BACKGROUND: Oral submucous fibrosis (OSF) is a chronic potentially malignant disorder that can progress to oral squamous cell carcinoma (OSCC). Although dysregulated non-coding RNAs have been implicated in oral carcinogenesis, the competing endogenous RNA (ceRNA)-mediated regulatory mechanisms underlying OSF-to-OSCC progression remain poorly understood. This study aimed to identify candidate regulatory molecules and construct a putative lncRNA-miRNA-mRNA network associated with malignant transformation.
METHODS: Publicly available microarray datasets (GSE117973 and GSE125866) were analyzed to

Fusobacterium nucleatum (F. nucleatum), a key periodontal pathogen, is increasingly detected in atherosclerotic plaques, yet its epigenetic regulatory mechanisms in atherosclerosis remain enigmatic. This study investigates how F. nucleatum reshapes the non-coding RNA landscape to drive atherosclerosis progression. Periodontal infection with F. nucleatum significantly increased atherosclerotic lesion area (p < 0.001) and necrotic core ratio, while reducing collagen content (p < 0.05) in ApoE-/- mice. Whole transcriptome sequencing of arterial tissues identified 847 differentially expressed mRNA

Circular RNAs (circRNAs), a class of non-coding RNAs, are critical regulators of liver injury repair. In this study, using a CCl4-induced mouse liver injury model, we conducted high-throughput sequencing to identify circRNAs enriched in the cell cycle pathway. Four circRNAs (circ_0000604, circ_0001350, circ_0001829, and circ_0001830) were significantly upregulated in early liver injury, with dynamic expression patterns closely linked to repair. Of these, circ_0001829 was selected for functional validation. Circ_0001829 overexpression promoted FL83B cell proliferation and alleviated G2/M phase

Long non-coding RNAs (lncRNAs) stand as newly-arisen molecular types that exert regulatory effects, able to operate as competitive endogenous RNAs (ceRNAs) to engage microRNAs (miRNAs) in interaction, resulting in the recovery of target mRNA expression and activity. Increasing evidences indicate that the ceRNA network affects various biological processes in mammals, including development, cellular differentiation, metabolism, immune response, and disease pathogenesis. In teleost fish, the lncRNA-miRNA-mRNA regulatory networks have been reported occasionally. However, up to now, the roles of ln

Nanos2 plays a key role in self-renewing spermatogenic stem cells (SSCs) and maintains the stem cell state during spermatogenesis. Alleles of the Nanos2 gene knockout showed germline ablated but otherwise structurally normal. To identify the probable ceRNA regulator involved in the process of spermatogenesis by Nanos2, whole transcriptome sequencing was performed in the testes between Nanos2 knock out mice and wild type mice. Finally, a total of 8644 Differentially expressed (DE) mRNAs,180 DE miRNAs, 9538 DE lncRNA and 481 DE circRNAs were identified. Three of each RNAs were selected randomly

Circular RNA (circRNAs) functions vital in the pathogenesis and progression of hepatocellular carcinoma (HCC). However, the expressions and functions of certain circRNAs on metastasis and proliferation of that cancer is still unclear. Bioinformation analysis and qRT-PCR indicated that CircC16orf62 was prominent upregulated in HCC of which the expression level was positively associated to cancer's malignant progression. Gain or loss-of-function studies indicated that the reduction of CircC16orf62 expression promotes the proliferation, invasion, and glycolysis of HCC in vitro and in vivo. The bi

BACKGROUND: Recent studies have revealed that circRNA can serve as ceRNA to participate in multiple autoimmune diseases. Our study aims to explore the key circRNA as ceRNA and biomarker for MG.
METHODS: We used circRNA microarray to explore differentially expressed circRNAs (DECs) from MG and compare with control. Then, we predicted the target miRNA associated with DECs and screened miRNAs by the algorithm of random walk with restart (RWR). Next, we constructed the circRNA-miRNA-mRNA ceRNA regulated network (CMMC) to identify the hub objects. Following, we detected the expression of hub-circR

Penile squamous cell carcinoma (PSCC) is becoming increasingly common and posing a severe threat to men's health, particularly in developing countries. The function of long non-coding RNAs (lncRNAs) in PSCC progression remains mysterious. Therefore, we explored the significance of lncRNAs in the competing endogenous RNA (ceRNA) network in PSCC tumor progression. The 5 healthy and 6 tumor tissue samples were subjected to lncRNA sequencing. Using miRcode, LncBase, miRTarBase, miRWalk, and TargetScan, we constructed a ceRNA network of differentially expressed lncRNAs, miRNAs, and mRNAs. Our analy

