Role of IscA1 in Low-Frequency Magnetic Field-Associated Changes in Monascus Pigments and Citrinin Levels in Monascus purpureus.

We constructed IscA1 overexpression (IscA1+) and RNA interference (RNAi-IscA1) strains to investigate the role of IscA1 in regulating the synthesis of Monascus pigments (MPs) and citrinin (CIT) in Monascus purpureus under low-frequency magnetic field (LF-MF) exposure. Both IscA1+ and RNAi-IscA1 strains exhibited altered nutritional growth and sexual reproduction; however, asexual reproduction, the ultrastructure of spores and mycelia, and biomass in liquid fermentation, remained unaffected. The production of yellow and orange pigments increased in the IscA1+ strain, while the production of red

Genome-Wide Analyses of Individual Strongyloides stercoralis (Nematoda: Rhabditoidea) Provide Insights into Population Structure and Reproductive Life Cycles.

The helminth Strongyloides stercoralis, which is transmitted through soil, infects 30-100 million people worldwide. S. stercoralis reproduces sexually outside the host as well as asexually within the host, which causes a life-long infection. To understand the population structure and transmission patterns of this parasite, we re-sequenced the genomes of 33 individual S. stercoralis nematodes collected in Myanmar (prevalent region) and Japan (non-prevalent region). We utilised a method combining whole genome amplification and next-generation sequencing techniques to detect 298,202 variant posit

Adapt or Die: Targeting Unique Transmission-Stage Biology for Malaria Elimination.

Plasmodium parasites have a complex life cycle that includes development in the human host as well as the Anopheles vector. Successful transmission of the parasite between its host and vector therefore requires the parasite to balance its investments in asexual replication and sexual reproduction, varying the frequency of sexual commitment to persist within the human host and generate future opportunities for transmission. The transmission window is extended further by the ability of stage V gametocytes to circulate in peripheral blood for weeks, whereas immature stage I to IV gametocytes sequ

Unraveling genetic diversity and population structure of pineapple germplasm using genome-wide SNP markers.

Pineapple (Ananas comosus (L.) Merr.) is one of the most important tropical fruits worldwide. It is primarily propagated clonally and exhibits high heterozygosity which can mask deleterious mutations, potentially leading to inbreeding depression and hindering breeding efforts. To address this, we conducted a comprehensive genome-wide analysis of 91 pineapple accessions and identified over 7.9 million high-quality SNPs. We utilized these SNPs to investigate the genetic structure and reproductive patterns of diverse cultivars and related varieties. Population structure analysis revealed a unique

Genetic diversity and structure of Bipolaris oryzae and Exserohilum rostratum populations causing brown spot of rice in Burkina Faso based on genotyping-by-sequencing.

In recent years, Brown spot disease of rice (BSR) has been observed on leaves and seeds of rice in all rice-growing areas of Burkina Faso. Bipolaris oryzae and Exserohilum rostratum are the main fungal species isolated from BSR infected tissues and they are frequently observed in the same field. However, we are lacking information on the genetic diversity and population structure of these fungi in Burkina Faso. The mode of reproduction is also unknown. The genetic diversity of isolates of B. oryzae (n=61) and E. rostratum (n=151), collected from major rice-growing areas of Burkina Faso, was es

Cloning and Selfing Affect Population Genetic Variation in Simulations of Outcrossing, Sexual Sea Stars.

AbstractMany sea stars are well known for facultative or obligate asexual reproduction in both the adult and larval life-cycle stages. Some species and lineages are also capable of facultative or obligate hermaphroditic reproduction with self-fertilization. However, models of population genetic variation and empirical analyses of genetic data typically assume only sexual reproduction and outcrossing. A recent reanalysis of previously published empirical data (microsatellite genotypes) from two studies of one of the most well-known sea star species (the crown-of-thorns sea star; Acanthaster sp.

A Comprehensive Characterization of Ecological and Epidemiological Factors Driving Perennation of Podosphaera macularis Chasmothecia on Hop (Humulus lupulus).

Hop powdery mildew, caused by the ascomycete fungus Podosphaera macularis, is a consistent threat to sustainable hop production. The pathogen utilizes two reproductive strategies for overwintering and perennation: (i) asexual vegetative hyphae on dormant buds that emerge the following season as infected shoots; and (ii) sexual ascocarps (chasmothecia), which are discharged during spring rain events. We demonstrate that P. macularis chasmothecia, in the absence of any asexual P. macularis growth forms, are a viable overwintering source capable of causing early season infection two to three orde

Human babesiosis: The past, present and future.

Human babesiosis is a disease transmitted by the bite of an infected tick or via blood transfusions involving contaminated blood products; in humans, it can lead to severe complications and even death, depending on the clinical history, age and health status of the affected patient. Babesiosis is caused by members of the Babesia spp., protozoan parasites whose life cycle includes sexual reproduction in the arthropod vector and asexual reproduction in the mainly mammalian host. Cases of human babesiosis have been rare, but there are increasing reports of human babesiosis associated with climati

In vitro production of cat-restricted Toxoplasma pre-sexual stages by epigenetic reprogramming.

Sexual reproduction of Toxoplasma gondii , which is restricted to the small intestine of felids, is sparsely documented, due to ethical concerns surrounding the use of cats as model organisms. Chromatin modifiers dictate the developmental fate of the parasite during its multistage life cycle, but their targeting to stage-specific cistromes is poorly described 1 . In this study, we found that transcription factors AP2XII-1 and AP2XI-2, expressed in tachyzoite stage that causes acute toxoplasmosis, can silence genes necessary for merozoites, a developmental stage critical for sexual commitment a

Asexual propagation of a virulent clone complex in a human and feline outbreak of sporotrichosis.

