INTRODUCTION: Siderophores are secondary metabolites with high affinity and specificity for iron and play a crucial role in iron acquisition. Vibrio fluvialis, an emerging foodborne opportunistic pathogen of increasing concern, produces the catechol siderophore fluvibactin. Quorum sensing (QS) is a process of bacterial communication vital for many biological processes. To date, the genes responsible for fluvibactin biosynthesis and the role of quorum sensing (QS) in its regulation remain unclear.
METHODS: In this study, we characterized the gene cluster involved in fluvibactin biosynthesis an

Lip care products such as balms, glosses, and lipsticks are widely used to enhance lip health, appearance, and protection. Lipsticks provide aesthetic appeal, social and psychological benefits, that act as a protective barrier against environmental stressors. However, growing concerns have emerged regarding chronic lip infections associated with multidrug-resistant bacteria, particularly Pseudomonas aeruginosa. This is especially concerning in individuals with conditions such as juvenile bullous pemphigoid, hypochromic microcytic anemia, and sickle cell trait. The persistence of P. aeruginosa

UNLABELLED: Vibrio species adapt to different niches by sensing and responding to environmental signals such as nutrients, host cues, and quorum sensing autoinducers. Vibrio campbellii uses the master quorum sensing transcription factor LuxR to control expression of hundreds of genes at high cell density. Furthermore, many gamma-proteobacteria use the global transcription factor Catabolite Activator Protein (CAP) to control numerous physiologically relevant pathways, many of which are also modulated by quorum sensing, such as competence, biofilm formation, and carbon metabolism. However, the e

Listeria monocytogenes causes listeriosis, a severe foodborne disease with a high hospitalization rate. This pathogen forms robust biofilms in food environments and poses significant health risks. Bacillus subtilis PS-216, a proven surfactin producer with antimicrobial activity against Gram-negative pathogens, was investigated for its anti-Listeria effects. We analyzed the antimicrobial and antibiofilm activities of wild-type B. subtilis PS-216 and knockout mutants lacking quorum sensing genes (ΔcomQ, ΔcomA) and QS-regulated antimicrobials (ΔsrfAA, ΔbacA, ΔsrfAAΔbacA) against L. monocytogenes

The emergence of multidrug-resistant (MDR) Pseudomonas aeruginosa is driven by diverse resistance mechanisms, including quorum sensing (QS) and biofilm formation. QS regulates virulence, antibiotic tolerance, efflux pumps, and biofilm development, enhancing the pathogen's adaptability. Among QS systems, the Pseudomonas quinolone signal regulator (PqsR) plays a central role in controlling virulence and biofilm-associated genes. This review critically examines the PqsR-regulated network and highlights the potential of PqsR inhibitors to reduce pathogenicity. Disrupting QS instead of targeting ba

Biochar addition promotes direct interspecies electron transfer (DIET) between syntrophic bacteria and methanogens and increases the secretion of N-acyl homoserine lactones (AHLs); however, the specific role of AHLs in biochar-promoted DIET remains unclear. In this study, biochar accelerated the degradation of volatile fatty acids, increasing the maximum methanation rate by 24.97 ± 0.23 %. Compared to the control system, biochar significantly enhanced the secretion of AHLs, resulting in an approximate 2.7-fold increase. The increase in AHLs subsequently promoted the synthesis of c-type cytochr

Anammox bioreactors face challenges of prolonged startup times and sensitivity to environmental fluctuations. Although quorum sensing (QS) has emerged as a potential solution, most studies rely on expensive N-acyl homoserine lactones. This study explored an alternative by isolating indigenous QS bacteria from anammox granules and evaluating their impact on bioreactor performance. Four QS isolates- Ensifer sp. CH9, Brucella sp. CH10, Pseudomonas sp. CH11 and Burkholderia sp. CH12 -were immobilized and introduced into anammox bioreactors. The QS bacteria improved nitrogen removal efficiency, enh

Phenotypic diversity in bacteria often results from adaptation to changing environmental conditions, exemplified by variable colony morphotypes. In Burkholderia pseudomallei, discrete genomic alterations and modulation of gene expression facilitate adaptation. Adapted variants of species within the Burkholderia cepacia complex (Bcc) often lose the pC3 virulence megaplasmid, impacting their colony morphology and their production of virulence factors. In this study, we characterize variants arising in Burkholderia ambifaria clinical isolates using proteomics and phenotypic tests and show that so

Quorum sensing (QS) inhibition is a promising novel approach to control bacterial infections. However, it is not clear whether QS inhibition will impose selective pressure for the spread of resistance against QS inhibition in pathogen populations. Previous research tried to answer this question by using synthetic growth media, and this revealed that whether or not resistance will spread completely depends on the environment in which it is studied. Therefore, the spread of resistance should be studied in the environment where it ultimately matters: in vivo during infection of a host. Here, usin