Alzheimer's disease (AD) is an increasingly neurodegenerative disorder that causes progressive cognitive decline and memory impairment. Despite extensive research, the underlying causes of late-onset AD (LOAD) are still in progress. This study aimed to establish a network of competing regulatory interactions involving circular RNAs (circRNAs), microRNAs (miRNAs), RNA-binding proteins (RBPs), and messenger RNAs (mRNAs) connected to LOAD. A systematic analysis of publicly available expression data was conducted to identify integrated differentially expressed genes (DEGs) from the hippocampus of

Exosomes encompass a great deal of valuable biological information and play a critical role in tumor development. However, the mechanism of exosomal lncRNAs remains poorly elucidated in bladder cancer (BCa). In this study, we identified exosomal lnc-TAF12-2:1 as a novel biomarker in BCa diagnosis and aimed to investigate the underlying biological function. Dual luciferase reporter assay, RNA immunoprecipitation (RIP), RNA pulldown assays, and xenograft mouse model were used to verify the competitive endogenous RNA mechanism of lnc-TAF12-2:1. We found exosomal lnc-TAF12-2:1 up-regulated in urin

Hepatocellular carcinoma (HCC) remains the third leading cause of cancer-related mortality worldwide, with limited treatment efficacy and poor prognosis, particularly in advanced-stage disease. Despite progress in diagnostic imaging and systemic therapies, early detection and effective targeted interventions remain major clinical challenges. Long non-coding RNAs (lncRNAs), a class of regulatory RNA molecules exceeding 200 nucleotides in length, have emerged as essential regulators of oncogenic signaling, metabolism, and tumor microenvironment in HCC. This review provides a comprehensive overvi

This study aimed to explore the expression pattern of circ_0092222 in Alzheimer's disease (AD) and its role in Aβ-induced neuronal apoptosis. circ_0092222 was identified from the GEO dataset. Serum level of circ_0092222 in 95 AD patients and 100 controls was detected by reverse transcription quantitative real-time polymerase chain reaction (RT-qPCR). AD cell model was achieved by treating SH-SY5Y cells with Aβ25-35. Cell viability, cytotoxicity, apoptosis and inflammation were detected. The targeting relationship between circ_0092222 and miR-331-3p, and between miR-331-3p and SEC61A1 was verif

BACKGROUND: Post cardiac arrest brain injury (PCABI) results from global ischemia-reperfusion. Circular RNAs (circRNAs) are stable regulators of post transcriptional networks that may modulate neuronal survival and metabolism after ischemic injury. We investigated the role and mechanism of mmu_circ_0000376 (circ_0000376) in PCABI.
METHODS: Through integrated bioinformatics analysis of circular RNA/microRNA/messenger RNA datasets via the GEO platform, a regulatory network involving circ_0000376/mmu-let-7b-5p/CCND1 was identified. We validated its circular structure, stability, and cytoplasmic

The lncRNA NEAT1 is overexpressed in multiple myeloma (MM) plasma cells and plays a key role in MM pathogenesis. NEAT1 is involved in ceRNA network in several cancers; however, data in MM are virtually absent. This study identified a NEAT1-driven ceRNA network involving 96 miRNAs and 40 target genes, selected as concurrently downregulated in NEAT1-KD AMO1 cells and upregulated in NEAT1-overexpressing AMO1 cells (AMO1-OVX). The co-expression of NEAT1 and the targets was validated in MM patients (GSE116294, GSE13591, GSE6477, CoMMpass), and in NEAT1-KD NCI-H929, LP1, and KMS27 cell lines, showin

Aortic dissection (AD) is a fatal cardiovascular emergency. This study explored the clinical value and mechanism of the LINC00152/miR-145-5p/KLF4 ceRNA axis in AD. 105 ad patients and 55 healthy controls were enrolled. LINC00152, miR-145-5p, and KLF4 levels were measured by qRT-PCR. ROC and Cox regression analyses assessed diagnostic and prognostic value. Dual-luciferase reporter and rescue experiments in Ang II-induced HASMCs validated the ceRNA axis. As a result, LINC00152 and KLF4 were upregulated, while miR-145-5p was downregulated in AD patients. The three molecules showed good diagnostic

Hypertrophic scars (HS) arise from abnormal wound healing characterized by excessive fibroblast proliferation, enhanced migration, and resistance to apoptosis. Although the long non-coding RNA GAS5 (LncRNA GAS5) has been implicated in several fibrotic disorders, its function in HS remains largely unclear. This study investigated whether GAS5 regulates fibroblast behavior through a competing endogenous RNA (ceRNA) mechanism involving miR-205. Human skin tissue fibroblasts (HSTFs) were transfected with a GAS5 overexpression plasmid (pcDNA-GAS5) or miR-205 mimics, and cell proliferation, migratio
