Transsynaptic degeneration of lateral geniculate bodies in blind children: in vivo MR demonstration.

PURPOSE: To investigate signal alterations in the thalamic lateral geniculate bodies of blind patients compatible with transsynaptic degeneration of these nuclei caused by pregeniculate or postgeniculate interruption of the visual pathway. METHODS: Six patients were selected from a group of blind children in our care. Four had cerebral palsy caused by periventricular leukomalacia, one had infantile neuroaxonal dystrophy, and one had Chiari I malformation and hydrocephalus, which was worsened by bilateral ischemic lesions of the occipital lobes. MR examinations (obtained at 0.5 T) were reviewe

Disruption of raphé serotonergic neural projections to the cortex: a potential pathway contributing to remote loss of brainstem neurons following neonatal hypoxic-ischemic brain injury.

Neuronal injury is a key feature of neonatal hypoxic-ischemic (HI) brain injury. However, the mechanisms underpinning neuronal losses, such as in the brainstem, are poorly understood. One possibility is that disrupted neural connections between the cortex and brainstem may compromise the survival of neuronal cell bodies in the brainstem. We investigated whether brainstem raphé serotonergic neurons that project to the cortex are lost after HI. We also tested if neuroinflammation has a role in disrupting brainstem raphé projections. Postnatal day 3 (P3) rats underwent unilateral carotid artery l

Motor conduction block and high titres of anti-GM1 ganglioside antibodies: pathological evidence of a motor neuropathy in a patient with lower motor neuron syndrome.

A patient with a progressive lower motor neuron syndrome and neurophysiological evidence of motor axon loss, multifocal proximal motor nerve conduction block, and high titres of anti-ganglioside GM1 antibodies. Neuropathological findings included a predominantly proximal motor radiculoneuropathy with multifocal IgG and IgM deposits on nerve fibres associated with a loss of spinal motor neurons. These findings support an autoimmune origin of this lower motor neuron syndrome with retrograde degeneration of spinal motor neurons and severe neurogenic muscular atrophy.

New aspects of facial nerve pathology in temporal bone fractures.

Electron microscopic examination of intratemporal facial nerve segments removed from 12 patients with persisting facial paralysis following temporal bone fractures revealed that traumatic injury at the geniculum induces retrograde degeneration through the labyrinthine and distal meatal segments of the facial nerve. Fibrosis may occur in the traumatized labyrinthine segment and block regenerating motor fibers. The surgical treatment of traumatic facial nerve injuries should be aimed to avoid or eliminate fibrosis within the labyrinthine segment of the Fallopian canal.

Persistence of cholinergic neurons in the basal nucleus in a brain with senile dementia of the Alzheimer's type demonstrated by immunohistochemical staining for choline acetyltransferase.

Immunohistochemically identified cholinergic neurons in the basal nucleus of Meynert in a brain with SDAT have been compared with those in two age- and sex-matched normal brains. The numbers of such cells at carefully matched levels are not significantly lower, but the cells in SDAT are substantially smaller than in the normal basal nucleus. The persistence of shrunken cholinergic neurons in the basal nucleus in the diseased brain is similar to that seen in an experimental study of retrograde cellular degeneration in the nucleus following damage of the cortex.

Responses of lateral geniculate neurons that survive long-term visual cortex damage in kittens and adult cats.

Damage to visual cortex (areas 17-19) in kittens or adult cats produces severe retrograde degeneration of neurons in the dorsal lateral geniculate nucleus (LGN). However, some neurons survive in otherwise degenerated portions of the LGN after a visual cortex lesion at any age. Previous studies have shown that there are well-defined differences in potential retinal inputs, soma size, synaptic connections, outputs, and physiological properties of output targets of the surviving LGN cells in cats that received visual cortex damage at different ages. The present experiment investigated the relatio

Basal forebrain parvalbumin neuron dysfunction links network oscillation deficits to hippocampal pathology in Alzheimer's disease.

