Quinone-amino acid conjugates targeting Leishmania amino acid transporters.

The aim of the present study was to investigate the feasibility of targeting Leishmania transporters via appropriately designed chemical probes. Leishmania donovani, the parasite that causes visceral leishmaniasis, is auxotrophic for arginine and lysine and has specific transporters (LdAAP3 and LdAAP7) to import these nutrients. Probes 1-15 were originated by conjugating cytotoxic quinone fragments (II and III) with amino acids (i.e. arginine and lysine) by means of an amide linkage. The toxicity of the synthesized conjugates against Leishmania extracellular (promastigotes) and intracellular (

Exploration of respiratory chain of Nocardia asteroides: purification of succinate quinone oxidoreductase.

Nocardia asteroides is a pathogenic bacterium that causes severe pulmonary infections and plays a vital role in HIV development. Its electron transport chain containing cytochromes as electron carriers is still undiscovered. Information regarding cytochromes is important during drug synthesis based on cytochrome inhibitions. In this study we explored the electron transport of N. asteroides. Spectroscopic analysis of cytoplasm and membranes isolated from N. asteroides indicates the presence of soluble cytochrome-c, complex-II and the modified a(1)c(1) complex as the terminal oxidase. The molecu

Protective effects of metallothionein against dopamine quinone-induced dopaminergic neurotoxicity.

Dopamine (DA) quinone as DA neuron-specific oxidative stress conjugates with cysteine residues in functional proteins to form quinoproteins. Here, we examined the effects of cysteine-rich metal-binding proteins, metallothionein (MT)-1 and -2, on DA quinone-induced neurotoxicity. MT quenched DA semiquinones in vitro. In dopaminergic cells, DA exposure increased quinoproteins and decreased cell viability; these were ameliorated by pretreatment with MT-inducer zinc. Repeated L-DOPA administration markedly elevated striatal quinoprotein levels and reduced the DA nerve terminals specifically on the

Quinone formation as a chemoprevention strategy for hybrid drugs: balancing cytotoxicity and cytoprotection.

Cellular defense mechanisms that respond to damage from oxidative and electrophilic stress, such as from quinones, represent a target for chemopreventive agents. Drugs bioactivated to quinones have the potential to activate antioxidant/electrophile responsive element (ARE) transcription of genes for cytoprotective phase 2 enzymes such as NAD(P)H-dependent quinone oxidoreductase (NQO1) but can also cause cellular damage. Two isomeric families of compounds were prepared, including the NO-NSAIDs (NO-donating nonsteroidal anti-inflammatory drugs) NCX 4040 and NCX 4016; one family was postulated to

In vitro studies indicate a quinone is involved in bacterial Mn(II) oxidation.

Manganese(II)-oxidizing bacteria play an integral role in the cycling of Mn as well as other metals and organics. Prior work with Mn(II)-oxidizing bacteria suggested that Mn(II) oxidation involves a multicopper oxidase, but whether this enzyme directly catalyzes Mn(II) oxidation is unknown. For a clearer understanding of Mn(II) oxidation, we have undertaken biochemical studies in the model marine alpha-proteobacterium, Erythrobacter sp. strain SD21. The optimum pH for Mn(II)-oxidizing activity was 8.0 with a specific activity of 2.5 nmol x min(-1) x mg(-1) and a K (m) = 204 microM. The activit

Blue Light Irradiated, Para Site-Selective C-H/C-H Cross-Dehydrogenative Coupling of Electron-Rich Arenes with Quinones.

A metal-free, regioselective C-H/C-H dehydrogenative arylation of electron-rich arenes with quinones has been developed. Utilizing Eosin-Y as a photocatalyst and K2S2O8 as the terminal oxidant, this protocol enables the para selective quinonylation of phenols and anisoles under mild, open flask conditions. The reaction is promoted by trifluoroacetic acid (TFA) and visible light, providing a sustainable route to biologically active arylated quinone derivatives, including those with potential therapeutic applications for Alzheimer's disease. The methodology is scalable to gram level synthesis an

A Versatile One-Pot Access to Cyanoarenes from ortho- and para-Quinones: Paving the Way for Cyanated Functional Materials.

