Investigation of the protective effect of melatonin against radiobiological damage in laryngeal tissue due to radiotherapy at different dose rates in rats.

BACKGROUND: In radiotherapy (RT), the dose given to the organs at risk (OAR) is as important as the dose received by the tumor. FF (Flattening Filtered) and FFF (Flattening Filter Free) treatment techniques used in RT offer different advantages in terms of their effects on target and critical organs. Treatment duration varies depending on the number of monitor units (MU) delivered per unit time. The aim of this study was to investigate biochemically and histopathologically the damage in healthy laryngeal tissue exposed to 600 MU/min FF and 2400 MU/min FF ionizing radiation under different frac

Comparative analysis of simultaneous integrated boost and sequential boost radiotherapy in node-positive cervical cancer: dosimetric and radiobiological considerations.

For locally advanced cervical cancer, the standard therapeutic approach involves concomitant chemoradiation therapy, supplemented by a brachytherapy boost. Moreover, an external beam radiotherapy (RT) boost should be considered for treating gross lymph node (LN) volumes. Two boost approaches exist with Volumetric Intensity Modulated Arc Therapy (VMAT): Sequential (SEQ) and Simultaneous Integrated Boost (SIB). This study undertakes a comprehensive dosimetric and radiobiological comparison between these two boost strategies. The study encompassed ten patients who underwent RT for cervical cancer

Three-dimensional dose prediction and validation with the radiobiological gamma index based on a relative seriality model for head-and-neck IMRT.

This study proposes a quality assurance (QA) method incorporating radiobiological factors based on the QUANTEC-determined tumor control probability and the normal tissue complication probability (NTCP) of head-and-neck intensity-modulated radiation therapy (HN-IMRT). Per-beam measurements were conducted for 20 cases using a 2D detector array. Three-dimensional predicted dose distributions within targets and organs at risk were reconstructed based on the per-beam QA results derived from differences between planned and measured doses. Under the predicted dose distributions, the differences betwe

The incorporation of the concept of minimum RBE (RbEmin) into the linear-quadratic model and the potential for improved radiobiological analysis of high-LET treatments.

PURPOSE: The formulation of relative biological effectiveness (RBE) for high linear energy transfer (high-LET) radiation treatments is revisited. The effects of changed production of sub-lethal damage with varying LET is now considered via the RBEmin concept, where RBEmin represents the lower limit to which RBE tends at high doses per fraction. MATERIALS AND METHODS: An existing linear-quadratic formulation for calculating RBE variations with fractional dose for high-LET radiations is modified to incorporate the twin concepts of RBEmax (which represents the value of RBE at an effective dose-p

Raman spectroscopy and atomic force microscopy as tools to evaluate the radiobiological sensitivity of breast cancer cells at different therapeutic doses.

BACKGROUND AND PURPOSE: Radiotherapy is widely used as a treatment for patients with breast cancer (BC). In vitro irradiation of BC cells provides a strategy to investigate early cellular and molecular events induced by ionizing radiation. Characterizing structural and membrane responses may identify biomarkers of radiosensitivity, improving understanding of treatment effects and enabling patient specific optimization. In this work, Raman spectroscopy and atomic force microscopy (AFM) were applied in vitro to investigate mechanisms relevant to BC radiotherapy. MATERIALS AND METHODS: Breast ca

Impact of ataxia-telangiectasia mutated (ATM) loss on radiobiological and immune response to radium-223 in prostate cancer in vitro models.

INTRODUCTION: Personalised medicine approaches are redefining the therapeutic landscape for men with metastatic castration-resistant prostate cancer (mCRPC). While poly(ADP-ribose) polymerase (PARP) inhibitors have demonstrated clinical benefit in patients with BRCA1/2 mutant tumours, the therapeutic efficacy of these inhibitors in Ataxia-telangiectasia mutated (ATM)-mutated prostate cancer patients remains modest, highlighting the need for alternative treatment strategies. This study aimed to elucidate the impact of ATM loss on radiobiological and immune responses to different radiation modal

Reevaluating BED in cervical cancer HDR brachytherapy: source decay and tissue-specific repair significantly impact radiobiological dose.

