In vitro production of lysine from 2,2'-diaminopimelic acid by rumen protozoa.

Rumen protozoa can produce lysine from free 2,2'-diaminopimelic acid (DAP). However, the quantitative importance of this transformation has been disputed; lysine contents of protozoal incubation supernatants reported by Onodera & Kandatsu and Masson & Ling show a 26-fold difference. The in vitro experimental methods of both groups were compared to determine the causes of this difference. Lysine production was proportional to DAP concentration. Results with rumen protozoa from sheep or goats were similar. The incubation medium and deproteinizing procedure of the Welsh group gave a two-fold incr

The copper-molybdenum antagonism in ruminants. I. The formation of thiomolybdates in animal rumen.

The formation of thiomolybdates, MoOxS4--x2--(x = 0, 1, 2, or 3), from molybdate and sulphide salts in aqueous media has been studied under conditions which simulate the fluid phase in the rumen. The influences of the sulphide:molybdenum ratio, pH and phosphate levels on the nature of the species formed were investigated. The thiomolybdates, in particular the MoS42-- ion, have been implicated as the active intermediates in the widespread molybdenum induced copper deficiency that affects ruminants. The results presented here show that, under physiological conditions, di- and trithiomolybdates w

Toxic effects of large amounts of DL-methionine infused into the rumen of sheep.

Four Merino wethers were fed ad libitum on a roughage diet comprised of equal parts of chopped lucerne hay and oaten chaff. The sheep received continuous infusions of DL-methionine into the rumen; the daily dose increasing from 0 g (control treatment) up to 30 g in 3 g amounts at weekly intervals. Dry matter intake (DMI) fell below the control level when 24 g/day or more of the amino acid was infused, while plasma free methionine levels increase substantially when 30 g/day was given. There was no effect of DL-methionine supplementation on the bodyweight of the animals. However, when 30 g/day o

Polystyrene microplastics impair sheep spermatogenesis by disrupting rumen microbiota-derived butyrate signaling.

BACKGROUND: Microplastics are emerging environmental contaminants with male reproductive toxicity; however, their effects on ruminants and underlying mechanisms remain poorly understood. RESULTS: In this study, male Hu lambs were exposed to dietary polystyrene microplastics (PS-MPs) at 0, 75, and 150 mg/kg diet for 120 d, followed by a 180-d recovery period. PS-MPs exposure caused persistent testicular injury, characterized by disrupted seminiferous architecture and enhanced germ cell apoptosis. Single-cell transcriptomic analysis revealed impaired spermatogonial differentiation, accompanied

Distinct Rumen Microbial Features and Host Metabolic Responses in Three Cervid Species.

Rumen microbiota is pivotal for nutrient metabolism and physiological adaptation in ruminants. This study investigated the rumen microbial community, fermentation parameters, and serum biochemistry of three Cervid species-Sika deer (Cervus nippon), Reindeer (Rangifer tarandus), and Milu deer (Elaphurus davidianus) (n = 5/group)-fed an identical diet. Using 16S rRNA sequencing and biochemical analyses, we found that while Bacteroidota, Firmicutes, and Proteobacteria were dominant phyla across species. Sika deer and Milu deer exhibited significantly higher microbial diversity and abundance of ca

Subacute Rumen Acidosis in Greek Dairy Sheep: Prevalence, Impact and Colorimetry Management.

Subacute rumen acidosis (SARA) is a current issue in intensive livestock farming, and it is particularly associated with feeding high-concentrate diets. This study investigated the prevalence and impact of SARA in forty-two Greek dairy sheep flocks by recording rumen pH, milk composition, and milk yield over a period of nine months. Moreover, it explored the use of computerized rumen colorimetry as a management and diagnostic tool for SARA in dairy sheep. In culled ewes, computerized rumen mucosal colorimetry was applied, and rumen wall samples taken for histological examination. SARA cases we

Divalent cations reduce the electrogenic transport of monovalent cations across rumen epithelium.

