Chronic intracellular infection of alfalfa nodules by Sinorhizobium meliloti requires correct lipopolysaccharide core.

Our analyses of lipopolysaccharide mutants of Sinorhizobium meliloti offer insights into how this bacterium establishes the chronic intracellular infection of plant cells that is necessary for its nitrogen-fixing symbiosis with alfalfa. Derivatives of S. meliloti strain Rm1021 carrying an lpsB mutation are capable of colonizing curled root hairs and forming infection threads in alfalfa in a manner similar to a wild-type strain. However, developmental abnormalities occur in the bacterium and the plant at the stage when the bacteria invade the plant nodule cells. Loss-of-function lpsB mutations,

XoxF and the Calvin-Benson cycle mediate lanthanide-dependent growth on methanol in Bradyrhizobium and Sinorhizobium.

Nodule-forming bacteria play crucial roles in plant health and nutrition by providing fixed nitrogen to leguminous plants. Despite the importance of this relationship, how nodule-forming bacteria are affected by plant exudates and soil minerals is not fully characterized. Here, the effects of plant-derived methanol and lanthanide metals on the growth of nitrogen-fixing Rhizobiales are examined. Prior work has demonstrated that select Bradyrhizobium strains can assimilate methanol only in the presence of lanthanide metals; however, the pathway enabling assimilation remains unknown. In this stud

Synchrotron micro-X-ray fluorescence shows sulfur accumulation in the middle cortex of N2-fixing legume nodules.

Sulfur (S) and phosphorus (P) are essential elements for plant growth and physiological functioning. Their deficiency can limit N2 fixation and nodule development in nodulated legumes. The location of S within nodule tissues could provide insights into S metabolism and its little-known relationship with N2 fixation. Determinate and indeterminate nodules were inoculated with specific rhizobia and grown hydroaeroponically under sufficient versus deficient P supplies. Cryogenic and freeze-dried thin sections of nodules at the flowering stage were mapped using synchrotron micro-X-ray fluorescence

Pivotal role of nitric oxide in regulating beneficial plant-microbe interactions under both physiological and stress conditions.

Plant-microbe interactions involve a complex communication network, where microbes can serve as either beneficial partners or harmful pathogens. Nitric oxide (NO) is a crucial signaling molecule that regulates plant growth and development, while enhancing tolerance to both biotic and abiotic stresses. It does this by interacting directly with reactive oxygen species (ROS) and regulating antioxidant enzyme activity through post-translational modifications (PTMs), such as S-nitrosation and nitration. Additionally, NO affects transcription factors that regulate gene expression and modulates phyto

Pesticide side effect on the symbiotic efficiency and nitrogenase activity of Rhizobiaceae bacteria family.

The laboratory experiments tested the influence of selected pesticides on the symbiotic efficiency and nitrogenase activity of Rhizobium leguminosarumin bv. trifolii KGL, Sinorhizobiuni melilotii Bp and Badyrhizobium sp. Ornithopus B bacteria entering into symbiosis with clover, lucerne and serradella, respectively. The results obtained indicate that the pesticides used in the experiments (Funaben T seed dressing and Pivot 100SL herbicide) caused reduced nitrogenase activity in active strains tested. In addition, a toxic effect of the applied pesticides on the nodulation and root growth of the

Is salt stress tolerance in Casuarina glauca Sieb. ex Spreng. associated with its nitrogen-fixing root-nodule symbiosis? An analysis at the photosynthetic level.

Casuarina glauca is an actinorhizal tree which establishes root-nodule symbiosis with N2-fixing Frankia bacteria. This plant is commonly found in saline zones and is widely used to remediate marginal soils and prevent desertification. The nature of its ability to survive in extreme environments and the extent of Frankia contribution to stress tolerance remain unknown. Thus, we evaluated the ability of C. glauca to cope with salt stress and the influence of the symbiosis on this trait. To this end, we analysed the impact of salt on plant growth, mineral contents, water relations, photosynthetic

Phosphatidylcholine biosynthesis and its significance in bacteria interacting with eukaryotic cells.