Outer membrane vesicle (OMV)-delivered Pseudomonas quinolone signal (PQS) plays a critical role in cell-cell communication in Pseudomonas aeruginosa. However, the functions and mechanisms of membrane-enclosed PQS in interspecies communication in microbial communities are not clear. Here, we demonstrate that PQS delivered by both OMVs from P. aeruginosa and liposome reduces the competitiveness of Burkholderia cenocepacia, which usually shares the same niche in the lungs of cystic fibrosis patients, by interfering with quorum sensing (QS) in B. cenocepacia through the LysR-type regulator ShvR. Pseudomonas aeruginosa uses quorum sensing (QS) to coordinate the expression of multiple genes necessary for establishing and maintaining infection. It has previously been shown that lasR QS mutations frequently arise in cystic fibrosis (CF) lung infections, however, there has been far less emphasis on determining whether other QS system mutations arise during infection or in other environments. To test this, we utilized 852 publicly available sequenced P. aeruginosa genomes from the Pseudomonas International Consortium Database (IPCD) to study P. aeruginosa QS mutational signatures. To study

The human gut is host to almost 3000 microbial species, of which 90% are bacteria. Quorum sensing (QS) molecules generated by intestinal bacteria are important in establishing species- and strain-level structures within the gut microbiome but are also used to communicate with the host. Although we do not know which QS molecules have the most direct interaction with intestinal and sensory neurons, it is clear they affect our physiological and mental health. Signals produced by bacteria are diverse and include autoinducers (AIs), homoserine lactones (HSLs), quinolines, peptides, toxins and prote

Salicylic acid (SAL) has recently been shown to induce biofilm formation in Staphylococcus aureus and to affect the expression of virulence factors. This study was aimed to investigate the effect of SAL on the regulatory agr system and its impact on S. aureus biofilm formation. The agr quorum-sensing system, which is a central regulator in S. aureus pathogenicity, plays a pivotal role in the dispersal of S. aureus mature biofilms and contributes to the creation of new colonization sites. Here, we demonstrate that SAL impairs biofilm dispersal by interfering with agr expression. As revealed by

Antibiotics are well known drugs which, when present above certain concentrations, are able to inhibit the growth of certain bacteria. However, a growing body of evidence shows that even when present at lower doses (subMIC, for sub-minimal inhibitory concentration), unable to inhibit or affect microbial growth, antibiotics work as signaling molecules, affect gene expression and trigger important bacterial stress responses. However, how subMIC antibiotic signaling interplays with other well-known signaling networks in bacteria (and the consequences of such interplay) is not well understood. In

Acinetobacter baumannii causes several nosocomial infections and poses major threat when it is multidrug resistant. Even pan drug-resistant strains have been reported in some countries. The intensive care unit (ICU) mortality rate ranged from 45.6% to 60.9% and it is as high as 84.3% when ventilator-associated pneumonia was caused by XDR (extensively drug resistant) A. baumannii. Acinetobacter baumannii constituted 9.4% of all Gram-negative organisms throughout the hospital and 22.6% in the ICUs according to a study carried out in an Indian hospital. One of the major factors contributing to dr

Many bacteria use population density to control gene expression via quorum sensing. In Vibrio cholerae, quorum sensing coordinates virulence, biofilm formation, and DNA uptake by natural competence. The transcription factors AphA and HapR, expressed at low and high cell density respectively, play a key role. In particular, AphA triggers the entire virulence cascade upon host colonisation. In this work we have mapped genome-wide DNA binding by AphA. We show that AphA is versatile, exhibiting distinct modes of DNA binding and promoter regulation. Unexpectedly, whilst HapR is known to induce natu

Cell-to-cell communication via gap junctions regulates airway epithelial cell homeostasis and maintains the epithelium host defense. Quorum-sensing molecules produced by Pseudomonas aeruginosa coordinate the expression of virulence factors by this respiratory pathogen. These bacterial signals may also incidentally modulate mammalian airway epithelial cell responses to the pathogen, a process called interkingdom signaling. We investigated the interactions between the P. aeruginosa N-3-oxo-dodecanoyl-L-homoserine lactone (C12) quorum-sensing molecule and human airway epithelial cell gap junction

The transfer of novel genetic material into the genomes of bacterial viruses (phages) has been widely documented in several host-phage systems. Bacterial genes are incorporated into the phage genome and, if retained, subsequently evolve within them. The expression of these phage genes can subvert or bolster bacterial processes, including altering bacterial pathogenicity. The phage phiCDHM1 infects Clostridium difficile, a pathogenic bacterium that causes nosocomial infections and is associated with antibiotic treatment. Genome sequencing and annotation of phiCDHM1 shows that despite being clos

N-acylhomoserine lactone (AHL)-mediated quorum-sensing (QS) regulates virulence functions in plant and animal pathogens such as Agrobacterium tumefaciens and Pseudomonas aeruginosa. A chemolibrary of more than 3500 compounds was screened using two bacterial AHL-biosensors to identify QS-inhibitors (QSIs). The purity and structure of 15 QSIs selected through this screening were verified using HPLC MS/MS tools and their activity tested on the A. tumefaciens and P. aeruginosa bacterial models. The IC50 value of the identified QSIs ranged from 2.5 to 90 µg/ml, values that are in the same range as