The rumen epithelium of sheep and goats showed an increase in short circuit current ( Isc) and transepithelial conductance (gt) upon mucosal removal of divalent cations. A divalent-sensitive Isc and gt were present in Na+, K+ or Rb+ buffer, but nearly abolished in mucosal NMDG+ (N-methyl-D-glucamine) buffer. High K buffer, addition of BaCl2 or of ouabain on the serosal side also reduced or abolished the divalent-sensitive Isc. Mucosal Ca2+ was more potent in blocking Isc, but had the same potency as Mg2+ in blocking gt. A prolonged mucosal deprivation of Mg2+ ions increased gt, potential diffe

Yeast β-glucan alleviates the subacute rumen acidosis-induced mitochondrial dysfunction and cell structure integrity injury in yak rumen epithelial cells via the TLR2/PI3K/mTOR signaling pathway.

Subacute rumen acidosis (SARA) is characterized by decreased rumen fluid pH and impaired rumen epithelial function. In this study, the underlying molecular mechanisms of β-glucan can alleviate SARA induced damage to yak rumen epithelial cells (YRECs), and its potential protective effects were investigated. The results demonstrated that β-glucan alleviated the decrease in cell viability and LDH release induced by SARA. Furthermore, SARA led to mitochondrial dysfunction, including disrupted mitochondrial morphology observed through fluorescence and transmission electron microscopy, decreased mit

Gene function adjustment for carbohydrate metabolism and enrichment of rumen microbiota with antibiotic resistance genes during subacute rumen acidosis induced by a high-grain diet in lactating dairy cows.

The high-grain diets fed to ruminants generally alters the structure and function of rumen microbiota, resulting in variations of rumen fermentation patterns and the occurrence of subacute rumen acidosis (SARA). To clarify the microbial mechanism for carbohydrate metabolism during SARA, 8 ruminally cannulated Holstein cows in mid lactation were selected for a 3-wk experiment. The cows were randomly divided into 2 groups, fed either a conventional diet (CON; 40% concentrate; dry matter basis) or a high-grain diet (HG; 60% concentrate; dry matter basis). Compared with the CON diet, the HG diet r

In Vitro Degradation of Chlorpyrifos by the Ruminal Microbes: Insights from the Rumen Metagenome.

In vitro studies were conducted in a series to investigate if the ruminal microbes are capable of degrading chlorpyrifos. This in vitro study presents the results from three experiments: Exp. I was conducted without feed, while Exp II and III were conducted with feed, either with or without methanol for dissolving chlorpyrifos, respectively. A basal diet comprising finger millet straw and concentrate was prepared. Incubation medium with feed but without chlorpyrifos served as the control. A total of six replicates each of control and chlorpyrifos spiked were used for the incubation. The pestic

Influence of host genetics on the functional composition of the rumen metagenome in beef cattle.

Cattle rely on the microorganisms in their rumen to break down plant matter into useable nutrients. Studies have demonstrated that the rumen microbiome plays a critical role in economically important traits. One factor that impacts rumen microbial community assembly is the host genome. Previous studies have demonstrated host genetics affect rumen microbial community composition and the association of microbiome features with production traits. However, gaps exist relative to the underlying host genetic influence on functional features of the rumen metagenome. Here we elucidated the relationshi

Associations of rumen and rectum bacteria with the sustained productive performance of dairy cows.

The gut bacterial community is essential for maintaining lifelong health and productivity in ruminants, but the relationship between the gut microbiota and the sustained productivity of ruminants remains inadequately understood. In this study, we selected long-lived dairy cows in mid-lactation (≥5 parities) with different levels of milk production (n = 10). Significant differences were observed in the rumen bacterial structures between the two groups of dairy cows, whereas no significant differences were detected in the rectum bacterial communities. Additionally, there were no significant diff

The dynamics of single stranded, positive sense RNA bacteriophages in the rumen virome.

We studied a selection of single stranded (ss)RNA bacteriophages (Class Leviviricetes) in rumen samples from dairy cows. These phages are non-enveloped, positive sense bacteriophages infecting Gram-negative bacteria, e.g. Escherichia, Salmonella, Pseudomonas, or other Gammaproteobacteria. Leviviricetes phages have small genomes and are of interest, as they likely predate on commensal or pathogenic Gram-negative bacteria. These phages attach to retractile pili on the bacterial surface, characterising them as male- or sex/conjugation-dependent phages and may interfere with bacterial conjugation

Role of the Rumen Epithelium and Associated Changes Under High-Concentrate Diets.