Phosphatidylcholine (PC), a typical eukaryotic membrane phospholipid, is present in only about 10% of all bacterial species, in particular in bacteria interacting with eukaryotes. A number of studies revealed that PC plays a fundamental role in symbiotic and pathogenic microbe-host interactions. Agrobacterium tumefaciens mutants lacking PC are unable to elicit plant tumors. The human pathogens Brucella abortus and Legionella pneumophila require PC for full virulence. The plant symbionts Bradyrhizobium japonicum and Sinorhizobium meliloti depend on wild-type levels of PC to establish an efficie

Relationships between chromium biomagnification ratio, accumulation factor, and mycorrhizae in plants growing on tannery effluent-polluted soil.

Heavy metal-contaminated land is increasingly becoming an important environmental, health, economic, and planning issue in Pakistan. The unplanned disposal of industrial effluent from tannery, for example, has resulted in a many fold increase in chromium (Cr) in the land near a tannery. This study was undertaken to compare the total and the DTPA-available Cr contents in the soil and the roots and leaves of tree species growing on it with those on the nearby noncontaminated reference site at Kala Shah Kakoo, Panjab, Pakistan. A very reduced plant cover on the tannery effluent-contaminated site

Spatiotemporal co-transcriptomics uncovers coordinated rhizobia-legume stress programs supporting nitrogen fixation in Medicago.

Rhizobial nitrogen fixation in legume nodules requires precise spatial and temporal coordination between host and bacterial gene expression. Here, we applied spatial enhanced resolution omics sequencing (Stereo-seq) to profile Medicago nodules across five developmental stages, generating a co-transcriptomic atlas of nodule development. Using a mixed-probe capture strategy, we simultaneously detected host and rhizobial transcripts within the same tissue sections, enabling analysis of host-microbe transcriptional coordination during symbiosis. We identified dynamic changes in host cell compositi

Tolerance to soil acidity of soybean (Glycine max L.) genotypes under field conditions Southwestern Ethiopia.

Soil acidity with associated low nutrient availability is one of the major constraints to soybean production in southwestern Ethiopia. Integrated use of lime and acid-tolerant crops is believed to reduce soil acidity and improve crop production. The experiment was conducted in the field condition of Mettu, southwestern Ethiopia during the 2017/18 main cropping season. The experiment comprised fifteen soybean genotypes and two soil amendment (lime and unlimed) treatments arranged in a split-plot design with three replications. For each treatment, four rows were planted per plot; data related to

Burkholderia sp. induces functional nodules on the South African invasive legume Dipogon lignosus (Phaseoleae) in New Zealand soils.

The South African invasive legume Dipogon lignosus (Phaseoleae) produces nodules with both determinate and indeterminate characteristics in New Zealand (NZ) soils. Ten bacterial isolates produced functional nodules on D. lignosus. The 16S ribosomal RNA (rRNA) gene sequences identified one isolate as Bradyrhizobium sp., one isolate as Rhizobium sp. and eight isolates as Burkholderia sp. The Bradyrhizobium sp. and Rhizobium sp. 16S rRNA sequences were identical to those of strains previously isolated from crop plants and may have originated from inocula used on crops. Both 16S rRNA and DNA recom

Post translational modifications of Trifolitoxin: a blue fluorescent peptide antibiotic.

Trifolitoxin (TFX, C41H63N15O15S) is a selective, ribosomally-synthesized, post-translationally modified, peptide antibiotic, produced by Rhizobium leguminosarum bv. trifolii T24. TFX specifically inhibits α-proteobacteria, including the plant symbiont Rhizobium spp., the plant pathogen Agrobacterium spp. and the animal pathogen Brucella abortus. TFX-producing strains prevent legume root nodulation by TFX-sensitive rhizobia. TFX has been isolated as a pair of geometric isomers, TFX1 and TFX2, which are derived from the biologically inactive primary amino acid sequence: Asp-Ile-Gly-Gly-Ser-Arg-

The preprotein translocase YajC mediates biofilm formation and symbiotic nitrogen-fixation in Mesorhizobium huakuii.

