Bluetongue virus vaccine: conventional to modern approach.

Bluetongue (BT) is an economically important, infectious and non-contagious disease of ruminant animals. BT disease is caused by bluetongue virus (BTV) of the genus Orbivirus (the family Reoviridae). BTV is transmitted by certain species of biting midges of the genus Culicoides. Although originally BT was restricted to African continent, now it is present in all the continents except Antarctica. Conventional BT vaccines such as live attenuated and inactivated vaccines showed different degree of success in BT control. However, conventional vaccines have certain disadvantages of reversion to vir

A viral metagenomic survey identifies known and novel mammalian viruses in bats from Saudi Arabia.

Bats are implicated as natural reservoirs for a wide range of zoonotic viruses including SARS and MERS coronaviruses, Ebola, Marburg, Nipah, Hendra, Rabies and other lyssaviruses. Accordingly, many One Health surveillance and viral discovery programs have focused on bats. In this report we present viral metagenomic data from bats collected in the Kingdom of Saudi Arabia [KSA]. Unbiased high throughput sequencing of fecal samples from 72 bat individuals comprising four species; lesser mouse-tailed bat (Rhinopoma hardwickii), Egyptian tomb bat (Taphozous perforatus), straw-colored fruit bat (Eid

Co-circulation of multiple Colorado tick fever virus genotypes.

Colorado tick fever (CTF) virus, family Reoviridae, genus Orbivirus, contains 12 genes distinguishable by polyacrylamide gel electrophoresis (PAGE). Multiple genotypes of CTF virus were isolated at 3 field sites in Colorado in 1985. Five genotypes were found at Campos Cabin, 2 at Drake, and 6 at Rocky Mountain National Park. Virus isolations were made in 1985 from 6 patients with CTF. These isolates were distinct from each other and the field isolates. Although the CTF isolates were different by PAGE profile, the majority of the 12 genes were highly conserved among the 1985 isolates and a Flor

Novel human reovirus isolated from children and its long-term circulation with reassortments.

Mammalian orthoreovirus (MRV), also known as reovirus, was discovered in the 1950s and became the first reported segmented double-stranded RNA virus. MRVs have since been found in a variety of animal species, including humans. However, reports on MRV infections are scarce due to the rarity of their symptomatic occurrence. In Japanese surveillance studies, MRVs have been detected as gastrointestinal pathogens since 1981, with a total of 135 records. In Osaka City, Japan, MRV was first isolated in 1994 from a child with meningitis, and then in 2005 and 2014 from children with gastroenteritis. He

Experimental reproduction of enteritis in bobwhite quail (Colinus virginianus) with Cryptosporidium and reovirus.

Five-day-old bobwhite quails were inoculated with reovirus and Cryptosporidium previously isolated from the intestinal contents of young, commercially raised bobwhite quails experiencing severe enteritis. Quails inoculated with reovirus alone did not develop clinically apparent disease, infection was localized principally in the intestinal tract, and no lesions were detected. Quails inoculated with Cryptosporidium, alone or with reovirus, developed severe enteritis with high mortality and marked growth depression. Cryptosporidia caused blunting of intestinal villi and provoked a mononuclear ce

Influence of feed on the course of tenosynovitis caused by REO virus infection in broilers.

The first outbreak of tenosynovitis caused by infection with REO virus in the Netherlands, involving 15 broiler flocks, is described. The disease could be reproduced easily by subcutaneous, oral and contact infection of susceptible broilers with the isolated virus. The 15 affected flocks all came from one broiler parent flock and were fed with feed from one mill (A). Twenty other flocks, also coming from this parent flock and reared in the same period on feed from another feed mill (B) did not suffer from tenosynovitis. The parent flock showed no disease symptoms but seroconversion to REO viru

Identification and analysis of lncRNA50877, which is involved in KRT8-mediated mitochondrial homeostasis and GCRV replication in grass carp (Ctenopharyngodon idella).

Grass carp (Ctenopharyngodon idella) reovirus (GCRV), the causative agent of hemorrhagic disease, causes substantial economic losses in the grass carp industry every year. However, the biological regulatory roles of long non-coding RNAs (lncRNAs) during GCRV infection remain largely unclear. In this study, high-throughput sequencing technology was used to analyze the expression profiles of lncRNAs in GCRV-infected and mock-infected liver and intestinal tissues of grass carp. In this study, compared with those in the control group, the transcriptomic profiles of grass carp after GCRV genotype I

Sexual dimorphism of antiviral immunity in grass carp (Ctenopharyngodon idella): TLR7 and TLR8b promote resistance to GCRV in males, whereas TLR8a increases susceptibility in females.

Sexual dimorphism of immune responses is widespread in vertebrates. In mammals, Toll-like receptor 7 (TLR7) and TLR8 are tandemly localized on the X chromosome and constitute the key molecular basis for sex-biased immunity. However, most teleosts lack typical sex chromosomes, and due to whole-genome duplication (WGD), the TLR family has expanded and functionally diverged. In grass carp (Ctenopharyngodon idella), CiTLR7 and CiTLR8a are tandemly located on chromosome 9, while CiTLR8b resides on chromosome 10. The two TLR8 paralogs have undergone functional divergence, yet their involvement in se

ATG5 negatively regulates grass carp reovirus-induced immune-inflammatory response by degrading RIG-I and MDA5.

UNLABELLED: Grass carp reovirus (GCRV) is the most virulent aquareovirus, inducing hemorrhagic disease that results in significant economic losses for the grass carp aquaculture industry in China. It has been reported that the cytokine storm, resulting from the viral-induced acute immune-inflammatory response, is the primary cause of death associated with viral infections. Therefore, exploring the negative regulatory factors of the immune response may provide important targets for virus prevention and control. In this study, we demonstrated that GCRV infection induces an acute immune-inflammat

The structural characteristics and expression characteristics of C1S in response to GCRV infection in grass carp.