BACKGROUND: Basal forebrain (BF) degeneration is a key pathological feature of Alzheimer's disease (AD) and is closely associated with cognitive decline. Although parvalbumin (PV)-expressing neurons are abundant in the BF and are important regulators of cortical network activity, their contribution to AD pathogenesis remains poorly understood. METHODS: To investigate the role of BF-PV neurons in AD, we examined the effects of PV knockdown in the BF of APP/PS1 mice. Electrophysiological recordings, behavioral analyses, retrograde tracing, hippocampal transcriptome profiling, and integrative co

Glaucoma and the brain: Trans-synaptic degeneration, structural change, and implications for neuroprotection.

A recent hypothesis to enter the literature suggests that glaucoma is a neurodegenerative disease. The basis for this has been the finding of central nervous system changes in glaucoma patients on histology and neuroimaging. It is known that retinal ganglion cell pathology of any cause leads to anterograde and retrograde retinal ganglion cell degeneration, as well as trans-synaptic (transneuronal) anterograde degeneration. Trans-synaptic degeneration has been demonstrated in a range of optic neuropathies including optic nerve transection, optic neuritis, and hereditary optic neuropathies. More

Further characterization of the effects of brain-derived neurotrophic factor and ciliary neurotrophic factor on axotomized neonatal and adult mammalian motor neurons.

Neurotrophins and neural cytokines are two broad classes of neurotrophic factors. It has been reported that ciliary neurotrophic factor (CNTF) and brain-derived neurotrophic factor (BDNF) prevent the degeneration of axotomized neonatal motor neurons. In addition, BDNF is transported retrogradely to alpha-motor neurons following injection into the muscle, and patterns of BDNF expressed in spinal cord and muscle suggest a physiological role for this factor in motor neurons. In the present study, we characterize the effects of BDNF on axotomized neonatal facial motor neurons and extend these obse

Large-scale mapping of the MCH network in ALS mice reveals the vulnerability of dopaminergic and GABAergic neurons in zona incerta.

Weight loss and hypermetabolism are early and prognostically significant features of amyotrophic lateral sclerosis (ALS) and are associated with hypothalamic atrophy and degeneration of melanin-concentrating hormone (MCH) neurons that regulate energy balance. To investigate whether MCH vulnerability arises from upstream network dysfunction, we performed whole-brain retrograde rabies tracing in SOD1G93A mice. We identified an early, selective loss of monosynaptic inputs from the zona incerta (ZI), a dopaminergic (DA)/gamma-aminobutyric acid (GABA)ergic nucleus that preceded MCH neuron degenerat

Dual Role of Microglial TREM2 in Neuronal Degeneration and Regeneration After Axotomy.

Ventral horn microglia in the spinal cord proliferate after nerve injuries and migrate towards the cell bodies of injured motoneurons surrounding them. However, the significance of microglia enwrapping axotomized motoneurons has remained unclear. Moreover, some injured motoneurons degenerate while others regenerate. In mice spinal cords we found that each motoneuron fate associates with microglia of different activation profiles. Microglia surrounding degenerating motoneurons form cell clusters that fully envelop the cell body and express high TREM2 and large CD68 granules, with female microgl

Microglial cell responses in the rabbit retina following transection of the optic nerve.

The response of retinal microglial cells, which accompanies retrograde degeneration of ganglion cell axons and perikarya (induced by transection of the optic nerve), was studied in whole-mounted rabbit retinae labeled enzyme-histochemically for nucleoside diphosphatase (NDPase), which is a microglial cell marker. A few days after transection, the number of microglial cells/mm2, as well as their staining intensity, began to increase in the inner plexiform layer. The mosaic-like distribution of the star-shaped microglial cells present in the inner plexiform layer of a normal rabbit retina was pr

Decreased lateral geniculate nucleus activation in retrogeniculate hemianopia demonstrated by functional magnetic resonance imaging at 4 Tesla.

Functional magnetic resonance imaging (fMRI) can detect lateral geniculate nucleus (LGN) activation. We studied LGN function in 5 patients with retrogeniculate homonymous hemianopia using fMRI at 4.0 Tesla during binocular visual stimulation. Decreased activation of visual cortex and LGN on the side of the lesion was observed in all 5 patients. These findings suggest that retrogeniculate lesions are associated with decreased activation of the LGN, due to retrograde degeneration or a functional decrease caused by decreased feedback from ipsilateral visual cortex.

Clinical and ultrastructural ocular histopathologic studies of adult-onset metachromatic leukodystrophy.

