A limited role for ghrelin in heroin self-administration and food deprivation-induced reinstatement of heroin seeking in rats.

Food deprivation (FD) or restriction augments the locomotor activating and reinforcing effects of drugs of abuse. It has been proposed that these effects might be mediated by FD-induced increase in plasma levels of ghrelin, a 28-amino acid orexigenic peptide demonstrated to functionally interact with the mesolimbic dopaminergic system. However, a role for ghrelin has been demonstrated only with psychostimulant drugs and alcohol associated behaviors. We therefore examined the role of ghrelin in ongoing heroin self-administration and FD-induced reinstatement of extinguished heroin seeking. As ex

Effects of methsuximide and mephenytoin on the responding of pigeons under a fixed-consecutive-number schedule with and without an external discriminative stimulus.

The effects of the antiepilepsy drugs methsuximide and mephenytoin were examined in pigeons responding under a fixed-consecutive-number (FCN) schedule with and without an added external discriminative stimulus. On this schedule, food was delivered whenever subjects responded between 8 and 12 times on one response key (work key), and then responded once on a second response key (reinforcement key). Under one variant of the FCN schedule (FCN 8-SD), an external discriminative stimulus signalled completion of the response requirement on the work key; no such stimulus change occurred under the othe

The extinction burst: Effects of alternative reinforcement rate.

Discontinuing reinforcement for an operant response can sometimes produce a transient increase in the rate of that response (i.e., an extinction burst). Although differential reinforcement of alternative behavior (DRA) can attenuate the extinction burst, the effects of rate of alternative reinforcement on the extinction burst have never been examined. Using a within-session transition from a variable-interval (VI) 1.5-s schedule to extinction, we examined how arranging different rates of reinforcement for an alternative response during extinction affected target and alternative responding of r

Response-contingent cocaine increases the reinforcing effectiveness of social contact.

Epidemiological studies report a high concordance rate of drug use within groups, suggesting an interplay between drug reinforcement and social cohesion. Preclinical studies reveal that (a) contingent access to a social partner increases cocaine intake and (b) experimenter-delivered cocaine increases the reinforcing effects of social contact. The purpose of this study was to determine if response-contingent cocaine increases the reinforcing effectiveness of social contact. Male rats were implanted with intravenous catheters and trained on a fixed ratio (FR1) schedule for 30-s access to a socia

Pre-exposure of rats to amphetamine sensitizes self-administration of this drug under a progressive ratio schedule.

Two groups of male rats were tested to determine whether pre-exposure to d-amphetamine would enhance the motivation to self-administer the drug under a progressive ratio schedule of reinforcement. In the first phase of the experiment, one group of rats received d-amphetamine (2 mg/kg IP), while a second group received saline on alternate days for a total of ten injections. Following a 21-day drug withdrawal period, behavioral sensitization was confirmed by a significant increase in amphetamine-induced stereotypy in the d-amphetamine-pretreated group, relative to the saline-pretreated group. Effects of chlordiazepoxide and flumazenil on preference for punished and unpunished response alternatives in rats.

Male food-restricted hooded rats were trained to respond on a two-component multiple schedule. Reinforcement density was several times higher in one component than in the other. However, responses were intermittently punished with shock in the richer reinforcement component (conflict situation). Shock intensities were adjusted to produce mild and strong suppression of responding in two separate phases. Half of the rats controlled which component was operating (Preference group) and half did not (Yoked group). The effect of chlordiazepoxide (CDZ; 0, 1, 3, and 10 mg/kg; IP) was measured on compo

Effects of antianxiety and antipsychotic drugs on DRL responding for brain stimulation.

Satiated rats could be trained to give stable rates of responding for rewarding stimulation of the lateral hypothalamus delivered on differential reinforcement of low rate (DRL) schedule requiring 2 to 8 sec interresponse intervals for reinforcement (DRL-2 to 8). The performance on a DRL-8 schedule was tested 30 min after the oral administration of benzodiazepines. Diazepam (5 and 10 mg/kg) and meprobamate (200 mg/kg) caused significant increases in response rates during the first 5 min of a session, but not thereafter. Bromazepam (1 and 5 mg/kg) also caused a significant increase in the rates

Assessing potency of high- and low-preference reinforcers with respect to response rate and response patterns.

Previous research has suggested that the availability of high-preference stimuli may override the reinforcing efficacy of concurrently available low-preference stimuli under relatively low schedule requirements (e.g., fixed-ratio 1 schedule). It is unknown if similar effects would be obtained under higher schedule requirements. Thus, the current study compared high-preference and low-preference reinforcers under progressively increasing schedule requirements. Results for 3 of the 4 participants indicated that high-preference stimuli maintained responding under higher schedule requirements rela

On the effects of noncontingent delivery of differing magnitudes of reinforcement.