Increasing the proportion of concentrate in diets can effectively improve ruminant production, and is therefore widely used. However, high-concentrate diets (HCD) enriched with rapidly fermentable carbohydrates can accelerate the production of lactate and short-chain fatty acids (SCFAs). The accumulation of lactate and SCFAs in the rumen leads to a reduction in rumen fluid pH, potentially resulting in subacute rumen acidosis (SARA), which can decrease dry matter intake (DMI), induce local and systemic inflammation, and cause other negative impacts on the host. The substantial prevalence of SAR

The effects of induced hindgut acidosis in sheep on rumen fermentation and gut permeability.

The objective was to determine the effects of induced hindgut acidosis in sheep on cecal pH, ruminal fermentation, and gut permeability. Eleven ruminally and cecally cannulated ewes (49 ± 4 kg) were assigned to one of two treatments: control (CON; n = 5) or induced hindgut acidosis (HGA; n = 6). To induce hindgut acidosis, 3 g wheat starch/kg BW per 24 h was continuously infused via the cecal cannula for 4 d. Control ewes received a constant infusion of deionized water. Ewes were fed a common diet at a set level of intake based on body weight. Chromium EDTA was dosed once daily via the cecal c

Milk Odd- and Branched-Chain Fatty Acids as Biomarkers of Rumen Fermentation.

Cow's milk and dairy products are the primary sources of OBCFAs, which have beneficial health properties. The goal of this study was to identify the factors that influence the content of OBCFAs in cow's milk and to indicate which OBCFAs can serve as biomarkers for fermentation processes. The content of OBCFAs in milk depends on the species of ruminants, with studies showing that this varies between 3.33% (in goat's milk) and 5.02% (in buffalo's milk). These differences also stem from the animals' energy balance, lactation phases, forage-to-concentrate ratio, and the presence of bioactive compo

Effect of feeding spent coffee grounds on the methane production in bovine rumen.

The environmental impact of methane, a greenhouse gas emitted from ruminants, is a pressing issue and methods to control methane emissions from ruminants are being investigated worldwide. In this study, we investigated the effects of the administration of spent coffee grounds (SCG) on methane production in the rumen in two cows. In the control condition (days 1 and 2), the cows were fed a basic diet twice daily (roughage and concentrate), and in the SCG condition (days 1 and 2) sequentially, the cows were fed the same basic diet and administered SCG into the rumen twice daily. The methane and

Rumen and Liver Fluke Infections in Sheep and Goats in Northern and Southern Germany.

Paramphistomidosis has recently been identified as an emerging parasitosis in Europe. This study estimated the prevalence of rumen flukes, Fasciola hepatica and Dicrocoelium dendriticum, in small ruminants in Germany and identified occurring rumen fluke species and potential predictors for fluke infections. Pooled fecal samples from 223 sheep farms and 143 goat farms in northern and southern Germany were examined by the sedimentation technique, and molecular species identification was performed on rumen-fluke-positive samples. In sheep, a flock prevalence of 2.2% was detected for rumen flukes.

Rumen effects of monensin in dry cow diets varying in energy density.

Controversy has existed as to whether monensin will provide equal or differential benefits in a higher-energy, lower-roughage close-up diet versus a higher-roughage, lower-energy diet. Our objective was to determine the rumen effects of a controlled-energy, high-fiber diet balanced to meet but not greatly exceed energy requirements during the dry period or a traditional 2-group approach of higher-energy close-up diet. The effects of added monensin in each diet type were determined. Multiparous Holstein cows (n = 17) were fitted surgically with ruminal cannulas. During the first 4 wk of the dry

Reconstruction and Analysis of Cattle Metabolic Networks in Normal and Acidosis Rumen Tissue.

The objective of this study was to develop a system-level understanding of acidosis biology. Therefore, the genes expression differences between the normal and acidosis rumen epithelial tissues were first examined using the RNA-seq data in order to understand the molecular mechanisms involved in the disease and then their corresponding metabolic networks constructed. A number of 1074 genes, 978 isoforms, 1049 transcription start sites (TSS), 998 coding DNA sequence (CDS) and 2 promoters were identified being differentially expressed in the rumen tissue between the normal and acidosis samples (
