Interaction of nitric oxide with photoexcited rose bengal: evidence for one-electron reduction of nitric oxide to nitroxyl anion.

The interaction of nitric oxide (.NO) with Rose Bengal (RB) in the presence of electron donors was investigated. Upon illumination of a mixture of RB and .NO with visible light, an enhancement in the rate of .NO consumption was observed that increased with increasing RB concentration. In the presence of electron donors (NADH, glutathione, or ascorbate), the rates of .NO depletion increased further. NADH enhanced .NO depletion to a greater extent than either glutathione or ascorbate. Photoactivated RB under anaerobic conditions reacts with NADH to form the RB anion radical (RB.-), which has a c

Towards scalable broad-spectrum photodynamic antimicrobial textiles: synergistic effect of Rose Bengal and commercial cationic fixative on polyamide fabrics.

The use of photoactive textiles to reduce infection transmission in healthcare facilities and hospitals is not on the market due to the lack of scalable and cost-effective processes to prepare these materials. To address this issue, a new photodynamic antimicrobial fabric of polyamide with Rose Bengal (RB) and a commercial cationic fixative (CF) was prepared with a simple and scalable procedure using a conventional and industrialized process for incorporating dyes into textiles. Both fabrics (with and without CF) produced more than 99% inactivation of Gram-positive and Gram-negative bacteria (

Comparison of rose bengal-green light scleral crosslinking in rabbit eyes using different infiltration protocols - An Ex Vivo study.

Different concentrations and infiltration times of rose bengal (Rb) were assessed for their impact on penetration depth and crosslinking efficacy in rabbit sclera. Fresh rabbit eyes were used. Rb solution with concentrations of 0.1%-0.9% were applied for 5-30 min to infiltrate the sclera. The penetration depth of Rb was observed with confocal microscopy. After infiltration, the sclera was irradiated by green light for crosslinking. The sclera's biomechanical stiffness and the resistance to enzyme digestion post-treatment were evaluated. Histopathological analysis and transmission electron micr

Trilayer dissolving polymeric microneedle array loading Rose Bengal transfersomes as a novel adjuvant in early-stage cutaneous melanoma management.

Melanoma remains a global concern, but current therapies present critical limitations pointing out the urgent need for novel strategies. Among these, the cutaneous delivery of drugs selectively damaging cancer cells is highly attractive. Rose Bengal (RB) is a dye exhibiting selective cytotoxicity towards melanoma, but the high water solubility and low permeability hinder its therapeutic potential. We previously developed RB-loaded transfersomes (RBTF) to mediate the RB dermal delivery; however, a platform efficiently delivering RBTF in the deepest strata is essential for a successful therapeut

Targeted chemo-sonodynamic therapy treatment of breast tumours using ultrasound responsive microbubbles loaded with paclitaxel, doxorubicin and Rose Bengal.

Mastectomy is a common surgical treatment used in the management of breast cancer but has associated physical and psychological consequences for the patient. Breast conservation surgery (BCS) is an alternative to mastectomy but is only possible when the tumour is of an appropriate size. Neo-adjuvant chemotherapy has been successfully used to downstage tumours and increase the number of patients eligible for BCS. However, the chemotherapies used in this approach are non-targeted and often result in significant side effects to the patient. In this manuscript, we evaluate the potential of ultraso

Evaluation of tear break-up time, Schirmer's-I test and rose bengal staining as confirmatory tests for keratoconjunctivitis sicca.

The tear break-up time (BUT), Schirmer's I test (S-I) and slit lamp examination after rose Bengal staining (RBS) are simple tests for evaluation of keratoconjunctivitis sicca (KCS). Discrepancies, however, regarding the range of normal values and the specificity and sensitivity of these methods prompted us to reevaluate them in 81 primary Sjögren's syndrome patients (Ss) and 276 normal volunteers. In normal volunteers, BUT values ranged from 4 sec. to 32 sec. (means +/- SD: 13 +/- 6 sec.) and in primary Ss from 1 sec. to 15 sec. (means +/- SD: 7 +/- 3 sec.). Ninety five percent (95%) of the va

Adjunct rose Bengal photodynamic antimicrobial therapy for refractory Pseudomonas aeruginosa infectious necrotizing scleritis after scleral buckling: a case report.