We conducted a parametric analysis of response suppression associated with different magnitudes of noncontingent reinforcement (NCR). Participants were 5 adults with severe or profound mental retardation who engaged in a manual response that was reinforced on variable-ratio schedules during baseline. Participants were then exposed to NCR via multielement and reversal designs. The fixed-time schedules were kept constant while the magnitude of the reinforcing stimulus was varied across three levels (low, medium, and high). Results showed that high-magnitude NCR schedules produced large and consi

Schedule-induced defecation.

Excessive defecation, typically considered to be a concomitant of stress, was experimentally induced or eliminated under specific schedules of positive reinforcement of lever pressing by rats. The schedules were, by and large, those under which polydipsia is typically induced. In the first of three experiments, rats under fixed-interval 32-second schedules and variable interval 32-second schedules for food and water reinforcers defecated profusely, but not under fixed-interval one-second schedules or other small interval schedules. Somewhat higher rates of defecation were observed on variable

Drug discrimination in rats under concurrent variable-interval variable-interval schedules.

Eight rats were trained to discriminate pentobarbital from saline under a concurrent variable-interval (VI) VI schedule, on which responses on the pentobarbital-biased lever after pentobarbital were reinforced under VI 20 s and responses on the saline-biased lever were reinforced under VI 80 s. After saline, the reinforcement contingencies programmed on the two levers were reversed. The rats made 62.3% of their responses on the pentobarbital-biased lever after pentobarbital and 72.2% on the saline-biased lever after saline, both of which are lower than predicted by the matching law. Delineating anticipatory arousal from reward consumption: evaluating fixed-intervals in cocaine seeking-taking response chains.

RATIONALE: Anticipation is a critical antecedent to drug use, in which the prospect of imminent drug availability can potently motivate instrumental actions directed to procure it. Models that capture the behavioral dynamics that precede drug access may allow for the dissociation of key neural mechanisms underlying appetitive or consummatory processes in drug self-administration. OBJECTIVES: We aimed to isolate measurements attributed to the procurement and consumption of a reward by defining distinct actions for each using a chain-schedule of reinforcement. METHODS: Male Long-Evans rats wer

Deep brain stimulation of the nucleus accumbens core but not shell reduces motivational components of heroin taking and seeking in rats.

BACKGROUND: Deep brain stimulation is explored as a new intervention for treatment-resistant substance use dependence. A candidate brain region is the nucleus accumbens, due to its involvement in reward and motivation. This study aimed to explore effects of NAcore and NAshell deep brain stimulation on aspects of heroin taking and seeking in a self-administration model for rats. METHODS: NAcore and NAshell deep brain stimulation was applied during 25 or 100 µg/kg/infusion heroin self-administration on an FR4 schedule of reinforcement and during cue- and heroin-induced reinstatement. Behavioral momentum in computer-presented discriminations in individuals with severe mental retardation.

Behavioral momentum was examined in 2 individuals with severe mental retardation via within-subject manipulations of obtained reinforcer rates. Subjects performed self-paced discrimination problems presented on a touch screen computer monitor. Two different problems, Tasks A and B, alternated in blocks of 15 trials on a multiple schedule. Reinforcers were snack foods. The reinforcement schedule for Task A was continuous (fixed-ratio 1) and the schedule for Task B was continuous in some conditions and variable ratio in other conditions. Behavioral momentum was assessed in test sessions by prefe

Application of operant demand in autism and developmental disabilities: An updated review (2017-2025).

Operant demand within behavioral economics provides a quantitative framework for evaluating reinforcer efficacy in applied research involving individuals with autism and those with related developmental disabilities. This systematic review extended the work of Gilroy, Kaplan, and Leader (2018) by examining applied operant demand research published between March 2017 and December 2025. Following PRISMA guidelines, 30 articles (31 experiments; 307 participants) were identified and coded for procedural features and methods, including operant arrangements, demand and work output analyses, open and

Contingency discrimination training and resurgence: Effects of nonzero alternative reinforcement rates during "off" sessions.

Resurgence is an increase in a previously reduced target behavior with worsening conditions for a more recently reinforced alternative behavior. Alternating sessions in which alternative reinforcement is available (i.e., "on" sessions) versus extinction (i.e., "off" sessions) during treatment (i.e., contingency discrimination training [CDT]) reduces resurgence. This experiment examined whether CDT could mitigate resurgence without complete extinction during "off" sessions. Target lever pressing of rats was reinforced in baseline and then extinguished during treatment. In treatment, all groups

Attack produced by intermittent reinforcement of a concurrent operant response.

Squirrel monkeys pressed a lever to produce food on several fixed-ratio schedules; they could also bite a rubber hose. Biting attack occurred during the postreinforcement pause and in early portions of the ratio response run. Also, biting attacks increased after transitions to higher values of the fixed-ratio requirement and in extinction. The results show that extinction-induced aggression effects occur in primates.
