The spleen-strengthening and liver-draining herbal formula treatment of non-alcoholic fatty liver disease by regulation of intestinal flora in clinical trial.

OBJECTIVE: As a metabolic disease, one important feature of non-alcoholic fatty liver disease (NAFLD) is the disturbance of the intestinal flora. Spleen-strengthening and liver-draining formula (SLF) is a formula formed according to the theory of "One Qi Circulation" (Qing Dynasty, 1749) of Traditional Chinese Medicine (TCM), which has shown significant therapeutic effect in patients with NAFLD in a preliminary clinical observation. In this study, we aim to explore the mechanism of SLF against NAFLD, especially its effect on glucolipid metabolism, from the perspective of intestinal flora. MET

Partially Hydrolyzed Guar Gum Combined with a Low-Fat Diet Ameliorates Type 2 Diabetes Mellitus via Modulating Gut Microbiota and Fecal Metabolites.

Background: Low-fat diet (LFD) is widely applied in type 2 diabetes mellitus (T2DM), but the limited efficacy and difficulty in maintaining it hinder its wider promotion. Partially hydrolyzed guar gum (PHGG) is well-known as a probiotic in modulating gut microbiota, which is crucial in T2DM. However, the combined effects of LFD and PHGG remain unknown. Methods: Mice with T2DM were divided into 4 groups: T2DM control (DM-high-fat diet), LFD alone (DM-LFD), or LFD combined with low or high doses of PHGG (PHGG-L/H, 2.5% and 7.5% (w/w)) for 12 weeks. Altered gut microbiota mediates the association between APOE genotype and amyloid-β accumulation in middle-aged adults.

BACKGROUND: The apolipoprotein E (APOE) ε4 allele is the strongest genetic risk factor for Alzheimer's disease, yet the mechanisms linking APOE to amyloid-β (Aβ) pathology remain incompletely understood. Emerging evidence suggests that the gut microbiome may modulate neurodegeneration; however, its role as a mediator in the APOE-Aβ relationship remains unclear. OBJECTIVE: Evaluate whether specific microbial taxa mediate APOE-related effects on brain Aβ burden in a population-based study. METHODS: This study involved 227 participants from the Framingham Heart Study with stool 16S rRNA sequenc

The dysbiosis of intestinal microbiota associated with isolated tricuspid valve regurgitation.

BACKGROUND: Isolated tricuspid regurgitation (TR) is an increasingly recognized yet frequently underappreciated valvular disorder that adversely affects both survival and quality of life, with progressively worse outcomes as severity increases. Advances in high-throughput sequencing technologies have considerably expanded our understanding of the human microbiome, uncovering its close associations with a range of cardiovascular diseases, including atherosclerosis, heart failure, and hypertension. However, the potential role of intestinal dysfunction and the gut–heart axis in TR pathophysiology

Multikingdom characterization of gut microbiota in patients with rheumatoid arthritis and rheumatoid arthritis-associated interstitial lung disease.

Rheumatoid arthritis-associated interstitial lung disease (RA-ILD) is a serious and common extra-articular disease manifestation. Patients with RA-ILD experience reduced bacterial diversity and gut bacteriome alterations. However, the gut mycobiome and virome in these patients have been largely neglected. In this study, we performed whole-metagenome shotgun sequencing on fecal samples from 30 patients with RA-ILD, and 30 with RA-non-ILD, and 40 matched healthy controls. The gut bacteriome and mycobiome were explored using a reference-based approach, while the gut virome was profiled based on a

Gut inflammation and dysbiosis in human motor neuron disease.

Amyotrophic lateral sclerosis (ALS) is a systemic disorder that involves dysfunction of multiple organs. Growing evidence has shown that neurodegenerative disorders with gut dysbiosis affect the central nervous system via pro-inflammatory mediators thus impacting gut-brain communications. We have demonstrated dysbiosis and increased intestinal permeability in the SOD1G93A ALS mouse model. In this study, we comprehensively examined the human gut microbiome in stool samples and evaluated infection and markers of intestinal inflammation in five patients with ALS and motor neuron disorders. Five p

Variability in chewing, ruminal fermentation, digestibility and bacterial communities between subacute ruminal acidosis-susceptible and acidosis-tolerant sheep.