A generally applicable direct synthesis of cyanoarenes from quinones is presented. Particular emphasis is placed on the preparation of precursors and target molecules relevant for organic materials, including halogenated cyanoarenes and larger cyanated acenes. The reaction and work-up protocols are adjusted for the challenges presented by the different substrates and products. Screening results of the initial reaction optimization are given to further facilitate adaptation to other synthetic problems. The universality of the reaction is finally highlighted by successful substitution of para-qu

Features of idebenone and related short-chain quinones that rescue ATP levels under conditions of impaired mitochondrial complex I.

Short-chain quinones have been investigated as therapeutic molecules due to their ability to modulate cellular redox reactions, mitochondrial electron transfer and oxidative stress, which are pathologically altered in many mitochondrial and neuromuscular disorders. Recently, we and others described that certain short-chain quinones are able to bypass a deficiency in complex I by shuttling electrons directly from the cytoplasm to complex III of the mitochondrial respiratory chain to produce ATP. Although this energy rescue activity is highly interesting for the therapy of disorders associated w

Effects of natural organic matter, anthropogenic surfactants, and model quinones on the reduction of contaminants by zero-valent iron.

Recent studies of contaminant reduction by zero-valent iron metal (Fe0) have highlighted the role of iron oxides at the metal-water interface and the effect that sorption has at the oxide-water interface on contaminant reduction kinetics. The results suggest that a variety of organic surface-active substances might enhance or inhibit contaminant degradation, depending on the degree to which they promote solubilization, sorption. and/or reaction. Of particular interest is the effect of natural organic matter (NOM), because of its ubiquitous presence in natural waters and amphiphilic properties;

Cyanide as a copper and quinone-directed inhibitor of amine oxidases from pea seedlings ( Pisum sativum) and Arthrobacter globiformis: evidence for both copper coordination and cyanohydrin derivatization of the quinone cofactor.

The interactions of cyanide with two copper-containing amine oxidases (CuAOs) from pea seedlings (PSAO) and the soil bacterium Arthrobacter globiformis (AGAO) have been investigated by spectroscopic and kinetic techniques. Previously, we rationalized the effects of azide and cyanide for several CuAOs in terms of copper coordination by these exogenous ligands and their effects on the internal redox equilibrium TPQ(amr)-Cu(II) right harpoon over left harpoon TPQ(sq)-Cu(I). The mechanism of cyanide inhibition was proposed to occur through complexation to Cu(I), thereby directly competing with O(2

Regioselective Nucleophilic Diamination of Quinones Enabling Access to 3,5-, 3,6-, and Mixed Diamino Derivatives: Synthesis and Photocurrent Characterization.

In this study, we disclose a Cu(OTf)2-catalyzed regioselective nucleophilic amination of quinones to synthesize 3,5-, 3,6-, and mixed diaminoquinones under air. The regioselectivity, controlled by the catalyst coordination and the steric hindrance of amines, follows different patterns: the reactions with anilines afforded the 3,6-diaminoquinones; the coupling with aliphatic primary amines led to the formation of a mixture of 3,6- and 3,5-isomers; and the use of sterically hindered secondary amines delivered the 3,5-isomers. Furthermore, we found that the synthesized diaminoquinones exhibited s

Lewis Acid Catalyzed Divergent Reaction of Bicyclo[1.1.0]Butanes With Quinones for the Synthesis of Diverse Polycyclic Molecules.

