Exonuclease Ribozyme Encoded in a Circular Genome: On the Initiation of the RNA World.

The "RNA World" hypothesis posits a reasonable scenario for the origin of life. A question central to this scenario is: which RNA species may have emerged as the first ribozyme, thereby initiating the earliest stage of life? To date, experimental and computational studies have focused on ribozymes playing a constructive role in RNA synthesis-such as those facilitating template-directed copying or nucleotide synthesis. However, the answer remains unconvincing, primarily because their functional complexity likely demands lengthy sequences, making de novo emergence improbable. Here, we propose an

Blood-based circular RNAs for early diagnosis of Alzheimer's disease.

Detection of Alzheimer's disease (AD) before the development of clinical symptoms is critical for enabling the use of new treatments. Circular RNAs (circRNAs) are highly stable non-coding RNAs enriched in the brain that can cross the blood-brain barrier. Here, analyzing blood data from 1,221 individuals with AD and healthy individuals, we identified 34 circRNAs associated with AD status. A predictive model including these 34 circRNAs was comparable to plasma phosphorylated Tau-217 (pTau217) in classifying AD based on biomarker-confirmed (amyloid-β and Tau) status and replicated in independent

The emerging role of circular RNAs in neurodegenerative diseases and viral infections.

Circular RNAs (circRNAs) represent a class of highly stable, covalently closed RNA molecules increasingly recognized as important regulators of brain aging and neurodegenerative disease. Growing evidence also implies circRNAs in viral infection, suggesting a potential intersection between viral neuropathogenesis and neurodegeneration. However, no studies have yet directly integrated circRNAs, neurotropic viral infections, and neurodegenerative disorders within a single mechanistic framework. To date, specific circRNAs have been linked to the progression of Alzheimer's disease and Parkinson's d

Circular RNAs and SIRT1 in post-transcriptional regulation and human disease.

Circular RNAs (circRNAs) have emerged as stable post-transcriptional regulators that influence gene expression, cellular adaptation, and disease pathogenesis through mechanisms including microRNA (miRNA) interaction, RNA-binding protein modulation, and signaling-network regulation. Generated through the back-splicing process that produce covalently closed RNA loops, circRNAs exhibit remarkable stability, evolutionary conservation, and tissue-specific expression patterns, enabling them to function as miRNA sponges, protein scaffolds, transcriptional modulators, and, in some cases, translational

Circular RNAs in Plasma and Beyond: Potential Biomarkers for Breast Cancer.

Breast cancer (BC) continues to be a major cause of cancer-related mortality among women, and early diagnosis remains critical for improving survival outcomes. Conventional tissue biopsy and imaging techniques are constrained by invasiveness and limited sensitivity in early-stage disease, whereas routine serum tumor markers lack sufficient specificity for reliable early detection. Circular RNAs (circRNAs) have increasingly been recognized as promising non-invasive biomarkers, owing to their remarkable stability and detectability in plasma. Here, we summarize the current landscape of plasma cir

Circular RNAs in cardiovascular disease: A paradigm shift in diagnosis and therapeutics.

Cardiovascular diseases (CVDs) remain the leading cause of global mortality, accounting for 32 % of deaths worldwide in 2021. The increasing prevalence of CVDs highlights the urgent need for novel diagnostic and therapeutic approaches. Circular RNAs (circRNAs), a class of single-stranded RNA molecules produced via back-splicing, have gained attention as potential biomarkers and therapeutic targets due to their distinctive features. Unlike linear RNAs, circRNAs lack 5' caps and 3' poly(A) tails, making them highly resistant to exonuclease degradation and granting them an extended half-life of 1

Circular RNAs in colorectal cancer: Emerging insights and future perspectives.

Circular RNAs (circRNAs) are a distinct class of long non-coding RNAs characterized by their covalently closed-loop structure, which confers high stability. Once dismissed as nonfunctional 'junk RNA' or aberrant splicing byproducts, circRNAs have undergone a dramatic reappraisal and are now recognized as crucial regulatory molecules, abundantly expressed in eukaryotic cells and exhibiting notable tissue specificity. CircRNAs perform a wide range of regulatory functions, including acting as microRNA sponges, interacting with RNA-binding proteins, modulating transcription and splicing, and parti

Circular RNA expression in ALS is progressively deregulated and tissue-dependent.

BACKGROUND: There is increasing evidence on the role of circular RNAs (circRNAs) in neuronal and muscular processes. Accordingly, their dysregulation is associated with neurodegenerative diseases and myopathies. We investigated circRNA expression in the central nervous system (CNS) and skeletal muscle, the two main tissues affected in amyotrophic lateral sclerosis (ALS). RESULTS: Based on circRNA sequencing analysis in spinal cord from ALS mice (SOD1G93A) followed by a literature search, 30 circRNAs potentially involved in ALS were tested. All selected circRNAs were downregulated in the SOD1G

Circular RNAs and their emerging roles in muscular immune-related diseases.