Sorting behaviour is a common phenomenon observed in ruminants when they are provided with a total mixed ration, which contributes to variations in the severity of subacute ruminal acidosis (SARA). Pelleted total mixed ration (PTMR) reduces sorting, but high-grain content increases acidosis risk. However, whether the variability in the severity of SARA exists in sheep fed the same high-grain PTMR is less understood. This study aimed to investigate SARA variability among individual sheep offered a high-grain PTMR, considering chewing activity, ruminal fermentation, bacterial communities and nut

Multi-omics analyses of the gut microbiota and metabolites in children with metabolic dysfunction-associated steatotic liver disease.

The development and severity of metabolic dysfunction-associated steatotic liver disease (MASLD) in children are closely related to alterations of gut microbiota. This study aims to investigate changes in the gut microbiota signature and microbial metabolites in children with MASLD. We collected fecal samples from children and adolescents aged 6-16 years, and the presence of MASLD was diagnosed by ultrasound. We performed 16S ribosomal DNA sequencing and targeted metabolomics in 36 and 25 subjects, consisting of healthy controls, children with obesity, and children with MASLD. The α-diversity

Impact of a High-Fat Diet on the Gut Microbiome: A Comprehensive Study of Microbial and Metabolite Shifts During Obesity.

Over the last few decades, the prevalence of metabolic diseases such as obesity, diabetes, non-alcoholic fatty liver disease, hypertension, and hyperuricemia has surged, primarily due to high-fat diet (HFD). The pathologies of these metabolic diseases show disease-specific alterations in the composition and function of their gut microbiome. How HFD alters the microbiome and its metabolite to mediate adipose tissue (AT) inflammation and obesity is not well known. Thus, this study aimed to identify the changes in the gut microbiome and metabolomic signatures induced by an HFD to alter obesity. T

Effects of exoskeleton-assisted walking on bowel function in motor-complete spinal cord injury patients: involvement of the brain-gut axis, a pilot study.

Evidence has demonstrated that exoskeleton robots can improve intestinal function in patients with spinal cord injury (SCI). However, the underlying mechanisms remain unelucidated. This study investigated the effects of exoskeleton-assisted walking (EAW) on intestinal function and intestinal flora structure in T2-L1 motor complete paraplegia patients. The results showed that five participants in the EAW group and three in the conventional group reported improvements in at least one bowel management index, including an increased frequency of bowel evacuations, less time spent on bowel managemen

Comprehensive Analysis of Fecal Microbiome and Metabolomics in Hepatic Fibrosis Rats Reveal Hepatoprotective Effects of Yinchen Wuling Powder From the Host-Microbial Metabolic Axis.

Hepatic fibrosis (HF) is a typical consequence in the development of multiple chronic liver diseases, which is intimately related to the composition and metabolic status of gut microbiota. A myriad of evidence has indicated that traditional Chinese medicine can treat HF by regulating gut microbiota. Yinchen Wuling powder (YCWLP) is a famous traditional Chinese medicine prescription, which has been used to relieve liver diseases for thousands of years. YCWLP has demonstrated protective function on HF, but its effect on the alterations of gut microbiota is still unclear, and its explicit therape

Protective effect of a combination of multiple strains of Lactobacillus acidophilus on collagen-induced arthritis.

Rheumatoid arthritis (RA) is a systemic, chronic autoimmune disease. Many studies have shown that microorganisms may be an important pathological factor leading to the onset of RA. Some infectious or non-infectious pathogenic microorganisms and their metabolites may be the initiating factors of the early onset of RA. The aim of this study was to alleviate RA by regulating the imbalance of the gut microbiota in the early stage of RA using mixed bacterial strains. The mixed strains of Lactobacillus acidophilus (BD18, NCFM, BD1802, BD3545, BD5032) significantly reduced the clinical score and swel