A twin brother and sister with adult-onset metachromatic leukodystrophy developed progressive central acuity loss and optic disk pallor. Both had normal electroretinograms and fluorescein angiography. Postmortem examination of the sister's eyes by histochemistry and electron microscopy revealed ganglion cell loss and optic atrophy. Cerebroside sulfate had accumulated in optic nerve glial cells. Optic atrophy was more advanced than in previously reported cases of infantile-onset metachromatic leukodystrophy. The pathologic process seemed to be retrograde optic nerve degeneration due to abnormal

Hippocampal area CA1 and remote memory in rats.

Hippocampal lesions often produce temporally graded retrograde amnesia (TGRA), whereby recent memory is impaired more than remote memory. This finding has provided support for the process of systems consolidation. However, temporally graded memory impairment has not been observed with the watermaze task, and the findings have been inconsistent with context fear conditioning. One possibility is that large hippocampal lesions indirectly disrupt (by retrograde degeneration) the function of areas that project to the hippocampus that are important for task performance or thought to be important for

Occipital cortex ablation in adult rat causes retrograde neuronal death in the lateral geniculate nucleus that resembles apoptosis.

The mechanisms of retrograde neurodegeneration following axotomy and target deprivation in the adult central nervous system remain poorly understood. We used a unilateral occipital cortex ablation model in adult rats to test the hypothesis that retrograde neurodegeneration in the dorsal lateral geniculate nucleus resembles apoptosis. Using the retrograde tracer Fluorogold, combined with nuclear dyes or the terminal transferase-mediated deoxyuridine triphosphate-biotin nick end labeling method for detecting nuclear DNA fragmentation, apoptotic geniculocortical projection neurons were identified

Valve-in-Valve TAVR for Degenerated Perceval Valves: From Entrapment to Success.

BACKGROUND: Valve-in-valve transcatheter aortic valve replacement (ViV-TAVR) is an established treatment option for bioprosthetic valve failure. The sutureless Perceval bioprosthesis presents unique technical challenges because of the valve's supra-annular nitinol ring, lack of radiopaque markers, and potential difficulty with central valve crossing and entrapment of devices within the supra-annular ring. CASE SUMMARY: We report 2 cases of Perceval degeneration managed with ViV-TAVR using balloon-expandable Edwards Sapien 3 Ultra valves. In the first case, standard retrograde crossing led to

The use of optical coherence tomography in neurology: a review.

Optical coherence tomography is a non-invasive, cost-efficient technique that provides high-resolution in vivo imaging of retinal tissue. The peripapillary retinal nerve fibre layer and macular ganglion cell complex are surrogate markers of neuroaxonal integrity in not only the eye, but also the CNS. Retinal atrophy may occur in tandem with CNS pathologies as a result of injury to ganglion cells, direct degeneration of the pregeniculate pathway, or retrograde trans-synaptic degeneration secondary to postgeniculate lesions. In this review, we outline the basic principles of optical coherence to

Cholinotoxicity of the ethylcholine aziridinium ion in primary cultures from rat central nervous system.

The cytotoxic effects of ethylcholine aziridinium ion (AF64A) were studied in primary cultures prepared from either whole brain, septum, or midbrain of fetal rats. AF64A, at concentrations up to 22.5 microM, significantly reduced the number of acetylcholinesterase-stained cells without affecting the number of dopaminergic neurons or their ability to take up and release [3H]dopamine. Many of the survived acetylcholinesterase-stained cells appeared with intact somata but damaged processes, indicating a retrograde degeneration starting at the nerve terminal. Higher concentrations of AF64A (greate

[Neuritic degeneration in Alzheimer-type dementia, with special reference to the presence of dark neurites].

Four cases of neuritic degeneration in the cerebral cortex of Alzheimer-type dementia patients have been examined by routine and Gallyas-silver electron microscopy. This examination has revealed neuritic changes described below. (1) Dendrites and axons with an increased electron density (dark neurites) were found scattered throughout the neuropils, these dark neurites thought to be either degraded neurites resulting from simple neuronal atrophy or retrograde and/or transsynaptic degeneration due to an axonal injury within the senile plaques. (2) Both dystrophic neurites and neuropil threads we