Bicyclo[1.1.0]butanes (BCBs) are highly strained hydrocarbons with unique structural properties and intrinsic reactivity, making them valuable building blocks for constructing complex molecular architectures. Herein, we report the Lewis acid-catalyzed divergent reactions of BCBs with quinones, yielding a diverse array of polycyclic molecules. Using Sc(OTf)₃ as a catalyst, pyrazole-substituted BCBs efficiently undergo formal (3 + 2) cycloaddition reactions with quinones, producing highly substituted bicyclo[2.1.1]hexanes featuring a caged framework. Monosubstituted BCB ketones undergo a sequent

Patterns, variability, and predictors of p-phenylenediamine quinones and parent p-phenylenediamines in pregnant women across three trimesters.

p-Phenylenediamine quinones (PPD-Qs), oxidative derivatives of rubber antioxidant p-phenylenediamines (PPDs), have garnered increasing concern due to their environmental ubiquity and toxicity. However, evidence regarding the exposure assessment of these emerging contaminants in humans, especially pregnant women, remains limited. Herein, we measured the levels of six PPD-Qs and their parent six PPDs in 1,278 urine samples collected from 426 pregnant women over three trimesters with the aim of evaluating the exposure profiles, variability, and predictors of PPD-Qs and PPDs during pregnancy. Catechol ortho-quinones: the electrophilic compounds that form depurinating DNA adducts and could initiate cancer and other diseases.

Catechol estrogens and catecholamines are metabolized to quinones, and the metabolite catechol (1,2-dihydroxybenzene) of the leukemogenic benzene can also be oxidized to its quinone. We report here that quinones obtained by enzymatic oxidation of catechol and dopamine with horseradish peroxidase, tyrosinase or phenobarbital-induced rat liver microsomes react with DNA by 1,4-Michael addition to form predominantly depurinating adducts at the N-7 of guanine and the N-3 of adenine. These adducts are analogous to the ones formed with DNA by enzymatically oxidized 4-catechol estrogens (Cavalieri,E.L

Efficient synthesis of pillar[5]quinone for rapid identification of aromatic nitro explosives.

An efficient synthesis of pillar[5]quinone (Q5) is reported for the rapid identification of aromatic nitro explosives. A record-high synthetic yield of 96% on a gram scale for Q5 can be achieved via a two-step reaction process involving the demethylation of pillar[5]arene to hydroxypillar[5]arene, followed by oxidation with Oxone to form Q5. In addition, by modifying amino silicon spheres with Q5 to prepare a stationary phase material (Q5-Sil) for high-performance liquid chromatography (HPLC), efficient separation of eight typical aromatic nitro-explosives was achieved using a single column. Enantioselective toxicokinetics drive the differential toxicity of chiral 6PPD-quinone in rainbow trout.

The tire-derived contaminant N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine quinone (6PPD-Q) is widespread in aquatic ecosystems and poses significant ecological risk. As a chiral compound, its enantiomers may exhibit differential toxicity, yet the underlying mechanisms remain limited. Here, we found that (+)-6PPD-Q was 2.4-fold more toxic to rainbow trout than (-)-6PPD-Q (p < 0.05). In addition, in vitro metabolism assays using liver S9 fraction showed that (+)-6PPD-Q was metabolized 2.5-fold faster than (-)-6PPD-Q. Semi-quantitative nontarget analysis revealed that hydroxylated metabolit

Mass Spectrometry Imaging Unveils the Metabolic Effect of 6PPD-Quinone in Exposed Mice.

N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine quinone (6PPD-Q) has gained widespread attention as an emerging significant environmental contaminant, but its biochemical toxicity in mammals remains inadequately explored. In this study, the systemic toxicological effects of 6PPD-Q in mouse models exposed to both high and low doses were investigated. Matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI MSI) was applied to evaluate its effects on major organs, including the liver, kidneys, spleen, and testes. Notably, both high and low doses caused similar multiorgan m

Development of a novel family of antifungal agents based on a quinone methide oxime framework.

Given the widespread occurrence of fungal infections, the phenomenon of fungal drug resistance, and the limited number of systemic antimycotic therapies, novel chemicals should be developed to control pathogenic fungi. We propose using quinone methide oximes as a novel framework for developing a novel class of antifungal agents. Compound 2 was destroyed mature biofilms at the concentration of 0.5 µg/mL (MIC/4) and prevented hyphal growth of Candida albicans at 0.125 µg/mL (MIC/16). The chemical applied at the concentration of 16-128 µg/mL inhibited the growth of the majority of the clinical is
