Two-pore channel 1 (TPC1) is an endosomal Na+/Ca2+-selective channel implicated in membrane trafficking, endosome tubulation, and excitability, but how TPC1 regulates membrane trafficking is unknown. Using TPC1-null human cells, we demonstrate that TPC1 drives transferrin receptor (TfR) trafficking and recycling via Ca2+, and not Na+ fluxes or endosomal pH changes, since channel-targeted Ca2+-buffers inhibited trafficking, whereas a Na+-deficient Ca2+-permeable TPC1 mutant fully supported trafficking. TPC1 was unique since other Ca2+ sources did not support TfR trafficking. TPC1 activity depen

BACKGROUND: Transferrin receptor 1 (TFR1), a major iron receptor for immune cells, could impair T cell metabolism and function in systemic lupus erythematosus (SLE), leading us to investigate the effects of TFR1 and possible mechanisms on lupus B cells.
METHODS: B cells from lupus mouse models and systemic lupus erythematosus (SLE) patients were evaluated using flow cytometry (FCM) for levels of TFR1, intracellular iron deposition, reactive oxygen species (ROS), lipid peroxidation, and B-cell subsets. Transcript levels of TFR1 were assessed using reverse transcription-quantitative polymerase

BACKROUND AND AIM: Lichen planus (LP) is a chronic inflammatory dermatosis with an unclear pathogenesis. Recent studies suggest that iron metabolism, oxidative stress, and ferroptosis mechanisms may play a role in the development of LP. Transferrin Receptor 2 (TFR2), an important receptor in iron homeostasis, may contribute to these processes.
METHODS: TFR2 immunoreactivity was evaluated using immunohistochemical methods on skin biopsies obtained from patients diagnosed with cutaneous LP (n = 45) and healthy controls (n = 30). Staining intensity was measured with QuPath image analysis softwar

Human transferrin receptor 1 (TfR) is essential for cellular iron homeostasis by internalizing the iron carrier proteins transferrin and ferritin. It is also an entry point for various pathogens, such as South American hemorrhagic fever caused by arenaviruses and the malaria parasite Plasmodium vivax, which utilize TfR to gain access to cells. The receptor is additionally upregulated in many aggressive cancers and at the blood-brain barrier. Altogether, the TfR is a highly relevant target for many medical applications, and novel protein-interacting partners are sought after. A protein design s

BACKGROUND: Progressive supranuclear palsy (PSP) is a major atypical parkinsonism. Because diagnosis based on the cardinal clinical features is often difficult, misdiagnosis with Parkinson's disease (PD) and multiple system atrophy (MSA) is common in PSP patients. Iron metabolism genes are reportedly involved in tau-accumulating neuronal cell death and ferroptosis in PSP, which is more severe than PD and MSA. The validity of transferrin receptor (TfR) expression as a biomarker of ferroptosis was also demonstrated.
OBJECTIVE: We investigated whether TfR and the TfR to ferritin ratio in the cer

Nitric oxide (NO) is a gasotransmitter that avidly binds both free and heme-bound iron, forming relatively stable iron nitrosyl compounds (FeNOs). We have previously demonstrated that FeNOs are present in the human placenta and are elevated in preeclampsia and intrauterine growth restriction. The ability of NO to sequester iron raises the possibility of the NO-mediated disruption of iron homeostasis in the placenta. In this work, we tested whether exposure of placental syncytiotrophoblasts or villous tissue explants to sub-cytotoxic concentrations of NO would elicit the formation of FeNOs. Fur

Affibodies targeting amyloid-beta (Aβ) could potentially be used as therapeutic and diagnostic agents in Alzheimer's disease (AD). Affibodies display suitable characteristics for imaging applications such as high stability and a short biological half-life. The aim of this study was to explore brain delivery and retention of Aβ protofibril-targeted affibodies in wild-type (WT) and AD transgenic mice and to evaluate their potential as imaging agents. Two affibodies, Z5 and Z1, were fused with the blood-brain barrier (BBB) shuttle single-chain variable fragment scFv8D3. In vitro binding of 125I-l

Iron homeostasis is essential for maintaining cellular function in a wide range of cell types. However, whether iron affects the thermogenic properties of adipocytes is currently unknown. Using integrative analyses of multi-omics data, transferrin receptor 1 (Tfr1) is identified as a candidate for regulating thermogenesis in beige adipocytes. Furthermore, it is shown that mice lacking Tfr1 specifically in adipocytes have impaired thermogenesis, increased insulin resistance, and low-grade inflammation accompanied by iron deficiency and mitochondrial dysfunction. Mechanistically, the cold treatm

Hydrogen sulfide (H2 S) has a significant effect on the regulation of interleukin-6 (IL-6) and signal transducer and activator of transcription 3 (STAT3) activities, while IL-6 directly regulates hepcidin expression via STAT3. We therefore hypothesized that H 2 S has a role in body iron homeostasis by regulating the expression of iron transport proteins via the IL-6/STAT3/Hepcidin pathway. Here, we investigated the effects of two H 2 S donors sodium hydrosulfide and GYY4137 on the expression of ferroportin-1 (Fpn1), transferrin receptor-1 (TfR1), hepcidin, IL-6 and pSTAT3 in the spleen of mice