Circular RNAs (circRNAs) have recently emerged as a highly stable and versatile class of non-coding RNAs that play critical roles in gene regulation, yet their involvement in immune-mediated muscle disorders remains largely underexplored. This review synthesizes how circRNAs influence key processes in both skeletal muscle and immune cells, from myogenesis, regeneration, and muscle stem cell function to inflammatory signaling and muscle wasting. Our aim was to identify circRNA insights across muscle immune-mediated diseases. However, we found no idiopathic inflammatory myopathy-focused circRNA

Biogenesis and function of circular RNAs and their implications in the Down syndrome brain.

Circular RNAs (circRNAs), a class of covalently closed, non-coding RNAs, have recently emerged as crucial regulators of gene expression. They exert their roles through microRNA (miRNA) sponging, transcriptional regulation, and interactions with RNA-binding proteins (RBPs). Increasing evidence suggests that circRNAs play important roles in neurodevelopmental disorders, including Down syndrome (DS). DS is a condition caused by trisomy of chromosome 21 and characterised by intellectual disability (ID), neuroinflammation, and increased risk of early-onset Alzheimer's disease (AD). Aberrant circRNA

Therapeutic application of circular RNA aptamers in a mouse model of psoriasis.

Efforts to advance RNA aptamers as a new therapeutic modality have been limited by their susceptibility to degradation and immunogenicity. In a previous study, we demonstrated synthesized short double-stranded region-containing circular RNAs (ds-cRNAs) with minimal immunogenicity targeted to dsRNA-activated protein kinase R (PKR). Here we test the therapeutic potential of ds-cRNAs in a mouse model of imiquimod-induced psoriasis. We find that genetic supplementation of ds-cRNAs leads to inhibition of PKR, resulting in alleviation of downstream interferon-α and dsRNA signals and attenuation of p

Evaluation of Circular RNA SMARCA5 as a Novel Biomarker for Hepatocellular Carcinoma.

BACKGROUND: Hepatocellular carcinoma (HCC) is the fourth most prevalent type of cancer in Egypt and the sixth globally. Most patients with HCC are typically diagnosed during the advanced stages of the disease due to the absence of biomarkers for early detection. Consequently, these patients miss the optimal timeframe for receiving therapy. OBJECTIVE: we aimed to assess the circular RNA SMARCA5 level and SMARCA5 mRNA gene expression as a potential biomarker for early detection of HCC. METHODS: The present study utilized a case-control design comprising 159 participants. Participants were sele

Identification of circular RNAs hosted by the RPGR ORF15 genomic locus.

Mutations in the retina-specific isoform of the gene encoding retinitis pigmentosa GTPase regulator (RPGRorf15) cause X-linked retinitis pigmentosa, a severe and early onset inherited retinal degeneration. The underlying pathogenic mechanisms and variability in disease severity remain to be fully elucidated. The present study examines structural features of the ORF15 exonic region to provide new insights into the disease pathogenesis. Using canine and human RNA samples, we identified several novel RPGR ORF15-like linear RNA transcripts containing cryptic introns (exitrons) within the annotated

Marek's Disease Virus Virulence Genes Encode Circular RNAs.

Circular RNAs (circRNAs) are a recently rediscovered class of functional noncoding RNAs that are involved in gene regulation and cancer development. Next-generation sequencing approaches identified circRNA fragments and sequences underlying circularization events in virus-induced cancers. In the present study, we performed viral circRNA expression analysis and full-length sequencing in infections with Marek's disease virus (MDV), which serves as a model for herpesvirus-induced tumorigenesis. We established inverse PCRs to identify and characterize circRNA expression from the repeat regions of

Circular RNAs and thyroid cancer: Closed molecules, open possibilities.

Thyroid neoplasms requiring differential diagnosis between thyroid cancer and benign tumors can be detected in more than half of the healthy population. A generally accepted method that allows assessing the risk of malignant potential and determining the indications for surgical treatment of thyroid tumor is a fine-needle aspiration biopsy followed by a cytological examination. Nevertheless, in patients with indeterminate categories of cytological conclusions according to Bethesda system, the positive predictive value of the cytology result is significantly lower than desired and often leads t

Genome-wide identification of circular RNAs in adult Schistosoma japonicum.

Circular RNAs (circRNAs) are a class of novel, widespread, covalently closed RNAs that have played an essential role in animal gene regulation. To systematically explore circRNAs in the blood fluke Schistosoma japonicum, we performed RNA sequencing and bioinformatics analysis, and found that hundreds of circRNAs showed gender-associated expression. Among these identified circRNAs, more than 77.54% and 74.73% were putatively derived from the exon region of the genome and some circRNAs showed gender-associated expressions. The functional prediction of circRNAs (circ_003826 and circ_004690) showe

Advances in the Identification of Circular RNAs and Research Into circRNAs in Human Diseases.

Circular RNAs (circRNAs) are a class of endogenous non-coding RNAs (ncRNAs) with a closed-loop structure that are mainly produced by variable processing of precursor mRNAs (pre-mRNAs). They are widely present in all eukaryotes and are very stable. Currently, circRNA studies have become a hotspot in RNA research. It has been reported that circRNAs constitute a significant proportion of transcript expression, and some are significantly more abundantly expressed than other transcripts. CircRNAs have regulatory roles in gene expression and critical biological functions in the development of organi