A 34-year-old man developed rapidly progressive infectious necrotizing scleritis following blunt ocular trauma and subsequent scleral buckle placement for rhegmatogenous retinal detachment. At presentation, best-corrected visual acuity was light perception, with conjunctival lysis, multifocal scleral thinning with uveal exposure, purulent discharge, and marked ocular inflammation. Microbiologic cultures grew Pseudomonas aeruginosa. Despite targeted topical and systemic antimicrobial therapy and surgical debridement with partial buckle removal and tectonic reconstruction, scleral necrosis conti

Photodynamic antimicrobial therapy with Erythrosin B, Eosin Y, and Rose Bengal for the inhibition of fungal keratitis isolates: An in vitro study.

INTRODUCTION: Fungal keratitis is a leading cause of corneal blindness, with current antifungal treatments having limited efficacy. One promising treatment modality is Rose Bengal (RB) photodynamic antimicrobial therapy (PDAT) that has shown mixed success against fungal keratitis. Therefore, there is a need to explore the antimicrobial efficacy of other green-light activated photosensitizers that have deep penetration in the cornea to combat the deep fungal infections, such as Erythrosin B (EB) and Eosin Y (EY). OBJECTIVE: This study will explore PDAT inhibitory effects with different photose

Comparative performance of lateral flow immunochromatography, iELISA and Rose Bengal tests for the diagnosis of cattle, sheep, goat and swine brucellosis.

BACKGROUND: Brucellosis is a world-wide extended zoonosis that causes a grave problem in developing economies. Animal vaccination and diagnosis are essential to control brucellosis, and the need for accurate but also simple and low-cost tests that can be implemented in low-infrastructure laboratories has been emphasized. METHODOLOGY: We evaluated bovine, sheep, goat and swine lateral flow immunochromatography assay kits (LFA), the Rose Bengal test (RBT) and a well-validated protein G indirect ELISA (iELISA) using sera of Brucella culture-positive and unvaccinated brucellosis free livestock. How do you get the Rose Bengal Test at the point-of-care to diagnose brucellosis in Africa? The importance of a systems approach.

Brucellosis is a major neglected zoonotic disease, whose burden both in animals and humans is severely under-reported. Diagnosis in humans identifies cases in order to treat the disease at the individual level. In animals diagnosis is implemented at the population level in the context of appropriate control or eradication strategies. Molecular and bacteriological diagnosis are rarely undertaken in sub-Saharan Africa, at least outside research projects, due to cost, skills and laboratory infrastructure issues. The brucellosis toolbox contains a wide range of serological tests, but the perfect t

Same-session dual chromophore riboflavin/UV-A and rose bengal/green light PACK-CXL in Acanthamoeba keratitis: a case report.

BACKGROUND: Acanthamoeba keratitis (AK) is the most challenging corneal infection to treat, with conventional therapies often proving ineffective. While photoactivated chromophore for keratitis-corneal cross-linking (PACK-CXL) with riboflavin/UV-A has shown success in treating bacterial and fungal keratitis, and PACK-CXL with rose bengal/green light has demonstrated promise in fungal keratitis, neither approach has been shown to effectively eradicate AK. This case study explores a novel combined same-session treatment approach using both riboflavin/UV-A and rose bengal/green light in a single

Effect of Green Light-Mediated Corneal Cross-Linking on the Delivery of Rose Bengal and Vancomycin in Staphylococcus aureus-Infected Eyes.

Rose Bengal (RB)-green light (GL) cross-linking (RGX), a phototherapy used to treat infectious keratitis, activates RB to generate reactive oxygen species (ROS), promoting bacterial killing and corneal strengthening through collagen cross-linking. Despite its promise, the way GL exposure alters RB behavior within the infected cornea remains poorly understood. This study investigates RB distribution and fluorescence changes following RGX in methicillin-susceptible Staphylococcus aureus-infected bovine corneas with denuded epithelium. Ex vivo RGX treatment resulted in a pronounced, infection-dep

Comparative assessment of brucellosis detection in dogs: In-house ELISA versus Rose Bengal Plate Test utilizing rough and smooth antigens.

Brucella canis and smooth Brucella species infections have been reported in dogs globally. In endemic countries such as Türkiye, dogs can be infected with both species. The exact incidence of canine brucellosis in Türkiye is unknown. This study aims to investigate the seroprevalence of brucellosis in stray dogs undergoing sterilization in Van province, eastern Türkiye. In this study, a comprehensive approach was used to ensure reliable incidence of Brucella spp in the dogs. We used the Rose Bengal Plate Test (RBPT) with both antigens from smooth Brucella abortus S99 (RBPT-S) and rough B. canis