The Pierre Robin Sequence (PRS), consisting of cleft palate, glossoptosis and micrognathia, is a common human birth defect. However, how this abnormality occurs remains largely unknown. Here we report that neural crest cell (NCC)-specific knockout of transferrin receptor (Tfrc), a well known transferrin transporter protein, caused micrognathia, cleft palate, severe respiratory distress and inability to suckle in mice, which highly resemble human PRS. Histological and anatomical analysis revealed that the cleft palate is due to the failure of palatal shelves elevation that resulted from a retar

As a possible route for invasion of the CNS, circulating poliovirus (PV) in the blood is believed to traverse the blood-brain barrier (BBB), resulting in paralytic poliomyelitis. However, the underlying mechanism is poorly understood. In this study, we demonstrated that mouse transferrin receptor 1 (mTfR1) is responsible for PV attachment to the cell surface, allowing invasion into the CNS via the BBB. PV interacts with the apical domain of mTfR1 on mouse brain capillary endothelial cells (MBEC4) in a dose-dependent manner via its capsid protein (VP1). We found that F-G, G-H, and H-I loops in

Machupo virus (MACV) is a highly pathogenic New World arenavirus that causes hemorrhagic fever in humans. MACV, as well as other pathogenic New World arenaviruses, enter cells after their GP1 attachment glycoprotein binds to their cellular receptor, transferrin receptor 1 (TfR1). TfR1 residues essential for this interaction have been described, and a co-crystal of MACV GP1 bound to TfR1 suggests GP1 residues important for this association. We created MACV GP1 variants and tested their effect on TfR1 binding and virus entry to evaluate the functional significance of some of these and additional

The transferrin (Tf) receptor of Trypanosoma brucei (TbTfR) is encoded by two expression-site-associated genes, ESAG6 and ESAG7. There are around 20 different expression sites containing different copies of these genes that encode TbTfRs with quite distinct affinities for Tf of various hosts. It was proposed that T. brucei has developed multiple expression sites encoding different TbTfRs to ensure sufficient iron uptake in the presence of antibodies competing for binding to Tf. Here it is shown that anti-TbTfR antibody titres produced during chronic murine trypanosomiasis are only one-tenth of

The objective of this investigation was to study the effects of sex steroids on levels of haemoglobin (Hb) and haematocrit (Hct) and to analyse whether these effects can be related to levels of the soluble transferrin receptor (sTfR), a marker of erythropoietic activity. Nineteen male-to-female transsexuals were randomly assigned to either oral ethinyl oestradiol (EE) (n=12) or transdermal 17beta-oestradiol (E2) (n=7); both treatments included the anti-androgen cyproterone acetate (CA). Six male-to-female transsexuals were treated with CA only. Fifteen female-to-male transsexuals were treated

Transferrin, a serum iron-binding protein, delivers iron to the cell after binding to specific receptors on the cell surface and is an important component of culture medium for virtually all cell lines, including the FRTL5 line of rat thyroid follicular cells. Therefore, we undertook studies in FRTL5 cells to examine the regulation of the transferrin receptor, the effects of transferrin on growth and differentiated functions, and the interactions of transferrin with several mitogenic pathways. FRTL5 cells possess one class of saturable transferrin receptors (Ka, 0.7 x 10(9) M-1). Binding of 12

The immunohistochemical expression of insulin-like growth factor I and the transferrin receptor was investigated in regenerating rat skeletal muscle. Muscle injury was induced in the extensor digitorum longus muscle in one hind limb by tourniquet ischemia. The regenerating muscles were investigated 3 to 7 days after the injury. Expression of IGF-I and the transferrin receptor was demonstrated in cryostat sections by a double-staining method. It was found that both markers were transiently expressed by the regenerating muscle cells. Expression of IGF-I immunoreactivity preceded that of the tran

When human erythroleukemic (K562) cells were incubated with 25 microM trifluoperazine (TFP), a drug that inhibits both calmodulin-dependent and calcium-activated phospholipid-dependent kinases, the number of transferrin receptors detected on the cell surface was reduced to approximately half with no change in the affinity of the remaining surface receptors. Removal of the TFP from the incubation medium reversed the loss of surface receptors and they returned to the cell surface in an apparently synchronous manner. As a result, the number of receptors detected on the cell surface exceeded the o

The distribution of an epitope of the transferrin receptor in the human uterine cervical epithelium has been investigated. Immunohistochemical staining, both immunofluorescent and immunoperoxidase, was performed on biopsy specimens and cytological samples from normal, dysplastic, and neoplastic cervical epithelia using the monoclonal OKT9 antibody. The results of staining 145 cervical biopsy specimens with OKT9 showed widespread staining in all malignant epithelia and most severely dysplastic epithelia. No such staining was seen in either normal epithelia or in mildly dysplastic epithelia apar

The transferrin receptor, present on reticulocytes and nucleated cells in tissue culture, has been measured with both immunoassay techniques and transferrin binding studies. The total cellular immunoreactive receptor is rapidly lost from erythrocytes during the process of reticulocyte maturation (from as many as 400,000 molecules to <20,000 molecules/reticulocyte). This event parallels the loss of cell surface transferrin binding sites and RNA content, and correlates with previous studies that have measured the decline in hemoglobin synthesis.Nonhemoglobin-producing normal human fibroblasts, w