The complement system, a critical component of innate immunity in fish, plays a pivotal role in the defense against Grass Carp Reovirus (GCRV) infection in grass carp. This study explores the structural characteristics of C1S, a crucial molecule in the classical pathway of the complement system, and its involvement in the response to GCRV infection. We found that the grass carp C1S gene comprises six domains similar to those in mammals: two CUB (Complement C1r/C1s, Uegf, Bmp1) domains, two CCP (Complement control protein) domains, one EGFCA (Calcium-binding epidermal growth factor) domain, and

Functional Identification of Complement Factor D and Analysis of Its Expression during GCRV Infection in Grass Carp (Ctenopharyngodon idella).

Complement factor D (Df) is a serine protease well known for activating the alternative pathway (AP) in mammals by promoting the cleavage of complement component 3 (C3), thus becoming involved in innate defense. In teleost fish, however, the functional mechanisms of Df in the AP and against pathogen infection are far from clear. In the present study, we cloned and characterized the Df gene, CiDf, from grass carp (Ctenopharyngodon idella) and analyzed its function in promoting C3 cleavage and expression changes after grass carp reovirus (GCRV) infection. The open reading frame of CiDf was found

The intestinal virome of malabsorption syndrome-affected and unaffected broilers through shotgun metagenomics.

Malabsorption syndrome (MAS) is an economically important disease of young, commercially reared broilers, characterized by growth retardation, defective feather development and diarrheic faeces. Several viruses have been tentatively associated to such syndrome. Here, in order to examine potential associations between enteric viruses and MAS, the faecal viromes of 70 stool samples collected from diseased (n = 35) and healthy (n = 35) chickens from seven flocks were characterized and compared. Following high-throughput sequencing, a total of 8,347,319 paired end reads, with an average of 231 nt,

Protective efficacy of multivalent replication-abortive vaccine strains in horses against African horse sickness virus challenge.

African horse sickness virus (AHSV) is an orbivirus, a member of the Reoviridae family. Nine different serotypes have been described so far. AHSV is vectored by Culicoides spp. to equids, causing high mortality, particularly in horses, with considerable economic impacts. For development of a safe attenuated vaccine, we previously established an efficient reverse genetics (RG) system to generate Entry Competent Replication-Abortive (ECRA) virus strains, for all nine serotypes and demonstrated the vaccine potential of these strains in type I interferon receptor (IFNAR)-knockout mice. Here, we ev

An oral multivalent fusion vaccine based on antigenic fragment VP56310-500 and FlaC adjuvant confers effective protection against grass carp reovirus.

Grass carp reovirus (GCRV) poses a serious economic threat to grass carp aquaculture and oral vaccination has become the strategy of choice for its control. In this study, multiple sequence alignment was conducted on the sequences of the VP56 of seven GCRV-II strains. The most frequent amino acid at each site was assigned to the fusion sequence to obtain the sequences of GCRV-FUSION (VP561-512). Through antigenic index analysis, the full-length sequence (VP561-512) was truncated, thereby obtaining five truncated antigen fragments: VP561-210, VP56100-209, VP56210-310, VP56310-500, and VP56410-5

Experimental infection in specific-pathogen-free chicks with avian reovirus and avian nephritis virus isolated from broiler chicks showing runting syndrome.

Avian reovirus (ARV) and avian nephritis virus (ANV) were individually isolated from runty 10-day-old broiler chicks. The ARV isolate, IR-R, the ANV isolate, IR-N, and the reference strain of ANV, G-4260, were inoculated orally into 1-day-old chicks of two specific-pathogen-free (SPF) chicken lines, 151 and PDL-1. Growth retardation without the presence of gross lesions was clearly observed at 7 and 14 days postinoculation (PI) in chicks of both lines inoculated with the IR-R strain. On the other hand, in chicks inoculated with IR-N strain, growth retardation was observed only in the chicks of

Is lactase the receptor and uncoating enzyme for infantile enteritis (rota) viruses?

Rotaviruses are now regarded as important causes of diarrhoea in man, cattle, pigs, mice, and possibly other animals. Characteristically, disease occurs in newborn and young animals, and infection seems limited to the differentiated gut epithelial cells. The major surface polypeptide of the calf scours rotavirus is glycosylated, and highly purified beta-galactosidase (lactase) interacts with the virus in vitro causing removal of the outer shell of the capsid (uncoating). It is suggested that lactase present in the brush border of the intestinal epithelial cell performs a similar function in vi

Immune Response to Rotavirus and Gluten Sensitivity.

Rotavirus is a double-stranded RNA virus belonging to the family of Reoviridae. The virus is transmitted by the faecal-oral route and infects intestinal cells causing gastroenteritis. Rotaviruses are the main cause of severe acute diarrhoea in children less than 5 years of age worldwide. In our previous work we have shown a link between rotavirus infection and celiac disease. Nonceliac gluten sensitivity (NCGS) is emerging as new clinical entity lacking specific diagnostic biomarkers which has been reported to occur in 6-10% of the population. Clinical manifestations include gastrointestinal a

Characterization of interleukin-20 from black carp and its role in the antiviral innate immunity.

Interleukin-20 (il20) is known to participate in inflammation and host defense in mammals, whereas its immunological role in teleost fish needs to be further explored. In this study, a black carp (Mylopharyngodon piceus) il20 homolog, designated bcil20, was cloned and characterized. The full-length coding sequence of bcil20 encodes a putative protein of 179 amino acids with a predicted signal peptide and a conserved IL-10 family domain. Phylogenetic and structural analyses indicated that bcil20 is evolutionarily conserved. Expression analysis showed that bcil20 was constitutively expressed in