OBJECTIVE: This study aims to investigate the influence of intrafraction DNA damage repair on biologically effective dose (BED) in Ir-192 high-dose-rate (HDR) brachytherapy (BT) for cervical cancer. Specifically, we examine BED variations resulting from source decay at various treatment time points across different tumor cell lines and normal tissues. METHODS: Instead of the simplified BED formula, which does not account for intrafraction and interfraction repair or tumor repopulation, we applied the generalized BED (BEDg) formula. BED values for various subtypes of cervical cancer tissues an

The radiobiological effect of using Acuros XB vs anisotropic analytical algorithm on hepatocellular carcinoma stereotactic body radiation therapy.

The purpose of this work was to study the radiobiological effect of using Acuros XB (AXB) vs Analytic Anisotropic Algorithm (AAA) on hepatocellular carcinoma (HCC) stereotactic body radiation therapy (SBRT). Seventy SBRT volumetric modulated arc therapy (VMAT) plans for HCC were calculated using AAA and AXB respectively with the same treatment parameters. Published tumor control probability (TCP) and normal tissue complication probability (NTCP) models were used to quantify the effect of dosimetric difference between AAA and AXB on TCP, NTCP and uncomplicated tumor control probability (UTCP).

The Radiobiological Effect of Flattening Filter Free (FFF) Beam Delivery Parameters on A549 and MCF-7 Cell Survival: An In Vitro Study.

BACKGROUND: Recent improvements, such as flattening filter-free (FFF) beams, enable the delivery of treatments at higher dose rates and in shorter times. However, the biological influence still remains uncertain. Here, we examined how dose rate and dose per pulse influence responses to FFF beams compared with flattened beams in cancer cells with different radiosensitivities. METHODS: A549 (lung, high α/β) and MCF-7 (breast, low α/β) cells were irradiated using VMAT (6 MV FF, 6 and 10 MV FFF; 600-2400 MU/min; 5-20 Gy). Cell viability was assessed by the WST-1 assay, and reproductive survival b

Constraints, Cumulative Doses, and Toxicity Outcomes in Glioma Reirradiation: A Systematic Review and Radiobiological Comparison From the Reirradiation Collaborative Group (ReCOG).

There remains considerable uncertainty regarding normal tissue tolerances to reirradiation for recurrent brain tumours. This systematic review aimed to collate reirradiation constraints, planned cumulative organ at risk (OAR) doses and corresponding toxicity outcomes for adults treated with reirradiation for recurrent glioma. The Medline, Embase, Cochrane, Web of Science and Scopus databases were searched for studies published up to 1st July 2025. To allow comparison between constraints, where necessary, equivalent doses in 2Gy fractions (EQD2Gy) were calculated. In total, 27 studies were incl

Radiobiological damage by space radiation: extension of the BIANCA model to heavy ions up to iron, and pilot application to cosmic ray exposure.

Space research seems to be object of a renewed interest, also considering that human missions to the Moon, and possibly Mars, are being planned. Among the risks affecting such missions, astronauts' exposure to space radiation is a major concern. In this work, the question of the evaluation of biological damage by Galactic Cosmic Rays (GCR) was addressed by a biophysical model called BIophysical ANalysis of Cell death and chromosome Aberrations (BIANCA), which simulates the induction of cell death and chromosome aberrations by different ions. While previously BIANCA has been validated for calcu

The Tissue-Sparing Effect of Spatially Fractionated X-rays for Maintaining Spermatogenesis: A Radiobiological Approach for the Preservation of Male Fertility after Radiotherapy.

Radiotherapy can result in temporary or permanent gonadal toxicity in male cancer patients despite the high precision and accuracy of modern radiation treatment techniques. Previous radiobiological studies have shown an effective tissue-sparing response in various tissue types and species following exposure to spatially fractionated radiation. In the present study, we used an ex vivo mouse testicular tissue culture model and a conventional X-ray irradiation device to evaluate the tissue-sparing effect (TSE) of spatially fractionated X-rays for the protection of male fertility from radiotherapy
