an impact on production of pigments. Additionally, results showed that erythromycin sub-MIC inhibited the growth-rate in 17 isolates. Of note, in six isolates, the inhibition of growth-rate was gr

PmiR senses 2-methylisocitrate levels to regulate bacterial virulence in Pseudomonas aeruginosa.

To adapt to changes in environmental cues, Pseudomonas aeruginosa produces an array of virulence factors to survive the host immune responses during infection. Metabolic products contribute to bacterial virulence; however, only a limited number of these signaling receptors have been explored in detail for their ability to modulate virulence in bacteria. Here, we characterize the metabolic pathway of 2-methylcitrate cycle in P. aeruginosa and unveil that PmiR served as a receptor of 2-methylisocitrate (MIC) to govern bacterial virulence. Crystallographic studies and structural-guided mutagenesi

New Pqs Quorum Sensing System Inhibitor as an Antibacterial Synergist against Multidrug-Resistant Pseudomonas aeruginosa.

Development of new bacterial biofilm inhibitors as antibacterial synergists is an effective strategy to solve the resistance of Pseudomonas aeruginosa. In this paper, a series of 3-hydroxy-pyridin-4(1H)-ones were synthesized and evaluated, and the hit compound (20p) was identified with the effects of inhibiting the production of pyocyanin (IC50 = 8.6 μM) and biofilm formation (IC50 = 4.5 μM). Mechanistic studies confirmed that 20p inhibits the formation of bacterial biofilm by inhibiting the expression of pqsA, blocking pqs quorum sensing system quinolone biosynthesis. Moreover, we systematica

Squalene-based nanostructured lipid carriers for quorum-sensing inhibitor delivery: a multifunctional platform to disrupt p. Aeruginosa biofilms and suppress virulence.

BACKGROUND: The alarming rise of multidrug-resistant Pseudomonas aeruginosa (P. aeruginosa) demands anti-infective strategies beyond conventional antibiotics. Quorum sensing inhibitors (QSIs) represent a promising anti-virulence approach, yet their clinical translation is often hindered by poor solubility and formulation challenges. Nanostructured lipid carriers (NLCs) represent a multifunctional platform to overcome these barriers. OBJECTIVE: Herein, we developed a squalene-based NLC encapsulating a rationally designed QS inhibitor (Y0-C10-HSL) and systematically evaluated its anti-virulence

The mucoid switch in Pseudomonas aeruginosa represses quorum sensing systems and leads to complex changes to stationary phase virulence factor regulation.

The opportunistic pathogen Pseudomonas aeruginosa chronically infects the airways of Cystic Fibrosis (CF) patients during which it adapts and undergoes clonal expansion within the lung. It commonly acquires inactivating mutations of the anti-sigma factor MucA leading to a mucoid phenotype, caused by excessive production of the extracellular polysaccharide alginate that is associated with a decline in lung function. Alginate production is believed to be the key benefit of mucA mutations to the bacterium in the CF lung. A phenotypic and gene expression characterisation of the stationary phase ph

Ketoprofen attenuates Las/Rhl quorum-sensing (QS) systems of Pseudomonas aeruginosa: molecular and docking studies.

BACKGROUND: Quorum sensing (QS) is the leading cause of persistent infections and recalcitrance to antibiotic treatment of Pseudomonas aeruginosa. Hence, QS inhibitors are promising agents for the potential treatment of P. aeruginosa infections. METHODS AND RESULTS: Herein, the reducing effect of ketoprofen on virulence factors production including protease, hemolysin, pyocyanin, hydrogen cyanide, biofilm, and motility of P. aeruginosa strains was investigated. Furthermore, the quorum quenching activity of ketoprofen at the molecular level was examined by real-time PCR assessment. Our results

Plumbagin inhibits quorum sensing-regulated virulence and biofilms of Gram-negative bacteria: in vitro and in silico investigations.

The global rise in antimicrobial resistance and lack of discovery of new antimicrobials have created serious concerns. Targeting quorum sensing (QS) and biofilms of pathogenic bacteria is considered a promising approach in antimicrobial drug discovery. This study explored the inhibitory effect of plumbagin against biofilms and QS of Chromobacterium violaceum, Serratia marcescens and Pseudomonas aeruginosa. Violacein production in C. violaceum 12472 was reduced by >80%. The virulent traits of P. aeruginosa PAO1 such as pyocyanin, rhamnolipid and proteases were also inhibited at sub-minimum inhi

Positive control of swarming, rhamnolipid synthesis, and lipase production by the posttranscriptional RsmA/RsmZ system in Pseudomonas aeruginosa PAO1.

In Pseudomonas aeruginosa, the small RNA-binding, regulatory protein RsmA is a negative control element in the formation of several extracellular products (e.g., pyocyanin, hydrogen cyanide, PA-IL lectin) as well as in the production of N-acylhomoserine lactone quorum-sensing signal molecules. RsmA was found to control positively the ability to swarm and to produce extracellular rhamnolipids and lipase, i.e., functions contributing to niche colonization by P. aeruginosa. An rsmA null mutant was entirely devoid of swarming but produced detectable amounts of rhamnolipids, suggesting that factors

Cephalosporins Interfere With Quorum Sensing and Improve the Ability of Caenorhabditis elegans to Survive Pseudomonas aeruginosa Infection.

Pseudomonas aeruginosa utilizes the quorum sensing (QS) system to strategically coordinate virulence and biofilm formation. Targeting QS pathways may be a potential anti-infective approach to treat P. aeruginosa infections. In the present study, we define cephalosporins' anti-QS activity using Chromobacterium violaceum CV026 for screening and QS-regulated mutants of P. aeruginosa for validation. We quantified the effects of three cephalosporins, cefepime, ceftazidime, and ceftriaxone, on (1) pyocyanin production using spectrophotometric assay, (2) bacterial motility using agar plate assay, and