UNLABELLED: In bacteria, the auxiliary complex SecDF-YajC participates in the Sec system, facilitating post-translational translocation of preproteins across the membrane through the SecYEG channel. Although YajC homologs are strongly conserved in rhizobial strains and may play important roles in many processes, the functions and mechanisms by which they are involved in the interaction between rhizobia and host legumes are unknown. By constructing Mesorhizobium huakuii yajC mutant, we observed an increase in the strain growth rate, but a reduction in both the biomass and average thickness of t

Effects of Ensifer meliloti and Rhizophagus intraradices on alfalfa growth indices under cadmium sulfide nanoparticle stress.

Cadmium sulfide nanoparticles (CdS-NPs) are increasingly applied across various industries because of their unique properties. However, their accumulation in soil ecosystems and subsequent uptake by terrestrial plants can negatively affect plant growth. Beneficial microorganisms in the rhizosphere play an important role in mitigating the toxic effects of nanoparticles, thereby supporting plant health. Nevertheless, the role of these microorganisms in alleviating CdS-NP-induced stress in alfalfa remains largely unexplored. This study investigated the effects of different CdS-NPs concentrations

Mineral nutrition of plants: a short history of plant physiology.

The development of the knowledge on the mineral nutrition of plants begins between the 17th and 18th centuries when some European naturalists gave the first experimental evidences of what had been empirically known for about two millennia. The works of Hales and Ingenhousz were of absolute importance in relation to the transport of water and solutes, and assimilation of "fixed air" (carbon dioxide), respectively. The early chemistry introduced by Lavoisier benefited the first physiologists Senebier and De Saussure to reject the "theory of humus", which imposed the soil as the unique source of

Biogeography and specificity of legume rhizobial symbiovars.

The majority of legume species (plant family Fabaceae) can fix atmospheric nitrogen (N2) via symbiotic bacteria (general term 'rhizobia') in root nodules. Symbiovars are defined as strains of rhizobia that can establish an N2-fixing symbiosis with distinct legume species and have specific symbiosis gene phylogenies. Here, symbiovar occurrence across rhizobial species was related to rhizobia and legume host taxonomy and geographic location of the symbiosis, and then their specificity was assessed in relation to legume species they did or did not nodulate. Sixty-eight rhizobial symbiovars occurr

Genome sequence of Rhizobium etli CNPAF512, a nitrogen-fixing symbiont isolated from bean root nodules in Brazil.

Rhizobium etli is a Gram-negative soil-dwelling alphaproteobacterium that carries out symbiotic biological nitrogen fixation in close association with legume hosts. R. etli strains exhibit high sequence divergence and are geographically structured, with a potentially dramatic influence on the outcome of symbiosis. Here, we present the genome sequence of R. etli CNPAF512, a Brazilian isolate from bean nodules. We anticipate that the availability of genome sequences of R. etli strains from distinctly different areas will provide valuable new insights into the geographic mosaic of the R. etli pan

Phosphate solubilization and multiple plant growth promoting properties of Mesorhizobium species nodulating chickpea from acidic soils of Ethiopia.

The main purpose of this study was to screen and select strains from seven Mesorhizobium spp. for efficient phosphate solubilizing and other plant growth-promoting traits. Mesorhizobium species were tested for their ability to dissolve inorganic phosphate sources and multiple plant growth-promoting attributes. From a total of 62 Mesorhizobium strains, 47(76%) strains formed clear zones with an average PSI of 1.9-2.7 on Pikovskaya's agar plate. The selected strains also released soluble phosphorus [125-150 P (µgml-1)] from tri-calcium phosphate and low level of phosphorous i.e., 15.4 µg/ml and