Sporotrichosis is one of the most frequent subcutaneous fungal infections in humans and animals caused by members of the plant-associated, dimorphic genus Sporothrix. Three of the four medically important Sporothrix species found in Brazil have been considered asexual as no sexual stage has ever been reported in Sporothrix schenckii, Sporothrix brasiliensis, or Sporothrix globosa. We have identified the mating type (MAT) loci in the S. schenckii (strain 1099-18/ATCC MYA-4821) and S. brasiliensis (strain 5110/ATCC MYA-4823) genomes by using comparative genomic approaches to determine the mating

Why are triploid quaking aspen (Populus tremuloides) common?

PREMISE: Quaking aspen is a clonal tree species that has mixed ploidy, often with high relative abundance of both diploids and triploids but no haploids or tetraploids. Triploids typically have low fertility, leaving their occurrence apparently unlikely from an evolutionary perspective, unless they provide a "triploid bridge" to generating higher-fitness tetraploids-which are not observed in this species. This study focused on how triploidy can be maintained in quaking aspen. METHODS: A computational model was used to simulate gamete production, sexual reproduction, asexual reproduction, pare

Oncogenic stress alters predator-prey interactions between a globally invasive and a native planaria species.

Oncogenic pressures, factors that increase tumor risk, are intensifying with human-driven environmental change, yet their ecological effects remain mainly unquantified. We examined how UVB-induced oncogenic stress affects interactions between two Australian freshwater planaria: the globally invasive predator Girardia tigrina and the native prey Cura pinguis. We hypothesized that DNA damage could either heighten prey vulnerability or impair predator performance. Native planaria showed less UVB-induced DNA damage, likely due to darker pigmentation, while damage in G. tigrina reduced predation ra

Metalloaminopeptidases of the Protozoan Parasite Plasmodium falciparum as Targets for the Discovery of Novel Antimalarial Drugs.

Malaria poses a significant threat to approximately half of the world's population with an annual death toll close to half a million. The emergence of resistance to front-line antimalarials in the most lethal human parasite species, Plasmodium falciparum (Pf), threatens progress made in malaria control. The prospect of losing the efficacy of antimalarial drugs is driving the search for small molecules with new modes of action. Asexual reproduction of the parasite is critically dependent on the recycling of amino acids through catabolism of hemoglobin (Hb), which makes metalloaminopeptidases (M

Discrete time model of sexual systems.

Small factors are the biggest contributors to the biggest changes in an ecosystem which may lead to its demise. Simple things such as tweaking the humidity or tampering with the temperature can be the downfall of the habitants and the ecosystem itself. These changes can be observed right now with the consistent temperature increase on Earth along with the slew of issues that come with global warming. A small change in a few degrees in temperature can perish an entire food supply and the habitat of many already endangered beings forcing them to either adapt or die. Within such an ecosystem the

Viable Triploid Honey Bees (Apis mellifera capensis) Are Reliably Produced in the Progeny of CO2 Narcotised Queens.

The haplodiploid system of sex determination of Hymenoptera acts as an exaptation for species to evolve novel forms of asexual reproduction including thelytoky (clonal offspring of the mother). During normal reproduction in Hymenoptera, three of the four products of meiosis that are present in newly-laid eggs are lost as polar bodies, while the remaining pronucleus either develops as a haploid male or fuses with a sperm nucleus to produce a diploid zygote. In contrast, in thelytokous reproduction, which is uncommon but taxonomically widespread, two of the four products of meiosis fuse, as if o

Polyethylenimine-Coated MXene Quantum Dots Integrate Anti-Oomycete Activity with Host Immunity Activation for Plant Oomycete Control.

Pathogenic oomycetes can infect hundreds of crops, leading to significant economic losses globally. This study synthesized polyethylenimine-coated MXene quantum dots (PEI-MQDs) and investigated their antioomycete potential using the vegetable oomycete pathogen Phytophthora capsici as a model system. PEI-MQDs demonstrated potent antioomycete activity against P. capsici by damaging cellular structures and subverting asexual reproduction (zoosporangium/zoospore formation). Transcriptomic profiling demonstrated that treatment with PEI-MQDs induced extensive transcriptional changes in P. capsici ge

A small Candida glabrata protein promotes hyphal growth in C. albicans during mixed-species invasive candidiasis.

During mixed-species invasive candidiasis-an emerging threat in healthcare-the presence of Candida albicans is nearly essential for host invasion by C. glabrata. However, the molecular communication underlying such intriguing inter-species fungal interactions is largely unexplored. Here, we show that C. glabrata secretes a unique small protein, Yhi1-encoded by a likely de novo gene-which induces hyphal growth via Tsa1 in C. albicans. Notably, this Yhi1-based inter-species interaction is specific to C. glabrata and C. albicans compared with other common species of Candida. Our structure-functio

Single-cell transcriptomics of the human parasite Schistosoma mansoni first intra-molluscan stage reveals tentative tegumental and stem-cell regulators.

Schistosomiasis is a major Neglected Tropical Disease, caused by the infection with blood flukes in the genus Schistosoma. To complete the life cycle, the parasite undergoes asexual and sexual reproduction within an intermediate snail host and a definitive mammalian host, respectively. The intra-molluscan phase provides a critical amplification step that ensures a successful transmission. However, the cellular and molecular mechanisms underlying the development of the intra-molluscan stages remain poorly understood. Here, single cell suspensions from S. mansoni mother sporocysts were produced
